Enhanced TGF-β signaling caused by mutations in Fibrillin-1 (FBN1) in patients with Marfan syndrome (MFS) leads to myxomatous degeneration of the mitral valve (MDMV). MDMV can result in mitral valve prolapse, severe regurgitation, and sudden cardiac death. However, it remains unknown whether lymphatic vessel (LV) dysfunction contributes to MDMV development in MFS. Here, we show that lymphangiogenesis in murine mitral valves (MVs) begins postnatally. However, this process is inhibited in a mouse MFS model, Fbn1 mutant (Fbn1C1039G/+) mice, accompanied by disrupted lymphatic cell-cell junctions,

Mitral valve prolapse (MVP) is associated with an increased risk of infective endocarditis (IE), particularly in patients with mitral regurgitation and leaflet thickening. Historically, a subset of patients with mitral valve prolapse were considered for antibiotic prophylaxis before dental procedures. Conversely, current recommendations limit prophylaxis to the highest-risk cardiac conditions, such as congenital heart defects, mechanical valves, heart transplant with valvular issues, and a history of IE. We present a case of a 59-year-old man with a history of MVP with mildly thickened posteri

BACKGROUND: The main goal of this study was to identify various brightness, motion and Doppler echocardiographic variables in small-breed dogs with myxomatous mitral valve disease.
ANIMALS: Sixty client-owned small-breed dogs with mitral regurgitation murmurs.
METHODS: Echocardiography was performed in brightness, motion and Doppler modality from the right parasternal view. On the basis of thoracic radiographs, the dogs were categorized into two groups: those with cardiomegaly and those without cardiomegaly. Additionally, on the basis of the jet area signal to left atrium ratio in colour Dop

BACKGROUND: Left ventricular (LV) remodelling in mitral valve prolapse (MVP) is usually induced by chronic mitral regurgitation (MR), however it can also be disproportionate to the volume load, generating the hypothesis of MVP cardiomyopathy. Changes in the sarcomeric protein titin could contribute to its underlying pathophysiology.
OBJECTIVES: To investigate the role of titin modulation as a mechanism of disproportionate LV remodelling in MVP patients.
METHODS: Myocardial biopsies from 16 patients with MVP and severe MR were compared with 6 controls. All patients underwent pre-operative tra

In this case report, we describe an atypical clinical course in a dog with preexisting myxomatous mitral valve disease (MMVD) subsequently complicated by the development of hyperadrenocorticism. Typically, hyperadrenocorticism can exacerbate cardiac conditions; however, this dog demonstrated paradoxical improvement. A retrospective analysis of the clinical, radiographic, echocardiographic, and hormonal data was conducted to investigate the marked reduction in cardiac size in a Maltese dog with MMVD following the diagnosis of hyperadrenocorticism. A 10-year-old spayed female Maltese dog present

OBJECTIVE: Mitral annular disjunction (MAD), frequently associated with Barlow's disease, links degenerative mitral regurgitation to postoperative ventricular arrhythmias and sudden cardiac death, even after technically successful valve repair. However, despite advances in mitral repair, the arrhythmic substrate associated with MAD remains insufficiently addressed in current surgical paradigms.
METHODS: A systematic search of PubMed and Google Scholar (2000-2025) was conducted using keywords including "mitral annular disjunction," "mitral regurgitation," "arrhythmic mitral valve prolapse," an

AIMS: Myxomatous mitral valve disease (MMVD) is a progressive cardiac disease characterized by extracellular matrix remodelling, leaflet thickening, and proteoglycan accumulation. Despite its significant contribution to cardiovascular morbidity and mortality, the underlying pathophysiological mechanisms involved in its development and progression remain unclear. Recent investigations have revealed a genetic component to the disease, such as mutations in the FLNA gene, and suggested potential involvement of immune cells in the pathogenesis of MMVD. In this study, we investigated the role of mac

BACKGROUND: Mitral valve prolapse (MVP) is a common valvulopathy associated, in a minority of cases, with heart failure, severe mitral regurgitation (MR), and sudden arrhythmic death. Digital tools hold promise for faster and more efficient recruitment of study participants into a large-scale MVP Registry.
OBJECTIVE: This study sought to evaluate the feasibility of surveying participants in an existing e-cohort to identify and clinically characterize MVP cases based on self-reporting and to recruit them in an MVP Registry at the University of California, San Francisco.
METHODS: We surveyed N

Mitral annular disjunction (MAD) is increasingly recognized as a structural abnormality associated with ventricular arrhythmias and sudden cardiac arrest, particularly in the setting of bileaflet mitral valve prolapse (MVP). A 43-year-old man with a history of migraines presented following a witnessed out-of-hospital cardiac arrest and was successfully resuscitated with a single AED shock. Transthoracic echocardiography showed mitral regurgitation with bileaflet prolapse and suspected MAD. An implantable cardioverter-defibrillator was placed for secondary prevention, and the patient was discha

BACKGROUND: Myxomatous mitral valve disease (MMVD) is a common cause of congestive heart failure (CHF) in small-breed dogs. Clinical observations suggest that discontinuation of loop diuretics may be feasible in a subset of dogs with MMVD and CHF.
AIM: To characterize dogs with MMVD-related CHF that achieved stability following loop diuretic withdrawal and to investigate potential contributing factors.
METHODS: We retrospectively analyzed 3,055 canine medical records of dogs diagnosed with MMVD (2012-2022) from the National Chung Hsing University Veterinary Medical Teaching Hospital database

Mitral valve prolapse (MVP) is a prevalent and traditionally benign heart valve disease; however, mounting evidence identifies a subset of patients at risk for malignant ventricular arrhythmias and sudden cardiac death. This narrative review critically examines the role of multimodality imaging in arrhythmic MVP, with an integrative overview of transthoracic echocardiography, cardiac magnetic resonance (CMR), computed tomography, and positron emission tomography. Particular attention is given to the complementary contribution of these modalities to structural, functional, and myocardial tissue

BACKGROUND: Cardiomyopathy and channelopathy (CC) gene variants have been linked to sudden cardiac arrest (SCA) or death (SCD) in small, selected pedigree or post-mortem studies of arrhythmic mitral valve prolapse (MVP). However, the utility of clinical whole exome sequencing (WES) panels as a risk stratification tool in unselected MVP samples is unknown.
OBJECTIVES: The goal of the study was to test the utility of clinical WES panels with CC variant screening for arrhythmic risk stratification in MVP.
METHODS: We performed research-based WES in 203 consecutive MVPs without other arrhythmic

AIMS: Pectus excavatum (PE) can mechanically displace and compress the heart, often altering right ventricular (RV) geometry and function. Comprehensive preoperative echocardiographic characterization in large adult cohorts is limited. This study describes structural and functional cardiac findings in a large cohort of adults with PE evaluated by transthoracic echocardiography (TTE) prior to surgical repair.
METHODS AND RESULTS: We retrospectively reviewed adult patients undergoing primary minimally invasive repair of PE (MIRPE) at a single tertiary center between January 2016 and July 2024 w

PURPOSE: Mitral valve prolapse (MVP) can lead to heart failure, arrhythmia, and death. Current treatments for MVP are strictly surgical, while alternative therapies remain elusive due to lack of knowledge of underlying processes. Importantly, our understanding of cellular mechanisms of post-MVP repair remodeling that can lead to secondary surgery remain limited. We therefore explored how MVP affects human mitral valve interstitial cell (MVIC) extracellular matrix (ECM) remodeling and basal contractility.
METHODS: Isolated MVP and physiologically normal MVICs were embedded in poly(ethylene) gl

INTRODUCTION: Primary cardiac tumors are exceedingly rare in neonates, with rhabdomyomas predominating in this age group. While myxomas are the most common primary cardiac tumor in adults, they remain a clinical rarity in newborns and typically arise in the right atrium. We report an exceptional case of a left atrial myxoma originating from the left atrial appendage, associated with an unexplained cardiorespiratory arrest and imaging features consistent with a "ball-valve" mechanism, a presentation that, to our knowledge, has not been previously reported in the neonatal population.

Valvular heart disease (VHD) and psychiatric disorders are each highly prevalent, yet their co-occurrence is underappreciated in clinical practice. Patients carrying both diagnoses present particular management challenges because the pathologic relationship between the two conditions is bidirectional and mechanistically diverse. This narrative review examines the mechanistic, clinical, and therapeutic interplay between VHD and psychiatric illness, with the aim of providing cardiologists and psychiatrists with an integrated framework for recognition and management. We conducted a systematic sea

BACKGROUND: Long-term durability of degenerative mitral regurgitation (DMR) reduction after transcatheter edge-to-edge repair (TEER) remains limited, and reliable anatomical predictors are still lacking.
METHODS: This study prospectively enrolled patients with symptomatic DMR undergoing TEER across 27 centers in China. All pre-procedural clinical and echocardiographic parameters were collected and analyzed. The primary outcome was MR ≤ 1+ at 3-year follow-up. Logistic regression and receiver operating characteristic (ROC) analyses were performed to identify predictors and optimal cutoff value
