Baicalin inhibits Chlamydia trachomatis growth by impairing elementary body infectivity.

Chlamydia trachomatis (C. trachomatis) infection is a prevalent sexually transmitted disease worldwide. Although antibiotics are the standard first-line treatment, the rising incidence of treatment failure highlights the need for alternative therapeutic strategies. Baicalin, a natural flavonoid compound extracted from Scutellaria baicalensis, is known to possess antimicrobial properties. This study aimed to evaluate the anti-chlamydial effects of baicalin. Our results showed that baicalin significantly inhibited the growth of multiple C. trachomatis serovars (A, D, and L2) in HeLa cells, as in

Poriferast-5-en-3β-ol improves pelvic inflammatory disease via reducing GADD45A-induced inflammation.

BACKGROUND: Pelvic inflammatory disease (PID), often triggered by pathogens like Chlamydia, is a major cause of female infertility. This study investigated molecular mechanisms in PID and the therapeutic potential of DanZhiYin (DZY) and its components. METHODS: Bioinformatic analysis of dataset GSE110106 identified differentially expressed genes in PID, validated clinically. A Chlamydia infection model in OE-E6/E7 cells was used. siRNA knockdown and overexpression revealed GADD45A's role. Anti-inflammatory effects of DZY, Salvia miltiorrhiza, and Poriferast-5-en-3β-ol were tested via quantita

Chlamydial membrane vesicles deliver the beta barrel outer membrane protein OmpA to mitochondria to inhibit apoptosis.

Chlamydiae are obligate intracellular bacteria that inhibit mitochondrial apoptosis to maintain integrity of the host cell. We have previously reported that a chlamydial outer membrane β-barrel protein, OmpA, can during ectopic expression inhibit mitochondrial apoptosis through direct interaction with the BCL-2-family effectors BAX and BAK. We here show that OmpA from Chlamydia trachomatis (Ctr) uses membrane vesicles for its delivery to the outer mitochondrial membrane during Ctr infection. Using a number of imaging and fractionation techniques, we show that OmpA during infection reaches mito

Distribution and trends of the global burden of female infertility attributable to sexually transmitted infections from 1990 to 2021.

BACKGROUND: Sexually transmitted infections (STIs) contribute to female infertility by causing pelvic inflammatory disease (PID) and fallopian tube damage. Utilizing the Global Burden of Disease (GBD) database, we aim to comprehensively evaluate the global disease burden and spatiotemporal trends of STIs-related infertility, which is critical for formulating targeted intervention strategies. METHODS: Using data from the GBD 2021 database, we extracted age-standardized prevalence rates (ASPR) and age-specific prevalences (in 5-year intervals from 15 to 49 years) for STIs-related primary infert

Chlamydia spp., Bordetella bronchiseptica, and Phascolarctid Gammaherpesvirus 1 and 2 Infections in Koalas (Phascolarctos cinereus) in South East Queensland, Australia: Detection in Healthy Individuals and Those with Signs of Respiratory or Other Disease.

The decline in koala (Phascolarctos cinereus) populations has been significantly driven by infectious diseases, with chlamydial disease contributing to this trend. Chlamydia pecorum is often codetected with viruses such as phascolarctid gammaherpesvirus 1 and 2 (PhaHV-1 and PhaHV-2). Koalas can also be infected with other bacteria, including Bordetella bronchiseptica, which causes sporadic respiratory disease outbreaks. Respiratory infections and respiratory disease in koalas remain under-investigated. This study reports the detection of C. pecorum, Chlamydia pneumoniae, Chlamydia psittaci, B.

Identification of 3-cyanoquinolines as broad-spectrum inhibitors of Monkeypox virus and other sexually transmitted pathogens.

Sexually transmitted infections (STIs) remain a major global health concern, contributing significantly to morbidity and facilitating the co-transmission of other pathogens. Recent outbreaks of Monkeypox virus (MPXV) have further underscored the urgent need for broad-spectrum antiviral agents effective against emerging and re-emerging sexually transmissible viruses. We report here the design and synthesis of a series of 3-cyanoquinoline-based Src inhibitors to evaluate their antimicrobial efficacy against sexually transmitted pathogens. Among them, compound 7d demonstrated potent inhibitory ac

Unveiling the prevalence of sexually transmitted infections and its etiology among married women of remote South Andaman Island in India.

Sexually transmitted infections (STIs) are a global public health concern, particularly among women of reproductive age and leads to severe health complications. Married women, despite being perceived as low-risk, are often excessively affected due to factors such as partner infidelity, and lack of negotiation power in sexual relationships. Therefore, the study aimed to understand the prevalence of STIs among married women of South Andaman Islands. A cross-sectional study with an analytical focus was conducted on STIs in South Andaman after obtaining the ethical approval. Written informed cons

Elevated levels of advanced glycation end-products AGE10 in patients with reactive arthritis caused by Chlamydia trachomatis and Epstein-Barr virus infection.

Advanced glycation end products (AGEs), and particularly the unique AGE10 epitope, may be a potential biomarker of immunopathology in rheumatic diseases. They may be associated with inflammation, joint damage and ossification processes. AGE10 present in human and animal tissues could be detected with monoclonal antibody against melibiose-derived glycation product MAGE synthesized in anhydrous conditions. This MAGE product was different from the classic synthesis in water solution. The epitope was determined in serum with ELISA using these anti-MAGE monoclonal antibodies. This work aims to dete

Identification of collaborative cross mouse strains susceptible to chlamydial induction of hydrosalpinx.

Sexually transmitted infection with Chlamydia trachomatis can cause pathology, such as hydrosalpinx, in the upper genital tract, leading to infertility. To investigate how genetic variation affects chlamydial pathogenicity, we screened five strains of Collaborative Cross (CC) mice for susceptibility to intravaginal infection and hydrosalpinx induction by C. muridarum, a mouse-adapted chlamydial species used extensively to reveal chlamydial pathogenic mechanisms. In terms of susceptibility to genital chlamydial infection, the five CC strains fell into two categories: CC011 and CC012 were resist

Pathobiont-triggered induction of goblet cell response drives regional susceptibility to inflammatory bowel disease.

The gastrointestinal tract varies in structure and function by region, yet the drivers of region-specific inflammatory disease remain elusive. Here, a TNF-overexpressing murine model (TnfΔARE/+) of Crohn's disease (CD) was used to investigate how pathobionts interact with host immune susceptibilities to drive region-specific disease. We identified the pathobiont Chlamydia muridarum, an intracellular bacterium and murine counterpart to the human sexually transmitted C. trachomatis, as a necessary and sufficient trigger for disease manifestation in the proximal/ascending colon, a common site of

Evaluation of four molecular detection assays for the diagnosis of Chlamydia pecorum infection in Victorian koalas (Phascolarctos cinereus) using Bayesian latent class analysis.

Chlamydiosis poses a significant threat to koala health and population viability. Accurate diagnosis of chlamydial infection in koalas at veterinary hospitals informs patient prognosis, biosecurity and determines treatment. However, there is no standardised approach to the diagnosis of chlamydiosis in wildlife hospitals. The performance of available diagnostic tests has not previously been rigorously assessed, and there is a lack of sufficient well-characterised reference samples. This study aimed to estimate the diagnostic accuracy of four molecular detection assays for Chlamydia pecorum infe

Comparative transcriptomics reveals genes commonly induced by distinct stressors in Chlamydia.

Chlamydia trachomatis is a leading cause of urogenital infections that can result in serious long-term complications. This obligate intracellular bacterium undergoes a biphasic developmental cycle alternating between the infectious elementary body and the replicative reticulate body and can enter a persistent state in response to adverse environmental conditions. Although transcriptomic reprogramming is central to chlamydial stress adaptation and persistence, how responses differ across biologically distinct stressors remains incompletely defined. Here, we performed a comparative reanalysis of

Narrative Review of Chlamydia trachomatis Culture, Viability Testing, and Sequencing Methods.

BACKGROUND: Chlamydia trachomatis (CT) is the most commonly reported bacterial sexually transmitted infection in the United States and is routinely detected using nucleic acid amplification tests (NAATs). However, its obligate intracellular life cycle creates challenges for culture, viability determination, and antimicrobial susceptibility testing (AST). Despite these challenges, culture remains important for select applications, including strain typing, test-of-cure, sexual assault investigations, and resistance monitoring. METHODS: We conducted a systematic literature search across 5 electr

Phage therapy for treatment of bacterial vaginosis.

A common vaginal dysbiosis, bacterial vaginosis (BV), has serious consequences for reproductive health, including an elevated risk of infertility and pelvic inflammatory disease. Its distinctive feature is a robust polymicrobial biofilm, mainly composed of Gardnerella species (spp.), which protects infections against antibiotic therapy and promotes high rates of recurrence. Concurrently, Chlamydia trachomatis (CT) is the most common bacterial sexually transmitted infection globally, with over 130 million new cases annually. A primary cause of tubal factor infertility, CT infection promotes adn
