Evaluation of protective efficacy of an inactivated Japanese encephalitis virus genotype Ⅴ strain in mice.

The detection of Japanese encephalitis virus (JEV) genotype V (GV) in humans in Korea in 2015 has raised concerns regarding its potential public health impact. Current JEV vaccines, based on genotype Ⅲ (GⅢ) strains, exhibit suboptimal neutralizing activity against JEV GV, thereby underscoring the need for genotype-specific vaccines. To address this, we developed the KNIH (GV) vaccine strain optimized for enhanced production efficiency. We evaluated its neutralizing activity and protective efficacy in a murine model. The currently available GⅢ-based vaccine (Beijing-1 strain) exhibited limited

A community-based focal serosurvey for West Nile virus infection following a surge in cases in 2024 in Kerala, India: a cross-sectional analysis.

BACKGROUND: From January to May of 2024, 27 cases of neuroinvasive disease due to West Nile Virus (WNV) were confirmed from Kerala. The number of cases were more than those seen over past three years combined. Cases were not clustered geographically and were primarily reported from four districts, viz, Kozhikode, Thrissur, Malappuram and Ernakulum. To understand the circulation of WNV, a focal serosurvey was conducted in the regions from where cases were reported. METHODS: A cross-sectional study was done in four districts of Kerala. Patients, family members and immediate neighbours were recr

Japanese encephalitis virus activates the NLRP3/caspase-1/GSDMD signaling pathway in dopaminergic neurons.

Japanese encephalitis virus (JEV) preferentially targets brain regions rich in dopaminergic neurons, including the thalamus, midbrain, and striatum, leading to severe neuroinflammation. However, the underlying pathological mechanisms remain unclear. This study hypothesizes that JEV-induced pyroptosis exacerbates neuroinflammation by activating the NLRP3/caspase-1/GSDMD signaling pathway. In vivo experiments using JEV-infected mice revealed virus-specific targeting of dopaminergic neurons, concurrent activation of the NLRP3 inflammasome, and induction of inflammatory responses. In vitro studies

Antiviral Drug Discovery from Typha angustifolia Pollen: Computational Analysis Targeting Flaviviridae Polymerases and Entry Proteins.

INTRODUCTION: For centuries, Traditional Chinese Medicine has been a subject of extensive research for its healing properties, including its effects against viruses. The pollen of Typha angustifolia emerges as a notable natural source of antiviral agents, with earlier investigations focusing on its antioxidant and antiinflammatory properties, which are associated with flavonoids and phenolics that facilitate electron transfer. These bioactive compounds could potentially disrupt viral entry and replication, thereby necessitating further studies. METHODS: Molecular docking analysis was conducte

Mitochondrial genome analysis and molecular identification of two Culex tritaeniorhynchus (Diptera: Culicidae) clades from the Republic of Korea.

Culex tritaeniorhynchus is the primary vector of Japanese encephalitis virus (JEV) and is thought to exist as 2 forms: the continental type (Ct-C) and the Japanese type (Ct-J), distinguished by variations in the cytochrome oxidase subunit 1 (COI) region of mitochondrial DNA (mtDNA). However, the extent of genetic variation within and between the 2 types remains largely unexamined. To address this gap in the literature, we performed mitochondrial genome sequencing and additionally developed a rapid and cost-effective identification method that may facilitate efficient vector management and rese

Preclinical evaluation of DENV1 mRNA vaccines in mice: Toward improved neutralizing antibody and T cell responses.

Dengue virus (DENV) infection represents a significant public health concern in tropical and subtropical areas, with increasing cases in parts of Europe. Currently, licensed vaccines, Dengvaxia® and Qdenga®, rely on live-attenuated platforms but show limited efficacy due to imbalanced immune responses and risks associated with antibody-dependent enhancement (ADE). In this study, we developed and evaluated nucleoside-modified mRNA vaccine encoding the dengue virus serotype 1 (DENV1) pre-membrane and envelope (prME) proteins. To reduce the potential for ADE, we incorporated the F108A mutation in

NUP98 regulates orthoflavivirus replication through interaction with vRNA and can be targeted for antiviral purposes.

The nuclear pore complex (NPC) is composed of multiple nucleoporins (NUPs) and enables the exchange of RNA and proteins between the nucleus and cytoplasm. NUP98 is one of the major components of the NPC, being involved in the RNA export pathway by interacting with several transport factors. Previous studies have suggested both proviral and antiviral functions of NUP98 in viral infection, yet little is known about its function in orthoflavivirus infection. In this study we show that NUP98 is a proviral cellular protein that is recruited to the cytoplasm during orthoflavivirus infection. We obse

Application of Orthoflavivirus Pseudovirus Technology in Antiviral Research.

Arthropod-borne orthoflaviviruses, including dengue, Zika, Japanese encephalitis, yellow fever and West Nile viruses, pose a significant global public health threat, causing hundreds of millions of infections annually with severe clinical symptoms. However, the lack of effective vaccines and antiviral drugs, coupled with the biosafety risks associated with handling live highly pathogenic strains, hinders progress in antiviral research. Pseudovirus technology, which uses single-round infectious viral particles lacking replication competence, has thus gained prominence as a safe and versatile to

Promoter-like activity on the minus strand supports sfRNA biogenesis in Japanese encephalitis virus.

Arthropod-borne flaviviruses produce subgenomic RNAs (sfRNAs) from the highly conserved 3'-untranslated region (UTR). While most flaviviruses generate sfRNAs by resisting degradation from the host exoribonuclease XRN1, Japanese encephalitis virus (JEV) maintains sfRNA production even when XRN1 is depleted, suggesting an alternative mechanism. Using an in vitro RNA-dependent RNA polymerase (RdRp) assay, we identified a promoter-like element on the antigenome, designated (-)sfP, which exhibits transcriptional activity comparable to the well-characterized 5' stem-loop A (5'-SLA) promoter of the v

Newcastle disease virus hijacks mitophagy to reprogram amino acid metabolism for enhanced replication.

Mitochondria serve as the cellular "power plants," supplying energy and regulating metabolism, signal transduction, and other physiological processes. To successfully replicate within host cells, viruses have evolved multiple strategies to hijack mitochondrial functions. The oncolytic Newcastle disease virus (NDV) causes severe organelle damage in tumor cells; however, how it manipulates mitochondrial architecture to facilitate its own replication remains poorly understood. Here, we provide evidence that NDV infection disrupts mitochondrial spatial distribution and imbalances mitochondrial fus

The application of DNA markers in population genetics of mosquitoes: a comprehensive review.

Mosquitoes are major vectors of pathogens causing diseases such as dengue, malaria, and Japanese encephalitis, imposing significant global public health and economic burdens. Traditional morphological approaches for mosquito research are limited by the reliance on specialized taxonomic expertise, the inability to distinguish cryptic species or immature life stages, and the susceptibility to environmental factors. DNA markers have emerged as indispensable tools to address these limitations. This review systematically summarizes the characteristics and applications of important DNA markers in mo

Downmodulation of cholesterol biosynthetic network governs activation of the innate immune response to Japanese encephalitis virus infection.

Viruses remodel metabolic processes and utilize host lipids for different stages of their life cycle. Our earlier studies have shown that the flavivirus Japanese encephalitis virus (JEV) downmodulates several key proteins involved in sterol and lipid biosynthesis. Through a lipidome analysis, we report that virus infection dysregulates nearly 41% of the cellular lipids in mouse embryonic fibroblasts (MEFs). This manifests as down-modulation of cholesterol, cholesterol esters, and glycerolipids and upregulation of ceramides and several phospholipids. Significant transcriptional downregulation o

Mosquito-capsid interactions contribute to flavivirus vector specificity.

Multiple mosquito species serve as competent vectors to carry and transmit numerous flaviviruses1,2. Several long-standing scientific questions remain to be answered, including identification of the fundamental factors that facilitate flavivirus infectivity in mosquitoes and the genetic basis that contributes to the naturally occurring interspecies specificity of mosquitoes to flaviviruses3-8, such as Aedes aegypti mosquitoes to dengue virus (DENV). Here we report that circulating mature virions are inactivated by the acidity of mosquito haemolymph; thus, extracellular vesicles carrying replic

Neutrophil depletion at the early stage of Japanese encephalitis virus infection affects CD8+ T cell infiltration into the mouse brain and causes severe encephalitis.

Neutrophils have been reported to have protective and detrimental functions in viral infections. However, the role of neutrophils remains unexplored in Japanese encephalitis virus (JEV) infection. In this study, we elucidated the dynamics of neutrophils and their influence on immune cell recruitment in subclinical and severe encephalitis in mouse models. Further, we depleted neutrophils from 3-4 week-old C57BL/6 mice using mAb1A8 (anti-Ly6G) antibody and studied their association with inflammation, viral replication, immune cell infiltration and disease outcome. We observed that an increase in

Review of selected mosquito-borne diseases: arboviruses (dengue, chikungunya, Zika, West Nile, Japanese encephalitis, yellow fever) and parasitic diseases (malaria, lymphatic Filariasis).

Mosquito-borne diseases (MBDs) are a major global public health concern, accounting for 17% of infectious diseases and causing ~700,000 annual deaths. Transmitted by Aedes, Anopheles, and Culex mosquitoes, they include viral (dengue, Zika, chikungunya), parasitic (malaria, lymphatic filariasis), and zoonotic (Japanese encephalitis) pathogens. This review outlines key MBDs' clinical features, global distribution-concentrated in tropics but expanding due to climate change and urbanization-and transmission dynamics shaped by environmental (temperature, humidity) and ecological (urban breeding sit

Serological evidence of West Nile virus circulation in Indonesia: Initial insights into an underrecognized flavivirus threat.

West Nile virus (WNV) is a mosquito-borne flavivirus primarily transmitted by Culex species and maintained in a bird-mosquito-bird cycle. Though WNV is not considered endemic to Indonesia, sporadic molecular and serological findings suggest possible under-recognized circulation. To examine this possibility, we conducted a cross-sectional serosurvey of 569 archived serum and plasma samples collected between 2021 and 2023 from non-febrile individuals, including blood donors in Papua and community-based participants in Kalimantan. Samples were initially screened using a commercial WNV IgM Enzyme
