Design of Nucleoside Prodrugs for the Inhibition of Tick-Borne Encephalitis Virus Reproduction.

The design of prodrugs for nucleoside antiviral agents is the primary approach to achieving target profiles not only in terms of safety but also efficacy. We have proposed a new chemotype of inhibitors of tick-borne encephalitis virus (TBEV) reproduction among N6-substituted adenosine derivatives, characterized by the presence of a hydrophobic aromatic substituent at the N6 position of adenine and benzoic acid residues attached via ester bonds to the ribofuranose scaffold. The compounds inhibited TBEV reproduction at micromolar concentrations only in their benzoylated form, while exhibiting no

SAR exploration for 4-aminotetrahydroquinazolines with adamantyl moiety against orthoflaviviruses.

4-Aminotetrahydroquinazoline N-oxides form a prominent chemotype of orthoflavivirus reproduction inhibitors, attractive for the search of lead compounds for development as direct-action antiviral agents. Here we explored the relationship between the structure and antiviral activity within an extended series of adamantyl-containing 4-aminotetrahydroquinazolines, synthesized via SNAr reactions of 4-chlorotetrahydroquinazolines with adamantyl-containing amines. Most of the obtained compounds inhibited the reproduction of tick-borne encephalitis, yellow fever, and West Nile viruses in phenotypic a

Performance comparison of rapid and native barcoding methods for Oxford Nanopore sequencing of Poliovirus Viral Protein 1 (VP1) amplicons.

Accurate and timely sequencing of poliovirus is critical for global eradication efforts, particularly for molecular epidemiology based on the typing region of the genome, viral protein 1 (VP1). While Oxford Nanopore Technologies (ONT) sequencing has expanded capabilities for poliovirus surveillance, the relative performance of different ONT library preparation methods, including ligation-based (Native Barcoding) and transposase-based (Rapid Barcoding) approaches, has not been systematically evaluated. In this study, we compared rapid barcoding and native barcoding workflows for sequencing VP1

Response to a vaccine-derived poliovirus type 1 event - Sichuan, China, 2024.

BACKGROUND: In September 2024, a 4-month-old infant in Liangshan, Sichuan, China was diagnosed with paralytic poliomyelitis that turned out to have been caused by a vaccine-derived poliovirus type 1 (VDPV1). This was the third VDPV outbreak detected by routine acute flaccid paralysis (AFP) surveillance in Sichuan since 2012. We conducted an epidemiological investigation into the event and implemented response measures to prevent further viral spread. METHODS: The investigation and response were conducted according to World Health Organization (WHO) and China's National Health Commission stand

French guidelines for post-polio syndrome and management of effects of aging in people with sequelae of acute anterior poliomyelitis.

People with sequelae of acute anterior poliomyelitis (AAP) may develop secondary complications related to aging, as well as a specific complication of the disease known as post-polio syndrome (PPS). These complications are new neurological and musculoskeletal manifestations that occur after a prolonged period of disease stability. The onset is typically insidious and slow, but can be more rapid following a period of immobilization, surgery, or another intercurrent condition. The initial assessment may involve consulting a neurologist to rule out neurological diagnoses. Electroneuromyographic e

[Research progress in combination vaccines containing sabin strain inactivated poliovirus vaccine].

Sabin strain inactivated poliovirus vaccine (sIPV), derived from attenuated Sabin poliovirus strains, has emerged as a promising alternative to both oral poliovirus vaccine (OPV) and conventional inactivated poliovirus vaccine (cIPV). It eliminates the risk of vaccine-associated paralytic poliomyelitis (VAPP) and the emergence of vaccine-derived polioviruses (VDPVs). In addition, sIPV demonstrates superior biosafety and vaccine safety during production and administration compared with cIPV. The integration of sIPV into combination vaccines with other routine immunization antigens can reduce th

Inactivation of Wild Poliovirus with β-Propiolactone.

OBJECTIVES: Antiviral vaccines are usually created by inactivating viruses using physical or chemical methods. Inactivation of poliovirus with β-propiolactone (BPL) has advantages, including the absence of a requirement for long-term incubation with the inactivating agent, which minimizes the risk of negative effects of the potentially dangerous substance on the virus and reduces the duration of the inactivation process. METHODS: BPL was applied at 0.2% (w/v) concentration under two conditions: 4 °C for 24 h and 37 °C for 3 h. Inactivation completeness was confirmed on Vero cell culture, whil

'When you hear such rumours, you get scared': A comparison of vaccine-related concerns between the oral polio vaccine and Covid-19 vaccines among communities in Cameroon and Ethiopia.

Despite widespread acceptance of the oral polio vaccine (OPV), vaccine hesitancy towards OPV in some countries presents a barrier to global eradication efforts. Between 2021 and 2022, the Global Polio Eradication Initiative implemented OPV campaigns concurrently with the roll-out of Covid-19 vaccines, for which vaccine hesitancy is well-documented. We sought to understand stakeholder perceptions of OPV and Covid-19 vaccines in Cameroon and Ethiopia. We implemented 32 focus group discussions (16 per country) with n = 216 caregivers of children under 5 and polio frontline workers to understand r

Toward the prevention of clinically manifest Parkinson's disease.

Parkinson's disease (PD) is the fastest growing neurological disorder worldwide and is projected to affect unprecedented numbers of individuals by 2050. At the same time, PD research is undergoing a profound conceptual transformation. Advances in molecular neuropathology, genetics, biomarker science, artificial intelligence, multimodal imaging, digital medicine, and precision therapeutics are reshaping PD from a clinically defined syndrome into a biologically measurable and potentially preventable process. This Perspective is not a report of new empirical findings and should not be read as an

Strategies and lessons learned from a type 1 circulating vaccine-derived poliovirus (cVDPV) outbreak affecting adult population: Madagascar, 2020-2023.

INTRODUCTION: From 2020 to 2023, Madagascar experienced an outbreak of circulating vaccine-derived poliovirus type 1 (cVDPV1) that notably affected adults and was associated with high mortality among paralytic cases. This atypical epidemiological pattern required adapted surveillance approaches and tailored outbreak response strategies, including vaccination campaigns targeting adult populations. METHODS: We conducted a descriptive analysis of the cVDPV1 outbreak in Madagascar between September 2020 and September 2023. Epidemiological data on confirmed and polio-compatible cases were reviewed

Case Report: Prolonged nOPV2 shedding in an immunocompetent child from Tunisia border with Algeria, North African region.

Following the detection of vaccine-derived poliovirus type 2 (cVDPV2) in Algeria in 2022, enhanced surveillance activities along the Tunisian border identified novel Oral Poliovirus type 2 Vaccine (nOPV2) excretion in an apparently healthy nomadic child. Immunological investigations showed no evidence of major humoral, cellular, or phagocytic immune deficiency. To investigate the kinetics of viral shedding and characterize the complete genome of the isolated strain in this case report, longitudinal follow-up stool samples were collected and analyzed using cell culture inoculation, real-time RT

Investigating the weak antibody response to vaccination with inactivated polio virus serotype 3 when delivered by microneedle patch.

Eradication of poliomyelitis requires vaccination coverage approaching 100%, which means vaccinating children at locations with limited healthcare resources. Microneedle (MN) patches can simplify vaccination by enabling administration by personnel with limited training and reducing reliance on refrigerated distribution. Here, we report that IPV serotype 3 (IPV3) was much less immunogenic when administered by MN patch compared to intramuscular (IM) vaccination, generating a neutralizing antibody titer similar to a 90%-lower dose given IM. To study this effect further, we found that microneedle

Viral infection and dementia risk: A systematic bidirectional Mendelian randomization analysis.

BACKGROUND: Viral infections have attracted increasing interest as potentially alterable risk factors for dementia; nonetheless, the results from observational research are variable and prone to confounding bias and reverse causation. This work seeks to systematically explore the potential causal associations between several viral infections and various forms of dementia by a Mendelian randomization methodology utilizing summary statistics and extensive genomic data from the Finnish FinnGen R12 database. METHODS: This study initially included 14 viral infections, including anogenital herpes,
