In silico post-hoc analysis of a clinically tested recombinant West Nile virus envelope protein vaccine.

The recombinant West Nile virus (WNV) envelope (Env) protein WN80E is one of the few WNV antigens that has been evaluated in humans and has demonstrated robust immunogenicity across animal models and in a Phase I clinical trial. Here, we performed a retrospective in silico analysis of WN80E and its variant containing a N-terminal methionine (WNM80E) to assess how accurately contemporary computational vaccinology tools can capture and differentiate their structural and immunological profiles. Physicochemical and sequence-based predictions classified both constructs as antigenic, non-allergenic,

Diagnostic value of RT-PCR for detection of tick-borne encephalitis virus RNA in cerebrospinal fluid.

PURPOSE: Tick-borne encephalitis (TBE) diagnosis is mainly based on serology, which however can be difficult to interpret in previously TBE-vaccinate patients and in immunosuppressed patients. The objectives of this study were to investigate if the detection of tick-borne encephalitis virus (TBEV) RNA in cerebrospinal fluid (CSF) can be used to diagnose TBE, and to assess the relation of TBEV RNA positivity to clinical characteristics and disease severity. METHODS: A retrospective study was conducted in TBE patients from Region Stockholm and Uppsala, Sweden, 2015–2020. CSF-samples were analyse

MyD88 signaling in myeloid cells induces gastrointestinal tract injury and systemic inflammation after West Nile virus infection.

Humans with autoantibodies to type I interferons (IFNs) are at higher risk for neurological disease after West Nile virus (WNV) infection. IFN signaling deficient WNV-infected mice develop greater inflammation in the brain, which correlates with loss of gut barrier integrity. Here, we investigate the immune pathways that mediate this virus-gut-brain axis of inflammation. Myd88-/- mice administered anti-IFNAR1 antibody and infected with WNV exhibit reduced GI tract damage and inflammation. GI tract injury caused by WNV infection requires gasdermins or IL-1 receptor expression in hematopoietic c

Mapping the bird risk index for West Nile virus in Europe and its relationship with disease occurrence in humans.

West Nile virus (WNV) is a zoonotic mosquito-borne Flavivirus, with bird populations reservoirs. Although often asymptomatic, infection in humans can cause febrile symptoms and, more rarely, severe neurological symptoms. Previous studies assessed environmental drivers of WNV infections, but most overlooked areas with potential WNV circulation despite no reported human case, and mixed mechanisms affecting hosts vs. vectors. Our objective was to generate a WNV Bird Risk Index (BRI) mapping the potential of WNV circulation in bird communities across Europe. We first used a bird traits-based model

Rift Valley fever virus activates multiple cell death pathways in neurons.

Neuronal injury and death contribute to long-term impairments and lethality during viral encephalitis. Rift Valley fever virus (RVFV), an arbovirus with epidemic potential, can manifest as late-onset encephalitis in humans, yet this disease outcome remains understudied. A lethal rodent model of RVF encephalitis is characterized by a dysregulated immune response, neuronal necrosis, and blood-brain barrier breakdown that precedes lethality. In this study, we built upon this prior work using both in vivo and in vitro models to interrogate the mechanism of cell death in neurons during RVFV infecti

Expert Elicitation on Exposure to Tick Bites and Tick-Borne Encephalitis Risk in Occupational and Recreational Forest Activities.

Background: Tick-borne encephalitis (TBE) virus is transmitted to humans via tick bites and occasionally via the consumption of unpasteurized milk products. According to the literature, the most important driver of TBE emergence and increase in incidence in humans is changes in human behaviour/activities. Method and principal findings: To compensate for the lack of data, expert opinions were gathered to identify the risk factors for exposure to tick bites linked to twenty-eight human activities (professional or recreational) in forests and to target prevention messages at the populations most

The Therapeutic Potential for Steroid Treatment Strategies in the Treatment of Murine Venezuelan Equine Encephalitis Virus (VEEV) Infection.

One highly consequential presentation of Venezuelan equine encephalitis virus (VEEV) infection is encephalitis. Here we considered anti-inflammatory interventions to limit the effects of this using a BALB/c subcutaneously challenged mouse model of disease. This disease model nearly ubiquitously presents with severe encephalitis, where viral neuroinvasion correlates with much of the outward clinical signs of disease. A selection of already licenced, commonly used anti-inflammatory drugs were tested in mice developing encephalitis (starting treatment at 24 h post challenge). Drug regimens were u

Genetic Diversity of Tick-Borne Encephalitis Virus in Kyrgyzstan.

Tick-borne encephalitis virus (TBEV) causes tick-borne encephalitis (TBE), a severe disease of the human central nervous system. Currently, the data on the genetic variants of TBEV in Kyrgyzstan are practically absent. Therefore, the aim of this study was to analyze and describe the genetic diversity of TBEV in this region. The complete genome sequences of seven TBEV strains from the collection of the Scientific Centre for Family Health and Human Reproduction Problems (Irkutsk, Russia) were determined. These strains, isolated from Ixodes persulcatus ticks from Kyrgyzstan, were sequenced using

First detection of West Nile virus in Belgium through wild bird surveillance, Belgium, 2025.

In August 2025, West Nile virus (WNV) was detected for the first time in Belgium through a monitoring programme in wild birds, with three corvids testing positive by RT-qPCR. In September and October, four additional infected birds were identified. Whole genome sequencing classified the strain as WNV lineage 2, consistent with strains circulating elsewhere in Europe. These detections provide evidence of local WNV circulation with important implications for animal and public health preparedness and surveillance during the 2026 mosquito season.

Molecular surveillance supports differing roles for Culex pipiens and Culex restuans (Diptera: Culicidae) in West Nile virus transmission in Chicago, Illinois.

The Chicago metropolitan area is a hotspot for human West Nile virus (WNV) cases. Despite extensive surveillance and research, predicting WNV cases in Chicago on a local scale is a major challenge. Most studies and mosquito surveillance efforts do not differentiate between the cryptic species Culex pipiens Linnaeus and Culex restuans (Theobald), key vectors of WNV, due to the challenge of distinguishing them morphologically. This obscures each species' respective role in transmission and may blunt the accuracy of local case forecasting. We used species-specific PCR diagnosis to identify Chicag

Epidemiology and risk analysis of Powassan virus infection, New York state, USA, 2013-2023.

Powassan virus (POWV), a tick-borne flavivirus, is an emerging public health threat in the United States. In New York State (NYS), incidence of human POWV infection has increased in recent years. We describe the epidemiology of confirmed and probable POWV infection cases reported in NYS from 2013 to 2023. A total of 44 human cases were reported over the study period, with the highest incidence rates in Columbia and Putnam counties. Most cases occurred in White, non-Hispanic males over age 50. Hospitalization was reported in 91% of cases, and 11% were fatal. Human case data and tick surveillanc

ERGA-BGE Genomes of Culex laticinctus, Culex modestus, Culex perexiguus, and Culex theileri: Unveiling the Genomes of the Key Vectors of West Nile Virus in the Mediterranean Basin.

The reference genomes of Culex laticinctus, Culex modestus, Culex perexiguus, and Culex theileri offer crucial resources for exploring the genetic basis of vector competence for different pathogens, including the emerging West Nile virus. Each genome was assembled into three contiguous chromosomal pseudomolecules. These chromosome-level assemblies encompass 834, 698, 436, and 934 Mb, respectively. They are composed of 3,284, 4,220, 1,917, and 2,704 contigs and 312, 979, 475, and 709 scaffolds, with contig N50 values of 434, 313, 473, and 770 kb, and scaffold N50 values of 292, 246, 150, and 28

Experimental evidence for milk-borne transmission of tick-borne encephalitis virus to suckling lambs.

Tick-borne encephalitis virus (TBEV) is a medically important orthoflavivirus endemic across Europe and Asia, transmitted primarily by Ixodes ticks but also through ingestion of unpasteurized milk from infected ruminants. While milk-borne human infections are well documented, experimental evidence for natural mother-to-offspring transmission in livestock remains scarce. Here, we experimentally assessed the potential for lactogenic transmission of TBEV in sheep. Eight ewes were infected subcutaneously with the European subtype TBEV strain Hochosterwitz and co-housed with 16 three-week-old lambs

Ecologic Factors Contributing to West Nile Virus Hyperendemicity in Central South Carolina: An Integrated Vector-Human-Environmental Study.

West Nile virus (WNV) is an endemic arboviral infection in the United States that has undergone phylogenetic evolution since its introduction 25 years ago. An integrated vector-human-pathogen study was conducted in the summer of 2023 to unearth contemporary Culex quinquefasciatus habitat patterns and human transmission spillover foci in South Carolina, a state with historically little WNV data. A serosurvey revealed WNV seroprevalence 10 times the national average (22% versus 2%, respectively), with unusual epidemiologic risk factors. Female Culex quinquefasciatus WNV positivity was low (2.7%)

Donor-Derived West Nile Virus Infection in Kidney Transplant Recipients, France, 2025.

We report 2 cases of donor-derived West Nile virus infection in kidney transplant recipients in France. Both recipients had mild disease develop and recovered without sequelae. A more proactive screening strategy in France, particularly during periods of highest risk for West Nile virus circulation, would help reduce risk for donor-derived infections.

Arthropod exosomal glycine-rich protein as a potential vaccine candidate effectively reduces tick blood-feeding and pathogen transmission.

During blood feeding, Ixodidae ticks secrete cement proteins, including glycine-rich proteins (GRPs), that facilitate attachment to the vertebrate host. However, the molecular mechanisms underlying exosomal GRP secretion at the feeding site and their roles in tick-pathogen interactions remain poorly understood. Here, we analyzed the Ixodes scapularis genome to identify salivary exosomal components involved in modulation of the tick-host skin interface. We identify an arthropod exosomal GRP ( XM_002400035 ) that promotes transmission of Langat virus (LGTV), a tick-borne flavivirus, from ticks t

Introduction of a European Central-South-Eastern West Nile Virus Lineage 2 Strain in Italy in 2023: Evidence from the First Locally Acquired Neuroinvasive Case in the Calabria Region.

West Nile virus lineage 2 (WNV-2) is a growing public health concern in Europe causing West Nile fever or West Nile neuroinvasive disease (WNND) with substantial morbidity and mortality; however, genomic data from southern Italy are limited despite recent expansion of autochthonous transmission. The aim of the study was to characterize the phylogenetic and molecular features of the WNV-2 strain responsible for the first autochthonous human infection reported in Calabria (2023), and two more additional WNND cases detected in 2024. Full WNV-2 genomes were generated from the three cases. Phylogen

Durable protection against lethal Rift Valley fever hepatitis and encephalitis following low-dose ΔNSsΔNSm vaccination in mice.

Rift Valley fever virus (RVFV) is a zoonotic arbovirus that infects and causes disease in both humans and livestock. In humans, RVFV infection mostly results in febrile illness but has the potential to cause severe complications such as hepatitis and encephalitis. There are currently no licensed vaccines for human use; however, several candidates have been shown to be safe and immunogenic in pre-clinical studies. However, there remain gaps in our knowledge, including the duration of immunity and efficacy of protection from divergent disease manifestations over time. This pilot study investigat
