Iran's strategic geographic position, its historical role in major trade routes such as the Silk Road, and its proximity to plague-endemic regions have repeatedly exposed it to plague outbreaks. The most recent documented epidemic occurred in 1966 in Sarab, East Azerbaijan Province. Historically, these outbreaks have significantly influenced demographic patterns, socioeconomic structures, and political developments. In response, extensive research has been conducted to investigate the diversity of Yersinia pestis-vectoring fleas (Siphonaptera) in Iran, resulting in the identification of 119 sp

Anthropogenic changes associated with urbanization can bring wildlife species into closer contact with livestock and humans, thereby increasing the risk of pathogen spillover. Recently, urban-adapted (synanthropic) hedgehog species, including Hemiechinus auritus (long-eared) and Paraechinus hypomelas (Brandt's), have been suggested as potential sources of zoonotic pathogens. The increasing number of tick-infested hedgehogs in urban areas and confirmed cases of human rickettsial infections in south-eastern Iran motivated this investigation to identify rickettsial species in ticks collected from

BACKGROUND: Bartonella henselae and Bartonella quintana are among the most clinically relevant Bartonella species infecting humans. While B. henselae is a zoonotic pathogen maintained in animal reservoirs, B. quintana is considered a human-restricted bacterium transmitted via the human body louse. Both species can cause a wide range of manifestations, particularly in immunocompromised individuals such as those with HIV/AIDS. This study aimed to estimate the prevalence of IgG and IgM seropositivity to these two species and to identify determinants of seropositivity.
METHODS: This cross-section

OBJECTIVES: Psoriatic arthritis (PsA) has been defined as a systemic, chronic, inflammatory arthritis, usually seronegative for rheumatoid factor (RF), associated with cutaneous psoriasis. The exact prevalence of PsA is unknown and its estimation has been difficult, partly due to the lack of a widely accepted classification criteria. Agreed and validated criteria will facilitate comparison between centres and different countries in the areas of epidemiology, outcome studies and therapeutic trials. A number of classification criteria have been published by Moll & Wright (M & W), Bennett's, Vase

Chronic Q fever is caused by persistent infection with the Gram-negative bacterium Coxiella burnetii. The mechanisms underlying this persistence remain elusive, but the presence of the bacteria in the bone marrow of C. burnetii-infected patients has been demonstrated. Therefore, we investigated the potential role of osteoclasts, the bone-resorbing cells, in harboring C. burnetii during infection. The histological analysis of bones from a murine model of Q fever revealed the presence of C. burnetii inside osteoclasts. In vitro infection assays confirmed that osteoclasts can be infected with C.

Hemophagocytic lymphohistiocytosis (HLH) is an inflammatory condition that may run a rapid fatal course and calls for prompt diagnosis. Early intervention with steroids and other immunosuppressive drugs can contain the disease process and favours positive outcome. Ferritin ≥500 ng/ml is a HLH diagnostic criterion. We evaluated the diagnostic potential of admission ferritin, in children with HLH. Pediatric patients of a referral teaching hospital from Feb 2010-Oct 2013 having been investigated for ferritin on admission were included. HLH was confirmed when patients had clinical features and met

BACKGROUND: Acute Q fever caused by Coxiella burnetii is often missed because clinical manifestations are nonspecific, routine cultures are unrevealing, and typical zoonotic exposure may be absent. Persistent fever after correction of diabetic ketoacidosis (DKA) should prompt reassessment for atypical or intracellular pathogens.
CASE PRESENTATION: A 56-year-old man with type 2 diabetes mellitus was admitted with polyuria, polydipsia, and DKA but was afebrile and had no obvious infectious symptoms on admission. Metabolic acidosis resolved within 24 hours, but persistent high-grade fever subseq

Coxiella burnetii is an environmentally stable intracellular bacterium responsible for severe global outbreaks of the zoonotic disease, Q fever. Q fever is transmitted by the inhalation of aerosols contaminated with the birth products and excretions of infected animals, mainly of sheep and goats. A 28-year-old cattle raiser presented with headache and fever without a significant medical history, although his cattle herd had previously been diagnosed with brucellosis. He spent time outdoors but reported no tick bites or eschar. Initial evaluation revealed mild thrombocytopenia and elevated aspa

Destructive spinal change that directly abuts an aneurysm or vascular graft should first be read as a vascular sign. The key distinction is noninfectious chronic contained rupture, in which pulsatile pressure or contained hematoma causes smooth vertebral scalloping, versus infected aneurysm or graft, in which contiguous spread produces osteomyelitis or spondylodiscitis. Coxiella burnetii is an important cause of the latter because chronic Q fever can produce indolent, culture-negative vascular infection with adjacent spinal extension. Reported Q fever vertebral osteomyelitis frequently coexist

Q fever, caused by Coxiella burnetii, rarely presents as severe disseminated disease, and timely diagnosis can be difficult because clinical manifestations are nonspecific and routine microbiological tests are often unrevealing. We report a 68-year-old man with rheumatoid arthritis receiving long-term immunosuppressive therapy who presented with persistent unexplained fever, pancytopenia, hepatic dysfunction, polyserosal effusions, and sepsis with multisystem involvement, without a clear epidemiological exposure history. Blood cultures and routine respiratory pathogen testing were negative. Pe

Current high-throughput sequencing platforms enable routine whole-genome sequencing of bacteria via clonal amplification from primary samples, followed by DNA extraction, library preparation, and sequencing. This approach enriches bacterial DNA while removing contaminating nucleic acids, allowing efficient genomic sequencing using minimal sequencing resources per sample. However, for fastidious or obligate intracellular bacteria, such as Coxiella burnetii, cultivation is often impractical, leaving pathogen DNA heavily diluted by host and environmental nucleic acids. In such cases, sequencing r

Coxiella burnetii is an obligate intracellular zoonotic bacterium that causes Q fever. Infections can be either acute or chronic. Of note, chronic Q fever can develop months or years after primary infection without clinical symptoms, suggesting bacterial persistence. Yet, information about the induction, regulation, and/or location of C. burnetii persistence is rare. We have shown that during infection of primary macrophages, hypoxia-induced citrate limitation results in inhibition of C. burnetii replication without affecting viability. Here, primary murine macrophages were infected with C. bu

Tick-borne rickettsioses are emerging and re-emerging zoonotic diseases worldwide that are caused by Gram-negative bacteria of the genus Rickettsia. Ticks act as both reservoirs and vectors for these bacteria, thereby increasing opportunities for the spillover of infections that they cause. This study aims to bridge the knowledge gap regarding the occurrence of zoonotic Rickettsia species in the Republic of Guinea. Ticks were manually collected from dogs, cattle, goats, and sheep. Morphological identification of 1213 ixodid ticks revealed five genera. Screening of individual DNA for Rickettsia

PURPOSE: Advances in drug discovery and early clinical development depends fundamentally on robust pre-clinical research and a comprehensive understanding of cancer biology. There is an international effort to reduce the use of animals in research and product development, with a focus on technical advances to build better in vitro models. Three-dimensional (3D) cell culture mimics the architecture of solid tumors and could be validated as an alternative method to animal use in research and development, promoting faster and more.
METHODS: A workflow of assays and decision points was establishe

Coxiella burnetii is an obligate intracellular Gram-negative bacterium that causes acute and chronic Q fever in humans. Neutrophils are considered the major innate cells with a primary role in controlling extracellular bacteria. Despite the obligate intracellular lifestyle of C. burnetii, our previous study demonstrated that neutrophils are crucial for host defense against C. burnetii infection in mice. Our recent study also showed that virulent C. burnetii Nine Mile phase I (NMI) and avirulent NM phase II (NMII) strains differentially modulate autophagy and apoptosis in murine neutrophils. Ho

Claw lesions in dairy cattle pose a significant risk to dairy farmers worldwide in terms of animal welfare concerns and economic profitability. However, the use of different data sources, classification systems, and definitions of reference groups limits the comparison across herds and decreases the usability of the recordings for phenotypic and genetic analyses. In South Africa, information on claw lesions is not routinely collected by dairy farmers and data are limited to hoof trimmers recording lesions during preventative trimming or as needed by producers. Records of the most common claw l

BACKGROUND: Diabetes mellitus (DM) and associated complications remain a global public health challenge despite many confrontational aspects of the disease, and its prevalence is projected to rise in the coming decades. Thus, there is an urgent need to intensify the current efforts to address both the prevalence and adverse effects of diabetes, including the use of natural products. Increasing evidence from the scientific literature has revealed the beneficial effects of gamma oryzanol for treating diabetes and its related complications.
AIM: To investigate the effectiveness of gamma oryzanol

BACKGROUND & OBJECTIVES: Alzheimer's disease (AD) and type 2 diabetes mellitus (T2DM) share considerable similarities in their proposed patho mechanisms. Autophagy, an intrinsic cellular process involved in the degradation of dysfunctional organelles and abnormal proteins, has been implicated in the pathogenesis of both AD and T2DM. This study aims to identify potential shared biomarkers related to autophagy in AD and T2DM by analyzing hub differentially expressed autophagy-related genes (DEARGs) and examining their potential functions.
METHODS: Gene expression profiles for AD and T2DM were a
