Carnitine plays an obligatory role in energetics owing to its role in the translocation of long-chain fatty acids into the mitochondrion for oxidation. Here, we determined the metabolic and behavioral consequences of systemic carnitine deficiency (SCD) in mice. Female C57BL/6J mice were randomized to receive normal drinking water (control, n = 8) or drinking water supplemented with mildronate 4g.L-1 (mildronate, n = 8) for 21 days. Body composition was assessed at baseline and post-treatment. Metabolic and behavioral phenotyping was performed continuously over 72 h following 14 days of control

Cardiomyopathy, like other major cardiovascular diseases, is becoming increasingly prevalent and represents a growing public health challenge. Although both genetic and acquired factors contribute to its development, obesity and diabetes remain among the most significant modifiable risks. These conditions share key pathological features, including oxidative stress, inflammation, and elevated lipid peroxidation, all of which can disrupt cellular metabolism and function. Lipid oxidation products, irrespective of their origin, are characterized by increased oxygenation and hydrophilicity compared

INTRODUCTION: Pontocerebellar Hypoplasia, Hypotonia, and Respiratory Insufficiency Syndrome, Neonatal Lethal (PHRINL Syndrome) is a rare genetic disorder caused by impaired oxidative phosphorylation due to reduced activity of mitochondrial complexes I, IV, and V. The condition results from homozygous or compound heterozygous pathogenic variants in the ATAD3A gene, and is inherited in an autosomal recessive manner. Affected individuals typically present in early infancy with hypotonia, encephalopathy, corneal clouding, cardiomyopathy, and respiratory failure, and often die during infancy.

BACKGROUND: Limited evidence is available from randomized trials to guide clinical decision-making for cardiovascular disease (CVD) prevention with statins among women with breast cancer. We leveraged real-world data to estimate the effect of statin therapy vs usual care on the 5-year risk of CVD among breast cancer survivors.
METHODS: We sequentially emulated target trials using the National Cancer Institute-Kaiser Permanente Breast Cancer Survivors Cohort. Women aged 40 - 84 years, diagnosed with stage 0-III breast cancer from 1993 - 2022, without CVD, no statins in the past 6 months, and l

Cardiovascular disease (CVD) remains the leading cause of morbidity and mortality worldwide. While studies suggest that berry fruits offer beneficial effects in CVD prevention, the bioactive compounds within different berries vary, and the specific mechanisms behind their protective effects are not yet fully elucidated. Our study aimed to evaluate and compare the cardioprotective and anti-atherosclerotic effects of Morus alba (white mulberries, WMex) and Vaccinium myrtillus L. fruit extracts (blueberries, BBex), focusing on acute myocardial infarction (AMI), CVD risk factors, atherosclerosis d

IMPORTANCE: Dilated cardiomyopathy (DCM) is a major cause of heart failure that disproportionately affects individuals of African genetic ancestry (AFR), among whom familial clustering of disease is also more pronounced relative to those of European ancestry (EUR). However, established monogenic DCM genes, identified primarily in EUR populations, explain a smaller proportion of DCM cases in AFR populations. A recent study identified a common AFR-specific nonsense variant in CD36 that accounts for a substantial burden of DCM in AFR. How the risk and population impact of this variant compare wit

BACKGROUND: Percutaneous coronary intervention (PCI) and coronary artery bypass grafting (CABG) are standard revascularization strategies for chronic coronary syndrome (CCS). However, their effects on myocardial reperfusion have not been assessed using biological markers. Adenosine A2A receptor (A2AR) function on peripheral blood mononuclear cells (PBMCs), evaluated through cAMP production and EC50-measurement, may reflect myocardial ischemia and reperfusion status.
OBJECTIVES: To compare the effects of CABG and PCI on A2AR pharmacological profiles assessed by cAMP-production (EC50) values.

Cardiovascular disease (CVD) accounts for most of the increased morbidity and mortality associated with in chronic kidney disease (CKD). Cardiovascular risk emerges early in CKD, initially driven by atherosclerotic events but later dominated by heart failure and sudden cardiac death. This transition reflects progressive arteriosclerosis and the development of CKD-associated cardiomyopathy. Although patients receiving dialysis experience the highest cardiovascular risk, established atherosclerotic therapies such as lipid-lowering agents offer limited benefit at this stage. Recent therapeutic ad

BACKGROUND: Mutations in the Four-and-a-Half LIM Domains 1 (FHL1) gene are increasingly recognized as a rare cause of inherited cardiomyopathies, often associated with skeletal myopathy and adverse cardiac outcomes. The phenotypic spectrum and clinical implications of FHL1 variants remain poorly defined.
OBJECTIVE: To systematically review published cases of FHL1-related cardiomyopathy and characterize the clinical, genetic, and pathological features.
METHODS: We conducted a systematic literature search in PubMed and EMBASE up to July 2025 using predefined criteria to identify studies report

BACKGROUND: Obesity and Type 2 Diabetes are major contributors to cardiac hypertrophy and dysfunction, yet the molecular mechanisms driving early myocardial alterations remain incompletely understood. Evidence from rodent models and end-stage human disease suggests that cytoskeletal remodeling and oxidative stress may contribute to early increases in cardiomyocyte stiffness and hypertrophy. Whether these processes are involved at earlier disease stages in translationally relevant large-animal models remains unclear.
METHODS: Heart tissue from male Göttingen minipigs subjected to a 13-month in

Propionic acidemia (PA) is a rare inherited metabolic disorder associated with recurrent metabolic decompensations and chronic multisystemic complications. Liver transplantation (LT) may improve metabolic stability, but its long-term impact on organ involvement remains debated. We retrospectively studied 20 patients with PA transplanted between 1993 and 2024 in three French reference centers. Clinical, biochemical, and organ-specific data were collected before and after transplantation. Eighteen isolated LT and two combined liver-kidney transplantations were performed. Median age at transplant

Hypertrophic cardiomyopathy (HCM), arrhythmogenic cardiomyopathy (ACM) and dilated cardiomyopathy (DCM) represent the most frequent structural heart disorders. Due to the complexity of their diagnosis and the frequent occurrence of sudden cardiac death as the first manifestation, these diseases impose a relevant socio-economic burden, highlighting the urgent need for novel disease-specific biomarkers. Here, we assess the potential of microRNAs (miRNAs) as non-invasive predictors for HCM, ACM, and DCM. Electronic databases were systematically searched using relevant keywords pertaining to the d

Celiac disease (CD), a chronic immune-mediated enteropathy triggered by gluten ingestion, is increasingly recognized as a systemic disorder with significant extraintestinal manifestations, including effects on the cardiovascular system. This review synthesizes recent epidemiological evidence demonstrating that individuals with CD face a modest but statistically significant increase in cardiovascular disease risk, including ischemic heart disease, myocardial infarction, and venous thromboembolism, despite a lower prevalence of traditional risk factors such as obesity, hypertension, and smoking.

BACKGROUND: Hypertension-induced cardiac fibrosis leads to heart failure. Large-conductance calcium-activated potassium (BK) channels regulate vascular tone, and their activation may mitigate fibrosis. This study explores the impact of BK channel activation on angiotensin II (Ang II)-induced hypertension and cardiac fibrosis and its effects on nuclear factor-kappa B (NFκB) signaling.
METHODS: Male C57BL/6 J mice were infused with Ang II for 4 weeks to induce hypertension and cardiac fibrosis. The BK channel activator BMS-191011 was administered via intraperitoneal injection. Cardiac tissue wa
