Mosquito-transmitted alphaviruses, including chikungunya (CHIKV), Mayaro, Eastern equine encephalitis, and Venezuelan equine encephalitis viruses, cause severe arthritic or neurologic disease in humans. There are currently no approved antiviral treatments against alphavirus infection. Here, we report that the purine ribonucleoside EIDD-3525 is a potent inhibitor of alphavirus replication in vitro with broad spectrum antiviral activity against the Alphavirus genus. Using a trans-replicase assay, we find that EIDD-3525 blocks alphavirus RNA replication. Mutational profiling suggests that EIDD-35

Toward a more detailed understanding of the Eastern equine encephalitis virus (EEEV) cycle, individual blood-fed Culiseta melanura (Coquillett) mosquitoes were collected at an enzootic focus in New York State. Blooded females were tested for EEEV by multiplex real-time quantitative reverse transcription polymerase chain reaction, qRT-PCR. Host bloodmeals were identified using DNA amplification and sequencing of the cytochrome B gene. In 2018, 577 individual mosquitoes were tested. Virus was detected in the bodies of 16, of which two had the virus in their legs. In 2022, 606 individual mosquito

We present an updated checklist of mosquito species in New York State, integrating historical records, modern surveillance, and recent literature. A total of 68 species across 10 genera are documented, including invasive taxa (Aedes albopictus, Ae. japonicus) and historically rare species (Orthopodomyia alba, Or. signifera). This synthesis emphasizes species of public and veterinary significance, such as Culex pipiens and Culiseta melanura, vectors of West Nile and eastern equine encephalitis viruses. Analysis of more than 3 million specimens collected since 1999 confirms the absence of Ae. ae

Coquillettidia perturbans (Walker) is a North American mosquito species of medical and veterinary importance that has been implicated as a vector of eastern equine encephalitis virus. Larval and pupal C. perturbans possess highly specialized respiratory appendages that enable them to respire from air vesicles in the hollow roots and shoots of emergent aquatic vegetation, which means that the larval and pupal stages have remained largely understudied. Field-collected C. perturbans larvae, pupae, and adults were studied under laboratory conditions in an attempt to establish a laboratory colony.

Self-replicating RNA (srRNA) technology has received its first approval with Kostaive and Emergency Use Authorization for GEMCOVAC COVID-19 vaccines. The effective doses for srRNA vaccines are far lower than for conventional mRNA, as srRNA drives greater protein expression and has attributes of viral vectors that can potentiate immunogenicity. Importantly, srRNA encodes for a viral replicase, frequently derived from non-structural proteins (nsP) from Venezuelan equine encephalitis virus (VEEV), that helps amplify the encoded transgene. nsPs also influence critical cellular processes, including

Forecasting zoonotic mosquito-borne viruses remains a critical challenge because transmission depends on dynamic, multitrophic interactions among vectors, hosts, pathogens, and the environment. We integrate long-term sentinel chicken surveillance in Florida with environmental data to build a predictive framework for eastern equine encephalitis virus (EEEV), a zoonotic mosquito borne disease of concern to human and equine health. Models captured environmental drivers and latent spatiotemporal structure, achieving strong predictive accuracy. Models also revealed nonlinear effects of moderate pre

Eastern equine encephalitis (EEE) virus (EEEV) is a mosquitoborne alphavirus maintained in an enzootic cycle with mosquitoes and birds. EEEV can be transmitted to humans and susceptible animals by mosquitoes that bite both mammals and birds. EEEV causes severe neuroinvasive disease in humans; although an EEE vaccine is available for horses, no human vaccine is currently licensed, and treatment is supportive. Approximately one third of human cases are fatal, and many survivors experience long-term neurologic sequelae (1). In the United States, a majority of EEE cases occur in states along the A

Madariaga virus (MADV), widely distributed in Latin America, can cause severe disease in humans and equids, yet key aspects of its transmission cycle remain unclear. To identify mosquitoes that could act as vectors of MADV, we assessed the vector competence of Aedes aegypti, Ae. albopictus, Ae. taeniorhynchus, Culex tarsalis, Cx. coronator, and Cx. quinquefasciatus, following oral exposure to MADV isolated in Panama (all species) or Brazil (Ae. taeniorhynchus only). We also evaluated temporal infection dynamics of MADV from Panama in Ae. aegypti and Ae. albopictus. MADV RNA and infectious viru

BACKGROUND: Eastern equine encephalitis (EEE) is a highly fatal neurologic disease of horses, but data describing clinical presentation, epidemiology, and prognostic factors are limited.
OBJECTIVES: Describe the clinicopathologic and epidemiologic features of horses with EEE and identify factors associated with survival.
ANIMALS: One hundred four horses diagnosed with EEE presented with signs of diffuse brain disease.
METHODS: Retrospective case series of horses evaluated at a Veterinary Teaching Hospital (1979-2017). Medical records were reviewed for clinical findings, diagnostic results,

West Nile virus (WNV) and eastern equine encephalitis virus (EEEV) are typically maintained via enzootic cycles between birds and mosquitoes (Diptera: Culicidae). In North America, the blood hosts of mosquitoes involved in pathogen circulation are poorly described for many regions, limiting our ability to predict disease outbreaks. Our objectives were to (i) identify local (Eastern Ontario, Canada) bloodmeal hosts fed on by mosquitoes using DNA barcoding and (ii) investigate if the bloodmeal hosts identified were representative of the entire host community using species accumulation curves and

Delivery of the viral genome into host cells is a critical step in successful viral infection. Alphaviruses achieve this step by fusing the viral and endosomal membranes under acidic conditions. This process requires significant structural changes in the alphavirus glycoprotein organization. Structural characterization of acidic pH-induced conformational changes in alphavirus virions has remained elusive due to the rapid, transient nature of these states, conformational heterogeneity, and particle aggregation. Antibody binding studies conducted at elevated temperatures or under acidic pH condi

Frequent use of resting boxes by Culiseta melanura (Coquillett) during the day-time may offer the opportunity for use of these artificial resting sites as delivery substrates for insecticides to manage populations of this eastern equine encephalitis virus (EEEV) vector. In field studies at EEEV-endemic foci in southwestern Michigan, abundance in black-painted resting boxes of adult male and female Cs. melanura varied within the 24-h diel cycle, by whether the interiors of the boxes were treated with a microencapsulated formulation of lambda cyhalothrin (Demand CSTM), and from before to after t

Infection by neurotropic alphaviruses such as the Eastern equine encephalitis virus (EEEV) causes extensive inflammation in the central nervous system and tissue damage, including disruption of the blood-brain barrier (BBB). Neuroinflammation and BBB disruption following infection are critical pathological considerations for the development of robust countermeasure strategies. Encephalitic disease resulting from EEEV infection currently lacks FDA-approved therapeutic intervention strategies, thus exposing a major capability gap in the ability to address the global health burden that could resu

Mosquito pool surveillance is a cornerstone of arbovirus risk assessment, yet traditional metrics such as the minimum infection rate (MIR) can understate infection probability and, as conventionally reported, do not provide uncertainty quantification that properly accounts for pooling. We analyzed 7 yr (2018-2024) of mosquito pool surveillance data using pooled maximum likelihood estimation (MLE) with transformed Clopper-Pearson confidence intervals to estimate per-mosquito infection probabilities. The dataset comprised 4,930 pools (246,500 mosquitoes) collected across 80 surveillance sites an

The Eastern equine encephalitis virus (EEEV) complex comprises mosquito-borne neurotropic alphaviruses maintained in nature by mosquitoes. Although rare, human infections caused by these viruses can lead to febrile illness that may progress to severe encephalitis, for which there are no vaccines for prevention and no specific therapeutics for treatment. Moreover, a high percentage of human cases show long-term neurological sequelae. Here, we review the literature on cases, diagnosis, and management. Current gaps in clinical care include an urgent need to develop rapid diagnostic tests, new the

Mosquito host-feeding patterns strongly influence pathogen transmission potential, but landscape drivers of host use remain poorly understood for many generalist vectors. We examined whether landscape composition and spatial configuration predict blood-feeding patterns of Culex erraticus collected from natural areas across Illinois, USA. Specifically, we tested whether land-cover composition and distance from key landscape features, including forest edge, are associated with host-use shifts relevant to zoonotic pathogen transmission. Blood-fed females were identified morphologically, vertebrat

Venezuelan equine encephalitis virus (VEEV) is a prototypical encephalitic alphavirus. Members of the Alphavirus genus are found across the globe, transmitted by arthropod vectors, and cause significant disease burdens in humans and animals. There are currently no FDA-approved antivirals for human use against any member of the Alphavirus genus. While a vaccine exists against chikungunya virus (CHIKV), a member of the arthritogenic alphaviruses, FDA-approved vaccines are not available for other members of this genus, particularly the encephalitic alphaviruses such as VEEV, Eastern equine enceph

Alphaviruses are a globally distributed group of arboviruses responsible for two distinct disease manifestations, namely inflammatory polyarthritis and encephalitis. Among the arthritogenic viruses, chikungunya virus (CHIKV) is the most prevalent and has caused epidemics in 119 countries. Notably, two human vaccines against CHIKV have now received FDA approval. In contrast, the encephalitic alphaviruses, including Venezuelan equine encephalitis virus (VEEV), eastern equine encephalitis virus (EEEV), and western equine encephalitis virus (WEEV), are associated with substantially higher case fat
