Overview of Delivery Methods for Gene Editing.

The clinical success of CRISPR-based interventions depends primarily on the efficient delivery of editing components into target cells. While base and prime editing have refined genomic precision, achieving therapeutic efficacy requires specialized vehicles that can navigate systemic circulation, escape endosomes, and ensure cell-specific entry. Delivery platforms are traditionally categorized into viral and nonviral systems. Viral vectors, namely adeno-associated vectors (AAV), lentiviral vectors, and adenoviral vectors, employ evolved mechanisms to achieve high transduction efficiency and pr

Production and Storage of High-Capacity Adenoviral Vectors for the Delivery of Advanced CRISPR Systems.

Genome editing based on engineered CRISPR systems is advancing rapidly, with the field increasingly moving toward approaches that avoid the induction of mutagenic double-stranded DNA breaks (e.g., RNA-programmable base editing, prime editing, and donor DNA transposition). These nuclease-free strategies often rely on large or multi-component molecular assemblies that can include gene-sized donor DNA substrates. There is, nonetheless, a paucity of vehicles capable of delivering such large and complex genome-editing components effectively and, ideally, in defined stoichiometric ratios. High-capac

Completed clinical trials of p53-targeted therapeutic strategies in cancer: A cross-sectional ClinicalTrials.gov registry analysis.

The tumor suppressor gene TP53 is one of the most frequently altered genes in human cancers and plays a central role in regulating cell cycle arrest, apoptosis, and genomic stability. Therapeutic strategies targeting the p53 pathway have been investigated to restore or exploit p53 function in human malignancies. However, the characteristics, reporting completeness, and clinical development patterns of completed trials evaluating p53-targeted therapeutic strategies have not been comprehensively summarized. This study aims to characterize the design, therapeutic strategies, reporting completenes

Host life history traits as drivers of viral infection in European bats.

Bats are recognized as natural reservoirs of a wide range of viruses. Understanding the factors that determine patterns of viral occurrence in reservoir species, particularly viral richness and viral detection frequency, is essential for explaining host-pathogen dynamics and anticipating public health risks. In this study, we evaluated the influence of life-history traits on viral frequency (defined as the proportion of positive detections across studies) in European bats for six important viral groups (Adenoviridae, Herpesviridae, Papillomaviridae, Filoviridae, Coronaviridae, and the genus Ly

No-known-vector flaviviruses exhibit diverse replication and virulence phenotypes in mice.

The Orthoflavivirus genus (flaviviruses) includes globally significant arboviruses, which cycle between arthropod vectors (mosquitoes and ticks) and vertebrate hosts. In contrast, no-known-vector flaviviruses (NKVFVs) have been isolated from rodents and bats, but not arthropods, so they are thought to spread by vector-independent routes. However, little is known about the host range and pathogenic mechanisms of these viruses. To evaluate NKVFV pathogenesis, we infected wild-type, Ifnar1-/-, and Ifnar1-/- Ifngr1-/- mice by footpad inoculation with 12 NKVFVs: Entebbe bat virus (ENTV), Sokuluk vi

Engineering human endogenous retrovirus-K for improving immunogenicity and broad cancer targeting.

BACKGROUND: HERV-K (HML-2) is the most studied transcriptionally active endogenous retrovirus in human cancers, yet immune tolerance and incomplete mechanistic understanding have impeded its clinical use as an immunotherapy target. We hypothesized that rational engineering of the HERV-K envelope putative immunosuppressive domain (ISD) could improve immunogenicity. METHODS: We performed a mutational screen across the ISD region, evaluated NF-κB activation and then analyzed antigen expression in human monocyte-derived dendritic cells (moDCs) using flow cytometry, RNA-seq, and TEM. Human PBMCs w

Detection, characterization, and evolutionary inferences regarding a novel virus identified in Amblyomma dissimile Koch, 1844 (Acari: Ixodidae) from the Amazon, Pará, Brazil.

Ticks are among the primary vectors of infectious agents of medical and veterinary importance, second only to mosquitoes in the number of pathogens transmitted to humans. These arthropods are capable of transmitting a wide diversity of microorganisms, including protozoa, bacteria, and viruses. Notable among tick-borne viruses are those associated with viral encephalitis, which cause severe acute and chronic neurological infections in humans. This study aimed to characterize the RNA virome of an Amblyomma dissimile specimen collected from a Boa constrictor rescued in the municipality of Ananind

Safety and immunogenicity of an adenoviral-vectored vaccine against Crimean-Congo haemorrhagic fever in the UK (CCHF01): an open-label, first-in-human, non-randomised, phase 1 clinical trial.

BACKGROUND: There are no approved vaccines for Crimean-Congo haemorrhagic fever virus (CCHFV), which remains a WHO priority pathogen for vaccine development. We evaluated the safety and immunogenicity of ChAdOx2 Crimean-Congo haemorrhagic fever (CCHF), a novel, non-replicating adenoviral-vectored vaccine, in a first-in-human phase 1 clinical trial. METHODS: CCHF01 was an open-label, first-in-human, non-randomised, phase 1 trial conducted at a single centre, the University of Oxford Centre for Clinical Vaccinology and Tropical Medicine in Oxford, UK. Healthy adults aged 18-55 years, with no co

A Modular Biomimetic Nanovaccine for TBEV: Orchestrating Cascade Immunoprotection via Programmed Dendritic Cell Targeting and Antigen-Adjuvant Co-delivery.

Tick-borne encephalitis (TBE) is an escalating global health threat, increasingly exacerbated by climate change, and underscores the need for innovative vaccine strategies. Conventional subunit vaccines are limited by suboptimal spatiotemporal dissociation between antigens and adjuvants, as well as insufficient specific targeting, resulting in compromised immunogenicity. Herein, we report a biomimetic nanovaccine platform engineered to overcome these limitations through the synergistic co-delivery of antigen and adjuvant. This platform is constructed by co-displaying the TBE virus envelope pro

In silico and in vitro immunogenicity risk assessment of process- and product-related protein impurities reported in adenoviral-vectored COVID-19 vaccines.

Process-related protein impurities (PRPIs) such as host cell proteins (HCPs) in biologic products have been associated with unexpected immune responses, adverse events, and the discontinuation of clinical programs. While an HCP threshold of 100 ng/mg (PPM) is considered to be 'acceptable', that threshold may be too high if individual impurities have strong intrinsic immunogenicity or, in contrast, may be overly cautious if many of the HCP impurities have low immunogenicity. In silico methods for predicting the immunogenicity of PRI and HCP may help triage higher risk from lower risk entities.

High Exposure of Intravenous Alemtuzumab is Associated with Increased Risk of Adenovirus reactivation in Patients with Non-Malignant Disease Undergoing Allogeneic HCT.

INTRODUCTION: Alemtuzumab, a recombinant humanized immunoglobulin G1 monoclonal antibody directed against human CD52, is a widely used serotherapy strategy in pediatric allogeneic hematopoietic stem cell transplantation (HCT) in non-malignant diseases (NMD) to help reduce the risk of graft rejection (GR) and graft versus host disease (GVHD). Excessive exposure to alemtuzumab potentially delays immune reconstitution and increases the risk of infections. METHOD: We conducted a retrospective analysis at a single institution of 95 pediatric patients with NMD who underwent HCT and received intrave

Molecular surveillance of adenovirus and coronavirus in Procellariiformes, Brazil: Detection of new divergent adenoviruses.

Adenoviruses are widespread DNA viruses that infect a broad range of vertebrate hosts and are recognized for their long-term host coevolution, remarkable genetic diversity, and broad pathogenic potential. Coronaviruses circulate among wild birds due to their evolutionary potential and are relevant to animal and public health. However, the diversity and ecological role of these viruses in pelagic seabirds remain poorly understood. This study provides the first molecular survey of adenoviruses and coronaviruses in Procellariiformes from the South Atlantic. Fifty stranded individuals representing

A novel mycovirus identified in clinical isolates of Cryptococcus neoformans.

Mycoviruses are widespread throughout the fungal kingdom and are known to infect diverse fungal taxa, including fungal species that are important plant and human pathogens. Although many mycoviruses have been found to have minimal effects on their host, several viruses have been found to modulate fungal physiology, and, as a result, impact fungal virulence. Screens for mycoviruses in clinically relevant fungi have identified numerous mycoviruses within several important human pathogens, but mycoviruses remain uncharacterized in the clinically relevant human pathogen Cryptococcus neoformans. C.

Infection-specific long-chain fatty acid metabolism as a broad anti-enterovirus target.

Enteroviruses are arguably the most numerous group of viruses infecting humans. While most enterovirus infections are benign and self-resolving, their sheer number inevitably increases the chances of multiple complications. The diversity of enteroviruses means that the development of vaccines is only economically feasible against a select few, and no direct-acting or host-targeted anti-virals are approved to treat enteroviral infections, largely due to the rapid development of resistance against all experimental drugs. Here, we explored a universal property of enterovirus infection-a massive u

Human astroviruses: molecular virology.

SUMMARYHuman astroviruses (HAstVs) are important etiologic agents of pediatric gastroenteritis and are increasingly recognized as pathogens associated with extraintestinal disease, including encephalitis and meningitis, particularly in immunocompromised individuals. Despite their clinical relevance, widespread prevalence, and expanding genetic diversity, no approved vaccines or antiviral therapies are available. Progress toward effective interventions requires a comprehensive understanding of HAstV molecular biology, including mechanisms of cell entry, viral replication, and immune recognition

Development of a risk score to predict encephalitis by Herpes Simplex Virus and Varicella Zoster Virus.

Encephalitis guidelines recommend repeating a lumbar puncture (LP) 3-7 days after an initial negative cerebrospinal fluid (CSF) herpes simplex virus (HSV) polymerase chain reaction (PCR) test in patients with high suspicion for HSV or varicella zoster virus (VZV), but this subgroup has not been specified. Retrospective observational study of 602 adults with encephalitis from 2002 to 2023 from two large U.S. healthcare systems. Multivariate logistic regression models were developed and internally validated to identify clinically relevant and statistically significant features predicting HSV, VZ

The hidden burden of viral meningitis in ethiopia: Etiology and clinical outcomes.

BACKGROUND: Viral meningitis remains a significant global public health problem, particularly in resource-limited settings where the diagnosis of viral etiology is mainly based on clinical assessment. This study aimed to identify the common viral etiologies of meningitis, describe their associated clinical features, and assess patient outcomes among individuals suspected of meningitis without evidence of bacterial meningitis. METHODS: We conducted a multicenter study involving 430 patients clinically suspected of meningitis. Clinical data and cerebrospinal fluid samples (CSF) were collected f

In-Hospital Outcomes of Acyclovir With or Without Corticosteroid for Herpes Simplex Encephalitis: A Nationwide Retrospective Cohort Study.

BACKGROUND AND OBJECTIVES: Acyclovir is the standard therapy for herpes simplex virus (HSV) encephalitis, yet mortality remains high. Secondary inflammatory responses triggered by viral infection potentially contribute to additional brain injury. Given their anti-inflammatory properties, corticosteroids are considered a biologically plausible adjunctive treatment for HSV encephalitis. However, evidence regarding their effects remains inconclusive, highlighting the need for large-scale evidence. METHODS: We conducted a retrospective cohort study using the Japanese national inpatient Diagnosis

Viral Infection-Induced Epilepsy and the Gut Microbiome Crosstalk.

Viral infections of the central nervous system (CNS) pose a significant risk for seizures, and in some cases, long-term epileptogenesis. Seizure development following viral infection involves diverse CNS mechanisms, with emerging evidence that peripheral factors, including gut microbiome composition and immune-inflammatory responses, also contribute. The gut microbiome is a multifaceted ecosystem with important roles in CNS health, and advances in sequencing have enabled investigation of host and microbial genomic changes associated with seizure development. While both animal and human studies
