A live-cell medium-throughput assay for quantifying lipid accumulation in hepatocytes.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a global health burden contributing to the high prevalence of associated metabolic diseases such as cardiovascular disease and thus global mortality rates. MASLD is diagnosed by ≥ 5 % lipid accumulation in the liver, and candidate therapies are often evaluated for lipid lowering effects. Current cell-based lipid imaging approaches rely on fixed-cell staining with dyes such as BODIPY 493/503, limiting the ability to dynamically monitor lipid accumulation in living hepatocytes. To gain a better understanding of the effect of dif

Preparation design and connector geometry influence the mechanical behavior of additively manufactured periodontal splints.

UNLABELLED: Structural design guidelines for additively manufactured periodontal splints remain poorly defined, particularly regarding relative influence of preparation and connector geometry. OBJECTIVE: This in vitro study evaluated the effect of tooth preparation design and connector cross-sectional geometry on the mechanical behavior of these splints, providing preliminary mechanical insights rather than clinical design recommendations. MATERIAL AND METHODS: Standardized specimens were fabricated from a mandibular digital model, combining three preparation designs (no preparation, enamel

Bottlenecks in cardiovascular prevention: Can artificial intelligence close the gap?

Cardiovascular diseases are a leading cause of morbidity and mortality in France, despite current preventive strategies and evidence-based therapies. Artificial intelligence (AI) has emerged as a promising approach to overcoming several of these barriers, with the potential to improve cardiovascular prevention across the entire spectrum of care. Upstream, AI can enhance risk stratification through the integration of clinical, biological, imaging, environmental, behavioural and longitudinal data, thereby moving cardiovascular prevention toward an AI-augmented precision medicine paradigm, in whi

Advances in Cardiovascular Pharmacotherapy. XI: Sotatercept in Pulmonary Arterial Hypertension.

Sotatercept is a groundbreaking new treatment for pulmonary arterial hypertension (PAH). The disease-modifying drug is a recombinant fusion protein that consists of the extracellular domain of the activin receptor 2A linked to the Fc fragment of human immunoglobulin G1. Sotatercept potently traps activins and growth differentiation factors to inhibit transforming growth factor-beta (TGF-β) signaling through its activin receptor 2A/2B pathway, thereby restoring the neutral balance between pro-proliferative and antiproliferative signaling in PAH and causing a striking reversal of the disease's p

OpenDVP: an experimental and computational framework for community-empowered deep visual proteomics.

Deep visual proteomics (DVP) is an emerging approach for cell-type-specific and spatially resolved proteomics. However, its broad adoption has been constrained by the lack of an open-source end-to-end workflow within a community-driven ecosystem. Here we introduce openDVP, an experimental and computational framework for accessible and scalable DVP. OpenDVP combines open-source image analysis tools (MCMICRO, QuPath and napari) with scverse formats (AnnData and SpatialData) for multiomics integration. It provides two workflows: a fast-track pipeline requiring no image analysis expertise, and an

Clinical heterogeneity in chronic inflammatory demyelinating polyneuropathy: data-driven subtypes challenge current classification.

BACKGROUND: Chronic inflammatory demyelinating polyneuropathy (CIDP) presents with substantial phenotypic heterogeneity, yet current classifications do not adequately capture clinically meaningful subgroups. Thus, we aimed to discover novel, clinically meaningful CIDP subgroups using unbiased clustering. METHODS: In this observational, multicenter retrospective cohort study we screened 425 patients diagnosed with typical CIDP or CIDP variants from 10 German centers. Standardized data including demographic, clinical, electrophysiological, laboratory, comorbidity, and treatment information were

Discovery of novel alleles in Kartagener's syndrome.

Kartagener's syndrome (KS) is a rare, typically autosomal recessive ciliopathy and a subtype of primary ciliary dyskinesia (PCD), characterized by situs inversus, bronchiectasis, and chronic sinusitis. KS accounts for approximately 50% of PCD cases and results from impaired motile cilia function. Defective embryonic nodal ciliary beating leads to stochastic left-right axis determination and situs abnormalities. Although numerous genes have been implicated in PCD, up to 30% of cases remain genetically unresolved, and studies specifically focused on KS cohorts remain limited. Here, we performed

Association between the triglyceride-glucose index and carotid-femoral pulse wave velocity in rural hypertensive adults.

BACKGROUND: Triglyceride-glucose (TyG) index is a surrogate marker of insulin resistance and has been linked with cardiovascular outcomes. However, its association with arterial stiffness is understudied, particularly among hypertensive adults. OBJECTIVES: To explore the association between TyG index and arterial stiffness, assessed by carotid-femoral pulse wave velocity (cfPWV), among a group of rural dwelling hypertensive adults. METHODS: A total of 12,751 hypertensive participants from the Hypertension and Stroke Prevention and Control Project (HSPCP) were analyzed. cfPWV was measured non

Percutaneous closure of an iatrogenic left atrial-aortic perforation complicated by cardiac tamponade and coronary vasospasm during transseptal puncture: a case report.

BACKGROUND: Iatrogenic atrial-aortic perforation during transseptal puncture is a rare but potentially life-threatening complication. Percutaneous closure using dedicated occluder devices has emerged as a less invasive alternative to surgical repair in selected patients. CASE PRESENTATION: A 68-year-old woman with hypertension and long-standing persistent atrial fibrillation (≥ 12 years) underwent catheter ablation. Pre-procedural imaging showed a markedly elevated left atrial (LA) volume (171 mL) with predominantly lateral enlargement. During a second transseptal puncture attempt, the sheath

Drug-coated balloons in coronary disease: meta-analysis comparing sirolimus- and paclitaxel-coated technologies.

BACKGROUND AND AIM: Drug-coated balloons (DCBs) are a key alternative to drug-eluting stents for coronary artery interventions, offering the advantage of drug delivery without permanent implants. Both paclitaxel-coated balloons (PCBs) and sirolimus-coated balloons (SCBs) are valuable treatment options. This systematic review and meta-analysis compare the efficacy and safety of sirolimus-coated versus paclitaxel-coated balloon catheters in patients with coronary artery disease. METHODS: We conducted a systematic review and meta-analysis following PRISMA guidelines, searching five databases. We

Neural mechanisms underlying loss of functional independence after stroke: EEG evidence of impaired motor-to-sensory directed connectivity on the affected side.

BACKGROUND: Post-stroke reorganization of the prefrontal-sensorimotor network is critical for functional recovery, yet single connectivity metrics fail to capture its multidimensional characteristics. This exploratory study investigates network reorganization patterns and their clinical relevance to motor function and daily independence using multimodal electroencephalography (EEG) connectivity analysis, including Coherence (COH), phase lag index (PLI), and Granger causality (GC). METHODS: Resting-state EEG from stroke patients (n = 22) and healthy controls (n = 22) was analyzed. COH, PLI, an

Exercise tolerance test in Kawasaki patients without coronary lesions.

INTRODUCTION: Kawasaki disease (KD) is a systemic vasculitis of unknown origin and the leading cause of acquired heart disease in children in developed countries. If untreated, it can cause coronary artery damage in up to 25% of cases. Lifelong follow-up is essential, even for patients without obvious coronary lesions (risk level-I KD). Since some cardiac abnormalities may not be apparent at rest, stress testing offers valuable insight. This study aimed to evaluate exercise tolerance test (ETT) outcomes in children with a history of risk level-I KD compared to a control group. MATERIALS AND M

Screening of fabry disease in patients with left ventricular hypertrophy: a single-center study in Mexico.

BACKGROUND: Fabry disease (FD) is an X-linked sphingolipidosis caused by enzyme α-Galactosidase A (α-Gal A) deficiency due to pathogenic variants in GLA gene. Progressive glycosphingolipid accumulation leads to subsequent myocyte injury, which frequently induces concentric remodeling and manifests as left ventricular hypertrophy (LVH). This study aims to describe the prevalence of FD in patients with LVH diagnosis in a Mexican population. METHODS: Consecutive and unrelated patients with echocardiographic evidence of left ventricular hypertrophy (LVH) without known cause were screened for FD i

SGLT2 inhibitor therapy following aortic valve intervention for aortic stenosis: a systematic review and meta-analysis of mortality, heart failure, rhythm, renal, and valve-related outcomes.

BACKGROUND: Residual mortality and heart-failure events remain common after aortic valve intervention for severe aortic stenosis. Sodium-glucose cotransporter-2 inhibitors (SGLT2i) improve cardiovascular and renal outcomes in heart-failure, diabetes, and chronic kidney disease populations, but their role following transcatheter or surgical aortic valve intervention remains uncertain. METHODS: PubMed, Scopus, ScienceDirect, Cochrane Library, citation tracking, and manually identified eligible studies were screened through June 2026 for randomized and observational studies comparing SGLT2i with

Cytotoxic activity of selinexor in BRAFV600E-driven pediatric low-grade glioma 3D cultures.

Pediatric low-grade gliomas (pLGGs) are the most common primary brain tumors in children and frequently harbor activating RAS/MAPK pathway alterations, including BRAFV600E. Although MEK inhibitors (MEKi) show clinical activity, responses are often incomplete and tumors commonly regrow after treatment discontinuation, highlighting the need for therapies that induce sustained regression. To systematically identify cytotoxic treatments, we initially screened 87 clinically relevant drugs in BRAFV600E-mutant BT40 3D multicellular aggregates and subsequently evaluated the prioritized XPO1-targeting
