No-known-vector flaviviruses exhibit diverse replication and virulence phenotypes in mice.

The Orthoflavivirus genus (flaviviruses) includes globally significant arboviruses, which cycle between arthropod vectors (mosquitoes and ticks) and vertebrate hosts. In contrast, no-known-vector flaviviruses (NKVFVs) have been isolated from rodents and bats, but not arthropods, so they are thought to spread by vector-independent routes. However, little is known about the host range and pathogenic mechanisms of these viruses. To evaluate NKVFV pathogenesis, we infected wild-type, Ifnar1-/-, and Ifnar1-/- Ifngr1-/- mice by footpad inoculation with 12 NKVFVs: Entebbe bat virus (ENTV), Sokuluk vi

A Modular Biomimetic Nanovaccine for TBEV: Orchestrating Cascade Immunoprotection via Programmed Dendritic Cell Targeting and Antigen-Adjuvant Co-delivery.

Tick-borne encephalitis (TBE) is an escalating global health threat, increasingly exacerbated by climate change, and underscores the need for innovative vaccine strategies. Conventional subunit vaccines are limited by suboptimal spatiotemporal dissociation between antigens and adjuvants, as well as insufficient specific targeting, resulting in compromised immunogenicity. Herein, we report a biomimetic nanovaccine platform engineered to overcome these limitations through the synergistic co-delivery of antigen and adjuvant. This platform is constructed by co-displaying the TBE virus envelope pro

Retrospective serological evidence of exposure to selected equine viruses in horses from Indigenous communities of Costa Rica, 2011.

Horses are essential for transportation and livelihoods in Indigenous communities of Costa Rica. However, information on their exposure to viral pathogens remains scarce. We conducted a retrospective cross-sectional serosurvey using samples collected from horses from four Indigenous communities (Alto Pacuare, Amubre, Paso Marcos, and Vereh) sampled between March and October 2011 (n = 278). Sera were tested in 2014 using commercial immunodiffusion or ELISA assays for antibodies against West Nile virus (WNV), equine herpesviruses 1 and 4 (EHV-1 and EHV-4), influenza A virus, equine arteritis vir

Immunogenicity and safety of an interchangeable vaccination schedule of two inactivated Japanese encephalitis vaccines: a multicentric, phase IV, open-label, randomized controlled trial.

BACKGROUND: In India, two inactivated Japanese encephalitis (JE) vaccines, JENVAC and JEEV, are used in childhood immunization program as a two-dose schedule. Vaccine supply interruptions and population mobility can result in children receiving different products for their first and second doses. We assessed immunogenicity and safety of mixed-product vaccine schedules. METHODS: In this multicentric, open-label, phase 4, non-inferiority randomized trial conducted during November 11, 2023-September 2, 2025, infants aged 9-12 months were assigned (1:1:1:1) to JENVAC-JEEV, JENVAC-JENVAC, JEEV-JEN

MicroRNA Responses Associated with Vector-Borne Diseases in Humans and Arthropod Vectors: A Systematic Review.

Vector-borne diseases (VBDs) represent a significant public health challenge. Early and accurate diagnosis is crucial for effective disease management and control, with host-encoded microRNAs (miRNAs) emerging as promising disease biomarkers. This systematic review surveys research conducted on miRNA responses in disease vectors and human hosts of VBDs. A systematic search of four databases and manual searching identified 93 qualifying articles. Numerous VBDs were described, with the most common focus on dengue (21%; n = 15) and Chagas disease (11%; n = 8). Twenty-eight in vivo human studies h

Screening human brain targets for Japanese encephalitis virus NS5 via yeast two-hybrid assay.

Japanese encephalitis (JE) is the most important vaccine-preventable acute meningoencephalitis in tropical and subtropical Asia with no known antiviral therapeutics. Available vaccines offer limited protection against emerging Japanese encephalitis virus (JEV) genotypes, posing a persistent public health threat. Disruption of the interactions between JEV non-structural protein 5 (NS5) protein and human brain cellular proteins could be explored to develop flavivirus druggable host targets. This study aimed to identify the human brain cellular proteins that interact with the JEV NS5 protein. A h

Effects of West Nile Virus on behavioral and cognitive performance, circadian body temperatures, cortical Aβ and p-tau pathology, and immune measures in human apoE and human amyloid precursor protein NL-G-F/human apoE mice.

INTRODUCTION: There are three human apolipoprotein E (E) isoforms: E2, E3, and E4. Compared with E3, E4 is an AD risk factor. West Nile Virus (WNV) can cause neurological disease, and it results in some neuropathology and neuroinflammation seen in Alzheimer's disease (AD). Previously, we reported on the effects of exposure to the Kunjin strain (KUNV) of WNV in middle-aged knock-in (KI) mice expressing human amyloid precursor protein (APP) containing the dominant NL-G-F mutations with human apoE targeted replacement (TR) mice on behavioral, immune, and pathological measures, and circadian body

Elucidating the mechanism underlying incomplete immune recovery in people living with HIV through CD4⁺ and CD8⁺ T cell multi-omics.

BACKGROUND: A subset of people living with HIV (PLHIV), known as immunological non-responders (INR), fails to achieve adequate CD4⁺ T cell recovery despite viral suppression with antiretroviral therapy (ART). These individuals face an increased risk of adverse clinical outcomes. This study aims to explore the intracellular mechanisms and molecular signatures underlying this incomplete immune recovery using a multi-omics approach. METHODS: The study analysed CD4⁺ and CD8⁺ T cells from 100 ART-naïve PLHIV. Participants were classified into controls (baseline CD4⁺ > 200 cells/µL, n = 49) and cas

A versatile fluorescent reporter for Usutu virus drug screening and infection tracking.

Usutu virus (USUV) is an emerging mosquito-borne orthoflavivirus closely related to West Nile and Japanese encephalitis viruses. Despite its increasing prevalence in Europe and reports of neuroinvasive disease in humans, the molecular and immunological determinants of USUV infection remain poorly understood, largely due to the absence of tractable experimental tools. Here, we report the generation and characterization of the first full-length fluorescent reporter USUV. Using a previously established infectious clone of the Vienna 2001 strain, we inserted the mScarlet-I gene into a permissive r

Phospholipid codes at virus-host membrane interfaces: mechanisms and therapeutic opportunities in animal viral diseases.

Animal viruses depend on host membranes at every stage of infection, yet membrane lipid states are often treated as background rather than as causal determinants of virus-host interactions. This Review develops a phospholipid-code framework in which headgroup identity, acyl-chain chemistry, leaflet topology, and lipid flux define infection-stage-specific membrane states. Evidence from Newcastle disease virus, infectious bronchitis virus, avian influenza virus, porcine reproductive and respiratory syndrome virus, African swine fever virus, Senecavirus A, Japanese encephalitis virus, and porcine

Environmental surveillance: harnessing environmental sample data to protect animal health.

Infectious diseases in domestic animals-including those with zoonotic potential-impose a substantial global burden on food security, livelihoods, and public health. Environmental surveillance (ES) offers a critical complement to conventional animal-level diagnostics by detecting pathogens or their genetic signatures directly in the matrices through which they persist and spread: water, soil, air, manure, fomites, and arthropod vectors. This review synthesizes evidence across three analytical dimensions-matrix suitability, detection method performance, and wastewater-based epidemiology (WBE)-fo

A global database of West Nile virus host prevalence and competence.

West Nile virus (WNV) is one of the most widespread arboviruses globally and is maintained primarily through a bird-mosquito-bird transmission cycle, while other vertebrates play more limited roles. Host contributions to transmission depend on both infection evidence in natural populations (reflecting exposure and susceptibility) and reservoir competence, determined by the magnitude and duration of viraemia sufficient to infect mosquitoes. Despite extensive surveillance and experimental research, no comprehensive, standardised resource has integrated evidence on host exposure and infection in

EIDD-3525 is a broad-spectrum antiviral against alphavirus infection that targets the nsP4 RNA-dependent RNA polymerase.

Mosquito-transmitted alphaviruses, including chikungunya (CHIKV), Mayaro, Eastern equine encephalitis, and Venezuelan equine encephalitis viruses, cause severe arthritic or neurologic disease in humans. There are currently no approved antiviral treatments against alphavirus infection. Here, we report that the purine ribonucleoside EIDD-3525 is a potent inhibitor of alphavirus replication in vitro with broad spectrum antiviral activity against the Alphavirus genus. Using a trans-replicase assay, we find that EIDD-3525 blocks alphavirus RNA replication. Mutational profiling suggests that EIDD-35

Species-specific susceptibility and low transmission of the historical Japanese encephalitis virus Nakayama strain in North American Culex mosquitoes.

Japanese encephalitis virus (JEV) is an emerging mosquito-borne flavivirus with potential for geographic expansion, yet the risk of establishment in North America remains poorly characterized. We assessed vector competence of three North American Culex species ( Cx. pipiens, Cx. quinquefasciatus, and Cx. tarsalis ) for the JEV Nakayama strain, isolated from human brain in 1934 in Japan, across five constant temperatures (15, 20, 25, 30, and 33°C) at 4, 7, and 14 days post-feeding, quantifying infection, dissemination, and transmission rates. Vector competence was low but non-zero across all sp

Molnupiravir, first small molecule therapeutic to demonstrate therapeutic efficacy against Venezuelan equine encephalitis virus in the cynomolgus macaque model.

Venezuelan equine encephalitis virus (VEEV) is an emergent arbovirus of major epidemic concern. Associated with periodic outbreaks in Central and South America, VEEV has a high morbidity rate in humans due to a 1:1 clinical-to-subclinical ratio and can cause neurological disease in up to 14% of cases. Concerningly, no FDA-approved vaccines or therapeutics for VEEV exist. Molnupiravir, an isopropyl ester prodrug of β-D-N4-hydroxycytidine, has previously showcased notable antiviral activity against VEEV in vitro and in rodents. While molnupiravir emergency use authorization has been granted for

The D389G mutation in E protein attenuates replication and virulence of Japanese encephalitis virus.

Japanese encephalitis virus (JEV) is a mosquito-borne Orthoflavivirus that causes reproductive disorders in pigs and severe neurological sequelae in humans. Although genotypes I (GI) and III (GIII) have been dominant worldwide, the recent emergence of GV in South Korea underscores the urgent need to identify cross-genotype virulence sites. Through comparative genomic analysis, we identified the conserved D389 site in E protein (E-D389), presented in both GIII and GV strains but divergent in the attenuated GV-Sangju stain, as a potential virulence site. Our study showed that E-D389G mutation en

Neutralizing antibody evidence of dengue and chikungunya virus exposure in macaques from an urban-forest fringe in Peninsular Malaysia.

Arboviruses such as dengue virus (DENV), chikungunya virus (CHIKV), Zika virus (ZIKV), and Japanese encephalitis virus (JEV) circulate between mosquito vectors and vertebrate hosts, with non-human primates (NHPs) playing an important role in sylvatic transmission cycles. In Malaysia, frequent interactions between long-tailed macaques (Macaca fascicularis) and humans at the Universiti Kebangsaan Malaysia (UKM) Bangi campus provide an opportunity to investigate arbovirus exposure at the human-wildlife interface. Forty free-ranging macaques were sampled from five locations within the UKM Bangi ca

Tembusu virus neuroinvasion and neuropathogenesis: lessons from blood-brain barrier dysfunction in neurotropic flaviviruses.

Central nervous system (CNS) pathogenesis remains a critical but incompletely resolved aspect of flavivirus infection. Neurotropic flaviviruses usually initiate replication in peripheral tissues but can subsequently access the CNS, causing encephalitis, neurological dysfunction, or developmental abnormalities. Because these events occur along a continuum from peripheral infection to CNS injury, their temporal and mechanistic relationships are often difficult to define. In many studies, brain viral detection, blood-brain barrier (BBB) dysfunction, neuroinflammation, and neurological signs are r
