Polio Survivors' Experiences of Corrective Surgery and Rehabilitation in 1950s-1960s Finland.

This study aimed to explore polio survivors' childhood experiences of corrective surgery and rehabilitation in post-war Finland during the 1950s and 1960s. Oral history research was used with a descriptive qualitative approach. Between September 29, 2018 and June 30, 2019, 40 participants were interviewed, while one wrote about their experiences (N = 41). The data were analyzed using reflexive thematic analysis, which produced the overarching theme 'The Ongoing Process of Recovery: Body, Mind, and Self, 'describing recovery as a holistic process shaped by bodily changes, emotional responses, a

A dual-receptor checkpoint primes enterovirus D68 for respiratory cell entry.

MFSD6 is a newly identified receptor that mediates the invasion of respiratory cells by enterovirus D68 (EV-D68), a non-polio enterovirus that causes severe respiratory disease and poliomyelitis-like illness in children. Here, we report near-atomic-resolution cryo-electron microscopy (cryo-EM) structures of historical and contemporary AFM-associated EV-D68 strains, together with their complexes bound to the third extracellular loop of MFSD6 (MFSD6-L3). These structures uncover a previously unrecognized "binary gating switch" mechanism of virus-receptor engagement that differs from the reported

The Involvement of the Cryptococcal Yeast Kexin Protease in the Proteolytic Activation of the Uncleaved Influenza Virus Hemagglutinin Glycoprotein.

Infectious diseases typically involve a single pathogen harming its host. However, in clinical settings, co-infections also occur. This study investigated the potential interaction between Cryptococcus (C.) neoformans and influenza virus by assessing whether the cryptococcal yeast kexin protease (Kex2p) could activate the uncleaved hemagglutinin glycoprotein. Protein-protein docking was performed using HADDOCK and independently validated using AlphaFold3. 6x His-tagged truncated KEX2 gene was expressed in E. coli BL 21 (DE3), and the recombinant protein was purified using nickel ion affinity c

Beyond Hesitancy: Current Multi-Level Polio Vaccine Refusals and Pathways to Acceptance in a High-Risk Region of Pakistan.

BACKGROUND: Pakistan is one of only two countries where wild poliovirus type 1 (WPV1) continues to persist. Vaccine refusals challenge efforts to end the virus, especially in high-risk communities. METHODS: A qualitative exploratory study took place from September to November 2025 in three High-Risk Union Councils (HRUCs) of Karachi, Pakistan. The study examined reasons for refusing the polio vaccine and identified specific communication and operational strategies to improve vaccine acceptance. A total of 23 in-depth interviews and 10 focus group discussions were conducted with 71 participant

From Oncolysis to Adaptive Immunity: Yellow Fever Virus 17D and Ruxolitinib Activate Antitumor Immune Responses in Pancreatic Cancer Models.

Background: Pancreatic ductal adenocarcinoma (PDAC) remains one of the most lethal cancers, with a five-year survival rate of less than 12%. Oncolytic viruses are considered a promising immunotherapeutic approach, but their effectiveness is often limited by the innate interferon response, which is the main antiviral barrier in resistant tumors. A combination of an oncolytic virus with JAK/STAT pathway inhibitors has been proposed to overcome this resistance. Methods: In this study, we investigated the oncolytic and immunotherapeutic potential of the attenuated yellow fever vaccine strain YFV 1

A Panel of VHH Antibodies Against Sabin Type 1 Poliovirus D-Antigen Reveals Strain-Specific and Cross-Serotype Reactivity.

Vaccination remains the primary strategy for poliomyelitis prevention. The D-antigen of poliovirus is a critical component of inactivated polio vaccines, as it induces the production of neutralizing antibodies and provides protective immunity. Therefore, the development of quantitative immunoassays for monitoring D-antigen content during vaccine production is an important task. In this study, we generated recombinant camelid single-domain antibodies (VHHs) specific for the D-antigen of Sabin poliovirus type 1 and evaluated their antigen specificity. The obtained VHH antibodies demonstrated str

A multi-center, open-labelled, randomized controlled extended phase III non-inferiority clinical trial to evaluate the immunogenicity and tolerability of poliomyelitis vaccine (Vero cells), inactivated, Sabin strains administered with or without routine infant vaccines.

BACKGROUND: Oral poliovirus vaccine (OPV) has been reported to cause vaccine-derived poliovirus and vaccine-associated paralytic poliomyelitis, and the limited global supply of conventional IPV has led many countries to rely on fractional-dose IPV regimens alongside OPV. The current study aims to assess the sIPV safety and immunogenicity when given concurrently with or in a staggered manner with routine immunization. METHODS: A multi-country, multi-center, open-label, randomized controlled, extended phase III non-inferiority clinical trial was conducted with 1442 healthy infants aged 6-8 week

Improved polio outbreak prediction using a Caputo fractional-order model with memory effects.

This paper presents a fractional-order mathematical model of the transmission dynamics of poliomyelitis based on the Caputo fractional derivative to address the memory effects present in biological systems. The model sub-divides human population into susceptible, exposed, infected and vaccinated groups and reflects essential processes of the disease progression and control. A strict theoretical examination is conducted, with the calculation of the basic reproduction number [Formula: see text], using next-generation matrix method. The local and global stability of both disease-free and endemic

Virucidal Efficacy of Holder Pasteurization of Breast Milk in a Portable Device.

Breast milk is the optimal nutrition for infants. However, a number of viruses can accumulate in breast milk and be transmitted to a newborn during breastfeeding. Holder pasteurization is a gold-standard method for breast milk treatment, achieving an acceptable compromise between microbiological safety and biological quality. We have evaluated the virucidal activity of Holder pasteurization of breast milk contaminated with RNA and DNA viruses in a portable home-use pasteurizer. Viruses were selected on basis of their ability to infect humans, contaminate breast milk, and pose a threat for a ne

XRN1-Resistant Elements Are Located in the 3'-Terminal Regions of Certain Genome Segments of Jingmenviruses.

Jingmenviruses have been identified over the past decade. During this period, significant advancements have been made in understanding their diversity, biology, and genomic functionality. Conventionally characterized orthoflaviviruses are distinguished by a specific class of subgenomic RNAs (sfRNAs) that contain exoribonuclease-resistant RNA (xrRNA) structures. These sfRNAs are the products of incomplete 5'-3' degradation of genomic RNA by the host exoribonuclease XRN1. Resistance to complete hydrolysis is mediated by highly conserved secondary structural motifs localized within the 3' untrans

M184V: The Major Determinant of Cooperative ISL Resistance When in Combination with Multiple TAM2 Mutations in HIV-1 Subtype C.

Islatravir is a novel nucleoside reverse transcriptase translocation inhibitor recently approved for the treatment of HIV-1 infection in virologically suppressed PLWH. While we previously documented high-level phenotypic resistance to islatravir (ISL) in HIV-1 subtype C variants harboring combinations of type 2 thymidine analogue mutations (TAM2) along with the M184V resistance-associated mutation, the primary contributor to the observed phenotype remains unclear. This report dissects the relative contribution of M184V to ISL resistance in the context of TAM2-containing variants through system

Case Report: Infant with vaccine-associated paralytic poliomyelitis unveils global disparities in care for inborn errors of immunity.

Severe combined immunodeficiency (SCID) is a life-threatening inborn error of immunity (IEI) that is potentially curable when diagnosed early but is associated with high morbidity and mortality when recognition is delayed. In settings without universal newborn screening (NBS), and where vaccination programs rely heavily on live vaccines, affected infants are at risk of preventable complications. We report an Indonesian girl, the first child of non-consanguineous parents, who at the age of three months, developed a generalized maculopapular rash, followed by febrile encephalopathy with acute fl

Prediction of recent HIV-1 infections using Shannon entropy analysis of HIV-1 group-specific antigen protein sequence.

BACKGROUND: Avidity assays often misclassify chronic HIV-1 infection as recent HIV-1 infection (false recency rate), especially in participants on antiretroviral therapy. The aim of this study was to use Shannon entropy to evaluate HIV-1 group-specific antigen (Gag) sequence diversity for the prediction of recent HIV-1 infections. METHODS: This was a retrospective study that characterised the complete HIV-1 Gag using Sanger sequences obtained from participants with confirmed recent or chronic HIV-1 infection. Shannon entropy was calculated for the entire HIV-1 Gag amino acid (aa) sequence (50
