Cardiac lymphatics: functional plasticity in development, disease, and precision-targeted therapies.

The cardiac lymphatic system plays a crucial role in maintaining myocardial homeostasis by regulating fluid equilibrium, immune surveillance, and metabolite clearance. This review highlights recent advances in understanding its development, molecular regulation, dual roles in pathophysiology, and translational potential. Cardiac lymphatic endothelial cells (LECs) develop from diverse progenitors, including venous endothelium and Isl1⁺ precursors from the second heart field (SHF) under sex-specific molecular guidance. Functionally, the Vascular endothelial growth factor C (VEGFC)/Vascular endot

Decoding Diabetes: Hub Genes as Pivotal Players in Cardiomyopathy and Kidney Disease.

INTRODUCTION: Diabetes mellitus (DM) induces systemic vascular dysfunction and leads to life-threatening complications, including diabetic cardiomyopathy (DCM) and diabetic kidney disease (DKD), whose shared mechanisms remain elusive. In this study, we performed an indepth bioinformatics analysis to identify shared therapeutic targets and key molecular players in DKD and DCM. METHODS: Integrated computational and experimental approaches were employed. Bioinformatics analysis of GEO datasets (GSE30122 and GSE197850) identified differentially expressed genes (DEGs). Hub genes were extracted via

GLP-1 RA initiation versus metformin and risk of cardiomyopathy in patients with cancer and diabetes treated with chemotherapy, radiation, or immunotherapy: a target trial emulation.

AIMS: Glucagon-like peptide-1 receptor agonists (GLP-1 RA) have demonstrated cardioprotective effects; however, their association with cardiomyopathy remains unclear among patients with cancer and type 2 diabetes mellitus (T2D) treated with chemotherapy, radiation, or immunotherapy. We evaluated whether GLP-1 RA initiation reduces cardiomyopathy risk compared with metformin. METHODS: We conducted a retrospective cohort study using a target trial emulation framework within a large global electronic health record database. Adults aged 18-75 years with cancer and T2D, and prior exposure to chemo

Impaired mitochondrial morphology and respiratory dysfunction in human induced pluripotent stem cells with mitochondrial tRNA mutations (m.3243A>G and m.14739G>A).

BACKGROUND: Mitochondrial DNA (mtDNA) mutations contribute to respiratory dysfunction and cause mitochondrial diseases. The pathologies of these multisystemic inherited diseases are poorly understood. Mutations in the mitochondrial tRNA gene are one of the most frequent mtDNA mutations and are associated with various clinical symptoms such as diabetes mellitus, hearing loss, cardiomyopathy, exercise intolerance, and found in patients with mitochondrial disorders. Human induced pluripotent stem cells (hiPSCs), generated by reprogramming patient-specific somatic cells are recognized as useful to

Identifying Diabetic Cardiomyopathy Biomarkers via Proteomic and Glycation Modification Analysis Using DIA and PRM.

Diabetic cardiomyopathy (DCM), a severe complication of type 2 diabetes mellitus (T2DM), lacks specific and effective biomarkers for early diagnosis. This study constructed a plasma-specific spectral library by integrating proteomic and nonenzymatic glycation data from eight pretreatment workflows via data-dependent acquisition. Data-independent acquisition was then applied to profile plasma proteomes and glycation modifications in controls, DM patients, and DCM patients, revealing clear disparities in protein abundance and glycation modification patterns among the three groups. Functional enr

Case Report: Euglycemic ketoacidosis in a non-diabetic patient: a rare adverse effect of SGLT2 inhibitor therapy.

BACKGROUND: Sodium-glucose cotransporter-2 (SGLT2) inhibitors are increasingly prescribed for heart failure and chronic kidney disease, irrespective of diabetic status. While their cardiovascular and renal benefits are well established, euglycemic ketoacidosis (EKA) remains a rare but potentially life-threatening complication that can occur even in non-diabetic individuals. CASE PRESENTATION: We report a 58-year-old man with ischemic cardiomyopathy (LVEF 35%) and stage 2 chronic kidney disease who developed nausea, vomiting, and fatigue two weeks after initiating dapagliflozin. Dihydromyricetin in the management of diabetes and its complications: a narrative review.

Managing diabetes mellitus (DM) and its long-term complications remains a major global health challenge. Dihydromyricetin (DHM), a natural flavonoid abundant in Ampelopsis grossedentata and Hovenia dulcis, has attracted increasing attention for its multi-target anti-diabetic properties. Growing evidence indicates that DHM improves glucose metabolism, alleviates oxidative stress and inflammation, regulates autophagy and cell death, and exerts beneficial effects in DM and a range of related complications, including diabetic nephropathy, cardiomyopathy, cognitive impairment, and wound healing imp

Cardiomyocyte-Derived USP20 Attenuates Diabetic Cardiomyopathy by Facilitating the Degradation of STING and Mitigating STING-Mediated Inflammation.

Although extensive clinical and basic research has been conducted on diabetic cardiomyopathy (DbCM), the therapeutic efficacy for this condition remains significantly limited. Ubiquitin-specific peptidase 20 (USP20), a deubiquitinating enzyme, plays an essential role in regulating protein ubiquitination and modulating various cellular processes. In this study, we aimed to investigate the effect of USP20 on the pathogenesis of DbCM, which may provide a novel therapeutic target for its treatment. The cardiomyocyte-specific USP20 conditional knockout (USP20CKO) mice were employed in this study. T

Small extracellular vesicle-mediated adipocyte-cardiomyocyte crosstalk exacerbates heart failure with preserved ejection fraction.

AIMS: Although growing evidence suggests that obesity/central adiposity predisposes to the development and exacerbation of heart failure with preserved ejection fraction (HFpEF), it remains to be clarified whether there is a causal relationship between adiposity and HFpEF pathogenesis. METHODS AND RESULTS: HFpEF was induced in male C57BL/6N mice using a high-fat diet + Nω-nitro-l-arginine methyl ester. Resection or transplantation of visceral adipose tissue (VAT) blunted or exacerbated HFpEF phenotypes, respectively, in mice. VAT from HFpEF mice displayed greater weight and secreted more smal

Association of Butyrate Supplementation with Cardiovascular Disease:A Narrative Review.

Cardiovascular diseases (CVDs) have long been a significant source of the global disease burden and a leading cause of death and disability. In recent years, the precise microbiome modulation of the gut has made remarkable progress as a new strategy for treating CVDs, especially its production of short-chain fatty acids, the primary way the gut flora affects the organism. Butyrate, one of the critical metabolites of short-chain fatty acids, butyrate uniquely restores intestinal barrier function, suppresses systemic inflammation, promotes immune tolerance, and regulates energy metabolism while

A resource of "bottom-line" variant associations for 1,281 complex traits by integrating data across published genome-wide association studies.

Through an analysis of 2,602 genome-wide association studies (GWAS) across 830 human traits, we find that most (56% of) well-studied traits have at least two published GWAS, and many (29%) have at least five. We show that the lack of an established approach for adjudicating variant association estimates across multiple published studies can lead to uncertainty and invalid inferences: using all associations ever published for a trait increases true positives (by 12%) but also false positives (by 55%) relative to using associations from the largest published GWAS for the trait. We employ a "bott

Glaucocalyxin B Attenuates Diabetic Cardiomyopathy by Suppressing the NLRP3 Inflammasome and Restoring Gut Microbiota Homeostasis.

OBJECTIVES: Glaucocalyxin B (GLB) has documented anti-inflammatory activity in several disease contexts; however, its role in diabetic cardiomyopathy (DCM)-associated inflammatory injury remains insufficiently defined. In this study we aimed to evaluate whether GLB alleviates myocardial inflammation and injury in DCM and to explore the underlying mechanisms. METHODS: We combined network pharmacology with experimental validation using a streptozotocin-induced diabetic rat model (8-week protocol) and high-glucose-challenged H9c2 rat cardiomyocytes. Oxidative stress-related indices (e.g. reactiv

USP14 and ELAVL1-induced stabilization of VDAC1 aggravates myocardial cell injury in diabetic cardiomyopathy.

BACKGROUND: Diabetic cardiomyopathy (DCM) is a major complication of diabetes mellitus, characterized by myocardial dysfunction. Voltage-dependent anion channel 1 (VDAC1), a key regulator of mitochondrial function and cellular metabolism, has been implicated in various cardiovascular diseases. However, its role in DCM and the underlying molecular mechanisms remain poorly understood. Given the increasing prevalence of diabetes and the lack of effective therapies targeting DCM, investigating VDAC1 regulatory pathways could provide novel insights into disease pathogenesis and potential therapeuti
