The Auditory-Cognitive Assessment of Speech Understanding: A Comprehensive Analysis of Construct Validity.

OBJECTIVES: Age-related hearing loss, the predominant global cause of hearing loss in middle-aged and older adults, presents a significant health and social problem, particularly affecting speech understanding. Beyond the auditory system, cognitive functions play a crucial role in speech understanding, especially in noisy environments. Although visual cognitive testing is commonly used as an intriguing alternative to mitigate the potential adverse effects of hearing loss on the perception of auditory test items, its efficacy within a hearing-related context is questionable due to construct dif

Mendelian non-syndromic and syndromic hearing loss genes contribute to presbycusis.

Age-related (AR) hearing loss (HL) is the most prevalent sensorineural disorder in older adults. Here we demonstrate that rare-variants in well-established Mendelian HL genes play an important role in ARHL etiology. In all we identified 32 Mendelian HL genes which are associated with ARHL. We performed single and rare-variant aggregate association analyses using exome data obtained from white-Europeans with self-reported hearing phenotypes from the UK Biobank. Our analysis revealed previously unreported associations between ARHL and rare-variants in Mendelian non-syndromic and syndromic HL gen

Overexpression of CX3CL1-CX3CR1 Signaling in the Stria Vascularis rather than the Basilar Membrane May Be the Trigger for Inflammaging in the Cochlea.

INTRODUCTION: Inflammaging is a key mechanism in presbycusis. CX3CL1-CX3CR1 pathway is critical for cochlear macrophage-hair cell cross-talk. However, its role in inflammaging remains unclear. METHODS: To investigate the role of CX3CL1-CX3CR1 signaling in cochlear inflammaging, single-cell RNA sequencing (scRNA-seq) data from young and aged mouse cochleae were analyzed to map CX3CL1-CX3CR1 distribution and aging-related trends. Findings were validated with immunofluorescence, real-time quantitative polymerase chain reaction, and Western blot. A migration assay assessed CX3CL1-CX3CR1's influen

Expression of SLC2A1 is upregulated in a mouse model of age-related hearing loss.

BACKGROUND: Age-related hearing loss (ARHL) is a common issue associated with aging. One of the typical causes of hearing loss is the irreversible death of hair cells. In addition, oxidative stress contributes to ARHL. However, the underlying molecular mechanism in ARHL is not well understood. METHODS: The ARHL samples were from GSE153882 datasets of the Gene Expression Omnibus database. The Limma R-package was used to identify differentially expressed genes. The hub gene was obtained via intersection of oxidative stress genes and cuproptosis genes. Hearing function was measured using the aud

TNFAIP8L2 maintains hair cell function and regulates age-related hearing loss via mTORC1 signaling.

Age-related hearing loss (ARHL) is one of the most prevalent and complex disorders. Our previous study demonstrated that abnormal activation of mammalian target of rapamycin complex 1 (mTORC1) signaling in the cochlear neurosensory epithelium causes auditory hair cell (HC) damage and contributes to ARHL. However, the underlying mechanism of mTORC1 activation remains unclear. In this study, we identified tumor necrosis factor-alpha-induced protein 8-like 2 (TNFAIP8L2), an immune regulatory gene, as a potential candidate. To elucidate the effect of TNFAIP8L2 on mTORC1 signaling in the neurosenso

Middle-aged CBA/CaJ mice exhibit auditory dysfunction in background noise.

Aging is associated with deficits in auditory functioning. Characterization of auditory deficits that originate in middle-age is crucial for understanding the initial age-related functional impairments and the spatio-temporal progression of age-related auditory pathophysiology. Early age-related deficits in auditory processing are evident in difficult listening conditions, such as background noise, before becoming evident in quiet. To investigate the effect of noise on age-related auditory dysfunction, we collected suprathreshold auditory brainstem responses (ABRs) from young, middle-aged, and

Transcriptomic analysis reveals the function of m6A regulators in aged cochlea.

OBJECTIVE: Presbycusis is a prevalent health issue among the elderly. Previous studies have shown mechanisms related to this condition, but the underlying mechanisms of presbycusis remain elusive. N6-methyladenosine (m6A) modification in regulating gene expression and cellular functions has been implicated in the development of various diseases. Nevertheless, the potential role of m6A regulators in presbycusis is still unclear. In this study, we aim to determine the expression of m6A regulators in the cochleae of young and old mice, and to investigate their potential role in aging. METHODS: W

Aging reduces excitatory bandwidth, alters spectral tuning curve diversity, and reduces sideband inhibition in L2/3 of primary auditory cortex.

Presbycusis, or age-related hearing loss, is caused by changes in both the peripheral and the central auditory system. Many of the peripheral structures that degrade with age have been identified and characterized, but there is still a dearth of information pertaining to what changes occur in the aging central auditory pathway that are independent of peripheral degradation. The primary auditory cortex (A1) of aging mice shows reduced suppressive responses and reduced diversity of temporal responses suggesting alteration of inhibitory processing. To gain a better understanding of how tuning fea

Modification and validation of a GAD-GFP mouse line without accelerated aging-related hearing loss.

GABAergic neurons in the inferior colliculus (IC) play a crucial role in auditory processing by extracting specific features of sounds (Ono et al., 2005). The Gad67-GFP mouse model developed by Tamamaki et al. in 2003 on a Swiss background facilitates studying these neurons by using a green fluorescent protein that is expressed endogenously via the GAD67 promoter. Unfortunately, this mouse suffers from accelerated aging-related hearing loss, limiting its utility in studying the auditory system. Here, we report the results of an 8-generation backcross of this line onto CBA/CaJ mice, which produ

Novel_circ_0004013 targeting miR-29a-3p affects age-related hearing loss in miR-29a mouse model by RNA-seq analysis.

Age-related hearing loss (ARHL) is a gradual, symmetrical sensorineural disorder. Exploring the pathogenesis of ARHL from a biological perspective is important for its treatment. In this study, we analyzed the circRNA expression profiles of 2-month-old miR-29a+/+ mice and miR-29a-/- mice by transcriptome sequencing to investigate the role of circRNAs in ARHL. We identified 12 differentially expressed circRNAs in the two groups. Our focus was on circRNAs predicted to regulate miR-29a, with novel_circ_0004013 identified as having a targeted binding relationship with miR-29a-3p. Dual luciferase a
