DETECTION OF WEST NILE VIRUS AND ST. LOUIS ENCEPHALITIS VIRUS IN THE TEXAS PANHANDLE AND HIGH PLAINS REGION, USA, IN 2024.

The Texas Panhandle (26 counties) and High Plains regions (41 counties) have one of the highest West Nile virus (WNV) and St. Louis encephalitis virus (SLEV) human neuro-incidence rates in the USA, yet surveillance for these pathogens is predominately performed through passive measures (e.g., detecting human cases at hospitals and other health clinics). There is an overall lack of active pathogen surveillance (e.g., mosquito surveillance) systems in these regions, leading to knowledge gaps in our understanding of arboviral dynamics in enzootic cycles. To address this limitation, we implemented

Ecological drivers of St. Louis encephalitis virus activity in mosquito communities from Argentine agroecosystems.

Saint Louis encephalitis virus (SLEV) is an endemic arbovirus in the Americas, maintained in enzootic cycles involving mosquitoes and birds. Agroecosystems influence vector ecology and SLEV dynamics. We analysed mosquito communities and SLEV activity in agroecosystems of northeastern La Pampa, Argentina, assessing environmental and biological drivers. Mosquitoes were collected at 15 sites (2017-2019) using Centers for Disease Control light traps. SLEV was detected by RT-nested PCR, and minimum infection rate was estimated. We collected a total of 3075 mosquitoes; Culex interfor Dyar (Diptera:

Geographically Distinct Circulation of Genotype II and III St. Louis Encephalitis Virus, Texas, USA, 2009-2024.

We conducted a retrospective genomic surveillance study of St. Louis encephalitis virus (SLEV) in Texas, USA, to determine the genotypes circulating in the region. By using a custom tiled-amplicon assay with Oxford Nanopore sequencing, we generated 63 genomes from SLEV-positive mosquito pools and viral isolates collected during 2009-2024. Phylogenomic analysis revealed temporal overlap of genotype II and III circulation, but with distinct geographic segregation. Genotype II was confined to Gulf Coast counties with sustained local transmission, whereas genotype III was only in north and west Te

Determinants of West Nile virus transmission in Miami-Dade County, Florida.

West Nile is a mosquito-borne disease transmitted by Culex spp. mosquitoes and is widely distributed throughout the Continental United States. The causative agent is the West Nile virus (WNV); Family Flaviviridae Genus Orthoflavivirus. However, Miami-Dade County, a subtropical region with abundant populations of Cx. quinquefasciatus and Cx. nigripalpus, has not experienced sustained WNV transmission. This integrative review examines ecological, climatic, and surveillance determinants of WNV transmission in Miami-Dade County to understand why WNV cases remain low despite abundant vector populat

Dual-host choice experiments reveal inconsistent seasonal blood-feeding preference in Argentinean Culex quinquefasciatus populations.

Mosquitoes of the genus Culex are important vectors of St. Louis encephalitis (SLEV) and West Nile (WNV) viruses, and their host-feeding patterns strongly influence transmission dynamics by shaping contact rates between avian and mammalian hosts. In the United States, several Culex species exhibit seasonal shifts in host use, feeding primarily on birds during summer and increasing mammalian feeding in autumn. This shift coincides with seasonal peaks in SLEV and WNV incidence, suggesting that changes in mosquito feeding behaviour may trigger viral spillover to mammals. However, no experimental

Comparative fitness of reemerging St. Louis encephalitis virus in vertebrate and mosquito cells, Culex tarsalis and Culex quinquefasciatus mosquitoes, and mice.

The human pathogenic orthoflavivirus St. Louis encephalitis virus (SLEV) reemerged in the western United States in 2015 after more than a decade of absence and has since expanded throughout California with sustained interannual transmission. This shift from the historically sporadic pattern of SLEV activity before 2003 raises the question of whether contemporary strains differ in fitness from earlier strains. To assess whether reemerging SLEV possesses enhanced infectivity or transmissibility, we compared five contemporary genotype III strains from California (2016-2023) with a historical geno

BEYOND MIR: POOLED MAXIMUM LIKELIHOOD ESTIMATION FOR MOSQUITO POOL SURVEILLANCE.

Mosquito pool surveillance is a cornerstone of arbovirus risk assessment, yet traditional metrics such as the minimum infection rate (MIR) can understate infection probability and, as conventionally reported, do not provide uncertainty quantification that properly accounts for pooling. We analyzed 7 yr (2018-2024) of mosquito pool surveillance data using pooled maximum likelihood estimation (MLE) with transformed Clopper-Pearson confidence intervals to estimate per-mosquito infection probabilities. The dataset comprised 4,930 pools (246,500 mosquitoes) collected across 80 surveillance sites an

From RAMP to Triplex RT-qPCR: Modernizing Arbovirus Surveillance and Confirming the First Aedes aegypti in Idaho.

West Nile virus (WNV) remains the most frequently reported locally acquired arboviral infection in the United States, yet many small and mid-sized mosquito abatement districts lack the diagnostic capacity and integrated data systems needed for rapid detection and response. The Canyon County Mosquito Abatement District (CCMAD) in southwestern Idaho undertook a multi-year capacity-building effort to expand arbovirus surveillance, standardize mosquito identification and pooling procedures, and implement triplex RT-qPCR testing for WNV, Western equine encephalitis virus (WEEV), and St. Louis encep

Cross-Reactivity of 15 JEV Antigen-Directed Rabbit Antisera with Three Clinically Important JEV Serogroup Members: WNV, MVEV, and SLEV.

Japanese encephalitis virus (JEV), the prototype member of the JEV serogroup within the genus Orthoflavivirus (family Flaviviridae), is closely related to West Nile virus (WNV), Murray Valley encephalitis virus (MVEV), and St. Louis encephalitis virus (SLEV). To characterize antigenic cross-reactivity within this group, we evaluated 15 region-specific rabbit antisera, previously generated against nearly the entire JEV protein-coding region, by immunoblotting whole-cell lysates from BHK-21 cells infected with WNV, MVEV, or SLEV, with JEV included as a reference. Six antisera (α-EN-term, α-NS2B,

Evaluation of the In2care Mosquito Station against Culex quinquefasciatus mosquitoes (Diptera: Culicidae) under semifield conditions.

Culex quinquefasciatus is an important mosquito vector responsible for the transmission of filarial worms, arthropod-borne viruses like Oropouche, St. Louis encephalitis, and West Nile and protozoans that cause avian malaria. Due to insecticide resistance documented in Cx. quinquefasciatus populations worldwide, integrated vector management programs can benefit from new strategies to control this species. The In2Care Mosquito Station (In2Care station), a commercially available dissemination station containing pyriproxyfen (PPF) and Beauveria bassiana spores, has been shown to be effective agai

Role of Aging in Ulcerative Colitis Pathogenesis: A Focus on ETS1 as a Promising Biomarker.

PURPOSE: An increasing proportion of the aging population has led to a rapid increase in the number of elderly patients with ulcerative colitis (UC). However, the molecular mechanisms by which aging causes UC remain unclear. In this study, we explored the role of aging-related genes (ARGs) in UC pathogenesis and diagnosis prediction. METHODS: Gene expression data were obtained from four independent datasets (GSE75214, GSE87466, GSE94648, and GSE169568) in the GEO database, and ARGs were derived from multiple public databases. After identifying UC-related ARGs, consistent clustering was perfor

Silent St. Louis encephalitis virus circulation evidence by a haemovigilance survey in a centralized blood bank.

BACKGROUND: Saint Louis encephalitis virus (SLEV) re-emergence and its geographical expansion, evidenced by its emergence in previously unaffected areas, have raised significant public health concerns. We aimed to show the usefulness of haemovigilance as an effective tool to fill arboviruses surveillance gaps to track trends and identify hotspots. METHODS: Within the framework of a blood bank haemovigilance program, we performed a survey to evaluate the potential threat of circulating mosquito-borne infections for transfusion safety. SLEV was detected by a PCR targeting the conserved NS5-gene

Assessing pyrethroid resistance mechanisms in individual Culex tarsalis (Diptera: Culicidae).

The evolution of pyrethroid insecticide resistance in Culex tarsalis Coquillett, a vector for West Nile and St. Louis encephalitis viruses, is a growing concern in Northern California. Common mechanisms of resistance include the target-site mutation, kdr, and increased levels of detoxification enzymes, such as mixed-function oxidases, GSTs, and esterases. The goal of this study was to compare the prevalence of kdr mutations (L1014F and L1014S) and detoxification enzymes between pyrethroid susceptible and resistant Cx. tarsalis individuals. Individual mosquitoes, categorized by resistance statu

Serologic Surveillance for Orthoflaviviruses and Chikungunya Virus in Bats and Opossums in Chiapas, Mexico.

We performed serologic surveillance for selected arthropod-borne viruses (arboviruses) in bats and opossums in the Lacandona Rainforest, Chiapas, Mexico, in 2023-2024. Sera were collected from 94 bats of at least 15 species and 43 opossums of three species. The sera were assayed by the plaque reduction neutralization test (PRNT) for antibodies to eight orthoflaviviruses (dengue viruses 1-4, St. Louis encephalitis virus, T'Ho virus, West Nile virus, and Zika virus) and one alphavirus (chikungunya virus; CHIKV). Twelve (12.8%) bats and 15 (34.9%) opossums contained orthoflavivirus-specific antib

Interannual changes in the association between land use, abundance of Culex quinquefasciatus and Culex tarsalis (Diptera: Culicidae), and occurrence of arboviruses in Maricopa County, Arizona.

West Nile virus (WNV) (Orthoflavivirus nilense) and Saint Louis encephalitis virus (SLEV) (Orthoflavivirus louisense) are transmitted by Culex quinquefasciatus Say and Culex tarsalis Coquillett in Maricopa County, Arizona, where a significant increase in the number of WNV cases was reported in 2021. We used data collected between 2014 and 2021 from a network of CO2-baited surveillance traps to assess whether particular land use categories may have contributed to this rise in WNV cases. For each vector species and year, we estimated the association between the areas of each of 10 land use categ

Standardized Diagnostic Assays for Omsk Hemorrhagic Fever Virus.

Omsk hemorrhagic fever is an acute zoonotic disease caused by Omsk hemorrhagic fever virus, a member of the genus Flavivirus (family Flaviviridae), with a reported case-fatality rate of approximately 3%. Historically confined to southwestern Siberia, ecological changes raise concerns about possible spread to non-endemic regions. Although no Omsk hemorrhagic fever cases have been reported in the Republic of Korea, the risk of accidental importation highlights the importance of establishing a reliable diagnostic protocol. We established and validated an institutionally developed diagnostic proto
