PTPRS mediates invasion of tetanus neurotoxin into the central nervous system.

Tetanus neurotoxin (TeNT) invades the central nervous system (CNS) via retrograde transport through motoneurons. Whether a host factor is needed to mediate TeNT trafficking has remained unknown. Here, using pull-down and mass spectrometry analysis, and by developing a luciferase-based in vivo transport assay, we identify receptor protein tyrosine phosphatase sigma (PTPRS; PTPσ) as the retrograde transport receptor for TeNT. Global or motoneuron-specific PTPRS knockout (KO) abolishes TeNT entry into the CNS in mouse models. PTPRS-TeNT interactions occur in endosomes, prevent toxin translocation

Pneumothorax Following Botulinum Neurotoxin Injection for Trapezius Hypertrophy.

Botulinum neurotoxin (BoNT) injection into the trapezius muscle is increasingly performed for aesthetic contouring. However, iatrogenic pneumothorax has not been previously documented for this cosmetic indication, despite the inherent anatomical risks. We report a 26-year-old woman who developed immediate severe radiating pain during bilateral trapezius BoNT type A (BoNT-A) injections using a 4.8-cm, 25-gauge needle. During the injection, the patient experienced immediate severe radiating pain, followed by pleuritic chest pain and dyspnea within 30 minutes. Computed tomography confirmed a 30%

Development of novel human monoclonal antibodies for botulinum neurotoxin type F subtype 1.

Different types of Botulinum neurotoxins (BoNTs) cause human botulism. Among them, type F botulism, caused by genetically diverse BoNT/F subtypes, is rare and may be overlooked despite its clinical importance. Toxin-neutralizing human antibodies, which carry the minimum risks of inducing serum sickness or anaphylaxis, are ideal for the treatment of human botulism. In this study, we generated human monoclonal antibodies (mAb) against BoNT/F using trans-chromosomic monoclonal antibody (TC-mAb) mice. A recombinant toxin fragment consisting of the light chain and the heavy chain N-terminal domain

Toward an Aptamer-Based Biosensor for Cross-Serotype Detection of Botulinum Neurotoxin: Binding Mechanism from a Comprehensive Computational Investigation.

Botulism is a potentially fatal illness in humans and many animals caused by the botulinum neurotoxins (BoNTs) produced mainly by Clostridium botulinum, which can be found in a wide variety of foods. The treatment of this illness requires antitoxin drugs given to the patients within a 48 h window to be effective. Therefore, there is a strong need for the development of rapid and sensitive biosensors for the early detection of the toxins in foods. In this work, by using comprehensive atomistic computer modeling and simulation, we investigate aptamers that bind BoNT serotypes A, B, and C to serv

Diagnostic stewardship as the heart of clinical microbiology: a position paper of the French Society for Microbiology (SFM).

Caring about the clinical impact of laboratory results, beyond their technical performance, has been part of the clinical microbiologist's mission since the very origins of the discipline. The term diagnostic stewardship is more recent, emerging from a renewed need created by powerful new diagnostic technologies: as testing becomes faster, more sensitive and more widely used, results grow harder to interpret and easier to misuse. A result reported without interpretation is not neutral, and diagnostic misuse readily translates into inappropriate antibiotic use, making the laboratory an unrecogn

Novel use of far UV-C light as a surface disinfectant in the poultry industry: trials on broiler farms show reduced bacterial burden in poultry anterooms and on wellington boots.

Osteoarthritis (OA) is condition which poses a main concern to the aging population and its severity is expected to increase with the increasing life expectancy. In the future, several possible targets for OA treatment need to be defined. Dickkopf‑related protein 3 (DKK3) is an atypical member of the Wnt‑antagonistic dickkopf‑related protein (DKK) family. The availability of research into the role of DKK3 in the abnormal remodeling of subchondral bone in human knee joints is currently limited. Thus, the aim of the present study was the evaluation of DKK3 expression in the abnormal bone remodel

Challenges for the effectiveness of infectious disease interventions during global change.

BACKGROUND: Sequential treatment of ankylosing spondylitis (AS) that includes tumour necrosis factor-α antagonists (anti-TNF agents) has been applied in most of the Western countries. Existing cost-effectiveness (CE) models almost exclusively presented the incremental CE of anti-TNF agents using a closed cohort while budget impact studies are mainly lacking. Notwithstanding, information on impact on total population health and societal budget as well as on actual incremental CE for a given decision time span are important for decision makers. This study aimed at quantifying, for different deci

Intestinal inflammation frequently occurs alongside dysmotility, which is characterized by altered myosin light chain phosphorylation levels. Curcumin, an active component from the ginger family, is reported to confer anti‑inflammatory effects. However, the effects of curcumin on both diarrhea and constipation associated inflammation remains to be elucidated. The present study was designed to investigate the effects of curcumin on diarrhea and constipation and to determine the related mechanisms. Sprague‑Dawley rats were used to establish diarrhea and constipation models via intracolonic aceti

The many journeys of botulinum neurotoxins and the bacteria that produce them-evolutionary, geographic, and research-related movements that have contributed to our understanding of a diverse pathogen.

SUMMARYBotulism is a neuroparalytic intoxication caused by a collection of large proteins, known as botulinum neurotoxins (BoNTs), that are related in amino acid sequence and structure. The extreme potency of BoNTs can be traced to their ability to access and enter cholinergic nerve terminals, their enzymatic nature, and their persistence within these cells. The extreme diversity seen among the BoNTs (7 serotypes and 44 subtypes) and the bacteria that produce them (7 species) stands in stark contrast to its close relative, tetanus toxin, which exists as a single protein entity produced by a si

Outbreak of equine botulism type C associated with consumption of baleage in Brazil.

An outbreak of botulism occurred in March 2024 among horses at a Quarter Horse stud farm in Central-West Brazil. After ingesting baleage, 22 of 26 (85%) horses housed in stables and fed baleage became ill. The affected horses had dysphagia, muscular weakness, fasciculations, and progressive recumbency; 13 of 22 (59%) died within a few days. The diagnosis of type C botulism was established based on clinical and epidemiologic findings and confirmed by mouse bioassays, which indicated botulinum toxin type C in liver samples and intestinal contents. Furthermore, PCR testing identified toxigenic Cl

Repetitive nerve stimulation findings in iatrogenic botulism compared with myasthenia gravis.

OBJECTIVE: The widespread use of botulinum toxin has been accompanied by an increase in complications, including iatrogenic botulism (IB). This study investigated repetitive nerve stimulation (RNS) findings in IB and myasthenia gravis (MG) patients. METHODS: Clinical and neurophysiological data were retrospectively reviewed in IB patients negative for both acetylcholine receptor and muscle-specific kinase antibodies, and in MG patients who were positive for either. RNS was performed on the ulnar nerve at 2, 3, 5, and 50 Hz, and on the facial and spinal accessory nerves at 2, 3, and 5 Hz. RES

Therapeutic potential of [(8-hydroxyquinolin-7-yl)(phenyl)methylamino] benzoic acid regioisomers against human-intoxicating botulinum neurotoxin serotypes: computational modeling to in vivo protection.

Botulinum neurotoxins are class I tier bioterrorism agent, accountable for causing rare but fatal illness ‘botulism’. Out of seven serotypes, A, B, E, and F are responsible for intoxicating humans. Despite knowing harmful effects on human health for centuries, there is no commercial antidote for post-neuronal intoxication is available. In the present study, we report efficacy of regioisomers of [(8-hydroxyquinolin-7-yl)(phenyl) methylamino]benzoic acid against zinc-dependent light chain activities of BoNT/A, /B, /E & /F by combining molecular modeling with in vitro and in vivo studies. Advances in Clostridial Neurotoxins: Passage of the Intestinal Barrier and Targeting of Specific Neuronal Cells.

Clostridial neurotoxins, botulinum neurotoxins (BoNTs), and tetanus neurotoxin (TeNT) are potent toxins responsible for severe diseases, botulism and tetanus, respectively. BoNTs associate with non-toxic proteins (non-toxic non-hemagglutinin, hemagglutinins, and OrfXs), which protect BoNTs against acidic pH and protease degradation and facilitate BoNT passage through the intestinal barrier. TeNT enters motor neurons and undergoes a retrograde axonal transport until the target inhibitory interneurons in the central nervous system. BoNTs and TeNT recognize specific cell surface receptors which c

Presynaptic Terminal Proteins and Nicotinic Receptors Are Depleted from Mouse Parasympathetic Ganglionic Junctions Paralysed with Botulinum Neurotoxin Type A.

Plasticity is fundamental to the development, strengthening, and maintenance of healthy synaptic connections and recovery from injury in both the central and peripheral nervous systems. Yet, the processes involved are poorly understood. Herein, using a combination of patch-clamp electrophysiology and immuno-fluorescence confocal microscopy in adult mice, it is shown that blockade of synaptic transmission at submandibular ganglion junctions exposed to botulinum neurotoxin type A was accompanied by a rapid and striking decline in the abundance of synaptic vesicle markers-SV2, vesicle-associated
