OBJECTIVE: To investigate whether genetically proxied educational attainment influences the risk of intracranial aneurysm.
METHOD: We conducted a two-sample Mendelian randomization analysis using genome wide association study summary statistics from individuals of European ancestry. The inverse variance weighted method was the primary estimator, complemented by MR Egger, weighted median, weighted mode, and simple mode analyses, with years of schooling and college completion as educational exposures combined with intracranial aneurysm GWAS data.
RESULTS: We selected two groups of genes associ

Swine influenza A viruses (IAV-S) caused the 2009 H1N1 pandemic and pose a future zoonotic and pandemic threat. Vietnam represents a critical hotspot for IAV-S emergence within East and Southeast Asia, with dense swine and human populations and intensive livestock trade. We conducted genomic surveillance of IAV-S in Vietnam during 2020-2024, extending previous surveillance from 2013-2019. We identified multiple co-circulating H1 and H3 clades, including pandemic H1N1, Eurasian avian-like, and European lineages, by conducting phylogenetic analysis of 56 IAV-S isolates (21 H1N1, 31 H1N2, and 4 H

The concept of immunity debt is a phenomenon resulting from the suppression of endemic pathogens during the COVID-19 pandemic due to non-pharmaceutical interventions. The reduced circulation of various pathogens during the pandemic, particularly respiratory syncytial virus (RSV), altered typical infectious disease dynamics by reducing levels of population immunity usually acquired through exposure to infection. This concept is demonstrated through the post-pandemic resurgence of diseases such as RSV and group A Streptococcus, and highlights the interplay between reduced pathogen exposure and i

Avian influenza H5N1 has pandemic potential and historically has caused more severe disease in pregnant women than the general population. With increasing transmission of H5N1 detected among placental mammals, animal models are necessary for testing countermeasures, including during pregnancy. Pregnant outbred mice were infected with a contemporary strain of bovine H5N1 during the second or third trimester equivalent. Second trimester infection caused in utero transmission, with infectious virus detected in the uterus, placenta, and fetus. Birth following third trimester infection resulted in

Swine influenza virus (SIV) is a highly contagious respiratory pathogen that causes significant economic losses in the swine industry and poses a potential health risk to humans. This study investigated the genetic diversity and evolution of the H1N1 subtype SIV across different regions of China over the past four decades. Using 959 whole-genome sequences collected between 1977 and 2020 from public databases such as GenBank and the Global Initiative on Sharing Avian Influenza Data (GISAID), we systematically analyzed the epidemiology, phylogenetics, genotypes, and molecular characteristics of

Influenza A virus poses a significant threat to global health due to its high mutability and potential for pandemics. Accurate prediction of influenza virus pandemic risk can mitigate their threat to public health systems and societal stability. Current sequence-based deep learning methods predict the risk of viral pandemics by extracting features from the viral genome sequence. However, these methods ignore the gene flow between different virus strains through genetic reassortment, which is essential for understanding the dynamics of viral evolution. To leverage the gene flow of virus strains

CONTEXT: Attention to the potential impact of highly pathogenic avian influenza (H5N1, or "bird flu") on human health increased following documentation of mammalian transmission in spring 2024 and the prevalence of H5N1 in the commercial dairy supply. The authors aim to quantify (a) the prevalence and (b) the sociopolitical correlates of public concern about H5N1 as well as support for policy action aimed at reducing its potential health risks.
METHODS: In a nationally representative survey of N = 831 US adults, the authors asked respondents to answer questions about their levels of concern a

Three critically ill or fatal avian influenza A(H5N1) human infections have been reported in North America since November 2024. Notably, all were infected with genotype D1.1 instead of B3.13, the dominant genotype before November 2024. Here, we demonstrated that D1.1 could replicate to higher titers in human nasal and airway organoid-derived transwell monolayers from 6 donors. D1.1 exhibited a better binding to α2,3- and α2,6-linked sialic acid than B3.13. No significant differences in most inflammatory or antiviral cytokines/chemokines were observed. These observations suggest that D1.1 is be

The classical swine (CS) H1N1 influenza virus, first isolated in 1930, is highly homologous to the 1918 Spanish influenza virus. CS H1N1 virus, which crossed the interspecies barrier to infect humans has become the dominantly prevalent strain in China's pig population, showing a trend of continuous transmission. However, whether subsequent adaptation of CS H1N1 to mammals would increase their pathogenicity toward humans is unknown. To address this, a mouse-adapted (MA) CS H1N1 virus (A/Swine/Guangdong/1/2011[G11-MA]) was generated through serially passaged in mouse lungs, exhibiting increased

In a new study, Zhao et al. (2025) obtain 959 whole genome sequences of H1N1 subtype swine influenza virus (SIV) isolated from China. Their analysis of the sequences, isolated between 1977 and 2020, reveals how H1N1 lineages have co-evolved and contributed to instances of zoonotic transmission within the region. This study's findings characterize the long-term evolutionary effects of frequent viral reassortment in SIV and highlight its potential to drive future pandemics.

Influenza A virus (IAV) causes annual epidemics and occasional pandemics, remaining a threat to public health and economy worldwide. It has been established that many cellular noncoding RNAs (ncRNAs) are differentially expressed during IAV infection. These ncRNAs vigorously modulate the productive infection of the virus through diverse mechanisms, such as directly affecting the virus life cycle, altering the cellular signaling pathways, or regulating the host's innate immune response to viral infection. Some of the ncRNAs are deemed to be essential for IAV replication, while others, conversely

BACKGROUND: Rodents are primary reservoirs for zoonotic viruses, posing significant public health threats. However, our understanding of how habitat heterogeneity influences viral community composition and the spillover potential remains inadequate.
RESULTS: Here, a comprehensive meta-transcriptomic analysis of rodent-associated viruses was conducted across four distinct ecological habitats in the northern foothills of the Tianshan Mountains, China. Multiple novel viral sequences belonging to families with known zoonotic potential were identified, including Nairo-, Arena-, Flavi-, Picorna-, a

CONTEXT: Influenza is a viral infection with pandemic potential that affects individuals of all ages. The immune response is key to resolving the disease; however, immune deregulation, such as cytokine storm, can cause tissue damage and death. Diethylcarbamazine (DEC), a drug with immunomodulatory effects, has demonstrated anti-inflammatory and antifibrotic properties. However, DEC's activity in viral respiratory infections remains unknown.
OBJECTIVE: This study aimed to analyze the effect of DEC in a model of respiratory epithelial cells infected with Influenza A (H1N1)pdm09 virus (IAV) to e

Avian influenza virus cross-species infection in humans poses a major threat to global public health. Species-specific differences between avian ANP32A and mammalian ANP32 proteins create a natural barrier against viral cross-species infection by directly impairing the functional interaction between the avian-origin viral RNA polymerase and mammalian ANP32 proteins, thereby restricting viral genome replication. The key to overcoming this barrier lies in the adaptation of viral RNA polymerase to host ANP32 family proteins. This mini-review summarizes the mechanisms and variations in influenza v

Since 2020, large-scale outbreaks of highly pathogenic avian influenza (HPAI) H5N1 in Great Britain have resulted in substantial poultry mortality and economic losses. Alongside the costs, the risk of circulation leading to a viral reassortment that causes zoonotic spillover raises additional concerns. However, the precise mechanisms driving transmission between poultry premises and the impact of potential control measures in Great Britain, such as vaccination, are not fully understood. We have developed a spatial transmission model for the spread of HPAI in poultry premises calibrated to infe

Enveloped viruses are significant zoonotic disease threats with the potential to cause global pandemics. We identified a class of small-molecule sulfur-containing antiviral compounds (XM series) that broadly inhibit enveloped viruses. Multidisciplinary approaches revealed that XM compounds alter the viral membrane lipid chemical composition, enhance membrane order within the hydrophobic bilayer, and increase membrane phase transition temperatures. This mechanism inhibits membrane fusion and viral entry, while leaving the viral glycoproteins and genomes largely unaffected. Leveraging these uniq

INTRODUCTION: Recent intercontinental spread of highly pathogenic avian influenza (HPAI) A(H5N1) among kept and wild birds, and transmission to mammalian hosts, including cattle and humans, has heightened the need to review public health risk assessments. Biosecurity measures (BMs) are essential for limiting disease spread, but how widely different practices are implemented is not fully known.
METHODS: Here, we report on the uptake of BMs and risk perception of avian influenza virus (AIV) in the context of preventing zoonotic transmission to persons potentially at high risk of exposure. Quest

Highly pathogenic avian influenza A (H5N1) clade 2.3.4.4b is now widespread in US birds and has spilled over into mammals, raising concern about pandemic emergence. We applied the Viral Trait Assessment for Pandemics (ViTAP) model to score 11 intrinsic viral traits using a structured literature review, expert elicitation, and sensitivity analyses. The resulting composite score for contemporary US H5N1 was 3.28/5 (moderate-high risk); transmission-related traits contributed most to score uncertainty, while the absence of sustained human-human spread currently limits risk. Thus, ViTAP provides a

To counteract host antiviral responses, influenza A virus triggers a global reduction of cellular gene expression, a process termed "host shutoff." A key effector of influenza A virus host shutoff is the viral endoribonuclease PA-X, which degrades host mRNAs. While many of the molecular determinants of PA-X activity remain unknown, a previous study found that N-terminal acetylation of PA-X is required for its host shutoff activity upon ectopic expression. However, it remains unclear how this co-translational modification promotes PA-X activity. Here, we report that PA-X N-terminal acetylation
