The Analytical Performance and Inter-laboratory Standardization of Next-Generation Sequencing (NGS) Panels for Genetic Risk Stratification in Acute Myeloid Leukemia: A Systematic Review and Meta-Analysis.

Genetic risk stratification is fundamental to the clinical management of acute myeloid leukemia (AML). While targeted next-generation sequencing (NGS) panels have replaced traditional single-gene assays, significant inter-laboratory variability in assay design and bioinformatic pipelines persists. This systematic review and meta-analysis evaluates the analytical performance and standardization of NGS panels used for AML risk stratification. Major databases were searched for studies reporting the analytical validation or inter-laboratory harmonization of NGS panels for AML compared to orthogona

Vascular Endothelial Growth Factor-D Improves Lung Vascular Integrity During Acute Lung Injury.

BACKGROUND: Disorders in pulmonary vascular integrity are a prominent feature in many lung diseases. Paracrine signaling is highly enriched in the lung and plays a crucial role in regulating vascular homeostasis. However, the specific local cell-cell crosstalk signals that maintain pulmonary microvascular stability in adult animals and humans remain largely unexplored. METHODS: In this study, we used single-cell RNA-sequencing-based computational pipelines to systematically profile ligand-receptor interactions within the lung microvascular niche and identified VEGF-D (vascular endothelial gro

Respiratory Support in Cardiogenic Pulmonary Edema: Clinical Insights from Cardiology and Intensive Care.

Cardiogenic pulmonary edema (CPE) is a life-threatening manifestation of acute heart failure characterized by rapid accumulation of fluid in the interstitial and alveolar spaces, leading to severe dyspnea, hypoxemia, and respiratory failure. The condition arises from elevated left-sided filling pressures that increase pulmonary capillary hydrostatic pressure, disrupt alveolo-capillary barrier integrity, and impair gas exchange. Neurohormonal activation further perpetuates congestion and increases myocardial workload, creating a vicious cycle of hemodynamic overload and respiratory compromise.

Comparison of Liquid Chromatography- and Nano-Electrospray Ionization-Mass Spectrometry Approaches for Single-Cell Metabolomics.

Live single-cell metabolomics is a rapidly growing area of research, which offers the potential to provide unique insights into cellular function and heterogeneity. Single-cell isolation approaches based on capillary sampling are in principle compatible with either nano-electrospray ionization-mass spectrometry (nano-ESI-MS), where the cell is lysed and sprayed directly into a mass spectrometer, or liquid chromatography-mass spectrometry (LC-MS) for metabolomics analysis. However, there are no data indicating which approach can provide the best performance (metabolite coverage, reproducibility

Advances in In Vitro Vascularization of Engineered Tissues: From Microvessel Formation to Hepatic Tissue Integration.

Advances in tissue engineering have enabled the construction of two-dimensional tissues such as skin and cornea, as well as avascular three-dimensional (3D) tissues such as cartilage. However, building fully functional 3D tissues and organs remains a major challenge. In 3D constructs, microvascular networks are indispensable for delivering oxygen and nutrients to the inner regions; thus, the formation of capillaries and microvessels and their integration with 3D parenchymal tissues--so-called vascularization--are of critical importance. In this review, we discuss the current status and future

Availability of diagnostic tools and stewardship protocols for managing infectious diseases in Italian paediatric emergency departments: a nationwide survey.

BACKGROUND: Infectious diseases are a leading cause of Paediatric Emergency Department (PED) visits worldwide, characterised by significant clinical variability in both presentation and severity. Literature highlights that infection biomarkers and rapid etiological tests, when integrated into standardised clinical protocols, are essential for timely diagnosis and appropriate treatment, reducing misdiagnosis and antibiotic misuse. However, these tools are not consistently available in Italian PEDs. The objective of this survey is to assess the availability of diagnostic tools and current diagno

Diabetic peripheral neuropathy identification using enface optical coherence tomography and multi-head attention deep learning algorithm.

OBJECTIVE: Diabetic peripheral neuropathy (DPN) is a common chronic complication of diabetes, but current diagnostic methods are limited by invasiveness, poor sensitivity, or subjectivity. This study aims to develop a non-invasive, reliable diagnostic tool using multimodal optical coherence tomography (OCT) images and a deep learning (DL) algorithm with multi-head attention for early DPN detection. METHODS: A multi-head attention-based DL model was constructed, with ResNet-18 as the feature extractor to fuse and classify enface OCT images from different retinal layers. A total of 3264 OCT ima

Nanoscopic analysis of tight junction organization in in vitro blood-brain barrier models.

BACKGROUND: The composition of tight junctions (TJs) in epithelia is well understood, whereas that one in endothelial cells at the blood-brain barrier (BBB) is controversial. Although freeze fracture EM remains the gold standard for assessing TJ nano-structures, advances in fluorescence microscopy, such as stimulated emission depletion (STED) together with image analysis tools, allow immunostaining-based quantitative analysis of the protein composition and the nano-organization of TJ strands. METHODS: Here, we present two approaches for quantitative analysis of the BBB TJ meshwork utilizing c

Comparative Evaluation of Glucose Levels in Gingival Crevicular Blood and Peripheral Blood Using Two Different Glucometers in Patients Suffering From Periodontal Disease.

INTRODUCTION: Given dentists' increasing role in healthcare, dental clinics may be essential sites for diabetes screening. Gingival bleeding is a hallmark of periodontal inflammation. Gingival crevicular blood (GCB) collection with a portable glucometer for glucose measurement is a safer, more convenient, patient-friendly, and time-saving option. The primary objective of the current investigation was to compare random blood glucose levels with those in gingival crevicular and peripheral blood, as determined using two different glucometers. The study's secondary goal was to assess the feasibili

Dual-targeted and ROS-responsive dexamethasone prodrug liposomes for enhanced therapy of acute lung injury.

The clinical application of dexamethasone (DEX) in acute lung injury (ALI) is constrained by systemic toxicity, poor localization to inflamed sites, and limited lipophilicity that hampers nano-encapsulation. To overcome these barriers, we engineered a novel DEX prodrug liposome that couples high encapsulation efficiency, active biological targeting with disease-triggered release. Inspired by the drug-loading paradigm of Onivyde®, we installed a weak-base handle (4-piperidinopiperidine) on DEX to generate a prodrug (DPD) to achieve efficient active loading into liposomes (LipDPD; encapsulation

Preferential crosstalk between perifollicular capillary vessels and dermal papilla cells during hair cycling homeostasis.

The tissue-specific capillary system supports the unique function of each organ and dynamically remodels in response to local requirements. In this study, we found that perifollicular vascularization flexibly adjusts to accommodate physiological changes. Notably, not all capillary vessels responded and migrated equally. However, those around the dermal papilla (DP) exhibited preferential mobilization. Treatment with minoxidil, a hair growth agent, significantly increased perifollicular vessel mobilization around the DP, whereas it was inhibited in experimental models of tissue aging, such as t
