Phytol from Sphaeranthus indicus: A Natural Insecticide Against Aedes aegypti and Culex quinquefasciatus.

BACKGROUND: Mosquitoes, as blood-feeding ectoparasites, are primary vectors for numerous pathogens that cause diseases such as dengue, lymphatic filariasis, malaria, chikungunya, and Japanese encephalitis in India. Due to the increasing resistance to synthetic insecticides, there is growing interest in developing phytochemical-based insecticides. This study aimed to evaluate the mosquitocidal potential of the hexane leaf extract of Sphaeranthus indicus, isolate its bioactive compound, and assess its ovicidal, larvicidal, and pupicidal efficacy against Aedes aegypti and Culex quinquefasciatus.

Porcine USP18 inhibits Japanese encephalitis virus replication by regulating the type I interferon signaling pathway.

Ubiquitin-specific peptidase 18 (USP18) possesses both deubiquitinating and deISGylating activities and plays a crucial role in regulating innate immune responses and antiviral defense. However, the function of USP18 in Japanese encephalitis virus (JEV) replication remains unclear. In this study, the expression pattern of the porcine USP18 gene following JEV infection was examined by quantitative real-time PCR (qRT-PCR) and Western blot analysis. The effects of USP18 overexpression and siRNA-mediated knockdown on JEV replication and the expression of type I interferon (IFN-I) signaling pathway

Association of parental vaccination readiness and descriptive norms with childhood vaccination status.

BACKGROUND: Maintaining high vaccination coverage requires understanding parental psychological factors, such as readiness conceptualized by the 7C model and descriptive norms, that shape decisions about childhood vaccinations. This study aimed to validate the Japanese short version of the Children-7C (C7C) scale, and examine how parental readiness and descriptive norms relate to vaccination status. METHODS: This cross-sectional study analyzed data from 5148 parents in a self-reported online survey. Structural validity and internal consistency were assessed using confirmatory factor analysis

NAD+ Homeostasis Attenuates Japanese Encephalitis Virus Infection Progression.

Nicotinamide adenine dinucleotide (NAD+) is a crucial molecule involved in numerous interconnected metabolic processes. Due to its implication in multiple viral infection responses, maintaining NAD+ homeostasis has become a promising target for host-directed therapies. Japanese encephalitis virus (JEV) causes severe, fatal encephalitis with irreversible brain damage and long-lasting neurological deficits in survivors. However, the potential interaction between JEV infection and NAD+ metabolism remains largely unclear. In this study, we found that JEV infection dysregulates NAD+ metabolism and

Genotype-resolved NS5 stability predicts Japanese encephalitis virus fitness.

Japanese encephalitis virus (JEV) remains a major health threat across Asia, yet the contribution of genotype-specific variation in the multifunctional NS5 protein to viral fitness is not fully resolved. This study evaluated how sequence differences among JEV genotypes G1-G5 shape NS5 stability and, in turn, replication potential. A unified in-silico workflow combined physicochemical profiling, residue-level substitution mapping, and atomistic molecular dynamics to compare structural stability and conformational behavior across genotypes, with a focus on substitutions predicted to modulate enz

Neuroinflammation and blood-brain barrier disruption in Japanese encephalitis virus infection: mechanisms, therapeutic targets, and intervention strategies.

Japanese encephalitis virus (JEV), as a neurotropic flavivirus, remains a primary cause of viral encephalitis and poses a significant public health challenge. Its pathogenic mechanism centers on the pathological interaction between blood-brain barrier (BBB) disruption and uncontrolled neuroinflammation, which collectively promote JEV neuroinvasion and result in irreversible brain damage. Therefore, elucidating the molecular mechanisms by which JEV disrupts the BBB and induces neuroinflammation is crucial for developing novel therapeutic strategies. This paper systematically elucidates the mech

Hydranencephaly and porencephaly predominated among neuropathologic findings in stillborn and neonatal piglets during the first major outbreak of Japanese encephalitis in Australia.

In 2022, piggeries in southeastern Australia experienced an increase in reproductive losses and occasional neurologic disease in neonates with up to 100% mortality in some litters. Molecular testing identified a genotype IV Japanese encephalitis virus (JEV) as the etiological agent, detected for the first time in Australia. Necropsy of 235 JEV-positive domestic piglets, mostly stillbirths with 30% mummified, revealed hydranencephaly-porencephaly impacting the dorsal cerebrum as the most common lesion. Microscopically, this was characterized by varying degrees of parenchymal collapse as a resul

Cytokine-based rapid process control of live virus production: MCP-1 as a cross-family biomarker in Vero cells.

Rapid determination of infectious virus titers is essential for efficient vaccine process development, yet conventional assays such as TCID50 and plaque assays require several days and limit throughput. Cytokine secretion represents a potential surrogate marker, enabling rapid detection of productive infection. Here, we evaluated cytokine secretion in Vero cells infected with live vaccine candidates from the Flaviviridae (Japanese encephalitis virus [JEV], yellow fever virus [YFV], Zika virus [ZIKV]) and Togaviridae (Chikungunya virus [CHIKV]) families. Cytokine profiling revealed consistent i

NS4B and NS5 mutations expand the temperature range permissive for the replication of dual-host affiliated insect-specific flaviviruses in mosquito cells.

The genus Orthoflavivirus within the family Flaviviridae includes mosquito-borne flaviviruses (MBFs) and dual-host affiliated insect-specific flaviviruses (dISFs). MBFs infect both vertebrates and mosquitoes, while dISFs are thought to infect only mosquitoes, despite their phylogenetic proximity to MBFs. Some dISFs have been shown to be capable of infecting mammalian cells in the setting of a suppressed immune response and subphysiological temperatures. Therefore, temperature sensitivity is considered one of the key factors restricting viral host tropism. To investigate the effect of temperatu

Transcriptomic insights into the neuropathogenic and immune response alterations in mouse neurons challenged with west nile and japanese encephalitis viruses.

West Nile virus (WNV) and Japanese encephalitis virus (JEV) are major mosquito-borne pathogens that cause severe neurological illnesses in humans, including encephalitis and meningitis. Neurons are the primary target of WNV and JEV infections in the brain. However, the pathogenic mechanisms and host immune response associated with WNV and JEV infections in the primary neurons are not fully characterized. Herein, using high-throughput RNA sequencing (RNA-Seq), we performed a comparative transcriptomic analysis of the primary mouse neurons following infection with WNV and JEV. We identified diff

Timeliness of Routine Vaccination, Catch-Up Completion, and Immune Function in Chinese Children with Special Healthcare Needs: A Retrospective Cohort Study.

BACKGROUND: Children with special healthcare needs (CSHCNs) face persistent barriers to timely immunization in China, but comparative evidence across disease groups and vaccines, and data on immune function, are limited. METHODS: We conducted a retrospective cohort study linking electronic medical records, vaccination records, and a structured telephone and questionnaire follow-up. We estimated timely vaccination by National Immunization Program (NIP) dose definitions, assessed catch-up completion at follow-up, and compared cellular/humoral/complement immune indices with published pediatric r

Cocrystals of telmisartan with ascorbic acid: Enhanced solubility and antiviral potency against Japanese encephalitis virus.

The significant impact of Japanese encephalitis virus (JEV) on society and the lack of any approved antiviral drugs demand urgent attention. Considering the prohibitive cost involved in new antiviral development, repurposing existing drugs is an alternative strategy. Telmisartan (TM) is a prime candidate for repurposing against JEV due to its ability to inhibit JEV infection. Poor aqueous solubility of TM can be a major hurdle in harnessing its repurposing against JEV. Cocrystallization with appropriate coformers has the scope to improve solubility and antiviral properties. Ascorbic acid (Asca

A Bovine Cell Line Resistant to Japanese Encephalitis Virus Entry but Permissive to Post-Entry Replication.

Japanese encephalitis virus (JEV) is a mosquito-borne zoonotic orthoflavivirus that poses a significant global health threat. It causes severe neuroinflammatory disease in humans and reproductive failure in swine. Because of the broad host range and cell tropism of JEV, identifying animal cell lines resistant to infection has been a persistent challenge. In this study, we demonstrate that Madin-Darby bovine kidney (MDBK) cells are resistant to JEV infection yet remain fully permissive to viral replication when transfected with viral genomic RNA. Using immunoblotting, immunofluorescence, and fl

mRNA Vaccine Against Japanese Encephalitis Virus Genotype IV Protects Against Lethal Infection.

In 2022, Australia saw an unprecedented outbreak of Japanese encephalitis virus genotype IV (JEV GIV). The outbreak involved 42 human cases with 7 fatalities, as well as affecting >80 pig farms in New South Wales and Queensland. Herein, we designed, constructed, and tested two JEV GIV mRNA vaccines encoding prME, which provided protection against a lethal JEV GIV challenge in an Ifnar-/- mouse model. The vaccines were not codon optimized and included either the Native (full-length) or a Shorter signal peptide, with the latter missing the N-terminal n-region. Two vaccinations with 5 µg of the S

1'- and 4'-Cyano Modified Adenosine Analogs Against Prototypic Flavivirus RNA-Dependent RNA Polymerases.

Flaviviruses are arthropod-borne RNA viruses associated with significant human diseases globally. There are no effective direct-acting antivirals approved to treat these viral infections. Given its critical role in viral replication, the RNA-dependent RNA polymerase (RdRp) is a logical target for antiviral drug development. Remdesivir (formerly GS-5734), a 1'-cyano modified C-adenosine monophosphate prodrug, was the first US Food and Drug Administration (FDA) approved antiviral for coronavirus disease 2019 (COVID-19) and was also shown to inhibit flavivirus replication. Japanese Encephalitis: Understanding Its Impact, Prevention, and the Role of Vaccination.

Japanese encephalitis (JE) is a mosquito-borne viral disease and remains an important cause of viral encephalitis in Asia and parts of the Western Pacific. Earlier WHO-supported global burden estimates published around 2014 suggested approximately 68,000 clinical cases and up to 20,400 deaths annually; however, the World Health Organization emphasises that the true burden of JE is difficult to quantify due to substantial underreporting and heterogeneous surveillance systems. Although vaccination programs have led to marked declines in incidence in several countries, JE transmission persists in
