Exercise intensity-dependent cardiac telocyte expansion is associated with physiological growth despite JAK/STAT pathway inactivity in male Wistar rats.

While exercise induces physiological cardiac growth, the underlying cellular mechanisms remain incompletely understood. This study investigated the role of cardiac telocytes (TCs) and the Janus kinase (JAK)/signal transducer and activator of transcription (STAT) pathway in mediating exercise intensity-dependent cardiac adaptation. Twenty-four male Wistar rats were assigned to control (CTRL), high-intensity interval training (HIIT) or low-intensity interval training (LIIT) groups for 8 weeks. Physiological hypertrophy was assessed via heart weight/body weight ratio, left ventricular wall thickn

Validation of the VasCog-2-WSO VCID Criteria in a Dementia-Free and Stroke-Free Community Cohort: Cardiovascular Risk and Mortality.

BACKGROUND AND OBJECTIVES: Vascular cognitive impairment and dementia (VCID), a major contributor of dementia, remains underrecognized, owing to the absence of universally accepted diagnostic criteria. The VasCog Society and WSO recently released updated criteria (VasCog-2-WSO); however, their applicability is unknown yet. We aimed to evaluate these criteria in a dementia-free and stroke-free community cohort and determine their associations with vascular burden and long-term mortality. METHODS: We conducted a community-based cohort study using data from the I-Lan Longitudinal Aging Study. Ad

Exploring the Role and Mechanism of DSF in HFpEF Based on IL-1β/IL-1βR/TAK1/RIPK1 Axis-Mediated PANoptosis.

Heart failure with preserved ejection fraction (HFpEF) has a high prevalence and a low quality of life, and there are limited medications for the treatment of this disease. In recent years, disulfiram (DSF), an FDA-approved drug for the treatment of chronic alcohol addiction, has been found to have anti-inflammatory properties. The present study was designed to investigate the cardioprotective effects of DSF on patients with HFpEF and its mechanism using a model of HFpEF induced in mice fed a high-fat diet (HFD, 60% of calories from fat) and Nω-nitro-L-arginine methyl ester (L-NAME, 0.5 g/L in

Modulation of Ferroptosis by the Irx3-Etfa Pathway Protects Against Cardiac Hypertrophy.

Pathological cardiac hypertrophy is a major contributor to heart failure and is often accompanied by ferroptosis and mitochondrial dysfunction. However, the upstream transcriptional mechanisms governing these processes remain poorly defined. We performed integrative bioinformatics analysis using transverse aortic constriction (TAC)-induced hypertrophic heart datasets to identify mitochondria-related differentially expressed genes (MitoDEGs), followed by transcription factor prediction and experimental validation in both in vivo and in vitro models. Adeno-associated virus-mediated overexpressio

Impact of core temperature management and postoperative continuous renal replacement therapy in extensive aortic arch repair for type A dissection.

BACKGROUND: Continuous renal replacement therapy (CRRT) represents a critical complication following extensive aortic arch repair for type A aortic dissection (TAAD), significantly influencing patient outcomes. This study evaluates the impact of different hypothermic circulatory arrest (HCA) temperature strategies on postoperative CRRT requirements. METHODS: A total of 1,283 TAAD patients who underwent total arch replacement (TAR) with frozen elephant trunk (FET) procedure were retrospectively enrolled [2010-2018]. Patients were divided into CRRT (N=123) and non-CRRT (N=1,160) groups. Core te

The relationship between serum ghrelin and cardiac geometry structure in children with left-to-right shunt CHD and its clinical significance.

OBJECTIVE: To investigate the variations of serum ghrelin levels in children with left-to-right shunt CHD, and analyse the correlation with the geometric structure of the heart. It may provide some clinical guidance for evaluating the early changes of cardiac structure and function in CHD children. METHODS: A total of 93 children with left-to-right shunt CHD who were hospitalised to Children's Hospital of Soochow University from September 2021 to February 2023 were included in the CHD group (of which 58 were ventricular septal defect and 35 were patent ductus arteriosus). In addition, 57 case

Extracellular matrix remodeling and internal elastic lamina alterations in cerebral arteriovenous malformations: Insights into vascular wall biology.

Cerebral arteriovenous malformations (cAVMs) are high-flow vascular lesions associated with a relevant risk of intracranial hemorrhage. Beyond angioarchitectural determinants, emerging evidence suggests that alterations of the internal elastic lamina (IEL) and elastic fiber network may contribute to vascular remodeling and structural instability. We conducted a structured qualitative literature review with narrative synthesis, without quantitative meta-analysis because of substantial methodological and biological heterogeneity across studies. We included histological, molecular, and genetic in

Hexokinase 2-Driven Cardioprotection in Ischemia, Hypertrophy, and Regeneration: From Underlying Mechanisms to Clinical Translation.

The heart is the most energy-demanding organ in the body and has a dynamically adaptable metabolic state. Under normal conditions, the adult heart primarily relies on mitochondrial oxidative phosphorylation for ATP generation, whereas glycolysis supplies a minor portion of the energy. However, under pathological conditions, glycolysis is increased, and mitochondrial oxidative phosphorylation is reduced. Hexokinase 2 (HK2) is the first rate-limiting enzyme in the glycolytic pathway in cardiomyocytes. Accumulating evidence in recent years has indicated that HK2 confers protection to the heart un

Efficacy of SGLT2 inhibitors, GLP-1 receptor agonists, and aerobic exercise for moderate-to-severe obstructive sleep apnea in overweight or obese patients: a network meta-analysis.

OBJECTIVE: Obstructive sleep apnea (OSA) is a highly prevalent sleep disorder strongly linked to obesity and substantially increases the risk of cardiovascular and metabolic diseases. Sodium-glucose cotransporter 2 (SGLT2) inhibitors, glucagon-like peptide-1 (GLP-1) receptor agonists, and aerobic exercise have shown potential in improving OSA through distinct metabolic and physiological mechanisms. However, direct comparative evidence of their efficacy remains limited. This network meta-analysis aimed to compare the effects of SGLT2 inhibitors, GLP-1 receptor agonists, and aerobic exercise on

Amentoflavone, a Preferentially TGF-β Receptor 1 Inhibitor, Alleviates Cardiac Remodeling and Dysfunction Through Modulating the TGF-β/Smad and MAPK Signaling Pathways.

Chronic cardiac stress induces cardiac hypertrophy and fibrosis, thereby promoting pathological heart remodeling, which is the core pathological process of heart failure. Amentoflavone (AMF) has beneficial therapeutic effects against several cardiovascular diseases; however, its role and underlying mechanisms in pathological cardiac remodeling remain unclear. Here, we investigated the protective role of AMF against pathological cardiac remodeling in transverse aortic constriction (TAC)-induced overloaded hearts. Mice with pressure overload caused by TAC were treated with AMF, and the effects o

Smad3 deficiency attenuates disease severity in a zebrafish model for Smad6-related aortic disease.

Thoracic aortic dissection (TAD) associates with a high mortality rate. Treatment options are limited and mainly consist of surgical repair at critical aortic diameters as current pharmacological interventions are unable to stop disease progression. Despite the existence of different mouse models for thoracic aortic aneurysm (TAA) and TAD, the underlying disease mechanisms remain elusive. In humans, loss-of-function of SMAD3 or SMAD6 increases the risk for TAA. We therefore targeted both ohnologs of smad3 and smad6 in zebrafish in order to further investigate their contribution to aortic homeo

Rare culprit and clinical dilemma: thrombolysis in a spontaneous anterior cerebral artery dissection.

Intracranial arterial dissection (IAD) is a rare cause of ischaemic stroke, with involvement of the anterior cerebral artery (ACA) being uncommon. We present the case of a woman in her mid-40s who developed acute right leg weakness associated with a headache. Initial non-contrast CT (NCCT) and CT angiography (CTA) scans were reported as normal; she was thrombolysed with intravenous alteplase for a clinical diagnosis of stroke. Post-thrombolysis NCCT showed an infarct in the left ACA territory. At the time of review, in our weekly neuroradiology meeting, a dissection of the proximal left ACA wa

Decoding the heart-saving power of Sanweidoukou decoction: Mitochondrial and molecular insights.

BACKGROUND: Inhibition of cardiac hypertrophy and improving cardiac function are the main aims for treating heart failure. Sanweidoukou decoction (SD-3) is a traditional Chinese remedy known for its heart-protective properties and effectiveness in treating cardiovascular and cerebrovascular conditions. However, the mechanisms of action of SD-3 have not been clearly established. We investigated the protective effects of water extraction of SD-3 against heart failure and elucidated on its potential molecular mechanisms. METHODS: Cardiac failure models were established in rats and H9c2 cells usi

Improvement of aphasia deficits following traumatic intracranial hemorrhage: a case report.

Aphasia is a common and disabling consequence of dominant-hemisphere brain injury, and the pharmacological options after traumatic intracranial hemorrhage are limited. A 69-year-old man developed severe aphasia after traumatic intracranial hemorrhage. On day 7 after traumatic brain injury (TBI), he was transferred for inpatient rehabilitation, where the Korean version of the Western Aphasia Battery (K-WAB) showed marked language impairment, with an Aphasia Quotient (AQ) of 66.2 (scale 0-100; higher scores indicate better language function). He received intravenous Cerebrolysin, 30 mL/day infus

Parent-of-origin effects in Alzheimer's liability dissociate neurocognitive and cardiovascular traits in at-risk individuals.

Alzheimer's disease (AD) has a higher prevalence in women than men and is more frequently inherited from mothers than fathers. Yet, while neuroimaging and biomarker studies link maternal family history to stronger AD-related alterations, epidemiological studies suggest that paternal history confers comparable or even greater risk. Here, we leverage the deeply profiled PREVENT-AD cohort to derive three intermediate phenotypes of AD susceptibility. Drawing on nearly 1,000 individual study visits, we quantify how these intermediate phenotypes vary as a function of maternal versus paternal AD line
