Circadian disruption exacerbates alcohol-induced cardiac injury through impaired cellular quality control and increased cardiomyocyte death.

Circadian rhythms orchestrate 24-h oscillations in physiology and gene expression in many tissues, including the heart. Disruptions to these rhythms have been linked to increased cardiovascular risk and impaired cellular homeostasis. However, the extent to which circadian disruption influences cardiac responses to environmental stressors such as alcohol, remains poorly understood. Herein, we performed RNA-sequencing and transcriptomic analysis on hearts from C57BL/6 J mice exposed to alcohol for ten weeks and collected at two distinct circadian time points (Zeitgeber Time (ZT)8 and ZT20). Cardioprotective Effect of Metformin Treatment in Mouse Model of Experimental Autoimmune Myocarditis.

BACKGROUND: Metformin is a well-established treatment for Type 2 diabetes. Beyond its role in glycemic control, metformin has gained attention for its pleiotropic effects, including cardioprotective properties. However, its role in autoimmune myocarditis and subsequent postinflammatory dilated cardiomyopathy remains poorly understood. METHODS: To investigate the role of metformin in autoimmune-mediated cardiac disease, we employed the experimental autoimmune myocarditis mouse model. Six to eight weeks old Balb/c mice were immunized twice with an α-myosin heavy chain peptide emulsified in comp

Ebselen restores mitochondrial function through metabolic rewiring in Barth syndrome and related mitochondrial diseases.

Despite major advances in the genetic diagnosis of mitochondrial disorders, effective disease-modifying therapies remain scarce. Here, we identify ebselen (EBS) as a promising therapeutic candidate through a phenotypic drug screen in a yeast model of Barth syndrome, a cardiomyopathy caused by defective cardiolipin maturation. EBS improved oxidative phosphorylation-dependent growth across diverse fungal models of mitochondrial diseases, including defects in complex I, complex IV, mitochondrial DNA maintenance, mitochondrial translation, and ATP synthase. Therapeutic efficacy was further validat

Cyclic azapeptide CD36 ligand attenuates cardiac injury and reduces long-chain fatty acid accumulation after myocardial ischemia-reperfusion in mice.

Ischemic heart disease remains a leading global cause of death. We investigated the cardioprotective effects of the selective cluster of differentiation-36 receptor (CD36) modulator azapeptide MPE-298 in a mouse model of myocardial ischemia-reperfusion. Given before reperfusion, a single intravenous dose of azapeptide MPE-298 reduced infarct size by 44% of the area-at-risk and transiently decreased left ventricular long-chain fatty acids (LCFA) accumulation, independently of saturation status. Metabolomic profiling identified changes in amino acids that may fuel the tricarboxylic acid cycle an

Clinical consensus guidelines for the management of Noonan syndrome in Japan.

Noonan syndrome (NS) is a multisystem RASopathy marked by characteristic facies, congenital heart disease, short stature, and variable neurodevelopmental and systemic complications. Although molecular and clinical knowledge has advanced, evidence-based clinical guidance in Japan has been limited. A multidisciplinary committee in pediatric endocrinology, clinical genetics, and pediatric cardiology developed consensus guidelines on diagnosis, comorbidity assessment, treatment, and transitional care. Clinical questions were defined, and the literature published through March 2024 was systematical

Therapeutic Targets for Myocardial Fibrosis: A Comprehensive Review of Current and Emerging Approaches.

INTRODUCTION: Cardiovascular disorders (CVDs) remain the leading cause of global mortality, surpassing other chronic illnesses. An alarming rise in CVD-related deaths, particularly among younger populations, has intensified research efforts to better understand the disease and its complications. Among these, myocardial fibrosis plays a central role in the development of cardiac dysfunction and heart failure. Given its multifactorial nature, diverse therapeutic strategies are required to manage its progression effectively. METHODS: A comprehensive literature review was conducted using database

Transcriptional readthrough at Atf4 locus suppresses Rps19bp1 and impairs heart development.

AIMS: Activating transcription factor 4 (ATF4) functions as a transcriptional regulator in various cell types and tissues under both physiological and pathological conditions. While previous studies have linked ATF4 activation with promoting cardiomyocyte (CM) death in dilated cardiomyopathy (DCM), atrial fibrillation, and heart failure, its role in developing CMs remains unexplored. METHODS AND RESULTS: We generated multiple distinct CM-specific (Atf4cKO(e2/3/pA) and Atf4cKO(e2)) and global Atf4 knockout (KO; Atf47del/7del and Atf41ins/1ins) mouse models targeting different Atf4 regions, as

Lactococcus lactis subsp. Cremoris reprograms systemic metabolism and protects against myocardial injury.

Therapeutic microbes are increasingly recognized as potent modulators of host physiology, yet their influence beyond the gut remains underexplored. While Lactococcus lactis subsp. Cremoris (LLC) has been shown to preserve gut epithelial integrity and counteract Western diet-induced metabolic syndrome in murine models, its effects on extraintestinal systems such as the gut-cardiovascular axis, are not well defined. In this study, we employed a multimodal experimental approach to investigate whether LLC confers cardioprotective benefits. We showed that LLC supplementation significantly preserved

Fucoxanthin prevents septic cardiomyopathy by targeting BRD2 in M1-polarized macrophages.

Septic cardiomyopathy (SCM) leads to heart failure, with few effective approaches for its intervention. Fucoxanthin (FX) possesses anti-inflammatory capacity, but its effect on SCM remains unknown. Herein, we report that FX protects against cardiac dysfunction in a lipopolysaccharide (LPS)-induced mouse model of sepsis with a dramatic reduction of inflammatory cell infiltration in the heart tissue. The analysis of human and mouse databases revealed M1-polarized macrophages as the most evidently infiltrated immune cells into the heart tissue under septic conditions. This phenomenon was found in

Micheliolide alleviates myocardial ischemia/reperfusion injury in rats via KEAP1/NRF2-mediated suppression of oxidative stress and inflammation.

BACKGROUND: After acute myocardial infarction (AMI), reperfusion therapy can help restore blood flow and nutritional support to the ischemic myocardium, thereby limiting ongoing myocardial injury. However, its effectiveness is increasingly challenged by myocardial ischemia-reperfusion (I/R) injury. In this regard, micheliolide (MCL) has been reported to exert beneficial effects in cardiovascular disease. Herein, we aimed to determine the mechanism underlying the cardioprotective effects of MCL in a rat I/R model. METHOD: Rats were randomly divided into a control group, an I/R group, and an I/

Genetic and epidemiological insights into the role of neutrophil percentage in heart failure risk: a Mendelian randomization and NHANES study.

Systemic inflammation contributes to the progression of heart failure (HF). However, the precise effects of platelet count (PCT), white blood cell count (WBC), neutrophil percentage (NEUT) on HF development and progression are poorly understood. Thus, in this study, we aimed to assess the associations between PCT, WBC, NEUT and HF. We utilized National Health and Nutrition Examination Survey (NHANES) data from 1999 to 2018, excluding the second round of test data from 2001 to 2002, to assess HF using questionnaires and measured blood parameters adjusted by the covariates. We employed weighted

Mitoregulin supports mitochondrial membrane integrity and protects against cardiac ischaemia-reperfusion injury.

AIMS: We and others discovered a highly conserved mitochondrial transmembrane microprotein, named Mitoregulin (Mtln), that supports lipid metabolism. We reported that Mtln strongly binds cardiolipin (CL), increases mitochondrial respiration and Ca2+ retention capacities, and reduces reactive oxygen species (ROS). Here, we extend our observation of Mtln-CL binding and examine Mtln influence on cristae structure and mitochondrial membrane integrity during stress. METHODS AND RESULTS: We demonstrate that mitochondria from constitutive- and inducible Mtln-knockout (KO) mice are susceptible to mem

Liraglutide Modulates Zinc Release and Improves Mitochondrial Function in Insulin-Resistant Senescent Cardiomyocytes.

Aging and insulin resistance are intertwined factors in the development of metabolic diseases such as type 2 diabetes and cardiovascular disorders. Liraglutide, a glucagon-like peptide-1 (GLP-1) receptor agonist, has shown promising cardioprotective effects in preclinical and clinical studies of metabolic diseases. Yet, its action on insulin-resistant aged subjects is not clearly defined. This study aimed to investigate the effects of liraglutide on intracellular zinc levels, including its modulation of oxidative stress, mitochondrial function, and Endoplasmic Reticulum (ER) stress in a novel
