Neuroligin-3-CSPG4 interaction maintains oligodendrocyte precursor cell progenitor state and promotes glioma proliferation through mechanotransduction.

Glioma pathophysiology is robustly regulated by interactions with neurons. Key to these interactions is the role of neuroligin-3 (NLGN3), a synaptic adhesion molecule shed in response to neuronal activity that functions as a paracrine factor crucial for glioma growth. Here we elucidate the mechanistic pathway whereby shed NLGN3 interacts with glioma and their normal glial counterparts. NLGN3 binds to chondroitin sulfate proteoglycan 4 (CSPG4, also known as NG2) on both glioma and healthy oligodendrocyte precursor cells (OPCs), facilitating CSPG4 shedding by ADAM10. NLGN3-CSPG4 interactions alt

APOE ε4 genotype-dependent reduction of amyloid-related imaging abnormality in real-world lecanemab use: Implications for risk-stratified prescribing.

Real-world amyloid-related imaging abnormality (ARIA) rates with lecanemab are consistently lower than in the Clarity AD trial, yet whether this reduction is genotype-specific is unknown. We compared published APOE ε4-stratified ARIA-E and ARIA-H rates from Clarity AD with a US real-world cohort and Japanese post-marketing surveillance. ARIA rates were largely unchanged in APOE ε4 noncarriers, whereas reductions were concentrated among carriers, especially homozygotes. This pattern was observed for both ARIA subtypes across both settings. These early real-world findings suggest that the appare

Model-based ordering of the ALPS index among Alzheimer's disease biomarkers.

Glymphatic dysfunction has been implicated in Alzheimer's disease (AD), but its position relative to amyloid and tau biomarkers remains unclear. We examined the relative position of the diffusion tensor image analysis along the perivascular space (ALPS) index within the AD biomarker cascade using Alzheimer's Disease Neuroimaging Initiative data. Amyloid PET, tau PET, plasma amyloid-β 42/40 ratio, plasma phosphorylated-tau 217, and ALPS index were analyzed with a data-driven event-based model. The ALPS index was ranked between plasma amyloid-β 42/40 alteration and amyloid PET positivity in the

Oral administration of melatonin modulates macrophage polarization via modulating the Vegf signaling pathway and ameliorates Alzheimer's disease in APP/PS1 mice.

BACKGROUND: Alzheimer's disease (AD) is a neurodegenerative disorder with a global prevalence, currently lacking effective treatments and posing a major public health challenge due to the burden it places on healthcare systems while affecting millions of people. Melatonin is a hormone with considerable potential for treating various neurodegenerative disorders, including AD. The mechanisms responsible for melatonin's therapeutic benefits in Alzheimer's disease (AD) require further elucidation. This study was designed to investigate the mechanisms by which melatonin exerts its effects in APP/PS

Cardiovascular risk factors and regional neuropathology in autopsy-confirmed Alzheimer disease.

Cardiovascular risk factors are implicated in Alzheimer disease (AD) progression and its neuropathological hallmarks but their specific contributions to regional brain pathology within AD remain understudied. In this cohort study (n = 276), we examined associations between diabetes, hypertension, and hypercholesterolemia (present vs absent), and neuropathologies in decedents with pathologically confirmed Intermediate/High AD from 3 Alzheimer's Disease Research Centers. Regional arteriolosclerosis, cerebral amyloid angiopathy (CAA), cored plaques (CPs), diffuse plaques (DPs), neurofibrillary ta

Presenilin 1 Mutations Converge on Aβ42/Aβ40 Elevation via Distinct Mechanisms.

Mutations in Presenilin 1 (PS1) are the most common genetic cause of familial Alzheimer's disease (AD). However, the exact mechanisms by which these mutations alter amyloid precursor protein (APP) processing and drive amyloid β (Aβ) production remain unclear. In this study, five PS1 mutations (M146V, G384A, C410Y, Y115H, and the Notch-sparing mutation ΔS169) were transfected into neuroblastoma cells lacking endogenous PS1/PS2 expression and human embryonic kidney cells stably overexpressing either wild-type APP or the APP Swedish mutation. We investigated the effects of these mutations on APP

What can narcolepsy teach us about hypocretin and dementia?

Sleep and circadian disturbances are associated with cognitive decline and dementia. Narcolepsy type 1 (NT1), characterized by severe sleep-wake disruption and loss of hypocretin, would therefore be expected to predispose individuals to premature brain aging. Yet available biomarker and neuroimaging findings do not consistently support this possibility, raising questions about the role of hypocretin in mechanisms linking sleep to neurodegeneration. Beyond maintaining wakefulness, hypocretin interacts with inflammatory circuits, metabolic regulation, synaptic plasticity, amyloid processing, and

Lateralized amyloid PET uptake and beta-band connectivity in relation to cognition in temporal lobe epilepsy.

We examined focus-relative amyloid PET asymmetry and its associations with resting-state EEG connectivity and processing speed in mesial temporal lobe epilepsy (MTLE). Thirty adults with clinically assigned MTLE (19 right-focus, 11 left-focus) underwent 18F-florapronol PET and resting-state EEG. Regional standardized uptake value ratios (SUVRs) and sensor-level weighted phase lag index (wPLI) were evaluated using paired comparisons, rank correlations and exploratory bootstrap indirect-association models. Cognitive data were available for 26 participants, including Digit Symbol Coding (DSC) for

miR-124 delivery to the hippocampus reduces excitability, enhances neurogenesis and affects microglia in a model of Alzheimer's disease.

MicroRNA-124 (miR-124) has been proposed as therapeutic candidate for Alzheimer's disease (AD), given its regulation of gene networks governing synaptic function, neurogenesis and inflammation. However, its functional role during early disease stages, and the cellular context-dependence of its effects, remain unclear. Here, we investigated the effects of targeted neuronal miR-124 delivery to the hippocampal dentate gyrus (DG) of AppNL-G-F mice. At an earlier 'presymptomatic' stage, miR-124 reduced intrinsic excitability of mature granule neurons and induced subtle changes in dendritic architec

Plasma proteomic signatures in bipolar disorder and their implications for data-driven patient stratification.

Bipolar disorder (BD) is a heterogeneous mood disorder that lacks robust biomarkers for biological stratification. We used high-throughput plasma proteomics to characterize case-control differences, identify proteomic subgroups within BD, and examine their associations with blood-based markers and clinical features. Plasma samples from the French I-GIVE (Immuno-Genetics, Inflammation, retroVirus, Environment) cohort, including 248 participants with BD and 76 healthy controls, were assessed using the Olink Reveal panel (1,034 proteins). Age-, sex-, and BMI-adjusted case-control analyses identif

[Carpal Tunnel Syndrome and Lumbar Spinal Stenosis: When to Suspect Amyloidosis and How to Proceed with the Diagnostic Work-up].

Transthyretin (TTR) amyloidosis, particularly the wild-type form (ATTRwt), is an age-related systemic disease characterized by the deposition of misfolded TTR in tendons and ligaments. Patients therefore frequently present initially to orthopaedic or hand surgeons with carpal tunnel syndrome (CTS) or lumbar spinal stenosis (LSS) rather than to cardiologists or neurologists. These musculoskeletal manifestations may precede ATTR cardiomyopathy (ATTR-CM) by several years to more than a decade, serving as "red flags" that provide an opportunity for early diagnosis, particularly now that disease-mo

Brain DNA methylation, APOE genotype and Alzheimer's disease neuropathology in a large postmortem cohort.

DNA methylation (DNAm) plays a critical role in Alzheimer's disease (AD). Previous epigenome-wide association studies using 450K arrays captured limited CpG coverage and often focused on a single neuropathological measure. Here we profiled DNAm in 1,250 postmortem prefrontal cortex samples from the Rush Alzheimer's Disease cohorts using the Infinium MethylationEPIC v2 array (~930,000 CpGs) and performed epigenome-wide association studies of multiple neuropathologic measures, including AD diagnosis, amyloid-β and tau tangles. We identified 107 differentially methylated probes associated with AD

Unraveling Shared Molecular Mechanisms in Alzheimer's, Parkinson's, and Neuropathic Pain: Targeting p38 MAPK and Wnt/β-Catenin Pathways.

Neurodegenerative disorders, including Alzheimer's Disease (AD), Parkinson's Disease (PD), and Neuropathic Pain (NP), share overlapping molecular and cellular mechanisms that contribute to progressive neuronal dysfunction, neuroinflammation, and impaired neural repair. Although these conditions present with distinct clinical manifestations, accumulating evidence indicates that dysregulation of the p38 mitogen-activated protein kinase (p38 MAPK) and Wnt/β-catenin signaling pathways represents a common pathogenic axis underlying disease initiation and progression. Activation of p38 MAPK amplifie

Intermittent Fasting as a Therapeutic Strategy in Alzheimer's Disease: Understanding of Brain-Derived Neurotrophic Factor Modulation.

Alzheimer's disease (AD) is a progressive neurodegenerative disease with amyloid-beta (Aβ) plaques, tau tangles, neuroinflammation, and oxidative stress, and it causes cognitive impairment and neuronal loss. The pharmacological treatment is of limited effectiveness, therefore the interest in novel approaches such as intermittent fasting (IF). IF, a nutritional regimen of periodic caloric restriction, has shown a potential to modulate neurodegenerative alterations by metabolic and molecular mechanisms. This review presents the implications of IF in the etiopathogenesis of AD, specifically in co

Association between early-life farming exposure and Alzheimer's disease neuropathological burden.

BACKGROUND: Farming populations may face a disproportionate share of adverse exposures influencing Alzheimer's disease (AD), yet farming data are rarely connected to cohorts with characterized AD neuropathology. Whether early-life exposure is related to AD neuropathology is limited. METHOD: Brain donors from two Alzheimer's Disease Research Centers (N = 1035; between 1986-2018) were included. US Census records were used to identify those with early-life farming exposure (birth to 17 years old) and examined alongside AD neuropathology assessments. RESULT: Early-life farming exposure (≈ 17% of
