BACKGROUND: To mitigate the risk of outbreaks of serotype 2 poliomyelitis after withdrawal of this serotype from oral poliovirus vaccine (OPV) in 2016, inactivated poliovirus vaccine (IPV) was introduced into the routine immunization (RI) programmes of all countries using OPV. Since 2022, WHO has recommended a 2-dose schedule, with a first dose at 14 weeks of age followed by a second dose at least 4 months later (e.g., 14-39 week schedule), although an earlier schedule may be adopted, despite lower immunogenicity, if vaccine coverage is low at older ages.
METHODS AND FINDINGS: We combined pub

Our knowledge of Cryptococcus neoformans and cryptococcal disease spans many years. Yet, despite numerous clinical studies and revisions to treatment protocols, the burden of cryptococcal disease remains alarmingly high, more so in endemic regions like Africa. Thus, the inclusion of this organism in the World Health Organization's (WHO) Fungal Priority Pathogens List (FPPL) in part underscores the unmet need to respond to cryptococcal infections and antifungal resistance adequately. To this end, this contribution interrogates the available literature, highlighting emerging treatment strategies

Accurate forecasting of infectious diseases drives modern public health interventions that reduce morbidity and mortality. However, accurate forecasting in real-time remains a challenge for the modeling community. Ensembling has emerged as a critical tool for accurate forecasting by leveraging multiple component (individual) models into a single weighted average. Traditional ensembling strategies have relied on bespoke component models that weight the contributions of individual models according to extensive historical data for specific diseases. This is impractical for an emerging disease, si

The ever-present threat of new viral epidemics makes the scientific community relentlessly work on the development of universal methods of antiviral therapy. The development of broad-spectrum antivirals (BSAs) focuses either on substances acting directly on viral proteins (direct-acting antivirals [DAA]) or on substances directed at the cell's own proteins (host-targeting antivirals [HTA]). Decades of development have led to the market entry of a number of DAAs with a wide range of antiviral activities; however, their clinical approval has been obtained for individual infections. HTAs have a n

Poliovirus (PV), a historically significant enterovirus responsible for severe paralytic diseases, has seen its incidence dramatically reduced through widespread vaccination efforts, propelling global eradication initiatives. Despite the success of traditional oral poliovirus vaccines (OPVs) and inactivated poliovirus vaccines (IPVs), challenges such as vaccine-derived virus reversion and biosafety concerns during vaccine production persist. Virus-like particle (VLP) vaccines, which mimic native viral structures without containing viral genomes, offer enhanced safety profiles and robust immuno

Background/Objectives: Sabin-strain inactivated poliovirus vaccine has been increasingly incorporated into routine immunization programs as part of the global strategy to eradicate poliomyelitis. As childhood immunization schedules become more complex, concerns persist regarding the safety of concomitant vaccination. Although randomized controlled trials and regional surveillance studies have demonstrated acceptable safety profiles, additional population-based real-world evidence remains valuable for evaluating the safety of sIPV administered concomitantly with other vaccines under routine pro

Hemorrhagic fever with renal syndrome (HFRS) is the most common zoonotic disease in Russia, with about a 98% abundance of Puumala virus in all HFRS cases. We report clinical manifestations and genomic characteristics of the Puumala virus strain that caused an unconventional course of HFRS with sudden death. The patient was admitted to the hospital on the third day from fever onset with hyperthermia, cough, shortness of breath, and severe weakness, and died 28 h after hospitalization despite intensive care. Further analyses of autopsy samples led to Puumala virus detection. The viral genome was

SARS-CoV-2, which causes COVID-19, continues to circulate around the world, making it necessary to study the impact of rapidly emerging mutations on escape from neutralizing antibodies and pathogenesis. While RBD mutations are well characterized, mutations in the N-terminal domain (NTD) of the spike protein remain comparatively understudied despite their relevance to antibody recognition. This study investigates two phenotypically distinct SARS-CoV-2 mutants, which exhibited differences in plaque morphology on Vero cells. Whole-genome sequencing via Illumina identified a novel 12-nucleotide in

Guillain-Barre' syndrome (GBS) is an acute onset disease of the peripheral nerves with a variable presentation and severity. It is the most common cause of acute flaccid paralysis worldwide after the eradication of poliomyelitis. Regarding the diagnosis of GBS, cerebrospinal fluid analysis and electrophysiological study plays a pivotal role. The objective of the study was to find out an association between cerebrospinal fluid (CSF) glucose, fasting plasma glucose (FPG) and hemoglobin A1C (HbA1C) with clinical severity in patients with GBS. This cross-sectional observational study conducted in

BACKGROUND Vaccination is one of the most effective tools available to protect against infectious diseases. In Italy, Law no. 119/2017 expanded the list of mandatory vaccinations to 10: against poliomyelitis, diphtheria, tetanus, hepatitis B, pertussis, Haemophilus influenzae type b (hexavalent vaccine), measles-mumps-rubella (MMR), and varicella (chickenpox). The present study focuses exclusively on 3 of them: MMR, hexavalent, and varicella. The emergence of the COVID-19 pandemic led to a significant reorganization of the healthcare organization. The aim of this study was to assess the potent

Poliovirus continues to pose a global public health threat, with both wild and vaccine-derived viruses detected in clinical and environmental samples, including in countries declared to be polio free. Although wastewater and environmental surveillance increasingly reveal silent poliovirus circulation, how these findings are translated into public health action remains unclear. We reviewed the literature to identify documented events in which poliovirus detection in wastewater triggered public health responses across WHO regions. 26 unique events across 21 countries and all six WHO regions were

In this article, I review highlights of my academic background and scientific journey in pursuit of understanding all things related to poliovirus and the molecular mechanisms that lead to an amazingly successful intracellular replication cycle. Although it is not possible to describe all my lab's research findings during the past five decades, I attempt to cover some of our major accomplishments and how they have influenced the field of virology as well as other areas of science.

Vaccination remains one of the most effective public health interventions; however, recent global trends indicate a reversal in routine immunisation coverage alongside resurgent outbreaks of vaccine-preventable diseases. This review synthesises current evidence on declining immunisation coverage, the re-emergence of diseases such as measles and poliomyelitis, and the growing influence of vaccine hesitancy and misinformation on vaccine uptake. A systematic search of PubMed, Scopus, and WHO/UNICEF databases was conducted using terms including 'vaccine hesitancy', 'immunisation coverage', 'vaccin

BACKGROUND: Distal forearm fractures are common pediatric injuries. While Closed Reduction and Percutaneous Pinning (CRPP) is a standard treatment with excellent outcomes, subsequent poliomyelitis is a less commonly reported but serious complication. This case highlights the diagnostic and therapeutic challenges of managing Methicillin-resistant Staphylococcus aureus (MRSA) fracture-related infection (FRI), particularly in the presence of subtle initial signs. CASE PRESENTATION: A 9-year-old male underwent CRPP for a right distal forearm fracture (Fig. 1). Intraoperative radiographs confirmed

UNLABELLED: The Central African Republic (CAR) is a high-risk country for poliomyelitis outbreaks because of difficulties in closing immunity gaps due to the lack of resources, logistics issues, and insecurity. The country experienced multiple poliomyelitis cases since 2019. A novel type 2 oral polio vaccine (nOPV2) engineered to lower the risk of reversion of the vaccine strain was deployed in the CAR in 2022-2023 to counter active serotype 2 poliovirus transmission. This study reports routine poliovirus surveillance conducted from November 2021 through October 2023 in the CAR. Polioviruses i

Human papillomavirus (HPV) can cause cervical cancer. Global viral eradication relies on specific criteria, including a single host species and effective vaccines, a feat successfully achieved with smallpox and rinderpest. Although measles is also a candidate for elimination, its progress has been hindered by vaccine hesitancy based on misinformation about vaccine safety. Similarly, HPV is an ideal candidate for eradication due to its strict human infectivity and the proven vaccine efficacy in reducing cancer rates and establishing herd immunity. We highlighted the growing global consensus on

Motor neuron diseases (MNDs) encompass a clinically heterogeneous group of neurodegenerative conditions with varying impact on dexterity, mobility, decision making, respiratory and bulbar dysfunction. While consensus best-practice recommendations exist for genetic screening, diagnostic work-up, pharmacological and respiratory management, disease-specific facets of driving safety, assessment approaches and intervention strategies to support patients for safe driving have not been comprehensively reviewed. MNDs have unique, phenotype-specific clinical features, which are distinct form other neur
