Botulism is a severe and often fatal disease in equine patients worldwide. Clostridium botulinum is a ubiquitous soil organism which produces a potent neurotoxin resulting in neuromuscular blockade and flaccid paralysis in affected animals. Definitive diagnosis is often impractical or impossible, leading to diagnosis and treatment based on clinical experience and presenting signs. Delay in case identification and treatment results in rapid deterioration of the patient. Treatment of recumbent equine botulism cases presents challenges due to patient size and requires intensive nursing care. This

UNLABELLED: Soil is a known reservoir of human pathogens, including botulinum neurotoxin-producing clostridia. However, the occurrence and distribution of botulinum neurotoxin types in French soils are unknown, prompting this study. Botulinum neurotoxin-producing clostridia were sought, and botulinum neurotoxin types were identified through an analysis of 486 soil samples representative of nationwide soil type and land use, provided by the French Soil Quality Monitoring Network. Botulinum neurotoxin-producing clostridia were detected in 45.3% of soil samples, with type B-producing clostridia b

BACKGROUND: Botulism is a neurological disease caused by the ingestion of the toxin produced by Clostridium botulinum. This bacterium is globally distributed, infects mainly mammals and birds, and is present in both the intestinal microbiota and the environment. The most frequently observed clinical manifestation is flaccid paralysis of the skeletal muscles, which can rapidly progress to death due to cardiorespiratory failure. Diagnosis is performed through clinical, environmental, and laboratory findings, mainly by mouse bioassay seroneutralization, which serves as the gold standard.

Recombinant monoclonal antibody (mAb) botulinum neurotoxin (BoNT) antitoxins, consisting of three mAbs that bind non-overlapping epitopes, are highly potent. However, the three-mAb mixtures pose unique development and manufacturing challenges. Combining even more mAbs to create multivalent antitoxin drugs multiplies those challenges. We previously reported that a single tri-epitopic IgG1-based mAb (TeAb) containing the variable domains of the three parental BoNT/A mAbs and an Fc was as potent as the combination of three IgGs in the mouse neutralization assay (MNA). Here, we extended the tri-ep

Clostridium botulinum produces one of the most potent biological toxins and causes botulism, a rare but potentially fatal neuroparalytic disease. In 2014, a foodborne botulism case was reported in Korea, and a strain (CB-2014001) was isolated. Initial characterization identified it as a BoNT/B-producing strain based on mouse bioassay and conventional PCR. However, subsequent genomic analysis revealed the presence of dual BoNT gene clusters, bont/B and bont/F, corresponding to subtypes B5 and F2, respectively. Therefore, we aimed to analyze the genetic characteristics and toxin expression profi

A 56-year-old male patient was presented to the neurology department of the emergency unit by the ambulance service with progressive gait instability, dysarthria and gastrointestinal symptoms. Clinical findings also included bilateral mydriasis, asymmetric bilateral ptosis, proximal paresis of the upper extremity and marked dry mouth. Imaging was unremarkable, but the differential diagnosis revealed evidence of a neurotoxic cause. The history of recent consumption of preserved food and the characteristic symptoms quickly led to a suspected diagnosis of foodborne botulism. After early administr

Foodborne botulism is a rare and potentially fatal illness caused by ingestion of food containing botulinum neurotoxin produced by the bacterium Clostridium botulinum or other neurotoxigenic Clostridium species. C. botulinum can grow in improperly prepared or stored food items such as home-canned or home-preserved vegetables. On June 25, 2024, the Fresno County Department of Public Health and California Department of Public Health, in collaboration with CDC and two local hospitals, initiated an investigation of a foodborne botulism outbreak linked to two related family gatherings in Fresno Cou

OBJECTIVE: We investigated if specific carbohydrate blood group antigens were associated with infant botulism (IB) among discordant twins and triplets.
STUDY DESIGN: Infants in the US and Canada from 2015 to 2022 who had laboratory-confirmed IB; were members of a multiple gestation birth; and had unaffected sibling(s), were eligible to participate. Blood specimens were tested for ABO, Lewis, H, and P1 carbohydrate blood antigens. FUT2 and FUT3 genes were sequenced to determine secretor and Lewis antigen status, respectively. In addition, characteristics of IB cases in the study were compared

Infant botulism, the most common form of botulism in the United States, occurs when botulinum toxin-producing clostridia colonize the infant's gut and produce botulinum neurotoxin (BoNT). This condition is treated with botulinum antitoxin and supportive care; antibiotics are avoided due to the risk of worsening symptoms by promoting toxin release. Consequently, antimicrobial resistance genes (ARGs) are rarely investigated in Clostridium botulinum strains. During 2020 through 2023, 226 C. botulinum isolates associated with infant botulism were sequenced in our laboratory using whole-genome sequ

Botulinum neurotoxins (BoNTs) are the most potent toxins known, causing life-threatening flaccid paralysis by blocking acetylcholine release at neuromuscular junctions. Clinical botulism results in respiratory failure, requiring prolonged artificial ventilation for survival. The only specific therapy is antitoxin, which neutralizes circulating toxin but cannot affect toxin within neurons, resulting in a narrow therapeutic window. Due to its high potency and lack of treatment options, large-scale BoNT exposures present profound risks to human life. Thus, there is an urgent need for rapidly acti

Botulism is a severe muscle paralysis disease mediated by the botulinum toxin. Here, we reported a foodborne botulism case caused by Clostridium botulinum subtype A5(b3) from self-packaged vacuum spicy rabbit heads. Treatment for this case was delayed due to misdiagnosis and insufficient diagnostic capacity in three hospitals, which resulted in progressive clinical deterioration, and eventually, the patient was transferred to Shandong Public Health Clinical Center for specialized therapy. The case was suspected as foodborne botulism by the Qilu Medical-Prevention Innovation Integration pathway

Fugate, a waste product from biogas production, regularly used in agriculture as a fertiliser, may contain bacterial pathogens that cause zoonoses. Anaerobic digestion (AD) can inactivate viable pathogens, including parasites, viruses, and pathogens containing antibiotic resistance genes. This study aimed to compare the numbers of pathogenic bacteria and diversity of potential bacterial pathogens in the fugate using three different types of slurry: cattle, swine, and poultry manure. The swine fugate showed higher numbers of Clostridium perfringens and Campylobacter sp. than the poultry and cat

A 6-month-old infant was hospitalised with suspected infant botulism after being given peanut butter to reduce their risk of developing peanut allergy. Clostridium botulinum type A was detected in their faeces and the peanut butter by PCR and culture. Whole genome sequencing confirmed identical strains, identifying the peanut butter as the source of infection. The infant was treated with human-derived antitoxin Botulism Immune Globulin Intravenous (Human) (BIG-IV) and recovered well. This case highlights potential botulism risk with early peanut introduction.

Botulinum neurotoxins (BoNTs), produced by Clostridium botulinum, exert their potent neuroparalytic effects by specifically targeting presynaptic cholinergic nerve terminals. BoNTs consist of a heavy chain that mediates high-affinity neuronal binding and endocytosis, and a light chain that, once translocated into the cytosol, acts as a zinc-dependent metalloprotease. The light chain cleaves SNARE proteins essential for synaptic vesicle fusion, thereby inhibiting acetylcholine release and leading to flaccid paralysis. This intoxication spans foodborne, wound, and infant botulism, all characteri

Foodborne botulism is a rare but highly lethal disease. The botulinum neurotoxin (BoNT) that causes botulism is produced by Clostridium botulinum (C. botulinum) and other clostridia. In this study, we characterized C. botulinum strains isolated during food poisoning events in Sichuan Province from 1990 to 2024 by analyzing whole-genome sequencing data. Statistical analyses of the geographical distribution food sources of strains suggested that the primary sources of contamination were associated with specific regions and food types. The isolates were further compared with reference strains usi

INTRODUCTION: Botulism, a severe neuroparalytic disease caused by the botulinum toxin produced by Clostridium botulinum, poses significant threats to wild birds. This study describes a natural outbreak of type C botulism in waterfowl in the surroundings of a lagoon in Saavedra, Buenos Aires province, Argentina, during January 2021. The outbreak, affecting approximately 300 birds, was attributed to environmental conditions that allowed the proliferation of C. botulinum. Clinical signs included progressive weakness, paresis, flaccid paralysis, difficulties in locomotion and swimming, "limbing ne
