Staphylococcal Extracellular Complement-Binding Protein Induces IL-4 Production in Murine Basophils.

Staphylococcus aureus is responsible for a wide range of pyogenic infections, food poisoning, and allergic inflammation. S. aureus produces a very wide variety of proteinaceous exotoxins, which are classified by structure as β-barrel pore-forming toxin, oligonucleotide/oligosaccharide-binding (OB)-fold/β-grasp proteins, and triple-helix bundle-containing proteins. We previously reported that three staphylococcal exotoxins belonging to the first two groups activate basophils and mast cells, which are innate immune cells that play a central role in allergic inflammation. In this study, we focuse

Identification of Nonclassical Enterotoxin Genes in Stored Staphylococcus aureus Isolates From Milk of Subclinical Mastitis Cows in Sumedang Regency.

Staphylococcal enterotoxins (SEs) are thermostable superantigenic proteins produced by Staphylococcus aureus (S. aureus) that are responsible for staphylococcal food poisoning (SFP) in humans. SEs are classified into classical and nonclassical types, with increasing attention given to the nonclassical due to their potential role in food safety. Therefore, this study is aimed at identifying the presence and distribution of selected nonclassical SE genes (sek, sel, sem, sen, seo, sep, seq, and ser) in 22 stored biological isolates of S. aureus obtained from milk of dairy cows with subclinical ma

Whole-genome analysis and phenotypic antimicrobial resistance of Staphylococcus aureus isolated from foods in Northern Kazakhstan.

Staphylococcal food poisoning is widespread, causing significant public health problems. This study aimed to characterize the genomic profiles and antimicrobial susceptibility of Staphylococcus aureus isolates obtained from food products in Northern Kazakhstan using whole-genome sequencing. The study included 42 food-derived isolates from raw food products (milk, chicken fillet, beef, horse meat, semi-finished meat products) and ready-to-eat (RTE) products (sour cream, butter, canned tuna, culinary dishes). All isolates were recovered from individual samples collected between June 2024 and Jun

Biocontrol of Staphylococcus aureus in dairy products using lytic bacteriophage UHP46: efficacy in milk, cheese, and yogurt.

Staphylococcus aureus is a major public health concern in the dairy industry. It is a well-recognized pathogen responsible for bovine mastitis and raw milk contamination; certain strains produce heat-stable enterotoxin capable of causing staphylococcal food poisoning in humans. The emergence of multiple drug-resistant strains of S. aureus has rendered ineffective the use of antibiotics for control purposes. This study determined the antibacterial activity of lytic bacteriophage UHP46 against a single, previously characterized multidrug-resistant mastitis isolate, S. aureus S46, in three dairy

Simultaneous optimization of film morphology and structure dimensionality in Cs3Sb2I9 through butylamine gas treatment.

Food safety is a critical public health issue worldwide as the consumption of unhygienic food causes millions of illnesses annually. The contaminated food is primarily unsafe due to the presence of microorganisms (i.e. bacteria and viruses), chemicals (i.e. pesticides and heavy metals), and physical hazards (i.e. glass, metal and plastic). Foodborne illnesses such as salmonellosis, campylobacteriosis, listeriosis, E. coli infection, botulism, cholera, norovirus infection, and others are commonly caused by the problems associated with food hygiene. From the modest beginnings rooted in basic hyg

Establishment of two Hypotheses for Gut Bacterial Alteration Process for Minimizing the Intestinal Communicable Diseases.

Intestinal communicable diseases like diarrhea, dysentery, nausea, vomiting, cholera, typhoid fever, paratyphoid fever, botulism, gastroenteritis, etc., are caused by different microbes. Intestinal microbial ecology is the most mysterious and complex environment for microbes. Bacteriophages are the dominant viruses in the intestinal microbiota. It is well established that gut microbial composition directly affects human health. Microbes use the gastrointestinal tract as niches there some factors or theories exist such as dietary, quorum sensing, and biosignaling, including biomass and biofilm

Harnessing Artificial Intelligence to Restore Standards in Medical and Dental Education in Pakistan.

The month of March is dedicated to Colon Cancer Awareness. Worldwide, colorectal cancer (CRC) incidence has been on the rise. It is currently the third most common cancer in men (746,000 cases, 10.0% of the total) and the second in women (614,000 cases, 9.2% of the total).1 Arecent meta-analysis reported a 61% risk reduction in CRC incidence with colonoscopy.2 Unlike screening programs for breast and prostate cancers, not only has CRC screening reduced mortality from colon cancer and detected early CRC, it has also decreased the incidence of CRC through detection and removal of pre-cancerous l

SAR317461 Inhibit JAK2/STAT3 Phosphorylation for Enhanced Anti-leukemia Activity.

Breast cancer is commonly diagnosed at late stages in countries with limited resources. Efforts aimed at early detection can reduce the stage at diagnosis, potentially improving the odds of survival and cure, and enabling simpler and more cost-effective treatment. Early detection of breast cancer entails both early diagnosis in symptomatic women and screening in asymptomatic women. Key prerequisites for early detection are ensuring that women are supported in seeking care and that they have access to appropriate, affordable diagnostic tests and treatment. We therefore propose the following seq

Role of hypothalamus function in metabolic diseases and its potential mechanisms.

In June 2008, three Dutch tourists participating in a mini-cruise in Turkey needed urgent repatriation for antitoxin treatment because of symptoms of botulism. Because there was a shortage of antitoxin in the Netherlands, an emergency delivery was requested from the manufacturer in Germany. An outbreak investigation was initiated into all nine cruise members, eight of whom developed symptoms. C. botulinum type B was isolated in stool culture from four of them. No other patients were notified locally. Food histories revealed locally purchased unprocessed black olives, consumed on board of the s

Organic electrolyte gated field effect transistor for detecting alpha synuclein longitudinally in Parkinsonism mouse model.

BACKGROUND: Enthesopathy is an evolving area for applied clinical research. MRI is the gold standard in the diagnosis of elbow joint pathology, but recent reports indicate that ultrasound imaging is more sensitive and accurate than MRI in detecting enthesopathy of the heels and knees. Too many patients are under-diagnosed and/or misdiagnosed because the early pathological changes of enthesitis in the different types of seronegative arthropathies are not detected. OBJECTIVES: This study was undertaken to describe the ultrasound features of elbow enthesitis in patients with seronegative arthrop

Cytotoxicity and Genome Characteristics of an Emetic Toxin-Producing Bacillus cereus Group sp. Isolated from Raw Milk.

The Bacillus cereus group frequently contaminates milk and dairy products. Some members of this group can produce the heat-stable pre-formed toxin cereulide, which causes emetic foodborne intoxication. This study characterised emetic B. cereus group isolates from raw cow's milk in the biochemical, genetic, and toxigenic aspects. Of the 158 B. cereus group isolates derived from 99 raw milk samples, 7 (4.43%) harboured cereulide synthetase A (cesA), which encodes a cereulide synthetase associated with the emetic phenotype. Heat-treated culture filtrates from the cesA-positive isolates demonstrat

Development and selection of candidate monoclonal antibodies for the detection of Clostridium botulinum neurotoxin serotype B light chain.

Botulinum neurotoxin, produced by the bacterium Clostridium botulinum, causes botulism, a severe, rapidly progressing, and potentially fatal condition. Swift detection of the toxin and timely administration of antitoxin antibodies are critical for effective treatment. The current standard for Botulinum toxin testing is the mouse lethality assay, but this method is time-consuming and requires live animals. Consequently, a key focus of research is the development of antibodies for both diagnostic purposes and toxin neutralization. Botulinum neurotoxin serotype B (BoNT/B), one of the most dangero

Development of a Recombinant Fusion Vaccine Candidate Against Lethal Clostridium botulinum Neurotoxin Types A and B.

BACKGROUND: Botulinum neurotoxins (BoNTs), produced by Clostridium botulinum, are potent protein toxins that can cause botulism, which leads to death or neuroparalysis in humans by targeting the nervous system. BoNTs comprise three functional domains: a light-chain enzymatic domain (LC), a heavy-chain translocation domain (HCN), and a heavy-chain receptor-binding domain (HCC). The HCC domain is critical for binding to neuronal cell membrane receptors and facilitating BoNT internalization via endocytosis. Accordingly, it may serve as a vaccine candidate, inducing anti-BoNT-neutralizing antibodi

An overview of biosensor advancements for detecting botulinum neurotoxins: Addressing food safety and biowarfare risks.

Botulinum neurotoxins (BoNTs) are lethal toxins produced by bacteria Clostridium botulinum. Ingestion of BoNTs contaminated foods causes botulism which affects individual's nervous system by blocking the release of neurotransmitters causing flaccid paralysis. This review article deciphers the comprehensive account on mechanism of action of BoNTs, pathogenicity, and various innovative analytical detection techniques of BoNTs in foods. Potential misuse of BoNT as a biowarfare agent is also a major concern. Hence, for the detection of deadly BoNTs various conventional techniques like mouse lethal

Insects as Natural Hosts, Vectors and Reservoirs of Botulinum Neurotoxin-Producing Clostridia and Their Non-Toxinogenic Counterparts: Preliminary Evidence.

Insects play a significant role in the transmission and spread of bacterial pathogens that cause various diseases in humans and animals. The relationship among insects, bacterial pathogens and diseases is complex and depends on the specificity of the pathogens. Some clostridial species produce botulinum neurotoxin (BoNT), which is responsible for paralytic botulism. However, the ecology of these bacterial species and their non-toxinogenic phylogenetic counterparts remains unclear. This study specifically explored in silico evidence of the interconnection between BoNT-producing Clostridia and t

The History of Neuromodulators in Dermatologic Surgery.

BACKGROUND: In 1817 and 1820, the German physician Justinus Kerner published a series of cases of lethal food poisoning that would unknowingly have a lasting impact on medical science. His compilation of over 75 cases linked the consumption of smoked sausages in the small town of Herrenberg in Württemberg, recounted a constellation of symptoms that today the authors call botulism. Now, over 2 centuries later, the discovery, study and refinement of the toxin causing clinical botulism has led to the acceptance of neuromodulators as a treatment for a wide variety of medical concerns.
