The association between vaccine coverage and herd protection: exploratory analyses of a cluster-randomised trial of Vi conjugate vaccine.

BACKGROUND: Indirect protection from the typhoid conjugate vaccine (TCV) has been negligible, possibly due to vaccination being restricted to children, which may have limited the overall vaccine coverage needed to sufficiently interrupt transmission dynamics across endemic populations. In this study herd protection was evaluated in households and schools with different TCV coverages by conducting secondary analyses of a double-blind cluster-randomised trial on TCV in Bangladesh. METHODS: TyVAC Bangladesh was a cluster-randomised trial conducted in Dhaka, Bangladesh between April 2018 and Marc

Japanese encephalitis virus NS1 and NS4B synergistically target TLR3 signaling to promote viral replication.

UNLABELLED: To establish effective infection, viral pathogens employ diverse strategies through encoded proteins to interfere with host antiviral responses. While previous studies have predominantly focused on elucidating the mechanisms by which individual viral proteins regulate type I interferon (IFN) responses, this study presents the first demonstration that Japanese encephalitis virus (JEV)-encoded NS1 and NS4B proteins cooperatively target the TLR3 receptor signaling pathway to suppress IFN production. Here, we first discovered JEV-encoded multifunctional glycoprotein NS1, which inhibits

25-Hydroxycholesterol Restricts Japanese Encephalitis Virus via Metabolic Suppression of the SREBP2-Mediated Signaling Axis.

Host lipid metabolism is a critical determinant of viral pathogenesis. Although the interferon-inducible cholesterol 25-hydroxylase (CH25H) typically acts as a broad-spectrum antiviral protein, its expression and regulatory patterns during Japanese Encephalitis Virus (JEV) infection display unique features. Here, we demonstrate that 25-hydroxycholesterol (25HC), the product of CH25H, potently inhibits JEV proliferation by suppressing SREBP2 activation. Distinct from the majority of viral infections that induce CH25H upregulation, JEV infection elicits a transient reduction in CH25H abundance i

The Host NADase CD38 Promotes JEV Replication by Targeting the NAD+/SIRT1 Axis.

The manipulation of host cellular metabolism is a key strategy for flaviviruses like Japanese encephalitis virus (JEV) to establish a productive infection. This study identifies the host NADase CD38 as a central regulator of this process. Using a CRISPR/Cas9-generated CD38 knockout (KO) TM3 cell model, we found that CD38 deficiency significantly restricted the production of infectious viral particles. While loss of CD38 also partially impaired viral entry, our central finding is that CD38 primarily promotes JEV infection by suppressing a host-intrinsic metabolic defense. We show that CD38 defi

Establishing and sustaining mosquito colonies: insights into morphology, bionomics, and advances in the rearing of Anopheles, Culex, and Aedes.

Mosquitoes of the genera Anopheles, Culex, and Aedes are of major medical importance as vectors of malaria, filariasis, Japanese encephalitis, dengue, chikungunya, and Zika. Understanding their morphology and bionomics is fundamental for vector biology, ecological studies, and the design of effective control strategies. Laboratory colonies of mosquitoes serve as an indispensable resource for investigating mosquito genetics, physiology, and vector-pathogen interactions, while also enabling the evaluation of new interventions such as Wolbachia-based control and the sterile insect technique. This

Importation risks and local transmission of Japanese encephalitis virus in Taiwan.

Japanese encephalitis (JE) remains a significant threat to the human, horse, and pig industries throughout the Asia-Pacific region. Emerging genotype V and IV JE virus (JEV) can evade vaccine-induced immunity and exacerbate the disease burden after spreading into JE-naïve or epidemic areas through the movement of mosquitoes, pigs, and birds. This study aimed to assess the risk of viral importation into Taiwan by conducting mosquito-based surveillance. Our phylogenetic results revealed that at least six JEV importation events occurred in Taiwan between 1958 and 2024. Over the past 16 years of m

Dual role of Japanese encephalitis virus fusion loop peptide antibodies in Zika virus infection.

Zika virus (ZIKV) is a key member of the Flavivirus genus that has emerged as a major global public health concern. The fusion loop region (residues 98-110), located within domain II of the envelope protein, is highly conserved among flaviviruses, including ZIKV and Japanese encephalitis virus (JEV). However, the functional consequences of such conservation for cross-reactive immunity remains unclear. Here, we integrated bioinformatic analyses, functional assays in vitro and mouse models in vivo to systematically determine the effects of antibodies directed against the JEV fusion loop (FL) reg

An orthoflavivirus inhibitor targeting multifunctional NS2A protein, a previously unidentified target.

Orthoflaviviruses, such as dengue virus (DENV), Zika virus (ZIKV), West Nile virus (WNV), Japanese encephalitis virus (JEV), and yellow fever virus (YFV), constitute a significant public health concern with billions of people at risk of infection. Climate change and the expanding geographical distribution of mosquito vectors transmitting orthoflaviviruses have increased their potential to cause large-scale disease outbreaks. The frequency and severity of disease outbreaks highlight the urgent need for a broad-spectrum antiviral agent targeting orthoflaviviruses. In this work, we conducted a co

Porcine CD161 promotes Japanese encephalitis virus entry: a potential host-directed antiviral target.

Japanese encephalitis virus (JEV) remains a major global zoonotic threat to both animal and human health. Viral attachment to host cells is an early and targetable step in infection. Here, we investigated the role of the C-type lectin-like receptor CD161 (encoded by Killer cell lectin-like receptor B1, KLRB1) in JEV infection using PK15 cells. The stable CD161-knockout PK15 cell lines (PK15-K.O.) and CD161-overexpressing PK15 cell lines (PK15-O.E.) were generated using CRISPR/Cas9 genome editing and lentiviral transduction, respectively. Following JEV infection, PK15-K.O. cells showed markedly
