In silico Exploration of Aegle Marmelos-Derived Compounds as Potential Inhibitors of NS5 RdRp in Japanese Encephalitis Virus.

INTRODUCTION: Japanese encephalitis virus (JEV) hijacks the host cell's endoplasmic reticulum (ER) to form assembly and replication complexes during replication, leading to the production of multiple copies of JEV negative-sense and positive-sense RNA strands. The nonstructural protein NS5 RNA-dependent RNA polymerase (RdRp) is conserved across all Flavivirus species, and its inhibition can suppress Flavivirus replication. The objective of this study is to evaluate the inhibitory potential of 126 Aegle marmelos-derived compounds on JEV NS5 RdRp using computational methods. METHODS: A computat

Interplay between flaviviruses and the interferon response: mechanisms of immune evasion and therapeutic implications.

The genus Orthoflavivirus encompasses a group of medically significant arthropod-borne viruses, such as dengue virus (DENV), Japanese encephalitis virus West Nile virus Zika virus (ZIKV), and others, which pose persistent global public health threats. The host interferon (IFN) system constitutes a pivotal first line of defense against these viral infections. However, orthoflaviviruses have evolved a remarkable array of sophisticated strategies to antagonize both the induction and signaling pathways of type I and III IFNs. This review systematically summarizes the mechanisms by which orthoflavi

Molecular evidence of Japanese encephalitis virus in poultry: Need to further elucidate JEV epidemiology.

Japanese encephalitis (JE) is a re-emerging viral zoonotic disease transmitted primarily by the bite of infected Culex mosquitoes with ardeid birds as reservoir host for JE virus (JEV). Swine being the amplifier host have been studied extensively, however the role of domestic poultry in JEV epidemiology remains unexplored. Therefore, the present study was undertaken with the aim of quantifying JE viral load and to identify the circulating genotype of JEV in domestic chicken and duck population of India. A total of 206 chicken and 76 duck samples collected across five JE endemic states of India

Biosensors for the detection of arthropod-borne veterinary viruses: A comprehensive review.

Veterinary arboviruses pose serious threats to animal health, especially in endemic areas. Traditional methods used for the diagnosis of these viruses are expensive, time-consuming, and require specialized personnel. This review discusses biosensor technologies for veterinary arboviruses, analyzing specific targets, mechanisms, sensitivity, specificity, and utility for use in the field. The remaining gaps were also mentioned. A comprehensive literature search was carried out in the major databases from 2015 to 2025. We used the keywords "Biosensing Technologies"Arboviruses," "Veterinary Medici

Japanese encephalitis virus envelope protein activates the TLR4/NF‑κB pathway to induce testicular inflammation.

Japanese encephalitis virus (JEV), a mosquito-borne flavivirus, remains a leading cause of viral encephalitis in Asia and poses serious threats to reproductive health. However, the molecular mechanisms underlying JEV-induced testicular inflammation remain incompletely understood. In this study, an integrated approach involving proteomic, cellular, and animal experiments was used to investigate the role of Toll-like receptor 4 (TLR4) in JEV-triggered inflammatory responses. Proteomic analysis and in vitro assays revealed that JEV infection upregulated TLR4 expression and activated the NF‑κB sig

Adult Japanese encephalitis in a non-endemic area: A case highlighting diagnostic limitations of targeted next-generation sequencing.

Japanese encephalitis (JE) is a mosquito-borne viral disease that is rarely reported in non-endemic regions. We describe an adult patient presenting with acute meningoencephalitis in northern China, a non-endemic area. Targeted next-generation sequencing (tNGS) performed twice on cerebrospinal fluid was negative because JE virus was not included in the detection panel. The diagnosis was confirmed by a strongly positive JE IgM antibody. This case highlights that tNGS has inherent diagnostic limitations depending on pathogen coverage. In non-endemic areas near endemic regions, JE should be consi

Biochemical, molecular, and in silico insights into the effects of cyphenothrin on Culex quinquefasciatus mosquitoes.

BACKGROUND: Cyphenothrin, a type II synthetic pyrethroid, is widely recommended by the World Health Organization (WHO) for control programs for mosquitoes such as Culex quinquefasciatus. This cosmopolitan mosquito species plays a pivotal role in transmitting several neglected tropical diseases, including lymphatic filariasis, West Nile virus, Japanese encephalitis, and so on. Besides treatment, vector control programs heavily rely on insecticides, leading to varying resistance due to prolonged exposure. METHODS: The present study assessed the susceptibility status of field-collected Cx. quinq

Spatial transcriptomics uncovers vasculature-centered cellular interactions driving Japanese encephalitis progression in a mouse model.

The cellular interactions that drive the neuropathology of most neuroinvasive viruses remain elusive. We used Japanese encephalitis virus (JEV) to infect female BALB/c mice and applied Stereo-seq to simultaneously capture host and viral transcriptomes in situ, thereby constructing a comprehensive spatiotemporal atlas of Japanese encephalitis (JE) pathogenesis. Our analysis pinpoints Ly6c2+ monocytes as the primary virus carrier, the most abundant infiltrating immune cell type, and a major source of IFN-γ in the infected mouse brain. Following infection, Ackr1+ endothelial cell activation is li

Isolation and characterization of Japanese encephalitis virus genotype I from pig provides evidence for the presence of the virus in nasal secretion and co-circulation of JEV genotypes in Assam, India.

BACKGROUND: Japanese encephalitis (JE) is a leading cause of viral encephalitis, among children, in many Asian countries despite the availability of effective vaccines. There are five JE virus genotypes (GI through GV), with GIII being the most prevalent. However, in the past three decades GI has emerged as the dominant genotype across several Asian countries, while the reappearance of GV is a concern due to the reduced cross-neutralization offered by existing GIII-based vaccines. Although both GI and GIII have been reported to co-circulate in India, all previous JEV isolations from pigs have

A time-varying geospatial model of habitat suitability for Japanese encephalitis virus vectors and vertebrate hosts in Australia.

In the austral summer of 2021-2022, Australia experienced an unprecedented Japanese encephalitis virus (JEV) outbreak, with detections over 3000 km south of previous occurrences. Given the limited knowledge of JEV transmission ecology in Australia, we developed geospatial models of transmission risk to support the public health response. We created time-varying habitat suitability models for suspected mosquito vectors and ardeid hosts using month-scaled occurrence and covariate data from 2000-2023. Ardeid host presence-absence data were obtained from eBird and BirdLife Australia, with habitat

Progress and Innovation in Japanese Encephalitis Vaccines: Toward Global Immunization.

Japanese encephalitis (JE) remains a major public health concern, especially in Asia and the Western Pacific, where more than 3 billion people are at risk. Despite advances in vaccine development, challenges such as viral strain diversity, limited crossprotection, and access barriers persist. JE directly impacts economic development because disease patterns are influenced by factors such as healthcare infrastructure, surveillance, vaccine availability, and agricultural trends across low-, middle-, upper-, and high-income countries. Current vaccines primarily target genotype III strains, but ge

Inactivated Japanese encephalitis virus vaccination imprints fusion loop-biased antibody responses that are attenuated by repeated live-attenuated dengue vaccination.

Immune imprinting, also known as immune history, is a core aspect of adaptive immunity that influences antibody responses to future antigen exposures. Nevertheless, the impact of sequential flavivirus vaccinations on epitope targeting and antibody activity in humans remains incompletely understood. This question is particularly important in regions where the inactivated Japanese encephalitis virus (JEV) vaccines and the live-attenuated dengue virus (DENV) vaccines are used, as both have been associated with an increased risk of symptomatic dengue infection and severe illness. We studied the im

Umbelliferone Inhibits Zika Virus (ZIKV) NS5 Activity and Replication In Vitro.

This study focused on the inhibitory effects of umbelliferone on Zika virus (ZIKV) NS5 activity and viral replication in vitro. Umbelliferone exhibited no detectable cytotoxicity and inhibited ZIKV replication while showing no activity against Japanese encephalitis virus (JEV), another member of the Flaviviridae family. Structure-activity relationship studies on structurally related derivatives of umbelliferone revealed that the hydroxyl group at the C7 position is critical for suppression of ZIKV replication. To identify the stage of the ZIKV life cycle targeted by umbelliferone, we performed

Knockdown resistance (kdr) mutations in the Japanese encephalitis virus vector Culex tritaeniorhynchus from the Republic of Korea.

BACKGROUND: Culex tritaeniorhynchus is the primary vector of Japanese encephalitis virus (JEV), a disease of global public health significance, and is widely distributed across Africa, Asia, Australia, and Europe. Despite its epidemiological importance as a JEV vector, studies on insecticide resistance in Cx. tritaeniorhynchus populations from the Republic of Korea (ROK) remain extremely limited. This study aimed to investigate the spatial and temporal occurrence of knockdown resistance (kdr) mutations in Cx. tritaeniorhynchus populations from the ROK. Additionally, the association between phe

A Review of Japanese Encephalitis Vaccines with Respect to Potential Veterinary Use.

BACKGROUND: Japanese encephalitis virus (JEV) is a reemerging vector-borne orthoflavivirus of significant public and animal health concern. Infection with JEV can cause inapparent, mild, or fatal encephalitis in humans and animals. The virus, which circulates primarily in Southeast Asia, has caused outbreaks in humans and livestock in nonendemic regions, leading to human disease and significant economic loss. The 2022 Australian outbreak of JEV led to renewed interest in JEV countermeasures available in the United States for veterinary use as the Australia outbreak primarily affected commercia

Alignment-Free Guided Design of a Pan-Orthoflavivirus RT-qPCR Assay.

The co-circulation and rapid expansion of the genus Orthoflavivirus, including dengue virus (DENV), Zika (ZIKV), and Japanese encephalitis virus (JEV), pose significant global health challenges. Developing inclusive pan-genus molecular diagnostics is hindered by high nucleotide divergence (>25%-30%) and the computational limitations of traditional multiple sequence alignment in detecting conserved motifs across large datasets. To overcome these limitations, we developed a systematic alignment-free design pipeline that uses rigorous k-mer analysis and compacted De Bruijn graphs. We analyzed 11,

CD4 Molecule Plays an Important Role in the Inflammatory Response Induced by Japanese Encephalitis Virus Infection.

Japanese encephalitis virus (JEV) is an important flavivirus that causes zoonotic and arboviral diseases. Infection with JEV not only induces acute central nervous system (CNS) infectious diseases but also leads to reproductive disorders. Currently, research on the pathogenic mechanism of JEV has mainly focused on CNS inflammation caused by infection, while studies on the pathogenic mechanism of JEV targeting the reproductive system are relatively scarce. This study used TM3 cells as a model to investigate the regulatory role of the CD4 molecule in JEV infection, the STAT1 signaling pathway, a

Japanese encephalitis virus NS1' subunit vaccine confers protection against viral challenge and mosquito-mediated transmission.

Japanese encephalitis virus (JEV) is a mosquito-borne pathogen that threatens human health and the livestock industry. Current JEV vaccines, including inactivated and live attenuated types, still present several limitations, underscoring the urgent need for new, safe, and broad-spectrum JEV vaccines. Antibodies targeting the orthoflavivirus NS1 protein, which is highly conserved among orthoflaviviruses, have been demonstrated to provide protection against viral infection, with NS1' being known as an extended form of NS1 in the Japanese encephalitis serogroup. In this study, we successfully con
