Material-specific modulation of microplastic biotoxicity by chlorination: Mechanistic insights into ER stress and associated health risks.

Chlorination, a widely used disinfection method in drinking water treatment plants, poses underexplored health risks through its interactions with microplastics (MPs). Here, we show that chlorination induces significant physicochemical changes in four common MPs-polyethylene (PE), polyurethane (PU), polytetrafluoroethylene (PTFE), and polymethyl methacrylate (PMMA)-focusing on particles smaller than 30 µm. PU and PE MPs exhibit the most pronounced changes including significant surface degradation and oxidation, releasing various organic compounds. In vitro assays show that disinfected PU MPs s

Detection of degradation products of chemical warfare agents in Halabja soil: In vitro genotoxicity and effects in exposed humans.

The town of Halabja in Iraqi Kurdistan remains a symbol of the devastating impact of chemical weapons. Almost four decades after the attack, inhabitants continue to suffer from increased cancer rates and severe respiratory, neurological, and psychiatric disorders. While the acute effects of chemical warfare agents have been widely studied, their long-term environmental persistence and associated health impacts remain poorly understood. Here, we investigated the presence of chemical warfare agent residues in the environment and assessed their potential genotoxic effects. Soil and water samples

Resveratrol alleviates hippocampal oxidative stress, neuroinflammation and pyroptosis and reduces anxiety-like behavior in broilers under cold exposure.

Resveratrol (Res) exhibits antioxidant properties and neuroprotection. To investigate its protective effect, 576 one-day-old broilers were assigned to eight groups, reared under normal temperature or cold exposure (CE) and received the basal diet with 0, 250, 500, and 750 mg/kg Res, respectively. Broilers underwent behavioral tests, and hippocampus were collected. The results showed that after CE, the latency to first move and vocalise in open field test, induction times in tonic immobility test, and the latency to move, eat and vocalise in attention bias test were increased, while the numbers

CD300f acts as an inducible inhibitory checkpoint that limits mast cell activation by suppressing MITF-dependent activation.

Mast cells (MCs) are key regulators of cutaneous immune responses and play a central role in allergic and non-IgE-mediated skin inflammation by activating the Mas-related G protein-coupled receptor X2 (MRGPRX2). The inhibitory receptor CD300f has been shown to modulate MC activation, but its role in regulating MRGPRX2-dependent signaling remains unclear. Here, we investigated CD300f expression and function in human skin MCs and its impact on MRGPRX2-mediated responses, with some comparison to IgE-mediated responses. CD300f was expressed on human skin MCs, and its surface expression increased f

Melatonin attenuates dermal fibroblast senescence with suppression of CXCL9-associated inflammatory signaling and mitochondrial stress.

Skin aging is driven by intrinsic and extrinsic factors that promote fibroblast senescence and dermal extracellular matrix degradation. Melatonin has potent antioxidant and anti-inflammatory activities. Here, we investigated the anti-aging effects of melatonin in human dermal fibroblasts and aged mouse skin, focusing on the pro-inflammatory chemokine CXCL9. Replicative-, irradiation-, and doxorubicin-induced senescence models of human dermal fibroblasts were used to assess the effects of melatonin. Melatonin significantly reduced the proportion of senescence-associated β-galactosidase-positive

Dual-functional hydrogel capable of scavenging reactive oxygen species and regulating macrophage polarization for repairing MRSA-infected wounds.

The healing of infected wounds presents a remarkable clinical challenge owing to bacterial infection-induced oxidative stress and persistent inflammation. To address these issues, a multifunctional hydrogel was prepared by means of Schiff base crosslinking between oxidized konjac glucomannan (OKGM) and sericin, followed by the incorporation of polydopamine nanoparticles (PDA NPs). The OKGM/Sericin-PDA NPs (KSP) hydrogel exhibited excellent antioxidant capacity, facilitating macrophage polarization to the M2 phenotype. Transcriptomic analysis revealed that the KSP hydrogel exerted immunomodulat

Restoring joint homeostasis: In vitro and in vivo efficacy of a sodium hyaluronate-lipoate-formate therapy for osteoarthritis.

Osteoarthritis (OA) is a multifactorial joint disorder characterized by inflammation, cartilage homeostasis disruption, and extracellular matrix degradation. Although hyaluronic acid (HA) viscosupplementation is widely used, available formulations mainly provide symptomatic relief without targeting underlying inflammatory and catabolic processes. Here, two innovative HA derivatives covalently functionalized with lipoic acid and formate (SHLF0.5% and SHLF1%) were developed to enhance viscoelastic properties and modulate the oxidative joint microenvironment. Biological performance was evaluated

A Dual pH‑Responsive Hydrogel with Site‑Specific Therapeutic Functions for Differential Pathological Microenvironment Remodeling and Accelerated Repair of Infected Diabetic Wounds.

Diabetic skin wounds have disrupted local acid-base balance, prone to bacterial adhesion and sustained chronic inflammation that hinders tissue repair. Benefiting from the unique properties of oxidized sodium alginate, we prepared a dual pH-responsive Schiff base crosslinked hydrogel loaded with herbal nanoparticles to remodel the defective wound microenvironment and speed up regeneration. Dynamic covalent crosslinking endows the hydrogel with favorable flexibility and shape adaptability to fit irregular wound surfaces. It exhibits obvious dual pH sensitivity: strong antibacterial activity in

FAM50A promotes esophageal squamous cell carcinoma progression via splicing-dependent oncogenic circSDK1- and DUSP22-mediated MAPK/FAK activation.

Dysregulated splicing factors play critical roles in tumor progression. Here, we identify family with sequence similarity 50 member A (FAM50A) as a pivotal oncogenic splicing factor involved in esophageal squamous cell carcinoma (ESCC). Multi-omics and multi-cohort analyses revealed that FAM50A is upregulated in ESCC and correlates with a poor prognosis. Functional studies demonstrated that FAM50A promotes ESCC proliferation and migration both in vitro and in vivo, primarily through the activation of mitogen-activated protein kinases (MAPKs) and focal adhesion kinase (FAK). We further characte

Microglia and border-associated macrophages play opposite roles in cerebrovascular and leukocyte responses under systemic inflammation in male mice.

Microglia closely associate with the vasculature, but their roles in leukocyte trafficking and cerebral blood flow (CBF) during systemic inflammation are unclear. We show that sustained systemic inflammation leads to altered microglia-vascular interactions, leukocyte recruitment to cerebral blood vessels and reduced hypercapnia-induced vasodilation. We find that microglia and border-associated macrophages (BAMs) play distinct roles in these processes: while BAM depletion with clodronate increases leukocyte recruitment, microglia/BAM depletion with PLX5622 and the absence of microglia, but not

Predicting long-term survival in primary myelofibrosis.

We interrogated data from 217 subjects with primary myelofibrosis (PMF) diagnosed before 2001 to identify covariates at diagnosis associated with long-term survival. Forty-one survived more than 25 years. Long-term survivors were more often female, younger than 50 years and less likely to have anemia, splenomegaly, increased blood CD34-positive cells, a JAK2V617F variant, and more likely to have grade-0 bone marrow fibrosis and increased platelets. In multivariable logistic regression analyses, age <50 years, platelet count >450 × 10⁹/L, and grade-0 bone marrow fibrosis independently predicted

Comparison of the Effects of Human Amniotic Fluid-Derived Cell Culture Medium and Adipose Tissue-Derived Stem Cells on Capsule Formation Around Silicone Prosthesis.

BACKGROUND: Peri-implant capsule formation is a physiological response to implanted biomaterials; however, excessive fibrotic reactions may contribute to capsular contracture and implant-related complications. Human amniotic fluid-derived cell culture medium (ACCM) and adipose tissue-derived stem cells (ADSCs) have demonstrated anti-inflammatory and antifibrotic properties in experimental models. This study investigated the individual and combined effects of ACCM and ADSCs on peri-implant capsule formation in a preclinical rat model. OBJECTIVES: To determine whether the combined application o

Single-cell eQTL Mendelian randomization reveals the monocyte-associated PRKCB-CTSL axis as a candidate regulatory pathway and potential therapeutic target in rheumatoid arthritis.

OBJECTIVES: Rheumatoid arthritis (RA) involves persistent immune dysregulation, but cell-specific genetically regulated pathways remain unclear. We integrated cell-specific genetic and multi-omics evidence to prioritize candidate regulatory pathways. METHODS: cis-eQTLs from 17 immune-cell populations were integrated with RA GWAS data by two-sample Mendelian randomization. Transcriptomics, WGCNA, and machine-learning feature selection identified convergent candidates. Single-cell and virtual perturbation analyses assessed cellular localization and downstream changes. CD47-SIRPα Axis as a Target for Neuroprotection.

Microglial "Off" signaling is crucial for maintaining microglia in a homeostatic state through the interaction of "Off" neuronal ligands with their corresponding microglial receptors. Dysregulation of "Off" signaling has been reported in both acute and chronic neuroinflammation; however, the influence of inflammatory mediators on this pathway remains underexplored. Here, we investigated whether chronic IL-6 or IL-10 overexpression modulates microglia-neuron "Off" signaling after FNA, thereby accounting for the previously observed differences in facial motoneuron (FMN) survival and microglial r

Coumarin-based Scaffolds as Multitarget Modulators of Chronic Inflammation: Insights into Inflammatory Signaling Networks.

A wide range of chronic illnesses, including metabolic, autoimmune, neurodegenerative, and malignant conditions, are caused by the dysregulation of inflammation, a complex, multi-scale biological defense mechanism. Despite their clinical efficacy, current pharmacotherapies such as NSAIDs, corticosteroids, and biologics are constrained by side effects, insufficient resolution of inflammatory cascades, and poor long-term appropriateness. Interest in natural-product scaffolds with multi-target anti-inflammatory potential has increased due to these therapeutic gaps. Among these, 7-hydroxycoumarin
