Dynamic brain functional connectivity in age-related hearing loss during auditory selective spatial attention.

Age-related hearing loss (ARHL) is a common health problem that impairs auditory perception. However, the dynamic patterns of brain functional connectivity in ARHL during auditory spatial selective attention have not been thoroughly investigated. In this study, 32 older adults were recruited to investigate the dynamic brain functional connectivity in ARHL. First, an experimental paradigm for auditory spatial selective attention was designed, and neural electrical signals were recorded using electroencephalography. Then, a multilayer time-varying brain network was constructed based on multiple

Study in Murine Model: Is There a Relationship Between Presbycusis and Frailty?

Age-related hearing loss, or Presbycusis, is the most frequent sensory deficiency in older adults and is associated with comorbidities such as falls, cognitive decline, and frailty. Frailty is related to poor health outcomes in old age. Recent research suggested that age-related hearing loss may be a potentially modifiable risk factor for frailty, although inconclusive. The use of animal models to study the correlation between age-related hearing loss and frailty is important to test future interventions to be translated into clinical practice. The aim of this study was to determine if there i

Sex-Specific Age-Related Changes in Excitatory and Inhibitory Intracortical Circuits in Mouse Primary Auditory Cortex.

A common impairment in aging is age-related hearing loss (presbycusis), which manifests as impaired spectrotemporal processing. Presbycusis can be caused by a dysfunction of the peripheral and central auditory system, and these dysfunctions might differ between the sexes. To date, the circuit mechanisms in the central nervous system responsible for age-related auditory dysfunction remain mostly unknown. In the auditory cortex (ACtx), aging is accompanied by alteration in normal inhibitory (GABA) neurotransmission and changes in excitatory (NMDA and AMPA) synapses, but which circuits are affect

Neural correlates of age-related hearing loss: An MRI and FDG-PET study.

AIM: Age-related hearing loss (ARHL) is a common problem among older adults and contributes to adverse health outcomes such as cognitive impairment. However, the neural mechanisms underlying ARHL remain unclear. We aimed to reveal the structural and metabolic (i.e., neural activity) correlates of ARHL using magnetic resonance imaging (MRI) and positron emission tomography with 18F-fluorodeoxyglucose (FDG-PET). METHODS: A total of 162 community-dwelling older adults without cerebral disorders or cognitive impairment (Mini-Mental State Examination score < 24) were included in this study. Audito

Impaired network organization in mild age-related hearing loss.

Age-related hearing loss (ARHL) is considered one of the most common neurodegenerative disorders in the elderly; however, how it contributes to cognitive decline is poorly understood. With resting-state functional magnetic resonance imaging from 66 individuals with ARHL and 54 healthy controls, group spatial independent component analyses, sliding window analyses, graph-theory methods, multilayer networks, and correlation analyses were used to identify ARHL-induced disturbances in static and dynamic functional network connectivity (sFNC/dFNC), alterations in global network switching and their

Administration of nicotinamide mononucleotide suppresses the progression of age-related hearing loss in mice.

Age-related hearing loss (ARHL) is a widespread problem in the elderly, significantly impairing their quality of life. Despite its high prevalence, no fundamental treatment for ARHL has been established. Nicotinamide adenine dinucleotide (NAD+) is required for various biological processes and tissue levels of the coenzyme NAD+ are known to decrease with age. A previous report suggested that declining NAD+ levels induce age-related diseases and NAD+ supplementation might be effective for treating or preventing age-related diseases. To clarify the effect of NAD+ supplementation on ARHL, C57BL/6J

Identification of therapeutic target genes for age-related hearing loss through systematic genome-wide mendelian randomization of druggable genes.

BACKGROUND: Age-related hearing loss (ARHL) is a common sensory disorder with significant public health implications. However, few effective treatment options are available. Mendelian randomization (MR) has been used to repurpose existing drugs and identify new therapeutic targets. Therefore, we performed a systematic genome-wide MR of drug-eligible individuals to explore potential therapeutic targets for ARHL. METHODS: We obtained data on the expression quantitative trait locis (eQTLs) of druggable genes, which were then subjected to two-sample MR analyses and co-localisation analyses with d

Innovative treatment of age-related hearing loss using MSCs and EVs with Apelin.

Utilizing single-cell transcriptome sequencing (scRNA-seq) technology, this study explores the viability of employing mesenchymal stem cells (MSCs) as a therapeutic approach for age-related hearing loss (ARHL). The research demonstrates MSCs' ability to differentiate into inner ear cell subpopulations, particularly hair cells, delivering Apelin via extracellular vesicles (EVs) to promote M2 macrophage polarization. In vitro experiments show reduced inflammation and preservation of hair cell health. In elderly mice, MSCs transplantation leads to hair cell regeneration, restoring auditory functi

Bioinformatics approach reveals the critical role of inflammation-related genes in age-related hearing loss.

Age-related hearing loss (ARHL) is the most prevalent sensory impairment in the elderly. However, the pathogenesis of ARHL remains unclear. This study was aimed to explore the potential inflammation-related genes of ARHL and suggest novel therapeutic targets for this condition. Initially, a total of 105 Inflammatory related differentially expressed genes (IRDEGs) were obtained by overlapping the differentially expressed genes from the GSE49522 and GSE49543 datasets with Inflammatory related genes. The IRDEGs were mainly enriched in MAPK, PI3K-Akt, Hippo and JAK-STAT pathways by analysis of Gen

Digital screening for cognitive decline in the Spanish language for older adults.

BackgroundRecent technological advances in digital assessment of auditory and cognitive function may be used to circumvent the costs associated to screening for cognitive decline in the general population. Pre-clinical cognitive screening could have a transformative impact for preventive care in our increasingly old world population. However, advances in digital assessment need to be adapted for Spanish-speaking populations as innovation occurs mainly in English.ObjectiveThis study explores the potential of a novel screening battery for cognitive decline that utilizes digital tests of cognitiv

How Aging and Age-Related Hearing Loss Affect the Recognition of Emotion in Whispered Speech.

Whispering is a common adverse hearing condition. However, it is still unclear whether older adults have more difficulty perceiving emotions in whispered speech, and whether hearing loss contributes to these difficulties. To fill this research gap, we compared emotional prosody perception under phonated and whispered conditions in three groups of participants (younger adults, and older adults with and without hearing loss). The results revealed that both older adult groups were less accurate when processing emotions in whispered speech. Moreover, older adults with hearing loss performed worse

RONIN/HCF1-TFEB Axis Protects Against D-Galactose-Induced Cochlear Hair Cell Senescence Through Autophagy Activation.

Age-related hearing loss is characterized by senescent inner ear hair cells (HCs) and reduced autophagy. Despite the improved understanding of these processes, detailed molecular mechanisms underlying cochlear HC senescence remain unclear. Transcription Factor EB (TFEB), a key regulator of genes associated with autophagy and lysosomes, crucially affects aging-related illnesses. However, intricate regulatory networks that influence TFEB activity remain to be thoroughly elucidated. The findings revealed that RONIN (THAP11), through its interaction with host cell factor C1 (HCF1/HCFC1), modulated

Transforming Growth Factor-β-Activated Protein 1 (TAK1) Regulates Necroptosis in Age-Related Hearing Loss.

Inflammation plays an important role in age-related hearing loss (ARHL). Transforming growth factor-β-activated protein 1 (TAK1), a key factor upstream of inflammatory pathways, mediates various cell death pathways, potentially influencing the survival and death of cochlear hair cells. The DBA/2 J mouse model and the HEI-OC1 cell line were used to investigate the mechanism of TAK1-mediated inflammation in ARHL. Hematoxylin and eosin staining revealed significant histological damage in the cochlea of 16-week-old mice, along with an increase in auditory-evoked brainstem response thresholds. Conc
