[Colchicine in Cardiology Practice: Use in Atrial Fibrillation, Inflammatory Diseases, Heart Failure, and Cardiac Complications of COVID-19].

Inflammation is an integral part of the pathophysiological processes leading to damage or regeneration of the heart and blood vessels. Interest to the "inflammatory theory" of cardiovascular disease is once again at the peak of scientific research, driven by the discovery of new laboratory and instrumental methods, as well as the emergence of new cardiotropic viruses, including SARS-CoV-2. Colchicine, the most effective and safe drug used to modulate excessive inflammation in heart disease, is included in guidelines for the treatment of perimyocarditis and ischemic heart disease with a high cl

Cardiac lesions in sheep and goats with spontaneous Clostridium perfringens type D enterotoxemia.

A 12-year retrospective search identified 106 sheep and goats diagnosed with Clostridium perfringens type D enterotoxemia, and in which the heart was histologically examined. Twenty cases (20/106, 19%), including 10 sheep and 10 goats, had cardiac lesions that were presumed to be associated with enterotoxemia. The lesions included myocardial degeneration and/or necrosis (n = 16, 80%), hemorrhage (n = 17, 85%), and proteinaceous interstitial edema (n = 6, 30%). Myocardial degeneration and/or necrosis was more frequent in goats (10/10, 100%) compared with sheep (6/10, 60%). Hemorrhage was more f

ACUTE MYOCARDIAL DEGENERATION AND DEATH IN AYE-AYES (DAUBENTONIA MADAGASCARIENSIS) FOLLOWING INGESTION OF AVOCADO (PERSEA AMERICANA).

Five of a cohort of 13 aye-ayes (Daubentonia madagascariensis) at the Duke Lemur Center (DLC) exhibited acute, nonspecific signs of lethargy and ataxia within 24 h of ingesting a meal containing avocado fruit (Persea americana) that included the skin and pit. Deaths rapidly ensued in four of the five animals within 13 h of initial symptoms. All animals that died had pericardial effusion on postmortem examination and microscopic findings of acute myocardial degeneration. After ruling out other causes, all evidence points to avocado toxicity as the cause of death. Following an analysis of the DL

Montelukast attenuates diclofenac sodium-induced cardiotoxicity in male rats via targeting Wnt/β-catenin pathway.

Diclofenac sodium (DCFN) is the most often prescribed NSAID, however its prolonged administration is linked to cardiotoxicity mediated by oxidative stress, inflammation, and apoptosis. The role of the Wnt/β-catenin pathway in cardiac injury is well documented; however, it remains unclear whether this pathway is upregulated in the heart during DCFN treatment. This work aims to investigate the cardiac activity of this pathway in DCFN-intoxicated rats and to explore the potential cardioprotective effects of montelukast (MNTL), a leukotriene receptor antagonist. Four groups of male rats were treat

Protective effects of vitamin E and quercetin against MDMA-induced cardiac and hematological dysfunction in male wistar rats.

3,4-Methylenedioxymethamphetamine (MDMA), a synthetic psychoactive amphetamine derivative widely abused as the recreational drug “ecstasy,” is associated with oxidative stress–mediated cardiotoxicity and hematological disturbances. Despite documented antioxidant properties of quercetin and vitamin E individually, limited evidence exists regarding their combined protective efficacy against MDMA-induced cardiovascular dysfunction. This study evaluated the combined and complementary effects of quercetin and vitamin E on cardiac, hematological, lipid, and electrocardiographic alterations induced b

Eugenol attenuates paclitaxel-induced cardiotoxicity by modulating autophagy-related markers in rats.

OBJECTIVES: Paclitaxel (PTX) is a commonly used chemotherapeutic agent that causes cardiotoxicity characterized by oxidative stress, inflammation, and mitochondrial dysfunction, which disrupts autophagy and apoptosis in cardiomyocytes. This study investigated the therapeutic potential of eugenol (EUG), a natural anti-oxidant and anti-inflammatory compound, against PTX-induced cardiac damage. MATERIALS AND METHODS: Thirty-six male Wistar rats were randomly assigned to six groups: Control, EUG5, EUG25, PTX, PTX+EUG5, and PTX+EUG25. Hemodynamic parameters (systolic and diastolic blood pressure a

Circulating cardiac troponin T is an early biomarker of cardiomyocyte injury in Myh6-Cre mice.

Cardiomyocyte-specific Cre driver lines such as Myh6-Cre are widely used in cardiovascular research, but Cre recombinase itself can induce cardiac toxicity independently of target gene deletion. Early biomarkers capable of identifying this injury before overt cardiac remodeling or dysfunction remain poorly defined. Here, we performed a longitudinal analysis of Myh6-Cre mice using circulating biomarkers, cardiomyocyte Ca²⁺ transient measurements, histology, echocardiography, organ pathology, and survival analysis. Circulating cardiac troponin T (cTnT) levels were unchanged at 7 and 8 weeks of a

DDX3X overexpression in mice can cause rapid tissue-specific toxicity and mortality.

DEAD-Box Helicase 3 X-Linked (DDX3X) is a ubiquitously expressed RNA helicase with diverse cellular roles implicated in a neurodevelopmental disorder called DDX3X syndrome. Although DDX3X is a leading genetic cause of intellectual disability in females, there is no treatment. While gene supplementation is a plausible therapeutic strategy, previous studies suggest DDX3X is carefully regulated and dose sensitive. To understand the consequences of overexpressing DDX3X with unregulated adeno-associated virus-mediated gene supplementation, we generated a vector driving strong ubiquitous DDX3X expre

Myocardial degeneration precedes fibrosis: The role of myocardial autophagy and its pathological evaluation.

Dilated cardiomyopathy (DCM) is characterized by progressive ventricular dilation and systolic dysfunction, yet the cellular damage that precedes intercellular fibrosis remains incompletely defined. Electron microscopy has long revealed autophagic vacuoles and myofilament lysis in cardiomyopathy, but their pathological significance has been debated for decades. Recent advances in ultrastructural analysis, including LC3 immunoelectron microscopy, have enabled more precise characterization of these changes and have renewed interest in the role of autophagy in cardiomyopathy. In this review, we i

Selenium deficiency-induced cardiomyopathy: a case report.

BACKGROUND: Selenium is an essential trace element involved in redox homeostasis, thyroid function, and immune regulation. Deficiency is uncommon in developed countries but occurs in patients with malabsorption, long-term parenteral nutrition, or chronic kidney disease on dialysis. Selenium deficiency is a rare but treatable cause of non-ischaemic cardiomyopathy and arrhythmias, which can be fatal if untreated. Due to its rarity, it is often unrecognized, its pathophysiology remains unclear, and the threshold for irreversible myocardial damage is not well defined. CASE SUMMARY: We present the

EPAS1 induction drives myocardial degeneration in desmoplakin-cardiomyopathy.

Arrhythmogenic cardiomyopathy (ACM) is frequently attributed to desmosomal mutations, such as those in the desmoplakin (DSP) gene. Patients with DSP-cardiomyopathy are predisposed to myocardial degeneration and arrhythmias. Despite advancements, the underlying molecular mechanisms remain incompletely understood, thus limiting therapeutic options. Here, we employed spatial transcriptomics on an explanted heart from a patient with a pathogenic DSP variant. Our transcriptional analysis revealed endothelial PAS domain-containing protein 1 (EPAS1) as a potential regulator of mitochondrial homeostas

Obtusifolin mitigates acute lung injury and improves cardio-pulmonary functions in acute lung injury model by modulating oxidative stress and NF-κB signaling.

Pulmonary-cardiac inflammation is a serious health condition affecting all age groups caused by either injury or infection. Acute or chronic lung/cardiac disorders cause several disturbances in their functions and architecture, leading to fatality by causing multi-organ failure. Obtusifolin (OBT) is a traditional phytomolecule known for its anti-inflammatory effects in arthritis models. This study aims to investigate the therapeutic effects of OBT against lipopolysaccharide (LPS)-induced lung and cardiac inflammation/injury using both in vitro and in vivo models.
