Exercise intensity-dependent cardiac telocyte expansion is associated with physiological growth despite JAK/STAT pathway inactivity in male Wistar rats.

While exercise induces physiological cardiac growth, the underlying cellular mechanisms remain incompletely understood. This study investigated the role of cardiac telocytes (TCs) and the Janus kinase (JAK)/signal transducer and activator of transcription (STAT) pathway in mediating exercise intensity-dependent cardiac adaptation. Twenty-four male Wistar rats were assigned to control (CTRL), high-intensity interval training (HIIT) or low-intensity interval training (LIIT) groups for 8 weeks. Physiological hypertrophy was assessed via heart weight/body weight ratio, left ventricular wall thickn

Exploring the Role and Mechanism of DSF in HFpEF Based on IL-1β/IL-1βR/TAK1/RIPK1 Axis-Mediated PANoptosis.

Heart failure with preserved ejection fraction (HFpEF) has a high prevalence and a low quality of life, and there are limited medications for the treatment of this disease. In recent years, disulfiram (DSF), an FDA-approved drug for the treatment of chronic alcohol addiction, has been found to have anti-inflammatory properties. The present study was designed to investigate the cardioprotective effects of DSF on patients with HFpEF and its mechanism using a model of HFpEF induced in mice fed a high-fat diet (HFD, 60% of calories from fat) and Nω-nitro-L-arginine methyl ester (L-NAME, 0.5 g/L in

Modulation of Ferroptosis by the Irx3-Etfa Pathway Protects Against Cardiac Hypertrophy.

Pathological cardiac hypertrophy is a major contributor to heart failure and is often accompanied by ferroptosis and mitochondrial dysfunction. However, the upstream transcriptional mechanisms governing these processes remain poorly defined. We performed integrative bioinformatics analysis using transverse aortic constriction (TAC)-induced hypertrophic heart datasets to identify mitochondria-related differentially expressed genes (MitoDEGs), followed by transcription factor prediction and experimental validation in both in vivo and in vitro models. Adeno-associated virus-mediated overexpressio

Hexokinase 2-Driven Cardioprotection in Ischemia, Hypertrophy, and Regeneration: From Underlying Mechanisms to Clinical Translation.

The heart is the most energy-demanding organ in the body and has a dynamically adaptable metabolic state. Under normal conditions, the adult heart primarily relies on mitochondrial oxidative phosphorylation for ATP generation, whereas glycolysis supplies a minor portion of the energy. However, under pathological conditions, glycolysis is increased, and mitochondrial oxidative phosphorylation is reduced. Hexokinase 2 (HK2) is the first rate-limiting enzyme in the glycolytic pathway in cardiomyocytes. Accumulating evidence in recent years has indicated that HK2 confers protection to the heart un

Limitations in Chest X-Ray Interpretation by Vision-Capable Large Language Models, Gemini 1.0, Gemini 1.5 Pro, GPT-4 Turbo, and GPT-4o.

Background/Objectives: Interpretation of chest X-rays (CXRs) requires accurate identification of lesion presence, diagnosis, location, size, and number to be considered complete. However, the effectiveness of large language models with vision capabilities (LLMs) in performing these tasks remains uncertain. This study aimed to evaluate the image-only interpretation performance of LLMs in the absence of clinical information. Methods: A total of 247 CXRs covering 13 diagnostic categories, including pulmonary edema, cardiomegaly, lobar pneumonia, and other conditions, were evaluated using Gemini 1

Amentoflavone, a Preferentially TGF-β Receptor 1 Inhibitor, Alleviates Cardiac Remodeling and Dysfunction Through Modulating the TGF-β/Smad and MAPK Signaling Pathways.

Chronic cardiac stress induces cardiac hypertrophy and fibrosis, thereby promoting pathological heart remodeling, which is the core pathological process of heart failure. Amentoflavone (AMF) has beneficial therapeutic effects against several cardiovascular diseases; however, its role and underlying mechanisms in pathological cardiac remodeling remain unclear. Here, we investigated the protective role of AMF against pathological cardiac remodeling in transverse aortic constriction (TAC)-induced overloaded hearts. Mice with pressure overload caused by TAC were treated with AMF, and the effects o

Cardiomyocyte-derived OTUD7B promotes cardiac hypertrophy by deubiquitinating SERCA2a.

Rationale: Pathological cardiac hypertrophy, triggered by persistent neurohumoral or hemodynamic stress, is a key precursor of ventricular dysfunction and heart failure. Deubiquitinating enzymes (DUBs) have emerged as critical regulators of cardiovascular biology. This study examined the function of a DUB, ovarian tumor domain-containing 7B (OTUD7B), in cardiac hypertrophy. Methods: Cardiomyocyte-specific OTUD7B knockout and overexpression mouse models were generated to evaluate myocardial hypertrophy and cardiac dysfunction in response to angiotensin II (Ang II) infusion or transverse aortic

Sodium-myo-inositol cotransporter-1, SMIT1, promotes cardiac hypertrophy and fibrosis induced by pressure overload in mice.

AIMS: Recent clinical studies have reported that myo-inositol is consistently elevated in plasma of patients with heart failure (HF), yet its role in cardiac dysfunction remains poorly understood. Myo-inositol is specifically transported into cells by the sodium-myo-inositol co-transporter-1 (SMIT1), a member of the sodium-glucose co-transporter (SGLT) family expressed in the heart. While myo-inositol is essential for phosphoinositide signalling, osmoregulation, and metabolic homeostasis, dysregulation of SMIT1-mediated myo-inositol transport may contribute to key pathological mechanisms in HF

T-cadherin, a major adiponectin binding partner, suppresses ERK signaling in metabolic tissues.

T-cadherin, which is a major adiponectin binding partner, exerts various organ-protective effects. However, the specific changes in intracellular signaling that are induced by T-cadherin in metabolic tissues/cells remain unclear. We demonstrated that T-cadherin suppresses ERK signaling in both cultured cells and murine tissues. T-cadherin knockdown increased ERK phosphorylation in C2C12 myocytes and F2 endothelial cells, whereas T-cadherin overexpression suppressed ERK phosphorylation. Proteomic analysis revealed that many proteins that are downstream targets of ERK signaling were upregulated

Structural Diversity of 2-O-(β-d-Glucopyranosyl)benzyl Quinates from Casearia tardieuae and Their Anti-Myocardial Hypertrophy Activity in Rat Cardiomyocytes.

Thirteen undescribed 2-O-(β-d-glucopyranosyl)benzyl quinates (1-13) and two reported derivatives (14, 15) were obtained from the twigs and leaves of Casearia tardieuae. The structures of the new compounds were elucidated by comprehensive spectroscopic analysis (1D/2D NMR, HRESIMS, IR, UV), coupled with acid hydrolysis and derivatization studies of the sugar moieties. Biological evaluation showed that compound 1 exhibited antihypertrophic potency comparable to the positive control prazosin, and its cytotoxicity was significantly lower than that of prazosin. Preliminary mechanism investigations

An automated vertebral heart scale measurement tool based on deep learning: Facilitating screening for prevention of canine cardiomegaly.

Automatically measuring the Vertebral Heart Scale (VHS) from canine thoracic X-ray images is an effective approach for the early screening and prevention of cardiomegaly. Establishing automated and precise measurement tools is crucial for advancing population-based research on canine cardiac health. In this study, an integrated framework based on an improved U-Net architecture (SWA-UNet) and YOLOv11 is proposed for precise heart segmentation and key point detection to enable automated and accurate VHS calculation. It provides an efficient, objective, and repeatable auxiliary diagnostic tool fo

Oxytocin ameliorates cardiac hypertrophy by inhibiting mitochondrial dysfunction and pyroptosis via AMPK/PGC-1α /TFAM pathway.

AIM: Oxytocin (OT) is increasingly recognized as a cardiovascular homeostatic regulator with anti-remodeling potential; however, the mitochondrial and innate immune mechanisms underlying its anti-hypertrophic action remain incompletely defined. We therefore investigated whether OT protects against pathological cardiac hypertrophy by preserving mitochondrial homeostasis and suppressing mitochondria-derived inflammatory signaling. MATERIALS AND METHODS: An isoproterenol (ISO)-induced rat model and ISO-stimulated H9c2 cardiomyocytes were used. Cardiac remodeling was assessed by echocardiography,
