Finite-time stability of the West Nile Virus model with Brownian motion.

West Nile virus disease is an acute infectious illness caused by infection with the West Nile virus, which was first isolated from the blood of a febrile patient in the West Nile region of Uganda in 1937. In recent decades, West Nile fever has continued to expand across endemic regions globally, with no specific antiviral treatment or vaccine currently available for prevention. This study examines the finite-time stability of a stochastic West Nile virus model. Initially, the boundedness, existence and uniqueness of the solution are established. Furthermore, by constructing a Lyapunov function

Bovine lactoferrin confers antiviral effect against La Crosse virus by interfering with virus-heparan sulfate proteoglycan binding.

La Crosse virus (LACV) is a mosquito-borne orthobunyavirus within the California serogroup that primarily causes neuroinvasive disease in pediatric populations in the upper Midwestern, mid-Atlantic, and southeastern states of USA. Despite its substantial impact on public health, no vaccine or effective antiviral treatment has been developed so far. In this study, we investigated the role of the cell-surface heparan sulfate proteoglycan (HSPG) in LACV cell attachment and assessed the in vitro antiviral potential of bovine lactoferrin (bLf), which interacts with both the cell surface molecules a

Increasing Seroprevalence In Horses Evidences Widespread Circulation Of West Nile Virus In Albania.

West Nile virus (WNV) is a zoonotic mosquito-borne flavivirus that causes West Nile disease, an emerging and re-emerging infection of major global importance. Horses are an important sentinel species for monitoring WNV circulation. Since viraemia in horses is brief and viral loads in the blood are low, serological assays are the primary tools used to monitor WNV exposure. This study aimed to estimate the seroprevalence of WNV antibodies in horses in Albania. Between 2017 and 2020, 509 serum samples were collected from apparently healthy, unvaccinated horses across different regions of the coun

Serological evidence of hidden circulation of three arthropod-borne diseases (Rift Valley fever, West Nile fever, and Crimean-Congo haemorrhagic fever) in dromedary camels from the Eastern Algerian Sahara.

Several arthropod vectors, including ticks, sand flies, and mosquitoes, play a prominent role in the introduction and spread of numerous viral diseases. Despite Algeria being at high risk of arbovirus emergence, data on the circulation of arboviruses remain limited, except for some reports on West Nile virus (WNV). Therefore, the aim of this study was to investigate the seroprevalence of antibodies against three arboviruses in several regions of the Algerian Sahara in order to document previous viral exposure. Serum samples collected from dromedaries between May and November 2023 were tested f

Molecular and Serological Screening of West Nile and Usutu Virus in Birds and Humans: A Sentinel Study in Finland.

BACKGROUND: The northward expansion of mosquito-borne viruses such as West Nile virus (WNV) and Usutu virus (USUV) in Europe, driven partly by climate-related changes affecting vectors and hosts, poses an increasing threat to human and animal health. Reliable diagnostic tools are therefore essential for early detection and effective surveillance. OBJECTIVES: This study aimed to develop and evaluate sensitive molecular assays for WNV and USUV, apply them to screen wild birds and human patients in Finland, assess bird exposure through serology, and evaluate national preparedness for emerging ar

Nuclear activities and interactome of the NS5 protein of tick-borne encephalitis virus.

Orthoflaviviruses are RNA viruses responsible for significant diseases in humans, domesticated animals, and wildlife. Their NS5 protein is central in viral replication, functioning both as an RNA-dependent RNA polymerase and a methyltransferase, while also modulating cellular processes, including the interferon response. Although viral replication is cytoplasmic, the NS5 protein of several mosquito-borne orthoflaviviruses cycles between the cytoplasm and the nucleus of infected human cells. However, the nuclear localization and function of NS5 of tick-borne orthoflaviviruses, such as tick-born

Opportunities and challenges for countermeasures against Rift Valley fever virus: A quintessential One Health pathogen.

Rift Valley fever virus (RVFV) is a zoonotic arbovirus transmitted by a wide range of mosquito vectors present across the African continent. RVFV has resulted in large-scale epidemics in East, West, southern, and northern Africa with additional epidemics affecting the Arabian Peninsula, Mayotte, and Comoros. In some areas, rather than large-scale isolated outbreaks, RVFV causes a fluctuating low-level endemic burden affecting livestock populations by resulting in pregnancy loss and significant production losses. RVFV risk extends into the human population when it is transmitted either by vecto

Viral and host determinants of arboviral disease severity in African populations: A systematic review with meta-analysis.

BACKGROUND: Arboviral diseases caused by dengue (DENV), chikungunya (CHIKV), yellow fever (YFV), Rift Valley fever (RVFV), and Zika (ZIKV) viruses are responsible for considerable morbidity and mortality across Africa. Clinical presentations range from self-limiting febrile illness to fatal haemorrhagic syndromes, encephalitis, and chronic arthropathy, yet the factors that govern who develops severe disease remain poorly defined in African settings. We set out to synthesise what is currently known about the viral and host immune determinants of clinical severity for all five arboviruses in Afr

Strengthening mosquito control to manage emerging mosquito-borne diseases.

Mosquito-borne diseases such as dengue, chikungunya and West Nile virus infection become increasingly frequent in Europe. These developments require preparedness and coordinated response mechanisms across countries and sectors. Vector control is a central component of public health response, both for containing outbreaks and for reducing the risk of disease emergence. While mosquito control is important, evidence on the cost-effectiveness of most methods remains limited, complicating the design of evidence-based vector management strategies. In addition, the range of authorised vector control

Arboviral infections of the central nervous system: Mechanisms of invasion and long-term consequences.

Arboviruses are a diverse group of vector-borne viruses that cause significant global morbidity and mortality. Beyond classical systemic and febrile symptoms, many arboviruses can induce a spectrum of neurological complications, ranging from mild encephalopathy to severe neuroinvasive disease. This review summarizes current knowledge on major arboviruses from the Flaviviridae, Togaviridae, Peribunyaviridae, and Reoviridae families, emphasizing mechanisms of CNS invasion, neurological manifestations, and relevant case studies or outbreaks. Mechanisms of central nervous system (CNS) invasion are

Exploring the Potential Human Kinase-Viral Substrate Network of West Nile Virus.

West Nile virus (WNV) is a mosquito-borne pathogen of escalating epidemiological importance and a growing global health concern, driven by the climate-associated expansion of its Culex mosquito vectors. Although WNV is an extensively studied flavivirus, most host-pathogen interaction studies focus on static and structural aspects rather than dynamic and functional ones. Delineating phosphorylation-mediated interactions between WNV proteins and human kinases bridges a critical gap by providing important insight into the molecular mechanisms underlying infection. In this study, we investigated p

Trends in Tick-Borne Virus Ecology: Vector Incrimination, Focal Distribution, and Reservoir Associations.

Ticks are prominent hosts for many viruses that are pathogenic to humans on almost every continent. Despite the documented historical association of ticks with viral disease in humans, clinical case reports are low, likely due to under-recognition of these pathogens. A key feature of understanding tick-borne virus transmission to humans lies in the natural invertebrate and vertebrate hosts supporting ecological maintenance. As humans are often infected as incidental hosts during the tick-virus life cycle, determining relevant ticks and mammals involved in maintenance can inform prevention stra

Mosquito diversity and arbovirus detection in Paraguay: First evidence of dengue and chikungunya virus infection in field populations.

Arboviruses are mosquito-borne viruses of major public health importance, particularly in tropical and subtropical regions. In Paraguay, dengue (DENV) and chikungunya (CHIKV) viruses are among the most epidemiologically relevant; however, studies integrating mosquito and arbovirus surveillance remain scarce. This study aimed to characterize mosquito assemblage composition across different ecological settings and investigate arbovirus circulation in field-collected mosquitoes. A total of 4,595 mosquitoes (609 pools) were collected between 2013 and 2024 from urban, rural, and Atlantic Forest env

Widespread circulation of West Nile and Usutu viruses in sedentary and migratory avifauna: A two-year study (2024-2025) of active surveillance in South of France.

West Nile virus (WNV) and Usutu virus (USUV) are neurotropic Orthoflaviviruses sharing a similar enzootic transmission cycle primarily involving Culex pipiens mosquitoes as vectors and birds as amplifying hosts. First identified in Africa, both viruses established endemicity across Europe over the past two decades, most likely introduced and spread by migratory bird species along Mediterranean flyways. In avian species, infection outcomes range from subclinical to fatal neuroinvasive disease, varying by viral strain, host immunity, and species sensibility. Southern France emerges as a key hots

Usutu virus promotes West Nile virus replication in Culex pipiens biotype molestus during simultaneous and sequential co-infections.

The epidemiological co-circulation of the two flaviviruses West Nile virus (WNV) and Usutu virus (USUV) in several European countries poses the risk of co-infections in vertebrate hosts and mosquito vectors. WNV is an important human pathogen, whereas USUV is primarily relevant in veterinary and wildlife health. Co-infections have been detected in birds and, occasionally, in humans. To determine the potential consequences of co-exposure and co-infections in mosquitoes on viral transmission, lab-reared Culex pipiens biotype molestus were orally infected with both viruses either simultaneously o

Laboratory Assessment of Wastewater as Surveillance Tool for West Nile Virus, United States.

West Nile virus (WNV) is the most common cause of domestic human arboviral infection in the contiguous United States. WNV surveillance across the United States is not comprehensive and is limited. Nonclinical WNV surveillance is variable, and WNV surveillance in humans is limited by the percentage of symptomatic human infections and time it takes to seek healthcare and diagnosis. Wastewater surveillance has been used to detect viruses shed from asymptomatic and symptomatic persons; therefore, we assessed wastewater as a surveillance tool for WNV tracking. We tested archived RNA and raw wastewa

Evaluation of the NovaplexTM Tropical Fever Virus Assay.

Arboviral infections represent a growing challenge for global public health due to vector expansion, globalization, and climate change. Dengue (DENV), Zika (ZIKV), Chikungunya (CHIKV), and West Nile virus (WNV) often co-circulate and present overlapping clinical manifestations, making rapid and accurate etiological diagnosis difficult. In this context, multiplex real-time RT-PCR assays can support clinical management and epidemiological surveillance. In this study, analytical and preliminary retrospective comparisons of the NovaplexTM Tropical Fever Virus Assay, a multiplex RT-PCR system desig
