Comparative genomics of Clostridium butyricum reveals a conserved genome architecture and novel virulence-related gene clusters.

Bacteria from the species Clostridium butyricum encompass a diverse range of phenotypes. While some strains are used as probiotics, others have been isolated from cases of botulism and necrotizing enterocolitis (NEC) in preterm neonates. We identify a unique genomic feature of this species, namely a highly conserved extrachromosomal element of ~0.8 Mb. This replicon satisfies the three principal criteria used to define a chromid, which include the possession of core genes that are encoded on the main chromosome in other species. Although C. butyricum is the type species of Clostridium, we find

Review of Foodborne Botulism in the UK: 2006-2024.

Food-borne botulism is a rare but serious disease caused by ingestion of botulinum neurotoxin pre-formed in food by Clostridium botulinum. Between 2006 and 2009, no foodborne botulism cases were reported in the UK. However, the period from 2010 to 2024 saw 13 cases, encompassing seven separate incidents and two outbreaks, with no reported fatalities. Cases were predominantly linked to imported, home-made, and artisanal foods, occasionally to commercial products. Diagnostic and public health challenges include delayed clinical diagnosis, delayed sample collection, inadequate specimen volumes, a

Microfluidic Nanobiosensor Detection of Botulinum Neurotoxin Serotype A.

Botulinum neurotoxin (BoNT) poses a significant threat to public health as the most potent toxin known to humans. BoNT intoxication can lead to botulism, a life-threatening disease that may result from ingesting food contaminated with biologically active BoNT. The gold standard method for BoNT detection, the mouse bioassay, is time-consuming and raises ethical concerns regarding the use of laboratory animals. Therefore, there is a critical need for alternative methods that are rapid and require minimal training for field use. In this study, with potential in-field use in mind, we developed a m

Iatrogenic botulism following botulinum toxin injection: a scoping review of clinical characteristics, risk factors, and dermal considerations.

STUDY PURPOSE: Botulinum toxin (BoNT) is commonly used for both cosmetic and medical purposes. However, improper use can lead to iatrogenic botulism. This scoping review aimed to evaluate the systemic and skin-related effects, identify risk factors, and highlight safety gaps in BoNT-related botulism. METHODS: We followed PRISMA-ScR guidelines to search for literature from 1997 to 2024 across PubMed, Scopus, EMBASE, and Google Scholar. Ten case reports (113 patient cases) met the inclusion criteria. RESULTS: Females accounted for 78.8% of cases. Systemic symptoms typically developed within fo

Cauda equina nerve root enhancement in adult intestinal toxemia botulism.

BACKGROUND: Adult botulism is a rare, life-threatening condition typically caused by exposure to preformed botulinum neurotoxin (BoNT). Acute intestinal toxemia botulism (AITB) is an uncommon subtype resulting from colonization of Clostridium botulinum in the intestines. Diagnosis is made by detecting BoNT in the patient's blood, stool, or gastric fluid. AITB is confirmed when C. botulinum is isolated in culture. Electrodiagnostic studies may support the diagnosis, while imaging-when performed-is generally used to exclude alternative conditions. CASE PRESENTATION: A 74-year-old man presented

Gene expression dynamics in Bacillus cereus and Bacillus subtilis treated with Thymus vulgaris and Origanum vulgare subsp. hirtum essential oils.

Essential oils (EOs) hold significant potential as antimicrobials in food, due to their high concentration of active phenolic compounds. These compounds can target bacterial cells through various mechanisms, such as membrane disruption, quorum sensing inhibition, and interference in virulence factors, affecting microorganisms at a genomic level. Bacillus cereus and Bacillus subtilis are key foodborne bacteria that could be managed using these natural preservatives. The present study investigated the effects of stress induced by applying Thymus vulgaris and Origanum vulgare subsp. hirtum EOs on

Nontoxic, multi-domain botulinum neurotoxin-LCHCN as vaccines against botulism.

Botulinum neurotoxins (BoNTs), the causative agents of botulism, are proteins consisting of an N-terminal catalytic Light Chain (LC) and a C-terminal Heavy Chain (HC), which comprises a LC-translocation domain (HCN) and a receptor binding domain (HCC). There are seven BoNT serotypes (A-G) and multiple subtypes. Currently, there is no approved vaccine against botulism available for human use. Several studies have investigated BoNT domains, multi-domains, or mutated holotoxins as candidate BoNT vaccines. However, residual toxicity has been observed in mutated full-length BoNTs and multi-domain (

Features of the First Case of Foodborne Botulism Caused by Dual-Toxin Clostridium parabotulinum Subtype A1(B5) in Spain.

The neurotoxin BoNT/B2 is the predominant Clostridium parabotulinum subtype in foodborne and infant botulism cases in Spain. This study characterizes a novel case of foodborne botulism in Spain caused by a dual-toxin A1(B5) strain. A 64-year-old male presented with acute, progressive flaccid paralysis including diplopia, dysphagia, and respiratory failure. Although botulism was not initially suspected, the patient recovered with supportive care and without antitoxin administration. Genomic characterization confirmed the presence of both bont/A1 and silent bont/B5 genes. The bont/A1 gene was as

Research Progress on the Detection Methods of Botulinum Neurotoxin.

Botulinum neurotoxins (BoNTs), produced by the anaerobic spore-forming bacterium Clostridium botulinum, are among the most potent known biological toxins. BoNTs cause lethal botulism via contaminated food, wound infections, or infant intestinal colonization, posing significant threats to public health. Although the mouse bioassay is still being considered as the gold standard for detecting BoNTs, its drawbacks, including the lengthy experimental duration, high costs, and ethical issues, highlight the urgent need to develop alternative methods to fulfill the detection requirements. In recent ye

Botulism in pregnancy: A clinical review.

Botulism is a rare but potentially life-threatening neuroparalytic condition caused by Clostridium botulinum toxin, which has significant maternal and fetal health consequences during pregnancy, necessitating a careful review of existing literature. We conducted a comprehensive literature review, searching databases such as PubMed, Scopus, and Embase for studies published through June 2025. This search was supplemented by hand-searching and the translation of foreign articles. We included studies describing clinical cases of botulism in pregnant patients or reporting on botulinum antitoxin saf

A local outbreak of iatrogenic botulism associated with cosmetic injections of botulinum neurotoxin-containing products, England, 2025.

In June 2025, 25 botulism cases were identified among recipients of botulinum neurotoxin-containing cosmetic injections in North East England. A case-control study indicated that cases were more likely to have attended two specific practitioners and received an unlicensed product (p < 0.001). Testing of seized product detected a potency (370 units/vial) that was higher than listed on its labelling (200 units/vial). Strengthened regulation of cosmetic procedures is necessary for mitigating public health risks, which are exacerbated by the availability of unlicensed products.

Phosphorylation of SNAP-25 at Ser187 is enhanced following its cleavage by Botulinum Neurotoxin Serotype A, promoting the dominant-negative effect of the resulting fragment.

Botulinum Neurotoxin Serotype A (BoNT/A), responsible for most human botulism cases, inhibits neurotransmitter release by cleaving the target protein SNAP-25. Previous literature demonstrated that BoNT/A mediated cleavage of a small subset of the SNAP-25 pool, resulting in SNAP-25 (1-197) fragments, is sufficient to block exocytosis. SNAP-25 (1-197) potentially competes against intact SNAP-25 for SNARE complexes and blocks neurotransmission through a dominant-negative mechanism. However, how a tiny fraction of cleaved SNAP-25 efficiently outcompetes a large pool of intact SNAP-25 remains unkno
