Comparison of Deep Learning Architectures for Cardiac Contour Segmentation in Catheterization Radiographs.

Introduction Accurate cardiac image segmentation is essential for quantitative assessment of cardiac anatomy and function. Manual segmentation, while considered the reference standard, is time-consuming and subject to inter- and intra-observer variability. Deep learning (DL) models, particularly convolutional neural networks (CNNs), have shown promise for automating this process. Among these, U-Net and DeepLabV3 are widely used architectures; however, direct comparisons between them for cardiac silhouette segmentation on cardiac catheterization radiographs are limited. Methods A supervised DL

Case-control study of mitragynine deaths in Florida.

Kratom use has increased in the USA, with its primary alkaloid, mitragynine, detected in postmortem toxicology, and implicated in reported fatalities. This study evaluated toxicologic concentrations and autopsy findings associated with deaths attributed to mitragynine. Autopsy and postmortem toxicology reports were obtained from the majority of Florida medical examiner districts over 5 years for decedents in whom mitragynine was listed as a cause of death (mitragynine-induced fatalities). A comparison group included cases from a single county in which mitragynine was detected but not deemed ca

Integrated spatial transcriptomics and single-cell RNA sequencing reveal Lars2-mediated spatiotemporal dynamics of myocardial remodeling in a mouse model of transverse aortic constriction.

INTRODUCTION: Pressure overload-induced myocardial hypertrophy is associated with complex spatial and temporal remodeling of cardiac cell populations. However, the spatial organization of these changes and their dynamic transcriptional programs remain incompletely understood. Integrating spatial transcriptomics with single-cell RNA sequencing enables a comprehensive characterization of cardiac remodeling at high cellular and spatial resolution. METHODS: Heart tissues were obtained from transverse aortic constriction (TAC) mouse models at different stages of disease progression (TAC-2w, TAC-4w

Cardiomyocyte Cyclin-dependent kinase 9 directly binds to and phosphorylates NF-κB p65 subunit to drive cardiac inflammation and remodeling.

Hypertensive heart failure highlights an urgent need for effective therapeutic strategies. Protein kinases regulate multiple pathways in cardiac pathophysiology and may provide promising therapeutic targets. Here, we identified a Cyclin-dependent kinase, CDK9, promoting inflammation and cardiac remodeling in terminally differentiated cardiomyocytes. Firstly, kinase enrichment analysis and experimental evidence revealed CDK9 phosphorylation at Thr-186 in both human and mouse hypertrophic heart tissues. CDK9 loss of function via T186A mutation in cardiomyocytes attenuated Ang II-induced heart re

Non-telomeric function deficiency of TERT enhances pressure overload-induced mouse cardiac remodeling by activation of CNBP-mediated THBS3/ITGB1 pathway.

Recent studies show that telomerase reverse transcriptase (TERT) possesses important new biological functions in gene transcription regulation, signal transduction, tumorigenesis, vascular development and mitochondrial DNA protection independent of the maintenance of telomere length. In this study we investigated the role and mechanisms of TERT in regulating the gene expression and signal transduction during pressure overload-induced cardiac remodeling. The first-generation TERT knockout (Tert-/-) and wild-type littermate control (Tert+/+) male mice were subjected to transverse aortic constric

Angiotensin-(1-7) Alleviates Isoproterenol-Induced Cardiac Hypertrophy by Suppressing Autophagy and Apoptosis Through the Synergistic Action of Mas Receptor and Angiotensin II Type 2 Receptor.

AIM: The aim of this study is to determine whether Angiotensin-(1-7) [Ang-(1-7)] alleviates isoproterenol (ISO)-induced cardiac hypertrophy by suppressing excessive autophagy and apoptosis through coordinated Mas receptor (MasR) and angiotensin II type-2 receptor (AT2R) signaling, and to elucidate the underlying mechanisms. METHODS: ISO-induced hypertrophy was established in mice and assessed by echocardiography, histology, and hypertrophic markers. H9c2 cardiomyocytes were exposed to ISO and treated separately with A-779 (MasR antagonist), PD123319 (AT2R antagonist), and a combination of bot

COX-2-derived prostaglandins in cardiac hypertrophy: Dual roles in injury and repair.

Cardiac hypertrophy represents a primary adaptive response of the heart to both mechanical stress and neurohormonal stimuli. However, beneficial hypertrophic adaptation eventually gives rise to maladaptive structural changes in the heart, ultimately leading to heart failure. Cyclooxygenase-2 (COX-2), a stress-inducible enzyme participating in the metabolism of arachidonic acid-derived eicosanoids, has been recognized as a central modulator of the adaptive changes of the hypertrophic heart through the production of bioactive prostaglandins. However, a growing number of studies have demonstrated

CYLD-mediated DNA damage coordinates pathological cardiac hypertrophy via RIPK1-dependent signaling.

Cardiac remodeling under pressure overload involves maladaptive changes that lead to heart failure. This study investigated the role of the deubiquitinating enzyme CYLD in this process. We found that CYLD expression was significantly upregulated in hypertrophic mouse myocardium and cardiomyocytes. Silencing CYLD markedly attenuated pathological remodeling, improved cardiac function, and reduced DNA damage. Mechanistically, CYLD drives apoptosis by deubiquitinating RIPK1, a key signaling kinase. Inhibition of RIPK1 synergized with CYLD silencing to alleviate hypertrophy-associated DNA damage. O

Strip1 Is a Novel Negative Regulator of Cardiomyocyte Hypertrophy.

Pathological cardiac hypertrophy is a critical factor leading to cardiomyopathy and ultimately heart failure. While several signaling pathways controlling cardiac hypertrophy have been identified, the molecular mechanisms underlying their precise regulation remain incompletely understood. Strip1, a structural component of STRIPAK complexes, has been implicated in various cellular functions; however, its role in cardiomyocytes is uncharacterized. Here we identify Strip1 as a potent anti-hypertrophic factor, controlling cell size and the hypertrophic gene program in neonatal rat ventricular card
