Brain-targeted extracellular vesicles for anti-cuproptosis therapy in subarachnoid haemorrhage.

BACKGROUND: Subarachnoid haemorrhage (SAH) is primarily caused by ruptured aneurysms with high mortality worldwide. Cuproptosis is a copper-induced cell death that regulates lipoylated tricarboxylic acid cycle proteins. The link between cuproptosis and SAH is unclear. To inhibit cuproptosis for SAH treatment, we designed brain-targeted delivery of siRNA to inhibit cuproptosis. METHODS: Transcriptome data of SAH related to cuprotosis were extracted from the Gene Expression Omnibus and defined using quantitative reverse transcription-PCR. We injected RVG-RBCEVs/siRNA (rabies virus glycoprotein-

Contextualizing the Association Between Maternal Incarceration and Teenage Motherhood.

BACKGROUND: Some preceding researches have observed that certain neurological disorders, such as Alzheimer's disease and multiple sclerosis, may affect breast cancer risk. However, whether there are causal relationships between these neurological conditions and breast cancer is inconclusive. This study was designed to explore whether neurological disorders affected the risks of breast cancer overall and of the two subtypes (ER+ and ER-). METHODS: In the course of this study, genome-wide association study (GWAS) data for nine neurological diseases (Alzheimer's disease, multiple sclerosis, Park

A Systematic Review of Historical and Contemporary Rabies Epidemiology and Management in Alabama, USA.

The history of wildlife pathogens in time and space impacts current understanding, detection, and management of infectious diseases, which may best be appreciated dynamically at a local level. As in the rest of the USA, over the past two centuries the epizootiology, prevention, and control of rabies in Alabama has changed significantly. Canine rabies was reported in Alabama during the USA civil war and predominated until after World War II. Appreciation of wildlife rabies grew only after the control of rabies in dogs. During the 1950s, rabies in gray foxes (Urocyon cinereoargenteus) spread thr

Properties and Application of Pseudotyped Lentiviral Vectors.

Lentiviral (LV) vectors derived from human immunodeficiency virus type 1 (HIV-1) mediate efficient gene transfer into various cell types, including neuronal, glial, and neural stem cells in the central nervous system. The HIV-1-based LV vectors commonly used are self-inactivating and pseudotyped with a variety of viral envelope glycoproteins. Pseudotyping with vesicular stomatitis virus glycoprotein (VSV-G) confers broad tropism to wide variety of cells and enhances vector stability of the vectors. Pseudotyping with other glycoproteins alters the tropism. In particular, the use of rabies virus

Production and Purification of Lentiviral Vectors.

Lentiviral vectors are efficient genetic tools for neuroscience research. Among several kinds of lentiviral vectors, human immunodeficiency virus type 1 (HIV-1)-based lentiviral vectors have shown promise. Envelope glycoproteins studding the surface of viral particles play a crucial role in viral infection of host cells. Conventional lentiviral vectors are pseudotyped with vesicular stomatitis virus glycoprotein (VSV-G), and efficiently transfer the gene to different neurons. Lentiviral tropism can be manipulated by genetically engineering glycoproteins. Analyses of specific neural pathways ar

Rabies Despite Postexposure Prophylaxis in High-Burden Settings: Vaccine Failure or Gaps in Healthcare Delivery?

BACKGROUND: Rabies, a fatal zoonotic encephalitis, is preventable through timely postexposure prophylaxis (PEP), which includes wound care, vaccination, and local infiltration of immunoglobulins. However, reports of rabies despite PEP raise concerns about vaccine efficacy and protocol adherence. This study aimed to assess the burden and underlying causes of such apparent rabies "breakthrough" infections. METHODS: This retrospective study analyzed laboratory-confirmed rabies cases from 2018 to 2024 who had received PEP. Cases with at least 1 documented dose of rabies vaccine were included. "Tr

Rabies in Europe: Epidemiology, Clinical Management, and Prevention.

BACKGROUND: Rabies is one of the oldest known zoonoses. In Europe, because cases are rare and medical experience is limited, there is often uncertainty about the indications and modalities of pre- and post-exposure prophylaxis (PrEP, PEP). METHODS: This review is based on studies of the epidemiology, diagnosis, treatment, and prophylaxis of rabies that were published from January 2000 to October 2025 and retrieved by a selective search in PubMed, supplemented by data from the WHO, CDC, ECDC, RKI, and FLI and the current recommendations of the German Standing Committee on Vaccination (STIKO).

Rabies post-exposure prophylaxis in India: a SWOT analysis.

Rabies remains a pressing public health issue, particularly in countries like India, where it inflicts a significant toll on marginalised communities. With approximately 20,000 deaths annually due to rabies, the disease underscores the urgency for effective prevention strategies. Despite the existence of post-exposure prophylaxis (PEP), rabies continues to claim lives, highlighting the importance of understanding and addressing the challenges in its implementation. This study aims to conduct a comprehensive analysis of strengths, weaknesses, opportunities and threats (SWOT) of India's rabies P

RVG-targeted extracellular vesicles loaded with echinatin attenuate dopaminergic neurodegeneration via the IGF-2/PI3K/Akt pathway in Parkinson's disease mice.

BACKGROUND: Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by the loss of dopaminergic (DA) neurons. The development of effective neuroprotective therapies is severely hampered by the blood-brain barrier (BBB), which restricts drug delivery to the central nervous system. This study aimed to develop a novel brain-targeted nanodelivery system by functionalizing extracellular vesicles (EVs) with a rabies virus glycoprotein (RVG)-derived peptide to deliver echinatin (Echi), and to systematically evaluate its therapeutic efficacy and underlying mechanisms in a mo

Challenges and potential opportunities for improving One Health surveillance in low-resource settings: Insights from rabies surveillance in Malawi.

The growing threat of zoonotic diseases has driven an increased interest in “One Health”, a framework that demands a more holistic approach to understanding, preventing and controlling the emergence and spread of health risks at the human-animal-environment interface. This approach requires systems that capture data across human, animal, and environmental health sectors. However, in resource-limited settings, critical actors, namely public health departments, veterinary services, and environmental monitoring agencies, are often underfunded and lack robust surveillance systems. Additionally, th

Comparison of serological tools for reliable diagnosis of brucellosis circulation in the West-African context.

BACKGROUND: Brucellosis is listed as a priority disease in low-income countries like Guinea, facing challenges in logistics, equipment, competence, and cost limitations for diagnosis. Serological diagnosis is mainly performed by the Rose Bengal agglutination test (RBT) in the veterinary sector. We have compared its discriminative capacity with more sophisticated and expensive serological tests, such as multi-species or species-specific ELISA kits and Complement Fixation test (CFT). METHODOLOGY/PRINCIPAL FINDINGS: A panel of 554 serum samples of pigs, goats, sheep, and cattle collected through

mRNA-Encoded antibodies as a next-generation therapeutic paradigm: a rapid and adaptive platform for the prevention and treatment of emerging and re-emerging infectious diseases - A critical review.

Messenger RNA (mRNA)-encoded antibodies represent a transformative therapeutic platform with the potential to rapidly combat emerging infectious diseases by enabling in situ expression of potent neutralizing antibodies directly in the patient's body. Unlike conventional monoclonal antibody (mAb) therapies, which rely on labor-intensive and time-consuming cell culture production, mRNA-encoded antibodies offer a faster, scalable, and cell-free approach that bypasses protein purification and cold-chain constraints. This strategy has demonstrated considerable promise during the COVID-19 pandemic,

Human safety of SPBN GASGAS for oral rabies vaccination of dogs.

Oral rabies vaccination (ORV) of dogs has been suggested as a complementary tool to injectable rabies vaccination, to access free-roaming dogs during mass dog vaccination campaigns. Stringent safety requirements have been developed and implemented for ORVs targeting wildlife and dogs. Part of the safety requirements involves conducting a risk assessment of the potential for human contact with the vaccine virus and the impact on human health. The former is predominantly determined by the bait distribution method and the latter by the safety profile of the vaccine construct. This risk assessment

Rabies post-exposure prophylaxis in children: adherence to national guidelines in a country at ongoing risk.

BACKGROUND: Rabies is a preventable but fatal zoonotic disease, and children account for a large share of exposures worldwide. Türkiye is regarded as rabies-endemic because of ongoing circulation in terrestrial animals, particularly dogs, although human cases are rare because of effective surveillance and post-exposure prophylaxis. Adherence to post-exposure prophylaxis guidelines is crucial for prevention. This study evaluated the appropriateness of initial rabies post-exposure prophylaxis administered to pediatric patients according to the 2019 Turkish Ministry of Health guideline. METHODS:

Imported Human Rabies - Kentucky and Ohio, 2024.

Human rabies cases are rare in the United States; most result from domestic wildlife exposure. U.S. residents can acquire rabies abroad, typically through contact with dogs in areas where dog-maintained rabies is endemic. In November 2024, a man from Haiti was admitted to a Kentucky hospital with an 8-day history of progressive lower extremity pain and weakness. Soon after admission, he experienced hypersalivation, dysphagia, agitation, and eventually, respiratory failure requiring invasive mechanical ventilation. Ten days after admission, he was transferred to a referral hospital in Ohio, whe
