Effects of the biocide cybutryne (irgarol) on freshwater invertebrates.

Cybutryne (irgarol) is a triazine biocide formerly used in antifouling paints worldwide and still detected in freshwater ecosystems in Europe, Asia, and North America despite its ban in antifouling paints in Europe in January 2017 and in the USA in April 2021. While primary producers are considered the most sensitive targets due to cybutryne's mode of action as a photosystem II inhibitor, data on freshwater invertebrates-particularly from standardized chronic tests-remain scarce. In this study, we investigated acute and chronic effects of cybutryne on three freshwater invertebrates representin

Not just 'Asian' but Japanese, Korean, Filipino, Chinese, and mixed Asian American: Examining depression in an ethnically heterogenous sample of Asian American young adults.

To date, most research focused on depression prevalence is with adolescents or adults and focuses mainly on risk factors, with few studies assessing factors related to depression for Asian American young adults. Using longitudinal data collected from web-based surveys administered over a 5-year period (wave 10: 2017-2018; wave 14: 2021-2022; and wave 15:2022-2023) on 460 young adults (22-28 years old at wave 15) who identify as Japanese, Korean, Filipino, Chinese, and mixed Asian American, we examined associations between risk and protective factors, such as discrimination, prior depression, s

Neuropsychiatric symptoms in sporadic Creutzfeldt-Jakob disease.

Although neuropsychiatric symptoms are not part of diagnostic criteria for sporadic Creutzfeldt-Jakob disease (sCJD), a few retrospective studies and our clinical experience have suggested they are prominent and often occur early. The objective of our study was to assess prospectively the neuropsychiatric features in sCJD (and their impact on caregivers) and compare them with five other neurodegenerative diseases: Alzheimer's disease (AD), dementia with Lewy bodies (DLB), progressive supranuclear palsy, behavioural variant frontotemporal dementia (bvFTD) and semantic variant primary progressiv

A fixed brain seeded amplification assay to complement neuropathological prion disease diagnosis.

Prion diseases are rare neurodegenerative disorders that share misfolding of the normal cellular prion protein into disease-causing isoforms known as "prions" as the critical pathophysiological event. Definite diagnosis can only be achieved through neuropathological confirmation. The neuropathological features of prion disease are well described; however, some molecular subtypes are typified by characteristic neuropathological features that are subtle or absent. Prion seeding assays have excellent specificity and have considerably improved premortem diagnostic accuracy but they have reduced se

Alpha-synuclein seeding activity in postmortem tissues from patients with diffuse and isolated Lewy bodies.

We have evaluated the diagnostic potential of the seeding amplification assay (SAA) in detecting α-synuclein seeding activity in postmortem brain and cerebrospinal fluid (CSF) samples from patients with primary and co-pathology α-synucleinopathies. Moreover, we investigated potential SAA positivity in control samples which may suggest unrecognized co-pathology. A total of 15 brain and 14 CSF samples with definite dementia with Lewy bodies (DLB, n = 6), Alzheimer´s disease with amygdala Lewy body (AD/ALB, n = 3), and patients with concomitant Creutzfeldt-Jakob disease and Lewy body pathology (C

Cerebrospinal Fluid Biomarkers in Creutzfeldt-Jakob Disease: Diagnostic Value, Limitations, and Future Multi-Omics Strategies.

Creutzfeldt-Jakob disease (CJD) is a rare but devastating neurodegenerative disorder characterized by the pathological misfolding of the cellular prion protein (PrPC) into the pathogenic isoform-scrapie prion protein (PrPSc), ultimately leading to fatal outcomes. Cerebrospinal fluid (CSF) biomarkers play a pivotal role in early diagnosis, longitudinal monitoring, and prognostic assessment, thereby enhancing the clinical management of this challenging disease. This review summarizes the established CSF biomarkers, 14-3-3 protein, tau protein (total tau), phosphorylated tau isoforms, α-synuclein

Rapidly progressive sensorineural hearing loss due to sporadic Creutzfeldt-Jakob disease.

A patient developed several weeks of progressive bilateral sensorineural hearing loss, initially suspected to be an ischaemic stroke due to right parietal diffusion restriction on MRI. However, the persistence and progression of symptoms, including left upper extremity weakness, word deafness and cognitive decline, were inconsistent with a vascular event. Repeat imaging revealed expanding cortical ribboning without infarct evolution. Extensive workup ruled out stroke, autoimmune encephalitis, infection and seizure-related processes. Cerebrospinal fluid analysis demonstrated significantly eleva

Genetic Creutzfeldt-Jakob disease linked to the E200K mutation: a large cohort study.

Creutzfeldt-Jakob disease (CJD), the most common human prion disease, is an invariably fatal neurodegenerative disorder affecting 1.5 cases per million individuals per year. About 10-15% of the human prion diseases are caused by a pathogenic variant in the prion protein (PrP) gene (PRNP), and the most common genetic human prion disease is CJD (gCJD) linked to a glutamic acid to lysine substitution at codon 200 (E200K) of PRNP. The polymorphic codon 129 methionine (M)/valine (V) genotype has a strong effect on disease phenotype. In the present study, we retrospectively evaluated many features o

Antibodies in Creutzfeldt-Jakob disease: A systematic review of patient characteristics, diagnostics, and clinical implications.

BACKGROUND: Creutzfeldt-Jakob disease (CJD) is a rare, rapidly progressive prion neurodegenerative disorder causing fatal dementia. Recently, reports have identified various autoantibodies in confirmed CJD patients, raising questions about their significance. We performed a scoping review of published CJD cases with co-occurring antibodies, including antineuronal (e.g., NMDAR, LGI1, CASPR2, VGKC, GAD65, GlyR), anti-thyroid peroxidase, antinuclear, and infection-related antibodies, to characterize patient demographics, clinical and diagnostic features, and outcomes, and explore the pathophysiol

Strain Traits of Intracranially Administered L-Type Bovine Spongiform Encephalopathy Prions Are not Significantly Modified During Intraspecies Transmission in Cynomolgus Monkeys.

Among the three prion strains of bovine spongiform encephalopathy (BSE), classical BSE (C-BSE) prions are known causative agents of variant Creutzfeldt-Jakob disease. By contrast, human infections with L-type (L-) or H-type (H-) BSE prions have not been reported. Nonetheless, the zoonotic potential of L-BSE prions is supported by their successful primary transmission from cattle to cynomolgus macaque (Macaca fascicularis) monkeys via intracranial challenge. To assess whether the defining strain traits of L-BSE prions remain stable following secondary intraspecies transmission, we prepared brai

Advancing prion diagnostics: full-length human E200K RT-QuIC substrate facilitates prion detection in tear fluid and improves sensitivity in cerebrospinal fluid.

The real-time quaking-induced conversion (RT-QuIC) assay has revolutionized prion disease diagnosis by detecting amyloidogenic PrP conformers in different tissue types and body fluids. Recently, we achieved a breakthrough by detecting amyloidogenic PrP conformers in tear fluid (TF), a non-invasive biofluid, warranting further evaluation. We refined our RT-QuIC protocol to assess seeding conversion efficiency using various recombinant substrates, including full-length human (FL Hu), and mutant versions linked to genetic prion diseases, such as E200K and D178N, in both CSF and TF samples. Peripheral Signatures of Multidimensional Pathology in Symptomatic and Asymptomatic Creutzfeldt-Jakob Disease.

BACKGROUND: Plasma markers of neuronal injury in Creutzfeldt-Jakob disease (CJD) are established, but peripheral biomarkers reflecting glial activation, synaptic dysfunction, and vascular impairment remain less explored. We systematically assessed these markers in symptomatic CJD and asymptomatic PRNP mutation carriers to improve diagnosis and identify early pathophysiology. METHODS: This prospective cohort study recruited CJD, frontotemporal dementia (FTD), healthy controls (HCs), and preclinical familial CJD pedigrees. Sixteen plasma proteins representing neuronal injury, glial activation,

Genetic causes and modifiers of prion diseases.

Prion diseases are transmissible neurodegenerative diseases caused by misfolding of prion protein (PrP). All inherited prion diseases, including genetic Creutzfeldt-Jakob disease, Gerstmann-Sträussler-Scheinker disease, and fatal familial insomnia, are caused by pathogenic mutations in the prion protein gene (PRNP). The mutation partly dictates age at disease onset and clinical phenotype, presumably by its effects on the misfolding of PrP, but the clinical features can be highly variable even within the same pedigree. In sporadic prion diseases, genetic factors also play pivotal roles, as poly

Enhanced Sensitivity of a Modified Quaking-Induced Conversion Diagnostic Test for the Broad Detection of Sporadic and Inherited Prion Diseases: A Retrospective Study.

OBJECTIVE: Quaking-induced conversion (QuIC) tests, which detect prion-seeding activity in cerebrospinal fluid (CSF), have markedly advanced the antemortem diagnosis of prion diseases such as Creutzfeldt-Jakob disease (CJD). These tests provide high diagnostic accuracy and enable timely differentiation from other rapidly progressive neurodegenerative disorders. However, a key limitation of current QuIC tests are the reduced sensitivity in detecting inherited prion diseases and rare sporadic subtypes, including variably protease-sensitive prionopathy (VPSPr). To address this gap, we evaluated a

[MRI as a key for early diagnosis of Creutzfeldt-Jakob disease].

We present a case of sporadic Creutzfeldt-Jakob disease (sCJD) in a 73-year-old patient, in which initial suspicion arose from unusual magnetic resonance imaging (MRI) findings and was later confirmed by cerebrospinal fluid analysis. This case report highlights the role of MRI in the early diagnosis of sCJD, facilitating timely palliative care, and highlights the key clinical and imaging characteristics of this rare and fatal prion disease.

Rapid generation of prion disease models using AAV-delivered PrP variants in knockout mice.

The study of prion biology has traditionally relied on transgenic mouse models, which, while valuable, require significant time and resources to develop. Here, we present a rapid and flexible alternative using adeno-associated virus (AAV) vectors to express modified prion proteins in PrP-knockout (PrP-KO) mice. Through systematic evaluation of multiple AAV constructs, we optimized vector design by comparing different CNS-specific promoters and regulatory elements to generate prion disease models capable of faithfully propagating the inoculated prion strain. We identified an optimized AAV const

Sporadic Creutzfeldt-Jakob disease in Palestine with early ataxia and dysarthria: a case report.

Creutzfeldt–Jakob disease (CJD) is a rare and fatal neurodegenerative disorder caused by misfolded prion proteins. It progresses inexorably to death once clinical symptoms emerge. Early manifestations are often nonspecific and may mimic other neurological conditions, making diagnosis particularly challenging. We report the case of a 54-year-old Palestinian male who initially presented with dysarthria, vertical gaze nystagmus, dizziness, and gait ataxia while maintaining preserved cognition. Over the following month, he developed short-term memory loss, hallucinations, and disorientation. Exten

The L108I polymorphism in mouse prion protein drives spontaneous disease and enhances transmission of atypical and classical prion strains.

Prion diseases are fatal neurodegenerative disorders that can be idiopathic, associated with genetic mutations, or acquired by infection with misfolded prion protein. We developed two complementary transgenic mouse models to investigate how the L108I substitution in mouse prion protein (PrP) influences spontaneous prion formation and transmission characteristics. The transgenic mouse model overexpressing the variant at approximately three times wild-type (WT) PrP levels (TgMo(L108I)3x) consistently developed a spontaneous neurodegenerative disorder between 219 and 536 days of age with 100% pen
