Oxaliplatin-chemotherapy induced cochlear synaptopathy.

Oxaliplatin is widely used as a first-line and reference anticancer agent for advanced colorectal cancer. However, it frequently causes peripheral neuropathy. It is generally accepted that oxaliplatin is significantly less ototoxic than the other platinum-based antineoplastic drugs, cisplatin and carboplatin. This study aimed to evaluate potential hearing damage induced by oxaliplatin in young male CBA/JRj mice. Mice received intraperitoneal injections of oxaliplatin (3 mg/kg) twice weekly for 3 weeks, for a cumulative dose of 18 mg/kg. Auditory function was assessed using electrophysiological

Normative Ranges for Wideband Middle Ear Muscle Reflex Magnitude: Limited Potential for Diagnosing Cochlear Deafferentation.

PURPOSE: Cochlear synaptopathy, the loss of the synapses between the inner hair cells and their auditory nerve fiber targets, is expected to be a common type of auditory deficit resulting from noise exposure or aging. Unfortunately, there is currently no means for diagnosing cochlear synaptopathy or other forms of cochlear deafferentation. Wideband middle ear muscle reflexes (MEMRs) have been proposed as a potential diagnostic indicator of cochlear deafferentation, but we lack normative ranges for MEMR magnitude. The objective of this study was to develop normative ranges for wideband MEMR mag

Impaired processing of time-critical language information in Parkinson's disease.

BACKGROUND: Parkinson's disease (PD) disturbs basal ganglia functioning and the precise coding of time-critical information. Beyond its known impact on movement capacities, it may therefore affect cognitive functions with high demands of temporal information coordination. We studied this via the processing of syntactic boundaries during auditory sentence parsing. OBJECTIVE: To analyze the processing of syntactic structure as a function of position and duration of prosodic breaks in persons with PD compared to controls. METHODS: 27 persons with PD and 26 healthy controls judged the grammatica

Acute Hallucinogen Intoxication Following Ingestion of a Commercially Available Mushroom Extract Product: A Case Report.

Psilocybin is a naturally occurring hallucinogen known to produce alterations in perception, cognition, and autonomic function through its effects on central serotonergic pathways. Products marketed as "mushroom" formulations may contain psilocybin or related psychoactive compounds despite ambiguous labeling and limited regulatory clarity, creating diagnostic challenges for clinicians in acute care settings. We present the case of an 18-year-old female with no prior medical or psychiatric history who presented to the emergency department with sudden-onset severe anxiety, agitation, tremors, di

Retinal Origins of Aberrant Salience in Schizophrenia: The Perceptual Theory.

In schizophrenia research, visual processing abnormalities have traditionally been viewed as downstream consequences of cortical dysfunction. However, the epidemiological observation that congenital cortical blindness appears to confer protection against schizophrenia, while peripheral blindness, regardless of its developmental timing, does not, indicates that visual input itself may play a more fundamental role in disease pathogenesis. Here we propose the Perceptual Theory of Schizophrenia (PerTh), a framework positing that instability in retinal dopamine signaling acts as an initiating mecha

Masseter Muscle in Somatosensory Tinnitus and Potential Therapeutic Role for Botulinum Toxin Type A: A Scoping Review.

Background: Increasing evidence supports the existence of a somatosensory subtype of tinnitus, in which craniocervical-craniomandibular structures can modulate tinnitus perception also through central auditory pathways. Despite growing mechanistic evidence, no targeted pharmacological intervention has been validated for this subtype. Objectives: This study aimed to synthesize the anatomical and neurophysiological rationale linking masseter hyperactivity to somatosensory tinnitus and to discuss BoNT/A as a hypothesis-generating therapeutic option requiring clinical validation. Methods: A PRISMA

Research Progress on Pathogenesis, Modern Interventions and Neuromodulatory Mechanisms of Acupuncture for Tinnitus.

Tinnitus refers to the subjective abnormal auditory perception occurring without external sound stimuli, characterized by prominent clinical heterogeneity and complicated pathophysiological mechanisms, which severely impair patients' mental health and quality of life. Adopting a standardized literature retrieval procedure, this paper systematically sorts out the epidemiological characteristics, clinical classifications and diagnostic criteria of tinnitus, and deeply analyzes its dual pathological mechanisms involving peripheral auditory damage (hair cell injury, auditory nerve lesions) and abn

Atg7-dependent autophagy is indispensable for mice spiral ganglion neurons.

Atg7-dependent autophagy is critical for the long-term maintenance of the post-mitotic cells, yet its role in spiral ganglion neurons (SGNs) remains incompletely defined. Here, we conditionally ablated Atg7 in SGNs by generating Bhlhe22Cre/+Atg7flox/flox mice and examined cellular p62 aggregates, ultrastructure, and auditory function at postnatal day (P)30 and P60. At P30, Atg7 was markedly reduced in SGNs. Immunofluorescence revealed progressive enlargement of p62-positive condensates from nano-scale at P30 to micron-scale by P60. Excess p62 activated the antioxidant transcription factor Nrf2

Binaural hearing, neurological disability and brain imaging in Multiple Sclerosis.

Multiple sclerosis is an inflammatory autoimmune disease characterized by focal destruction of myelin within the central nervous system. Auditory deficits are a relatively frequent, but often underdiagnosed symptom. This study examined sound detection threshold and functional hearing ability in a cohort of patients with multiple sclerosis. Thirty adults with relapsing-remitting multiple sclerosis and 30 age, gender and hearing-level matched controls underwent peripheral and central auditory assessments. These included audiometric (sound detection threshold) measurement, binaural speech percept

Binaural Fusion Sharpens on a Scale of Octaves During Pre-adolescence in Children with Normal Hearing, Hearing Aids, and Bimodal Cochlear Implants, but not Bilateral Cochlear Implants.

PURPOSE: The breadth of binaural pitch fusion, the integration of sounds differing in frequency across the two ears, can limit the ability to segregate and understand speech in background noise. Binaural pitch fusion is one type of central auditory processing that may still be developing in the pre-adolescent age range. In addition, children with hearing loss potentially have different trajectories of development of central auditory processing compared to their normal-hearing (NH) peers, due to disruption of auditory input and/or abnormal stimulation from hearing devices. The goal of this stud

Time moving 100-fold slower: time distortion as a diagnostic clue in anti-NMDA receptor encephalitis.

BACKGROUND: The primary symptoms in the early stages of anti-NMDA receptor encephalitis are psychiatric manifestations, making it difficult to distinguish from psychiatric disorders. While anti-NMDA receptor encephalitis requires a completely different treatment approach, the specific psychiatric features of the condition remain poorly identified. Although previous studies have suggested that altered perceptions may be characteristic, few case reports focus on altered perceptions of time or time distortion, a phenomenon closely linked to NMDA receptor dysfunction as seen in individuals using N

Loudness perception deficits during altered and absent auditory feedback in Parkinson's disease.

Patients with Parkinson's disease (PD) present with speech difficulties including abnormal speech intensity regulation. It is possible that the neural circuitry in speech may be unique and more complex compared to the other major motor symptoms. The current study aimed to provide a better understanding of the sensorimotor integration and loudness perception deficits in PD using an altered intensity feedback (AIF) paradigm. Twenty-six participants with PD and 26 neurologically healthy control participants completed a magnitude production task (normal loudness, 2× louder, 4× louder, and max loud

Altered auditory feature discrimination in a rat model of Fragile X Syndrome.

Atypical sensory processing, particularly in the auditory domain, is one of the most common and quality-of-life affecting symptoms seen in autism spectrum disorders (ASD). Fragile X Syndrome (FXS) is a leading inherited cause of ASD and a majority of FXS individuals present with auditory processing alterations. While auditory hypersensitivity is a common phenotype observed in FXS and Fmr1 knockout (KO) rodent models, it is important to consider other auditory coding impairments that could contribute to sound processing difficulties and disrupted language comprehension in FXS. We have shown pre

The neuroscience of music perception: a narrative review.

The present review article explores the neuroscience of musical perception, examining the roles of specific brain regions in decoding and interpreting music. Musical perception engages multiple cortical and subcortical areas that work in an integrated manner to process musical elements such as melody, harmony, and rhythm. The paper reviews the current knowledge about the brain circuits involved, as well as pathological conditions that result in abnormalities of musical perception. In addition, the relationship between musical perception and neurological conditions such as epilepsy and Alzheime

Comparison of One-Year auditory rehabilitation outcomes by etiology in pediatric patients with bilateral severe hearing loss (70-90 dB): enlarged vestibular aqueduct vs. Other causes.

PURPOSE: This study aimed to compare short-term language outcomes following hearing aid rehabilitation in pediatric patients with severe bilateral hearing loss (70-90 dB), with a particular focus on differences according to etiology. We hypothesized that children with enlarged vestibular aqueduct (EVA) exhibit more favorable speech development compared to those with other genetic or structural causes of hearing loss, and explored the potential presence of a "hidden" air-bone gap associated with EVA. METHODS: We retrospectively reviewed 36 children under five years of age diagnosed with bilate
