Evaluation of the effectiveness of cerium nanoparticles as a potential adjuvant in veterinary rabies vaccine.

BACKGROUND AND AIM: Rabies remains a major public health and veterinary concern worldwide, underscoring the need for improved vaccine immunogenicity. Nanomaterials are being looked at for their role in shaping early immune reactions, along with boosting targeted responses to antigens. In this work, particles made of cerium were tested as possible helpers in a vet-approved dead rabies vaccine using mice. Using a method that formed cerium oxide nanoparticles, scientists checked how they looked and behaved. These tiny particles were tested to understand their makeup plus shape. MATERIALS AND MET

A curvilinear coordinate flatmap for visualizing hippocampal structure and development.

The hippocampal formation is a highly curved and topographically complex forebrain structure. This complex geometry presents persistent challenges for analyzing subregional, laminar, and connectivity patterns. Here, we present a computational workflow that generates curvilinear-coordinate flatmaps from Common Coordinate Framework (CCF) registered hippocampal and retrohippocampal regions by solving the Laplacian equation to derive geodesic streamlines. This transformation unfolds the hippocampus into a planar slab, bounded by the meningeal and ventricular surfaces, with the depth defined along

Changes in feral dog populations during the COVID-19 pandemic in Arequipa, Peru.

BACKGROUND: Feral dogs in Arequipa, Peru, inhabit caves in periurban areas and they may access and rely on organic waste from landfills and livestock from backyard farms. These feral dogs frequently attack small and medium farm animals and occasionally people, posing significant public health risks. Beyond the physical injuries resulting from the attacks, these dogs pose a threat for dog-mediated human rabies as there is active virus transmission in the dog population of Arequipa city. The COVID-19 pandemic restrictions in Arequipa, including restaurant closures, led to a decline in backyard f

Modelling factors associated with the probability of seeking traditional care after dog bites in Sierra Leone.

Evidence suggests a rising incidence of dog bites in Sierra Leone despite ongoing efforts to prevent rabies. However, little is known about the factors influencing the decision to seek medical care following a dog bite. To address this gap, we developed a probabilistic model to examine factors associated with the likelihood of seeking traditional care in Sierra Leone. Among the 2558 respondents who completed the survey, 31 % (782/2558) indicated that they would seek traditional care after a dog bite. The posterior distributions of our model estimates indicated that the probability of seeking t

Developability assessments with four mRNA-LNP vaccine formulations comparing mouse immunogenicity, structural attributes, and stability profiles.

A standardized developability workflow to rank-order the stability profiles of mRNA-LNP vaccine formulations is described. As proof-of-concept, three experimental mRNA-LNP formulations with varying lipid compositions were benchmarked against a Spikevax® mimic via mouse immunogenicity studies and various stability studies using a "routine analytical toolbox" to monitor (1) chemical integrity of the mRNA and lipid components, (2) structural integrity of the assembled nanoparticles, and (3) functionality (i.e., cell-based transfection assays). Although the four mRNA-LNP formulations displayed ove

The Role of Companion Animals as 'Sentinels' From the One Health Perspective.

Companion animals, particularly dogs and cats, are becoming essential members of modern households. They consistently share both indoor and outdoor spaces with their owners, resulting in a substantial overlap in living environments. Although public health concerns associated with companion animals receive considerable attention, their potential role as sentinel species remains underestimated. This role aligns with the One Health concept, which emphasizes the interconnection and mutual influence of human, animal and ecosystem health. In this review, we discuss the sentinel role of companion dog

An anti-inflammatory neuroenhancer mitigates amyloid-β pathology to improve Alzheimer's disease therapy.

β-amyloid (Aβ) inhibition significantly attenuates the early-stage Alzheimer's disease (AD) progression, but the improvement in cognitive function remains limited by neuroinflammation. Here, we developed a bioinspired neuroenhancer that concurrently targets both Aβ aggregation and neuroinflammation. Rutin and small interfering RNA targeting beta-site amyloid precursor protein cleaving enzyme 1 (siBACE1) were co-loaded into the calcium phosphate core, which was further coated with lipid bilayers and Angiopep-2/rabies virus glycoprotein 29 peptides to form the multifunctional neuroenhancer (RB@L

Evaluation of Risk Factors for Failed Seroconversion in the Management of Potential Lyssavirus Exposures.

BACKGROUND: Although failure of rabies postexposure prophylaxis is rare, several factors, including improper vaccine or rabies immunoglobulin administration, may increase the risk. Where there is the potential for postexposure prophylaxis failure, public health units may recommend serology to confirm a therapeutic immune response. This study evaluates which factors increase the risk of failed seroconversion in cases where serology was indicated after rabies postexposure prophylaxis. METHODS: Potential lyssavirus exposure notifications where serology was collected were identified from 5 Southe

Stray dog bites and rabies in India: A silent epidemic.

In 2024, over 37 Lakh incidents of Dog Bites in India were reported, primarily due to stray dogs with the result that 54 persons died from rabies. Surely this is a public health crisis. The Municipality's fragile health infrastructure and its failure to provide a prompt and appropriate medical responses has aggravated ethical tension between animal rights and human safety issues and exposed the inadequacies in the system. The present Article analyses India's medico-legal framework for dog bite cases from various perspectives, including ethical, statutory and judicial. The present Article also

Early immunogenic response to intradermal, subcutaneous, and intramuscular rabies vaccination in cattle and buffaloes: a field study.

Rabies is a fatal zoonotic disease of major global health significance and continues to cause substantial economic losses in livestock in endemic regions, particularly in countries like India where surveillance and reporting remain limited. Preventive vaccination is the most effective means of reducing bovine rabies risk; however, practical considerations such as dose optimization and administration route require further evaluation under field conditions. This study assessed the early humoral immune response to post-exposure rabies vaccination delivered through different routes in cattle and b

The rabies glycoprotein C-terminal peptide attenuates LPS-induced synaptic dysfunction and cognitive impairment in male rats.

The rabies virus exhibits a specific neurotropism for neurons within the central nervous system. It encodes five proteins, one of them is glycoprotein RVG, which integrates into the host cell's plasma membrane. Our previous research found that RVG expression in hippocampal cells enhances memory and potentiates synaptic plasticity, dependent on its PDZ-binding motif (PBM). In this study, we aim to express a peptide from the carboxyl end of RVG, named Neurovita 2 (NV2), in the hippocampus, to investigate its effects on learning, memory, and synaptic plasticity. 2 μl of a lentiviral vector (108T.

Pediatric Conditions for Which Skin Biopsies of Clinically Normal Skin Have Diagnostic Yield: A Review for the Pediatric Dermatologist.

While skin biopsies are typically performed from lesional or pathologic skin, biopsies of non-lesional, healthy skin can be diagnostically valuable in certain pediatric conditions. This approach may offer a less invasive alternative to other diagnostic procedures, which is especially important in minimizing discomfort and risk in children. We evaluate the diagnostic utility of performing a biopsy of normal appearing, healthy skin in pediatric populations and provide guidance on disease-specific biopsy sites, specimen handling, and histopathologic analyses to consider in such evaluations. Rabies, host population structure, and cross-species transmission to the migratory bat Tadarida brasiliensis in Chile.

Although many bat species migrate, the consequences of migration for bat population structure and rabies virus transmission remain poorly understood. Understanding the spatiotemporal dynamics of bat rabies (Lyssavirus rabies) transmission is important to prevent rabies spillovers to dead-end hosts, including humans. Here, focusing on the long-distance migratory Brazilian free-tailed bat (Tadarida brasiliensis) in South America, we identify the extent of host population and rabies virus structure across Chile. The analysis of 112 cytochrome b sequences from T. brasiliensis individuals and 144 r

Rabies virus matrix protein hijacks the TGF-beta activated kinase 1 binding protein 2-p38 mitogen-activated protein kinase pathway to inhibit apoptosis and promote viral replication.

Rabies virus (RABV) is a neurotropic virus that infects nearly all warm-blooded mammals and is almost invariably fatal once symptoms appear. Previous studies indicate that RABV uses its structural proteins to hijack host factors or modulate host signaling pathways to promote viral replication and pathogenicity. In this study, we observed upregulated expression of the host factor TGF β activated kinase 1 binding protein 2 (TAB2) in N2a cells following RABV infection. Loss and gain of function experiments confirmed that TAB2 overexpression enhances RABV replication, whereas TAB2 knockout suppres
