Causal associations of white blood cell subtypes with age-related hearing loss: A Mendelian randomization study.

While elevated white blood cell (WBC) counts are associated with auditory decline, the causal nature of this relationship remains elusive due to potential residual confounding. We utilized 2-sample Mendelian randomization (MR) to assess potential causal relationships between 5 genetically predicted WBC subtypes and the risk of age-related (ARHL) and sensorineural hearing loss. Two-sample MR analyses were conducted using summary statistics from large-scale genome-wide association studies. Causal estimates were assessed via inverse variance weighted, weighted median, weighted mode, and MR-Egger

Epigenetic aging is associated with hearing loss and multi-system disease risk independently of chronological age.

INTRODUCTION: ARHL (Age-related hearing loss) affects over one-third of people aged 65 and older worldwide. It is often viewed as an isolated sensory deficit, but whether it is associated with systemic biological aging remains unclear. METHODS: We analyzed NHANES (n = 2530) and HRS (n = 2849) cohorts. GrimAgeAcc (GrimAge acceleration) was used to assess biological aging. GO/KEGG (Gene Ontology/Kyoto Encyclopedia of Genes and Genomes) and disease association analyses were performed on GrimAge-related proteins. Prospective associations between ARHL and dementia, CVD (Cardiovascular Disease), an

High-Frequency Pseudo-Cohort Threshold Change and Severity-Dependent Sex Disparity in 4-kHz Audiometric Notching in Korean Adults.

OBJECTIVES.: Using contemporary 2019-2023 Korea National Health and Nutrition Examination Survey (KNHANES) audiometry, this study estimated population-level pseudo-cohort hearing-threshold change and the prevalence and correlates of 4-kHz audiometric notching in Korean adults aged ≥40 years. METHODS.: We included 12,576 adults aged ≥40 years with valid harmonized otolaryngologic/audiometry examination weights and sufficient audiometric data. Survey-weighted regression estimated pseudo-cohort hearing-threshold slopes within seven reconstructed five-year birth-cohort groups (1944-1978), after e

Advances in Research on the Association between Age-Related Hearing Loss and Risk of Falls and Fractures.

With the rapid aging of the global population, age-related hearing loss and fall-related fractures have become major public health problems that are seriously affecting the quality of life and health expectations of older adults. Traditionally, fall risk has been attributed to factors such as muscle weakness and vision loss. However, in recent years, increasing evidence has suggested that age-related hearing loss is an independent and significant risk factor for falls and related fractures. The aim of this study is to systematically review the epidemiological evidence supporting the associatio

Neurovascular-metabolic coupling links hearing loss to cognitive impairment: Evidence from GABA and cerebral blood flow in presbycusis.

Hearing loss is recognized as the leading modifiable risk factor for dementia. However, the neurobiological pathways bridging auditory dysfunction and cognitive decline remain poorly understood. We hypothesized that a neurovascular-metabolic cascade, involving neurotransmitter imbalances and perfusion deficits, underlies this link. We recruited 124 patients with presbycusis and 91 age-matched health controls. Auditory GABA and glutamate levels were quantified using macromolecule-suppressed MEGA-PRESS, and cerebral blood flow (CBF) was quantified using pseudo-continuous arterial spin labeling.

Protective effects of circPVT1 against D-galactose-induced senescence in HEI-OC1 auditory cells.

BACKGROUND: Age-related hearing loss (ARHL) is a progressive, bilaterally symmetric sensorineural hearing disorder characterized by predominant high-frequency threshold elevation, and it represents the most prevalent chronic sensory impairment in the geriatric population worldwide. Circular RNA PVT1 (circPVT1) has been identified as an endogenous inhibitor of cellular senescence in multiple cell types, but its expression profile and functional role in auditory cell senescence remain unknown. METHODS AND RESULTS: In this study, we established a D-galactose-induced senescence model in HEI-OC1 a

Speech perception deficits and neural network alterations during cocktail-party listening in age-related hearing loss.

Age-related hearing loss (ARHL) is associated with difficulty understanding speech in complex listening environments, yet its neural basis remains unclear. We combined behavioral measures with functional near-infrared spectroscopy (fNIRS) to examine speech perception and neural responses during cocktail-party listening. Sixty-eight participants (Young-NH, Older-NH, ARHL) completed a spatialized speech-recognition task under different masker types and signal-to-noise ratios, together with resting-state and task-based fNIRS recordings. ARHL showed poorer speech perception and reduced self-report

Hearing healthy: physical activity moderates the association between age-related hearing loss and brain volume.

Hearing loss is one of the most common age-related health conditions and is associated with a number of adverse psychological, physical, and social outcomes. Chronic under-stimulation, due to reduced auditory input from the ear is hypothesised to result in cascading neural consequences, including atrophy in auditory and associated cortices. One key factor known to protect healthy brain ageing is exercise, potentially by providing an alternative source of stimulation and environmental enrichment. As such, we tested whether engaging with physical activity moderates the association between hearin

Investigating the Relationship Between Personality Traits and Treatment Adherence With the Mediating Role of Coping Styles in Adults With Presbycusis: A Cross-Sectional Study.

BACKGROUND: Presbycusis, an age-related hearing disorder, affects communication, psychosocial functioning, and quality of life. Because its management requires long-term engagement with rehabilitation and medication routines, psychological factors such as personality traits and coping styles may influence treatment adherence. This study investigated the relationship between personality traits and adherence in adults with presbycusis, with coping styles examined as a mediating variable. METHODS: A descriptive cross-sectional study was conducted among adults diagnosed with presbycusis recruited

The decline of progranulin expression in cochlear Microglia-Like Cells accelerates the progression of age-related hearing loss.

Accumulating evidence indicates that crosstalk between immune cells and intrinsic cochlear cells is critical for maintaining auditory homeostasis. However, the activation profile of cochlear microglia-like cells (MLCs) and their mechanistic role in age-related hearing loss (ARHL) remain poorly defined. Here, we show that MLCs are robustly activated in the cochlea of aged mice, and that pharmacologic depletion of cochlear MLCs significantly attenuates ARHL progression. To dissect the mechanisms underlying MLC-driven cochlear damage in ARHL, we leveraged the single-cell transcriptomic landscape

PRMT6 drives hearing loss by promoting asymmetric arginine dimethylation-mediated FOXG1 degradation and suppressing mitophagy.

OBJECTIVE: Age-related hearing loss (ARHL) is a progressive and irreversible sensorineural impairment with incompletely understood mechanisms. This study investigates the role of protein arginine methyltransferase 6 (PRMT6) and its regulatory mechanisms in ARHL. METHODS: In vivo, D-galactose (D-gal)-induced aging mouse models were established. Auditory brainstem response (ABR), cochlear β-galactosidase activity, and PRMT6 expression were measured, and the therapeutic effects of the PRMT6 inhibitor EPZ020411 were evaluated. In vitro, D-gal-treated HEI-OC1 cells were employed. PRMT6 was silence

Age-related hearing loss begins with gain, not silence: physiological compensation in dorsal cochlear nucleus fusiform cells during auditory aging.

Age-related hearing loss (ARHL) is the most common cause of sensorineural hearing loss. The cochlear nucleus, the first central auditory structure to receive input from the cochlea, has been shown to be disrupted by ARHL. Fusiform cells (FCs), the principal output cell of the dorsal cochlear nucleus (DCN), mature physiologically during hearing onset. Specifically, FCs increase in the rate of action potential (AP) rise and decay, stabilizing by postnatal day 14 (P14) in mice. However, whether FC intrinsic electrophysiological properties or morphological characteristics continue to change throug

Evaluating perceptual evidence of cross-modal plasticity in older adults with hearing loss using an audio-visual word mismatch task.

Older adults with age-related hearing loss (OAHL) are more strongly influenced by vision during audiovisual perception tasks compared to typical-hearing older adults, suggesting a greater weighting of visual input in the former group. This visual "bias" has been argued to arise through compensatory, top-down cross-modal neuroplasticity that strengthens visual perceptual processing in response to hearing loss. However, current behavioural evidence does not convincingly demonstrate that cross-modal plasticity confers a visual benefit to OAHL, and existing methods do not untangle top-down influen

Assessment of age-related hearing loss and related everyday functional and psychosocial consequences in older adults: A systematic review of self-report instruments.

To detect age-related hearing loss (presbycusis), beyond objective audiometric measurements, self-report instruments are commonly employed. However, clear evidence regarding the methodological and measurement quality of self-report instruments for assessing hearing loss in older adults is lacking. This systematic review, for the first time, aims to identify and evaluate self-report instruments for assessing age-related hearing loss and related functional and psychological consequences in everyday life among older adults. Following PRISMA and COSMIN guidelines, we preregistered in PROSPERO (CRD

Does occupational noise exposure aggravate age-related hearing loss? A population-based study.

PURPOSE: To determine whether occupational noise exposure aggravates the progression of age-related hearing loss and to analyze frequency-specific annual threshold shifts. METHODS: We analyzed cross-sectional data from 3,719 participants (aged ≥ 40 years) in the 9th Korea National Health and Nutrition Examination Survey (KNHANES, 2023). Occupational noise exposure was defined as working in an environment with loud noise for ≥ 3 years. Linear regression with an interaction term (age × noise exposure) was used to compare the annual rate of threshold deterioration (dB/year) for each frequency.
