Abdominal ultrasound mitigates neurodegenerative pathology and alters specific gut microbial taxa in senescence-accelerated mice.

Age-related changes in gut microbiota composition have been implicated in brain aging and cognitive decline, acting through the gut-brain axis on amyloid accumulation, tau pathology, synapse damage, and glial activation. Live microbial approaches such as probiotics, prebiotics, and fecal microbiota transplantation address this mechanism, but they still suffer from low efficacy and unknown safety issues. We tested whether abdominal ultrasound, an approach that does not involve microbes, can alleviate age-related neurodegeneration in the senescence-accelerated mouse strain SAMP8. We treated 6-mo

Atrial fibrillation in stroke patients: facts, numbers and current challenges.

Atrial fibrillation (AF) is a leading, modifiable risk factor for ischaemic stroke, with 20-30% of stroke patients harbouring underlying AF. Management of AF, however, differs substantially depending on clinical context. In ischaemic stroke patients with pre-known AF, direct oral anticoagulants (DOACs) are the cornerstone of secondary prevention, with initiation timing guided by infarct severity following a risk-adapted approach. In patients with no history of AF, prolonged cardiac rhythm monitoring after stroke or transient ischaemic attack improves AF detection rates. However, it remains unc

Alzheimer's disease risk gene Wwox controls amyloid pathology through metabolic reprogramming.

Genome-wide association studies have identified multiple loci that contribute to late-onset Alzheimer's disease risk, yet the functional mechanisms of many candidate genes remain unclear. WW-domain containing oxidoreductase (WWOX) is one such gene associated with disease and downregulated in patient brains, but its role in neurodegeneration is poorly understood. Here, we use Drosophila models to investigate the cell-type-specific and mechanistic roles of Wwox in ageing and amyloid-β42 (Aβ42)-induced pathology. Neuronal and glial knockdown of Wwox reduced lifespan and increased sleep drive, ind

Paratonia is inversely correlated to premotor areas perfusion.

OBJECTIVE: Paratonia is defined as an inability to relax the muscles during tone assessment, resulting in participants either opposing or facilitating the examiner's movement involuntarily. This phenomenon is frequently found in patients with cognitive impairment and has been attributed to frontal lobe dysfunction, although a direct correlation has not been established. This study aimed to explore the correlation between frontal lobe function, as assessed by functional brain imaging, and paratonia, as assessed by electromyography. METHODS: Memory clinic outpatients with suspected Alzheimer's

Amyloid PET Changes During Lecanemab Treatment in Patients With and Without Amyloid-related Imaging Abnormalities.

Lecanemab reduces cerebral amyloid-β burden in Alzheimer disease, but whether amyloid-related imaging abnormalities (ARIA) are associated with greater amyloid clearance remains unclear. We report 3 patients with comparable clinical backgrounds and similar baseline cognitive performance who underwent amyloid-β PET before and after lecanemab treatment. Quantitative analysis using Centiloid showed a greater reduction in amyloid burden in the patient who developed ARIA than in the 2 patients without ARIA. These observations suggest ARIA may be associated with more pronounced treatment-related amyl

NMR characterization of Tacrine in aqueous solution and insights into its interaction with amyloid-β1-40 aggregates revealed by STD.

The deposition of β-amyloid peptide (Aβ) aggregates in the brain is widely accepted as one of the major factors driving the development of Alzheimer's disease (AD). Tacrine (9-amino-1,2,3,4-tetrahydroacridine) is one of the most potent acetylcholinesterase inhibitors approved for the symptomatic treatment of AD, but its clinical use was discontinued due to hepatotoxicity. Recent studies have reported that tacrine can interact with Aβ and inhibit its aggregation, although the underlying mechanism is not yet fully elucidated, highlighting the need for detailed spectroscopic characterization unde

Micromotor-enabled platform for early, minimally invasive monitoring of amyloid pathology along a putative plasma-brain-retina axis in a transgenic islet amyloid polypeptide mouse model.

Growing evidence indicates that metabolic dysfunction contributes to neurodegeneration, leading to the "type 3 diabetes" hypothesis. In this context, human islet amyloid polypeptide (hIAPP), a highly amyloidogenic peptide, has emerged as a potential molecular link between peripheral metabolic dysfunction and neurodegeneration. Here, we developed a hybrid ex vivo platform integrating micromotor (MMGO-AuNPs)-based electrochemical aptassay for the detection of circulating amyloid-β42 oligomers (AβO42) in human IAPP-overexpressing transgenic (Tg) and wild-type (Wt) mice, under high-fat diet (HFD).

Serratol, a cembrane diterpene from Boswellia serrata attenuates α-synuclein and fibrillar amyloid-β aggregation in Caenorhabditis elegans.

ETHNOPHARMACOLOGICAL RELEVANCE: Boswellia serrata oleo-gum resin, known as Kundur in Unani medicine, has traditional applications in neurological and cognitive disorders, including Majoon-e-Nisyan and Dawa-ul-Kibreet for Nisyan (amnesia) and Muqawwi-e-Asab (nerve-strengthening) respectively. Serratol is a cembrane diterpene constituent of B. serrata resin, while related frankincense cembranes exhibit neuroactive properties. However, the contribution of serratol to the neurological activities of B. serrata remains poorly explored. AIM OF THE STUDY: To isolate serratol from B. serrata resin, ev

Plasma p-tau181, Aβ1-42, and GFAP across Alzheimer's disease, mixed-type Alzheimer's disease, mild cognitive impairment, anxiety disorders, and depressive disorders.

BACKGROUND: Blood-based biomarkers including phosphorylated tau181 (p-tau181), amyloid β1-42 (Aβ1-42), and glial fibrillary acidic protein (GFAP) have been widely investigated for Alzheimer's disease (AD) detection. However, their utility for discriminating neurodegenerative cognitive impairment from anxiety and depressive disorders remains poorly established. AIMS: To compare plasma p-tau181, Aβ1-42, and GFAP concentrations across clinically-defined patient groups with AD, mixed-type Alzheimer's disease (MAD), mild cognitive impairment (MCI), anxiety disorders (ANX), and depressive disorders

Gut-brain axis in Alzheimer's disease: neuroinflammatory mechanisms and immunopharmacological interventions.

Despite being defined pathologically by amyloid-β plaques, chronic neuroinflammation and neurofibrillary tangles, the drugs that target these processes directly (lecanemab and donanemab) have only modest effects on the clinical course of Alzheimer's disease (AD) and significant safety concerns have been identified, so there is renewed interest in upstream, systemic contributors to disease. This review aims to provide a synthesis of the latest mechanistic and clinical evidence that gut microbiota dysbiosis is associated with Alzheimer's disease with a specifically immunopharmacological focus. The use of three murine genetic models of Alzheimer's disease reveals efficacy of 4',5,7-trihydroxyisoflavone in alleviating cognitive deficits and reducing levels of amyloid-beta and hyperphosphorylated tau protein.

BACKGROUND: Despite intensive studies on developing therapies for Alzheimer's disease (AD), there is still a need to improve the treatment efficacy. Results of recent studies indicated that genistein (4',5,7-trihydroxyisoflavone) was effective in an autophagy stimulation-dependent improving of cognitive deficits and reducing levels of amyloid-beta and hyperphosphorylated tau protein in brains, when employing a streptozotocin-induced rat model of the sporadic form of AD. Despite those encouraging results, the above-mentioned model has its limitations, mainly related to the rapid and inducible c

The "Glyco-Remodeling" Hypothesis in Alzheimer's Disease: Linking Protein N-Glycosylation to Pathogenesis and Translation.

Alzheimer's disease (AD), an age-associated neurodegenerative disorder characterized by cognitive decline and accumulation of amyloid-β (Aβ) plaques and tau tangles, has long been studied through the lens of protein aggregation and phosphorylation. However, emerging evidence suggests that aberrant protein N-glycosylation, a key post-translational modification governing protein folding, trafficking, and signaling, plays an equally pivotal role in AD pathogenesis. This review consolidates current knowledge on N-glycosylation, and very briefly about the prominent O-Glycosylation dynamics in the h

Metal Ions and Protein Aggregation: Unraveling Molecular Interactions and Theranostic Prospects in Proteinopathies.

Protein aggregation is a pathological process associated with several human diseases termed proteinopathies, including neurodegenerative and metabolic disorders such as Alzheimer's disease, Parkinson's disease, type II diabetes, amyotrophic lateral sclerosis, and Huntington's disease. In these conditions, specific proteins misfold and accumulate into insoluble aggregates, disrupting normal cellular function and contributing to disease progression. Many studies have investigated the molecular mechanisms that underlie the aggregation pathways, the structure of aggregates, such as oligomeric inte

Transient focal neurological episodes in cerebral amyloid angiopathy-related convexity subarachnoid hemorrhage.

BACKGROUND: Convexity subarachnoid hemorrhage (cSAH) is recognized as a cause of transient focal neurological episodes. These episodes can closely mimic transient ischemic attacks (TIAs). We report a case of cerebral amyloid angiopathy (CAA)-related cSAH presenting with recurrent transient monoparesis. CASE DESCRIPTION: An elderly patient with a history of hypertension was admitted to our hospital because of recurrent episodes of transient left upper limb monoparesis. Initial diffusion-weighted magnetic resonance imaging (DWI) demonstrated a focal hyperintensity over the right frontal convexi

APOE ε4 accelerates plasma p-tau217 trajectory: a Bayesian joint longitudinal analysis of ADNI.

BACKGROUND AND OBJECTIVE: Plasma GFAP and p-tau217 are leading blood biomarkers for Alzheimer's disease (AD), but whether APOE ε4 modulates their longitudinal rates of change - directly or via amyloid burden - is unresolved. We tested APOE ε4 modulation of plasma GFAP and p-tau217 trajectories and their coupling with cognitive/structural decline in ADNI participants. METHODS: A total of 325 ADNI participants aged ≥ 65 (CN 181/MCI 106/AD 38) with ≥ 2 plasma GFAP and ≥ 2 p-tau217 (UPENN-Fujirebio) measurements over ≤ 10 years were analyzed. Linear mixed-effects models tested APOE ε4 × time inte

Metabolic Dyshomeostasis, neuroinflammation, and glymphatic clearance deficits as convergent drivers of Alzheimer's disease: an exposome-based conceptual framework for prevention.

The repeated failures of anti-amyloid therapies in Alzheimer's disease (AD) have exposed critical limitations of the linear amyloid cascade hypothesis and underscored the need for a broader etiological framework. The objective of this review is to synthesise mechanistic evidence for an exposome‑based model of sporadic AD that integrates metabolic, inflammatory, and sleep/circadian contributions to brain aging. Epidemiological evidence indicates that up to 40% of AD risk is attributable to modifiable environmental and lifestyle factors. These factors, collectively termed the aging exposome, rep

Purine metabolism disruption links pathology and cognitive decline in Alzheimer's disease.

Purine metabolism dysregulation has been implicated in Alzheimer's disease (AD), but its temporal dynamics and mechanistic links to core pathologies remain unclear. This translational study integrated a longitudinal preclinical AD mouse model and clinical cohort investigations to define the role of purine metabolism in AD progression. In APPswe/PSEN1dE9 (APP/PS1) mice, serum-untargeted metabolomics identified purine metabolism as the most notably altered pathway, with metabolite levels (e.g., adenine, adenosine) shifting dynamically across disease stages alongside increasing amyloid-β (Aβ) bur

Cannabidiol Regulates Amyloid-β Plaque Deposition and Astrogliosis in the 5xFAD Transgenic Mouse Model.

Alzheimer's Disease (AD) is marked by the accumulation of amyloid-beta (Aβ) plaques, neurofibrillary tangles, and neuroinflammation. Cannabidiol (CBD), the major non-psychoactive compound of Cannabis, has demonstrated significant pharmacological potential for the treatment of AD. CBD is known to act on various receptors, including cannabinoid receptor 1 (CB1R) and 2 (CB2R), and peroxisome proliferator activated receptor γ (PPARγ). However, the precise changes in receptor expression in AD, as well as following CBD treatment, remain unclear and inconsistent in literature. To address this, we per
