Disulfiram Exhibits Potent Copper-Dependent Antimicrobial Activity Against Mycoplasma genitalium.

BACKGROUND: Mycoplasma genitalium (Mgen) is a sexually transmitted urogenital pathogen. Studies in sexual health clinic settings have demonstrated that Mgen infection, with similar prevalence to Chlamydia trachomatis infection, is becoming increasingly resistant to the limited classes of antibiotics with in vitro and clinical activity. Disulfiram (DS) is a Food and Drug Administration-approved drug for treatment of alcohol dependence that shows promise for repurposing as an antimicrobial agent. METHODS: Minimum inhibitory concentration was performed via standard methods for human mycoplasmas

Exploring the clinical characteristics and etiological diagnosis of Chlamydia psittaci pneumonia confirmed by targeted next-generation sequencing: A retrospective observational study.

Chlamydia psittaci (C psittaci) is an important causative agent of community-acquired pneumonia (CAP). However, conventional pathogen detection methods are often ineffective in the early diagnosis of CAP, making C psittaci pneumonia challenging to detect. C psittaci ranges from mild, self-limited to acute respiratory failure, and the early identification of this disease can be challenging. Targeted next-generation sequencing (tNGS) of bronchoalveolar lavage fluid has the potential to improve the pathogen identification in C psittaci pneumonia. This retrospective study aims to systematically ev

From models to medicine: systematic development and evaluation of animal models for Chlamydia psittaci respiratory and genital tract infections.

Chlamydia psittaci (C. psittaci) is a zoonotic pathogen causing severe respiratory infections and systemic complications. Animal models are a crucial approach for studying C. psittaci. They can simulate the natural infection process and reveal the interaction between the host and the pathogen. This review examines the roles and limitations of various animal models in understanding the infection mechanism, developing vaccines and therapies, and researching the evolution and transmission of the pathogen. Mouse and guinea pig models are widely used in immunological research because they have nume

BACKGROUND: Gynecological cancers are among the most common cancers in women that affect female reproductive organs. The most common gynecological cancers are ovarian, cervical, uterine/endometrial, vaginal, and vulvar cancer. Women's reproductive organs have a dynamic and relative microbial balance. The disruption in the balance of the microbiome could result in numerous gynecological diseases, as well as, gynecological cancers. In this study, we aimed to review new findings on the role of different bacterial infections in various types of gynecological cancers. RECENT FINDINGS: The role of

Pelvic Inflammatory Disease: A Narrative Review of Disparities Between Black and White Women in the United States and Strategies for Improvement.

Pelvic Inflammatory Disease (PID) is an inflammatory syndrome of the upper female reproductive organs caused by infection, most commonly via Neisseria gonorrhoeae or Chlamydia trachomatis, spreading to one or more reproductive organs. With PID responsible for many of the major morbidities of sexually transmitted infections (STIs), such as ectopic pregnancy and chronic pelvic pain, the urgency is high to mitigate the impacts of untreated PID, especially in higher-risk populations. This paper discusses the current disparities in PID diagnosis, treatment, and management between Black and white wo

Towards precision management of Mycoplasma genitalium: a real-world cohort study identifying key predictors for treatment failure and the superiority of sequential therapy.

BACKGROUND: The management of Mycoplasma genitalium (MG) infection is challenged by rising macrolide resistance, leading to high failure rates with azithromycin. Evidence on the long-term effectiveness of alternative initial regimens, particularly sequential therapy, and easily obtainable predictors for poor outcomes remains scarce. METHODS: We conducted a retrospective cohort study at a tertiary hospital in China (2018-2024). Sexually active adults with nucleic acid amplification test (NAAT)-confirmed MG infection and available treatment records were included. The primary outcomes were treat

Epidemiological characteristics and environmental surveillance of human psittacosis in Lishui City, Zhejiang Province, China (2021-2024).

INTRODUCTION: Psittacosis, caused by Chlamydia psittaci, is an underdiagnosed zoonosis that can lead to severe pneumonia and fatal outcomes. In China, traditional poultry farming poses substantial risks for avian-to-human transmission, yet comprehensive epidemiological evidence is scarce. To address this gap, we aimed to define the local epidemiology, risk factors, and environmental reservoirs of human psittacosis in Lishui City, Zhejiang Province. METHODS: We conducted a multi-source epidemiological study (2021-2024) integrating surveillance data, clinical records, contact investigations, an

Clinical Management and Outcomes of Chlamydia pneumoniae Pneumonia Diagnosed by Targeted Next-Generation Sequencing: A Case Report and Insights.

Chlamydia pneumoniae is a significant cause of community-acquired pneumonia, yet its diagnosis remains challenging due to non-specific symptoms and the limitations of traditional methods like serology, which can cause delays. We present the case of a 14-year-old female admitted with cough and right-sided chest pain. Initial laboratory tests were unremarkable, except for a positive C. pneumoniae IgM result. A chest CT scan revealed right upper lobe infiltrates. Despite empirical azithromycin, her improvement was limited. Bronchoscopy was performed, and bronchoalveolar lavage fluid was analyzed

Genetic diversity of collaborative cross mice enables the establishment of a novel Chlamydia muridarum female genital tract infection model.

Chlamydial infection in women displays wide variation in bacterial burden, persistence, and risk of upper genital tract pathology, yet the host genetic factors underlying this heterogeneity remain poorly defined. We evaluated genital tract infection with Chlamydia muridarum across 20 Collaborative Cross (CC) strains, a recombinant inbred mouse panel that captures broad genetic diversity with high within-strain reproducibility. CC strains exhibited striking differences in early bacterial burden, time to clearance, and oviduct pathology, including prolonged low-inflammatory infections and burden

Epidemiology and antimicrobial resistance of Mycoplasma genitalium among female sex workers in China: a cross-sectional study.

BACKGROUND: Mycoplasma genitalium (MG) is an important sexually transmitted pathogen associated with cervicitis, pelvic inflammatory disease and adverse reproductive outcomes. Female sex workers (FSWs) are at elevated risk; however, data on MG epidemiology and antimicrobial resistance in this population in China remain limited. This study assessed the prevalence of MG infection, associated risk factors and resistance-related mutations among FSWs. METHODS: This cross-sectional study was conducted from April to August 2022 in four cities in Guangdong Province. FSWs were recruited through venue-

Neonatal conjunctivitis due to uncommon pathogen Streptococcus mitis/oralis: a case report.

Neonatal conjunctivitis (NC), historically attributed to Neisseria gonorrhoeae, is now known to be caused by various pathogens, including Staphylococcus, Streptococcus, and Chlamydia trachomatis. Although viridans group streptococci are generally regarded as commensal organisms, they may cause opportunistic conjunctival infections. Here, we present a case of NC caused by Streptococcus mitis/oralis, a member of the Mitis group of viridans group streptococci. A 15-day-old female neonate presented with bilateral severe eyelid edema and moderate hemopurulent discharge that precluded anterior segme

Epithelial polarity: a key player in Chlamydia trachomatis-host interactions.

Chlamydia trachomatis is a major human pathogen responsible for the most prevalent bacterial sexually transmitted infections. Given its high prevalence, potential for serious complications and high economic burden, understanding the pathogenesis of C. trachomatis infections is crucial for developing effective prevention and treatment strategies. Mucosal epithelial cells serve as the primary replicative niche and play a central role in infection. Historically, research has relied heavily on cancer-derived cell line models to study the interactions of C. trachomatis with host cells. Although the

Small intestinal organoids as a model to study interactions of C. suis with its porcine host.

Chlamydia suis is an obligate intracellular bacterium endemic in pig populations and is detected in the gastrointestinal tract, suggesting that the intestine may be an important site of chlamydial colonization. Despite this, intestinal chlamydial infections remain poorly understood, largely due to the lack of models that accurately mimic the interaction of C. suis with the gut epithelium. The aim of this study was to evaluate whether porcine jejunum-derived enteroids constitute a suitable in vitro model to investigate intestinal infection by C. suis and to compare infection dynamics with the c
