A non-invasive test for stria vascularis dysfunction.

Age-related hearing loss is common in the human population but it is highly heterogeneous in aetiology which has hampered efforts to develop ways of stopping its progression. Three major sites of the initial dysfunction are the sensory hair cells, their innervation, and the stria vascularis which generates the high potassium endolymph maintained at a high endocochlear potential bathing the apical surface of hair cells. Treatments aimed at the initial site-of-lesion may be useful, and diagnostic tools to distinguish the primary site would help stratify clinical trials and facilitate selection o

The ABHD10-KCMF1 Complex Mitigates Cochlear Aging by Regulating Mitochondrial Lipid Metabolism.

Presbycusis is caused by multiple factors, the mechanisms of which are not fully understood. This study investigated mitochondria-related genes in presbycusis, a condition with multifactorial and incompletely understood mechanisms, by combining Mendelian randomization analysis and functional validation. Genetically predicted higher ABHD10 expression was identified as a protective factor against presbycusis and was regulated by methylation sites such as cg15684481, whereas methylation at other sites inhibited ABHD10 expression and alleviated the condition. In contrast, ABHD10 was upregulated in

Vocal emotion perception in hearing loss - feasibility of an online training program to improve emotion perception in older adults.

BACKGROUND: Age-related hearing loss (ARHL) represents a frequent decline in hearing abilities reducing older adults' quality of life. Human communication not only depends on mere sound detection but also on extralinguistic and paralinguistic information like emotions. Understanding the emotions of a conversation partner is vital for connection, but is oftentimes reduced in ARHL. METHODS: A feasibility study was conducted on N = 18 (14 female) participants aged 74.6 ± 6.7 years with ARHL. Basic effectiveness and applicability of an online emotion-perception training was assessed in a pre- and

Chromatin and genomic instability in the cochlea contributing to age-related hearing loss: Insights from in vitro and in vivo models.

Age-related hearing loss (ARHL) is one of the most common causes of hearing impairment in older adults. However, the cellular and molecular mechanisms underlying ARHL remain unclear. Chromatin and genomic instability, including DNA damage, heterochromatin loss, and telomere attrition, are hallmarks of aging. In the present study, we investigated DNA damage levels, chromatin accessibility, telomere length, and telomerase changes in vitro and in vivo mouse models of ARHL. To further explore changes in outer hair cells (OHCs), one of the prominent auditory sensory cells in the cochlea, we constru

Advances in pathogenesis, novel therapeutic strategies and interventions for age-related hearing loss.

Age-related hearing loss (ARHL), or presbycusis, represents one of the most prevalent sensory dysfunctions in the elderly people. It is characterized by progressive hearing deterioration that significantly impairs quality of life and increases the risk of comorbidities including cognitive impairment, depression, and social isolation. The pathogenesis of ARHL involves complex structural alterations such as degeneration of cochlear hair cells, loss of spiral ganglion neurons, and stria vascularis atrophy, coupled with molecular and cellular dysregulations including oxidative stress, inflammation

Association between lifetime noise exposure and audiometric thresholds using the noise exposure structured interview.

Noise exposure is pervasive in modern society and a major contributor to hearing loss across the lifespan. This study examined how cumulative lifetime noise exposure, measured using the Noise Exposure Structured Interview (NESI), associates with age to affect audiometric thresholds. Fifty-four adults (23-82 years; 25 female) completed the NESI (occupational, recreational, and firearm noise exposure), hearing function self-reports, pure-tone audiometry, and speech-reception testing. Multivariate regression analyses evaluated independent and interactive effects of age and noise exposure on audit

Hearing Loss as a Potentially Modifiable Marker of Dementia Risk: Neurological Evidence, Uncertainty, and Clinical Interpretation.

BACKGROUND: Hearing impairment has emerged as a consistently associated and potentially modifiable risk factor for cognitive decline and dementia, including Alzheimer's disease (AD). Epidemiological studies demonstrate an increased dementia risk among individuals with hearing loss, with evidence of dose-response relationships across levels of auditory decline. Despite the robustness of this association, the mechanisms linking auditory dysfunction to cognitive deterioration remain multifactorial and incompletely understood. SUMMARY: By integrating biological, cognitive, and psychosocial perspe

Impact of Age-Related Hearing Loss on Brain Connectivity and Cognitive Performance: A Systematic Review.

Adult-onset hearing loss is associated with cognitive decline, although the mechanisms underlying this relationship remain unclear. Emerging evidence suggests that the early stages of hearing loss can alter brain network dynamics, which may be detectable before other neurocognitive markers. This systematic review aims to present evidence for altered brain connectivity in age-related hearing loss, and its subsequent relationship with cognitive performance. The review was preregistered in the Prospective Register of Systematic Reviews and conducted in accordance with the Preferred Reporting Item

Cortical activation during auditory working memory varies with hearing aid use status in age-related hearing loss.

Age-related hearing loss (ARHL) is associated with increased listening effort due to greater cognitive demands needed for supporting effective communication. Verbal working memory (WM) is an important mechanism supporting speech understanding in adults with ARHL. Using functional near-infrared spectroscopy and an auditory N-back task (0-, 1-, 2-back load), we examined whether hearing aid status was associated with cortical activation in adults with ARHL. Three groups were compared: typical-hearing adults, experienced hearing aid users, and newly fitted users. Linear mixed-effects models reveal

Presbycusis Across the Lifespan: Genetic, Molecular, and Multi-Omics Contributions.

Presbycusis, or age-related hearing loss (ARHL), is a multifactorial disorder characterized by a gradual, bilateral sensorineural decline in hearing sensitivity, predominantly affecting high-frequency sounds. It is one of the most common chronic conditions in the aging population and represents a major public health concern due to its high prevalence and progressive nature. Presbycusis significantly impairs speech perception, especially in noisy environments, leading to communication difficulties, reduced social participation, increased risk of social isolation, and a decline in quality of lif

Age-Related Changes in Speech Recognition Performance-Intensity Functions and Auditory Brainstem Responses.

OBJECTIVES: While the mechanisms underlying age-related changes in speech recognition are thought to be multifactorial, cochlear synaptopathy has been proposed as a possible peripheral auditory contributor. Although evidence from animal and human temporal bone studies supporting the presence of age-related cochlear synaptopathy appears compelling, findings from prospective studies in humans have been less conclusive. The goals of this study were to investigate the effect of aging on measures historically used to assess retrocochlear function-speech recognition performance-intensity functions i

G1 regulation of BKCa channel leads to decreased migration of senescent pericytes and improved age-related hearing loss.

The pathogenesis of age-related hearing loss (ARHL) remains incompletely understood. The blood labyrinth barrier (BLB) play a crucial role in maintaining cochlear homeostasis, yet in the stria vascularis (SV) of aged C57BL/6J mice, pericytes (PCs) undergo migration, potentially increasing BLB permeability and contributing to vascular leakage, which may exacerbate ARHL. The underlying molecular mechanisms, however, remain elusive. Studies have suggested gender disparities in auditory function linked to estrogen levels, with G1, an activator of the estrogen membrane receptor GPER, capable of act

Estimation of hair cell loss from audiograms.

Age-related hearing loss arises from multiple cochlear pathologies. However, linking audiometric thresholds to their underlying cellular mechanisms remains a challenge. We present a biophysically grounded framework linking audiograms to the survival of inner and outer hair cells (IHCs and OHCs) along the cochlear partition. This approach uses a nonlinear, time-domain model of the peripheral auditory system, integrating cochlear mechanics, hair-cell transduction, and auditory-nerve responses. Using postmortem human data combining audiograms and histological hair-cell counts (Wu et al., 2020), w
