DNA-damaging chemotherapy reshapes cardiac-resident macrophage composition and function.

Heart failure and ischemic heart disease represent prevalent causes of death among cancer survivors. Despite extensive use of conventional chemotherapies, a limited understanding of how these agents affect the cardiac immune landscape exists. Using mouse models, we show that DNA-damaging agents selectively deplete cardiac-resident macrophages through activation of p53 signaling and resultant necroptosis and apoptosis. Genetic lineage tracing, transcriptomic profiling, and functional studies revealed that recruited monocytes progressively reconstitute the cardiac-resident macrophage compartment

Cardioprotective Role of Neuregulin1-ErbB2 Signaling Pathway: Its Physiological and Onco-Cardiological Roles in the Heart.

Neuregulin 1 (NRG1), a member of the epidermal growth factor (EGF) family, regulates the development, differentiation, proliferation, and plasticity in multiple tissues through its binding to ErbB3 and ErbB4 receptors. In the cardiovascular system, NRG1 plays a crucial role in cardiac development, physiological function, and cell survival. Since NRG1 exerts cardioprotective effects through its interaction with ErbB2/ErbB4 and ErbB4 homodimers on cardiomyocytes, the administration of recombinant human NRG1 has the potential for the treatment of heart failure. ErbB2 is known as human epidermal g

Systematic review and meta - analysis of risk prediction models for heart failure after PCI in patients with acute myocardial infarction.

BACKGROUND: The incidence of heart failure (HF) following percutaneous coronary intervention (PCI) in patients with acute myocardial infarction (AMI) remains relatively high, severely impairing long-term prognosis and quality of life. In recent years, advances in biomarker identification and imaging technologies have driven growing research into developing HF risk prediction models for AMI patients post-PCI. However, significant heterogeneity exists across current studies in terms of model construction methods, variable selection, and validation strategies, and the predictive performance and c

Swedish snus use in non-smoking middle-aged men and risk of cardiovascular disease: a population-based cohort study with 27-year follow-up.

AIMS: The potential health hazards associated with snus use are less studied compared with the health risks related to smoking. However, during the past decade there is growing evidence on increased risk for mortality, cardiovascular disease (CVD) and cancer following snus use. The aim of the present study was to examine the possible association between snus use at the age of 40 and 50 years, and the development of coronary heart disease (CHD), stroke, hypertension, or heart failure later in life in men with no history of smoking. METHODS: Data from a population-based cohort with data from he

J-shaped association between non-high-density lipoprotein cholesterol to high-density lipoprotein cholesterol ratio (NHHR) and heart failure: a cross-sectional study.

AIMS: Non-high-density lipoprotein cholesterol to high-density lipoprotein cholesterol ratio (NHHR), a burgeoning lipid parameter, demonstrates significant predictive value for atherosclerotic and metabolic diseases. However, the correlation of NHHR with heart failure (HF) is still not well established. METHODS: Between 2011 and 2018, this study analyzed NHHR and HF data from 11,639 individuals in the National Health and Nutrition Examination Survey. The relationship between HF and NHHR was examined using a multivariate logistic regression model. Results were graphically represented through r

Mesenchymal stromal cells alleviate pulmonary arterial hypertension by suppressing pulmonary arterial adventitial fibroblast activation and extracellular matrix remodeling via the SOCS3/STAT3 pathway.

BACKGROUND: Pulmonary arterial hypertension (PAH) is a fatal condition characterized by progressive vascular remodeling in the pulmonary arteries, eventually leading to right heart failure and death. Dysregulated extracellular matrix (ECM) remodeling is central to PAH pathogenesis and represents a potential therapeutic target. Mesenchymal stromal cells (MSCs) have shown promise in preclinical studies; however, the optimal therapeutic window, dosing frequency, and mechanistic basis for their regulation of vascular ECM remain unclear. METHODS: We employed a monocrotaline (MCT)-induced rat model

Gut Microbiota in Pulmonary Arterial Hypertension: Murine Models and Human Microbial Signatures, Pathogenic Mechanisms, and Emerging Therapeutic Avenues.

Pulmonary arterial hypertension (PAH) is a chronic, severe cardiopulmonary disease characterized by the progressive increase in pulmonary vascular resistance (PVR) because of the proliferation and fibrosis of the pulmonary arterioles. Although the disease originates in the pulmonary vasculature, it ultimately leads to right heart failure and death. PAH is associated with high mortality rates and poor prognosis, with no therapies currently available to reverse pulmonary vascular remodeling, imposing substantial socioeconomic burdens. Growing interest in the gut-lung axis has highlighted the rol

Low and High Pressor Doses of Ang II Lead to Two Distinct Phenotypes of Hypertensive Heart Disease in Mice.

Hypertension is a major contributor to cardiovascular diseases, being the most common comorbidity and the biggest risk factor in heart failure with preserved ejection fraction. Angiotensin II (Ang II) is a known hypertension and heart failure inducer in mice, but its role in the causality in phenotype development remains unclear. Here, hypertension was induced with low (LowA) or high (HighA) pressor doses of Ang II in mice. Both LowA and HighA groups demonstrated equal levels of hypertension with aortic dilatation and decreased aortic wall strain, but only HighA developed left ventricular hype

Association of peak expiratory flow rate with clinical outcomes in pulmonary arterial hypertension.

Pulmonary arterial hypertension (PAH) is a rare disease characterized by a progressive increase in pulmonary arterial pressure, leading to right heart failure. In clinical practice, reduced exercise tolerance is commonly observed in patients with PAH. Several studies have identified skeletal muscle abnormalities and muscle weakness as factors contributing to impaired exercise capacity in these patients. Recently, peak expiratory flow rate (PEFR) has been shown to correlate with skeletal muscle mass. Given that PEFR can be easily and noninvasively measured using respiratory function tests, we i

Elucidation of mechanisms underlying the therapeutic effects of cordycepin on pulmonary hypertension, with a focus on cell senescence and gut microbiota.

INTRODUCTION: Pulmonary hypertension (PH) is a progressive cardiopulmonary disorder characterized by excessive pulmonary vascular remodeling and aberrant proliferation of pulmonary artery smooth muscle cells (PASMCs). Emerging evidence suggests that gut microbiota dysbiosis contributes to PH development. Cordycepin, a natural adenosine analogue derived from Cordyceps militaris, has demonstrated antiproliferative and microbiota-modulating properties; however, its mechanism of action in PH remains unclear. OBJECTIVE: Elucidate the mechanisms underlying the therapeutic effects of cordycepin on P

Linking Energetic Inefficiency to Ventriculoarterial Uncoupling in Pulmonary Hypertension.

BACKGROUND: Right ventricular-pulmonary arterial (RV-PA) uncoupling is central to prognosis in precapillary pulmonary hypertension (PH). While the tricuspid annular plane systolic excursion tosystolic pulmonary artery pressure (TAPSE/sPAP) ratio is an established bedside surrogate of RV-PA coupling, novel energetic markers such as three-dimensional (3D)-derived right ventricular (RV) global wasted work (GWW) may provide complementary insight into maladaptive RV remodeling. OBJECTIVES: To contextualize the prognostic value of RV GWW relative to TAPSE/sPAP and clarify their mechanistic and clin

Evolution and Prognostic Value of Right Ventricular to Pulmonary Artery Coupling During Guideline-Directed Medical Therapy Up-Titration.

BACKGROUND: Up-titration of guideline-directed medical therapy (GDMT) is known to enhance left ventricular function in heart failure (HF) with reduced ejection fraction. However, data regarding its effect on right ventricular (RV) function remain sparse. We aimed to assess the impact of GDMT up-titration on the RV, especially RV to pulmonary artery coupling, and its prognostic value in these patients. METHODS: All consecutive patients (n=291) with left ventricular ejection fraction <50% followed for GDMT up-titration in a dedicated HF clinic in a tertiary center from January 2019 to June 2022

Atrial Strain Outperforms Ventricular Strain for Detecting Hypertensive HFpEF Using CMR Feature Tracking.

BACKGROUND: Heart failure with preserved ejection fraction (HFpEF) accounts for ≈50% of heart failure, and hypertension often coexists. Although strain imaging detects subclinical dysfunction, the relative diagnostic value of left atrial (LA) versus left ventricular (LV) strain for identifying hypertensive HFpEF remains uncertain. We compared LA and LV strain using cardiac MR feature tracking to determine optimal markers for HFpEF detection. METHODS: A single-center, retrospective study included 191 participants: 71 with HFpEF and hypertension (HFpEF-HTN), 60 with essential hypertension, and

Rewriting the Cardiovascular Transcriptome: Emerging Roles of PIWIInteracting RNAs as Epigenetic Modulators and Precision Biomarkers.

PIWI-interacting RNAs (piRNAs), a class of small non-coding RNAs originally identified in germ cells, are now increasingly recognized as pivotal regulators of gene expression in somatic tissues, including the cardiovascular system. Cardiovascular diseases (CVDs) remain the leading global cause of morbidity and mortality, yet sensitive and specific molecular biomarkers and effective RNA-based therapeutic targets are still lacking. However, compared to microRNAs and lncRNAs, the roles of piRNAs in CVD are only beginning to be elucidated, highlighting an important knowledge gap. The objective of
