Comparative Evaluation of Carbohydrate, Amino Acid, and Ionic Liquid Excipients for Flavivirus Vaccine Stabilization.

Live-attenuated flavivirus vaccines (yellow fever, dengue, and Japanese encephalitis) exhibit poor thermal stability in liquid formulations, requiring lyophilization and storage between 2 and 8 °C to maintain potency. We evaluated carbohydrates, amino acids, and choline-based ionic liquids as preservatives for vaccine-like flavivirus. Among the carbohydrates tested, trehalose and sucrose provided greatest stabilization, while histidine demonstrated the strongest stabilizing effect among amino acids. Trehalose-histidine and sucrose-histidine combinations produced synergistic effects, preserving

Japanese encephalitis virus degrades TRAF3 to suppress type I interferon and promote viral replication through NS5 and host TUFM proteins.

Japanese encephalitis (JE), caused by the Japanese encephalitis virus (JEV), remains a major global health concern. A deeper understanding of how JEV manipulates host antiviral responses is critical for developing more effective therapeutic strategies. In this study, we identified a novel immune evasion mechanism through which JEV suppresses host innate immunity to facilitate viral replication. Specifically, JEV infection induces the expression of the host translation elongation factor TUFM by activating the transcription factor JUN. We demonstrated that TUFM promotes JEV replication, while TU

Antimicrobial peptide WK-13-3D inhibits Japanese encephalitis virus infection by interacting with viral particles, potentially targeting the envelope protein.

Despite causing fatal encephalitis and long-term neurological, cognitive, and behavioral impairments in survivors, no antiviral drugs are currently available to treat Japanese encephalitis virus (JEV) infections, highlighting the urgent need for effective therapies. Antimicrobial peptides (AMPs) exhibit broad-spectrum antibiotic activity; LL-37 is one such AMP that demonstrates a broad antiviral effect against various viruses. However, some viruses resist LL-37 and its analogues, emphasizing the importance of studying the impact of specific antimicrobial peptides on different viruses. In this

Japanese encephalitis among children in India: proportion of cases, case fatality, and epidemiological patterns - a systematic review and meta-analysis.

BACKGROUND: Japanese Encephalitis (JE) remains a major cause of viral encephalitis in India, disproportionately affecting children in endemic regions. Although vaccination programs have reduced incidence in several districts, seasonal outbreaks and pediatric mortality continue to be reported. This systematic review and meta-analysis aimed to estimate the pooled proportion of children among laboratory-confirmed JE cases and the pooled case fatality proportion among pediatric JE cases in India. Secondary objectives included narratively synthesizing morbidity patterns and vaccination coverage. ME

Pestiviruses utilize pyrimidine metabolism to regulate mitophagy for viral replication.

DHODH (dihydroorotate dehydrogenase (quinone)) has been demonstrated as a critical regulator of programmed cell death, yet its role in macroautophagy/autophagy remains poorly defined. Flaviviridae pose a significant threat to global public health, and their replication is closely associated with autophagy. Building upon our previous findings that DHODH was a broad-spectrum target for Flaviviridae and a key regulator of Pestiviruses replication, this study employed RNA-seq screening coupled with functional validation to demonstrate that DHODH affected Pestiviruses replication by regulating mito

Genetic variations in three insecticide targets in the disease vector Culex quinquefasciatus from Mianyang, China: simultaneous detection of novel mutations RDL A296G and VGSC A1007T.

Culex quinquefasciatus is a major vector of filariasis and Japanese encephalitis in China. The application of insecticides is a regular measure for prevention and control of such mosquito-borne diseases. However, insecticide resistance, often caused by target-site mutations, threatens control efforts. In Mianyang City, a region previously reporting high pyrethroid resistance, the current genetic basis of resistance remains unclear. To address this gap, resistance-related mutations in three key insecticide targets, including acetylcholinesterase (AChE, encoded by ace1, conferring organophosphat

Development of a Reverse Transcription Recombinase Polymerase Amplification CRISPR/Cas12a Assay for Visual and Highly Specific Identification of Zika Virus.

Zika virus (ZIKV), a single-stranded positive-sense RNA virus of the Flaviviridae family (Flavivirus genus), causes acute febrile illness and severe congenital anomalies. Serological cross-reactivity with Dengue virus (DENV) and Chikungunya virus (CHIKV) complicates diagnosis, underscoring the urgency of developing specific point-of-care tests for early detection, outbreak mitigation, and reduced misdiagnosis risks. This study established and optimized an RPA-CRISPR/Cas12a assay for the rapid, visual, and highly specific detection of ZIKV. Primers and crRNAs targeting the highly conserved caps

Microbiome Simplification During Metamorphosis in Larva and Adults of Armigeres subalbatus (Coquillett, 1898) (Culicidae) Revealed by Shotgun Metagenomics.

Armigeres subalbatus is medically significant vector for filarial worms and the Japanese encephalitis virus. Shotgun metagenomic sequencing was employed to investigate the bacterial communities in A. subalbatus mosquitoes. The diversity metrics (Shannon H', Simpson 1-D, Berger-Parker) were calculated for larval and adult stages. De novo assembly and binning were used to recover metagenome-assembled genomes (MAGs) with > 82% completeness and < 4% contamination. Functional profiling assessed gene expression via transcripts per million (TPM) and clusters of orthologous groups (COG) categories. La

Investigating antibody cross-reactivity and transmission dynamics of alphaviruses and flaviviruses using a multiplex serological assay.

Accurate serological tools are essential for monitoring the transmission of arboviruses with pandemic potential, yet cross-reactivity between closely related viruses hampers diagnostics and surveillance. Here, we develop a high-throughput multiplex serological assay to quantify antibody responses to 28 antigens from nine arboviruses (dengue, Zika, yellow fever, West Nile, Usutu, Japanese encephalitis, chikungunya (CHIKV), Mayaro (MAYV), and O'nyong-nyong virus) and apply it to over 4000 samples from epidemiologically distinct sites on four continents. We implement a flexible analytical method

Susceptibility of wild and domestic songbirds to Usutu virus.

Usutu virus (USUV) is an emerging mosquito-borne orthoflavivirus that can cause neuroinvasive disease in humans and wild birds. USUV clusters phylogenetically within the Japanese encephalitis virus serocomplex, sharing antigenic and ecological similarity with West Nile virus (WNV). USUV is maintained in an enzootic cycle primarily involving passerine birds and Culex spp. mosquitoes. USUV was first isolated in South Africa in 1959 and has since spread throughout Africa and Europe, causing mortality and disease in several wild bird populations, specifically the Eurasian blackbird (Turdus merula)

Japanese encephalitis virus orchestrates GLUT4-mediated glucose metabolism to potentiate viral replication via insulin receptor signaling.

Источник пометил публикацию как отозванную

Flaviviruses intricately rewire host metabolic networks to establish a replication-permissive environment; however, the role of glucose transporter-mediated uptake, particularly via glucose transporter 4 (GLUT4), remains insufficiently defined. Japanese encephalitis virus (JEV) infection induces extensive remodeling of glucose metabolism, exemplified by the coordinated upregulation of critical metabolic effectors. Pharmacological blockade of glucose metabolic pathways markedly attenuates JEV replication, whereas exogenous glucose supplementation enhances viral propagation in a concentration-de

THE MOSQUITO FAUNA OF K'GARI (FRASER ISLAND), AUSTRALIA, THE WORLD'S LARGEST SAND ISLAND.

K'gari (Fraser Island) is the largest sand island in the world and a popular tourist destination. Despite its importance as a United Nations Educational, Scientific and Cultural Organization (UNESCO) World Heritage Site, there is a paucity of information regarding its mosquito fauna. From 1968 to 1976, several qualitative entomological surveys were carried out on K'gari, but no published work has been undertaken since. This study presents quantitative findings from three surveys undertaken using CO2-baited light traps in April and October 2024, and May 2025. In total, 3,356 mosquitoes, represe

Early Detection of Asymptomatic Human Infections in Vector-Borne Diseases: A Programmatic Model for Enhanced Surveillance.

Global control strategies for vector-borne diseases (VBDs) remain largely anchored in the detection and management of symptomatic cases, despite substantial evidence that a considerable proportion of infections-often exceeding 60% in several major arboviral diseases-are asymptomatic or minimally symptomatic. These asymptomatic human reservoirs can sustain low-level pathogen circulation during interepidemic periods and may amplify transmission once ecological, immunological, or entomological thresholds are crossed. Because vector-dependent transmission is shaped by environmental variability, ve

Using electron microscopy to observe ultrastructural changes occurring in BHK-21 cells during cellular infection by Japanese encephalitis virus.

Japanese encephalitis virus (JEV), a mosquito-borne flavivirus and a leading cause of viral encephalitis in Asia, remodels host cellular membranes extensively during its life cycle. Although classical literature describes JEV budding into the endoplasmic reticulum (ER), accumulating within ER-derived vesicles, and maturing through the secretory pathway, detailed electron microscopy (EM) analyses have been severely limited. BHK-21 cells are widely used for propagating JEV and have served as a model for several virological studies. However, comprehensive ultrastructural characterization of steps
