Neuromuscular Junction Disorders in Children: Approach to Diagnosis and Management.

Neuromuscular junction disorders in children present with fatigable weakness and encompass genetic, autoimmune, and toxin-mediated subtypes. Timely and correct diagnosis is crucial as many subtypes respond to specific treatments. Congenital myasthenic syndromes may show therapeutic responses to targeted medications, but they can be misdiagnosed as congenital myopathy, neuropathy, or muscular dystrophy. Furthermore, the treatment landscape for autoimmune myasthenia gravis is rapidly expanding with complement inhibitors and neonatal fragment crystallizable receptor antagonists. In this review, w

Development and Humanization of a Camelid-Derived Neutralizing Nanobody Targeting Botulinum Neurotoxin Type A.

PURPOSE: Botulinum neurotoxin (BoNT), produced by Clostridium botulinum, is one of the most potent neurotoxins, causing botulism, a potentially fatal paralytic disease. Among different serotypes, BoNT/A is the most potent and the primary cause of botulism. The receptor-binding domain (Hc) of BoNT is crucial for targeting host cell receptors, making it a key therapeutic target. This study aims to develop neutralizing antibodies against BoNT/A by isolating camelid-derived nanobodies (Nbs) targeting BoNT/A-Hc (AHc). METHODS: A panel of Nbs specifically targeting AHc was isolated from a camelid i

Isolation and Characterization of Botulinum Neurotoxin-Producing Clostridium caccae from Meat and Soil - China, 1990-2025.

WHAT IS KNOWN ABOUT THIS TOPIC?: Botulinum neurotoxin (BoNT)-producing clostridia are categorized into six physiologically and phylogenetically distinct groups (Groups I-VI). Increasing genomic evidence indicates that bont genes are distributed across multiple clostridial lineages, highlighting the taxonomic complexity of BoNT-producing clostridia. Clostridium caccae (C. caccae), first described in 2020 from human stool, was originally described as a strain lacking the bont gene; however, its association with BoNT production has not been previously recognized. WHAT IS ADDED BY THIS REPORT?: S

Proteolytic Clostridium botulinum (group I) - evaluation of predictive models for time-to-toxicity and identification of product characteristics that prevent growth and toxin formation in different types of foods.

Preventing toxin formation by proteolytic Clostridium botulinum is critical for food safety but remains a challenge for example when developing or reformulating various products including sodium-reduced or nitrite-free foods. Validated predictive food microbiology models can support this process by identifying combinations of product characteristics and storage conditions that prevent growth and toxin formation. An existing cardinal parameter growth and growth boundary model (Koukou et al., 2022b), including the effect of 11 environmental factors and their interactions, was extended to predict

A Bioinspired Cellulosic Elastomer with Ultrahigh Robustness Enabled by Structure-Induced Reconstruction for Impact Protection.

BACKGROUND: Xanthogranulomatous cholecystitis (XGC) is an uncommon variant of chronic cholecystitis, characterized by marked thickening of the gallbladder wall and dense local adhesions. It often mimics a gallbladder carcinoma (GBC), and may coexist with GBC, leading to a diagnostic dilemma. Furthermore, the premalignant nature of this entity is not known. This study was undertaken to assess the p53, PCNA and beta-catenin expression in XGC in comparison to GBC and chronic inflammation. METHODS: Sections from paraffin-embedded blocks of surgically resected specimens of GBC (69 cases), XGC (65)

Impact of a national Smarter Medicine guideline on outpatient proton-pump inhibitor prescription in Switzerland: a nationwide interrupted time-series analysis.

BACKGROUND: Staphylococcus aureus is a human colonizer with high potential for virulence, and the spread of the virulent strains from the colonized hosts to non-carriers in the community is on the increase. However, there are few reports on comprehensive analysis of staphylococcal enterotoxin (SE) genes with clonal lineage in S. aureus in Africa. This is essential because of diversity of cultures and habits of the people. This study analyzed spa types and enterotoxin genes in S. aureus strains previously isolated from the human nostrils, poultry and clinical samples in Southern Nigeria. METHO

Anti-interference Photoelectrochemical Immunosensing of Staphylococcal Enterotoxin A Using a Dendritic Peptide Tetramer-Modified HsGDY/Cu2O Photocathode.

Staphylococcal enterotoxin A (SEA) is a major foodborne pathogen toxin capable of causing severe food poisoning even at nanogram levels. However, as widely reported, accurate quantification of SEA in complex food matrices remains challenging due to matrix effects from coexisting proteins, lipids, carbohydrates, and reducing small molecules. Herein, we report an innovative anti-interference photoelectrochemical (PEC) immunosensing platform constructed from a hydrogen-substituted graphdiyne (HsGDY)/Cu2O photocathode further functionalized with a dendritic peptide tetramer (DPT) bearing four term

Inhibitory effect of acetic acid on the toxicity of Staphylococcus aureus and its membrane vesicles.

Staphylococcus aureus is a bacterium present on the human skin and produces enterotoxins (SEs) when it grows in food, resulting in toxin-type food poisoning. The SEs and virulence factors from S. aureus, which are considered encapsulated and concentrated in membrane vesicles (MVs) composed of the bacterium's own cell membrane, reach host target cells and trigger inflammation. This study was conducted to investigate the effects of acetic acid, used as a food additive, on the expression of virulence factors and MV-induced inflammation in S. aureus. Results showed that 40 mM acetic acid inhibited

Development of a dual-nanobody sandwich immunochromatographic strip based on a site-directed conjugation strategy for ultrasensitive detection of staphylococcal enterotoxin B (SEB).

BACKGROUND: Staphylococcal enterotoxin B (SEB), a highly potent and stable biotoxin, is a leading cause of food poisoning and poses a significant threat to food safety and public health. While lateral flow immunoassay (LFIA) is valued for its low cost and rapid on-site detection capability, it faces challenges in detecting SEB due to the nonspecific binding of staphylococcal protein A to the Fc region of conventional antibodies. To overcome this limitation, a series of paired nanobodies against SEB were successfully selected from an immunized phage-display nanobody library. RESULTS: The scree

Identification of targetable epitope surfaces from the high resolution structure of the superantigen Staphylococcal enterotoxin L.

Emetic exotoxins secreted by Staphylococcus species (Staphylococcal enterotoxins, SEs) are a major cause of food poisoning cases worldwide and many are additionally classified as superantigens-able to potently activate T cells in an antigen independent manner. Fewer than half of the gene products of the known SE genes have been extensively characterized. The gene for Staphylococcal enterotoxin L (SEL) occurs in both foodborne and clinical isolates but no detailed structural characterization has yet been available. We report here the crystal structure of SEL and confirm its function as a supera

Interactive Effects of Temperature and Nutrient Conditions on Growth and Virulence Factor Expression of Staphylococcus aureus Under Model Food-Relevant Environments.

Staphylococcus aureus is a major cause of foodborne intoxication through the production of heat-stable enterotoxins (SEs) and is also an important opportunistic pathogen of humans and livestock. Meat and meat products are major vehicles for this pathogen because their protein-rich composition supports bacterial growth and toxin production. However, the combined effects of temperature and nutrient composition on S. aureus growth and virulence expression under food-relevant conditions remain unclear. In this study, we investigated the interactive effects of temperature and nutritional context on

In Vitro Detection of Biologically Active Staphylococcal Enterotoxins Type B and C1 as an Alternative to In Vivo Testing.

Staphylococcus aureus is a major bacterial pathogen that can cause clinical infections and foodborne illnesses through the production of 25 exotoxin types. The most frequently implicated toxins in food poisoning outbreaks are Staphylococcal enterotoxins type A-E (SEA-SEE), which are the first enterotoxins discovered. While in vitro detection methods are available to identify the presence of enterotoxins, they cannot distinguish between biologically active and inactive forms of the toxins. Detection of biologically active enterotoxins currently relies on in vivo testing, using the emetic respon

Anatomical site-dependent distribution and toxigenic potential of Staphylococcus aureus in spent laying hens.

Staphylococcus aureus in poultry is a potential source of food contamination and staphylococcal food poisoning. However, its carriage patterns and toxigenic potential at the slaughterhouse stage remain insufficiently characterized in Japan. This study investigated the prevalence and molecular characteristics of S. aureus in spent laying hens entering the processing line over a one-year period. Three anatomical sites (head, wing, and leg) were sampled per carcass (n = 800). S. aureus was detected in 649 of 800 birds (81.1%), and 995 isolates were obtained. Virulence gene profiling showed that 5
