Development and Application of a Pseudovirus-Based Assay for Modelling SARS-CoV-2 Spike Protein Mediated Drug Screening.

Requirements for novel effective antiviral agents against SARS-CoV-2 emphasizes the importance of robust in vitro screening platforms. We developed a test system based on spike-pseudotyped lentiviruses, carrying either luc+ or EGFP reporter genes as a payload, and a human non-small cell lung carcinoma (NSCLC) cell line, overexpressing ACE2 (H1299-hACE2). The cell origin makes our system resemble lung epithelium infection. Transmission electron microscopy confirmed that the spike glycoproteins on the pseudotyped lentiviral particles resemble native SARS-CoV-2 spike glycoproteins, thus validatin

The Airway Microbiome as a Modulator of Influenza Virus Infection: Mechanistic Insights and Translational Perspectives-Review.

Outcomes of influenza virus infection vary widely across individuals, reflecting not only viral genetics and host factors but also the composition and function of the airway microbiome. Over the past few years, mechanistic work has clarified how specific commensals (for example, Staphylococcus epidermidis and Streptococcus oralis) restrict influenza replication by priming epithelial interferon-λ programs, reshaping intracellular metabolite pools (notably polyamines), dampening host protease activity, and maintaining barrier integrity; meanwhile, pathobionts (notably Staphylococcus aureus and S

Changing Trends of Respiratory Viruses in Hospitalized Children During and After the COVID-19 Emergency Phase in Yongin, South Korea (2020-22 vs. 2023-24).

The COVID-19 pandemic and subsequent non-pharmaceutical interventions (NPIs) significantly disrupted the epidemiology of pediatric respiratory viruses. This study compared infection patterns among 3658 hospitalized children in South Korea during the pandemic (2020-2022) and the post-emergency phase (2023-2024), following the relaxation of mandatory NPIs. Of 4419 eligible tests, the most frequently detected viruses overall were rhinovirus/enterovirus (HRV/HEV) (27.9%), influenza (14.5%), and respiratory syncytial virus (RSV, 11.9%). The post-emergency phase was marked by a dramatic surge in inf

Vaccination Against Respiratory Infections in Adults with Cancer: A Concise Guide for Clinicians.

Global cancer incidence reached 20 million new cases across 185 countries in 2022, with approximately 10 million cancer-related deaths annually. Among adults with solid tumors and hematological malignancies, infections are a major contributor to morbidity and mortality, with respiratory infections playing a particularly significant role. These infections not only reduce life expectancy but can also delay cancer therapy, negatively affect treatment outcomes, and increase healthcare costs. In recent years, the burden of respiratory infections in this population has been driven by influenza virus

Repeat Influenza Vaccination Effects in 2021/22 and 2022/23 in a Community-Based Cohort in Hong Kong.

BACKGROUND: Repeated influenza vaccination has been associated with attenuated immune responses and reduced clinical effectiveness. Most studies have focused on the responses at 30 days post-vaccination. As influenza did not circulate in Hong Kong during the COVID-19 pandemic, we were able to investigate vaccine responses in the absence of infections. METHODS: In this community-based cohort study in Hong Kong, we investigated the impact of repeated annual influenza vaccination on antibody titer boosting and waning rates of 8 contemporary vaccine strains in 2021/22 and 2022/23, using hemagglut

A Malaysian study on COVID-19 (SARS-CoV-2) vaccination immune response in haemodialysis patients: A prospective, multicentre, cohort study.

INTRODUCTION: Patients with end-stage renal disease (ESRD) have compromised immune systems, possibly reducing their vaccine-induced antibody responses compared to the general population. COVID-19 remains a persistent threat, with the virus continuing to mutate into new variants. Similar to influenza, there is a possibility that COVID-19 could resurge significantly, underscoring the importance of understanding vaccine-induced immunity in vulnerable populations. This study aims to determine the immunogenicity of this group to the COVID-19 vaccine. MATERIALS AND METHODS: Haemodialysis patients r

SARS-CoV-2 T-cell vaccine VB10.2210 induces broad T-cell responses in a phase 1/2 open-label clinical trial.

The repeated emergence of highly transmissive SARS-CoV-2 variants requires a broadly protective vaccine. We developed a T-cell vaccine VB10.2210 that targets SARS-CoV-2 viral antigens to antigen presenting cells comprising 96 validated immunogenic T-cell epitopes covering a global HLA diversity. We report results from a first in human open-label dose-escalation phase 1/2 clinical trial evaluating safety, reactogenicity and immunogenicity of VB10.2210. The study investigated three dose levels (0.3, 1.0 and 3.0 mg), delivered intramuscularly as DNA plasmid by jet injection (NCT05069623), in 34 h

Epidemiological diversity of seasonal influenza and post-COVID-19 pandemic changes in the UK, 2015-2025: a surveillance-based analysis.

BACKGROUND: Research is lacking on whether seasonal influenza epidemiology differs across geographically and climatically similar regions and whether the COVID-19 pandemic has caused changes to influenza dynamics that transcend these differences. METHODS: We adopted two complementary databases covering ten consecutive years of seasonal influenza epidemics across the four nations of the United Kingdom (UK). The Global Influenza Surveillance and Response System (GISRS) database mainly provides weekly numbers of tested individuals and infected cases, while the UK Health Security Agency (UKHSA) da

Association between adjunctive corticosteroid therapy and clinical outcomes in children with severe viral community-acquired pneumonia: A retrospective cohort study.

The therapeutic role of corticosteroids (CS) in severe community-acquired pneumonia (CAP) remains contentious, particularly in pediatric populations with viral etiology. Limited pediatric-specific evidence exists regarding CS efficacy in viral CAP, despite widespread clinical utilization. This retrospective cohort analysis examined the association between adjunctive CS therapy and clinical outcomes in children with severe viral CAP. A retrospective cohort analysis was conducted at a tertiary pediatric intensive care unit from January 2016 to December 2019. Inclusion criteria comprised positive

The syndemic nexus: How vaccine hesitancy and COVID-19 interact to influence vaccine uptake among university students.

This study examined whether COVID-19 Vaccine Hesitancy (VH) and COVID-19-related factors interact to influence COVID-19 booster doses uptake among university students in Canada, from a syndemic perspective. A cross-sectional survey was conducted among 4453 students at the University of Waterloo in 2024. VH was measured toward both COVID-19 primary and booster doses. Change in VH scores were computed to capture shifts in hesitancy over time. Logistic regression models assessed the main effects of VH and COVID-19-related factors on booster uptake. Interactions were tested using additive and mult

An adaptive method of emergency department syndromic surveillance to nowcast the frequency of presentations that will have a severe 28-day outcome following influenza or COVID-19 infection: a retrospective analytical record linkage study.

During epidemics, emergency department (ED) syndromic surveillance of patient arrivals provides timely but nonvirus-specific assessment of epidemic intensity. Surveillance of severe infection outcomes (intensive care admission or death) is less timely because outcomes can take weeks to occur. Time series models can be used to estimate the frequency of severe infection outcomes due to viruses. We developed and evaluated daily time series modeling applied to linked ED, infection, and outcomes data from Australia to better predict population and health system burden during acute respiratory virus

Designing a broad-spectrum multi-epitope vaccine against influenza A and Mycoplasma pneumoniae: an immunoinformatics approach.

INTRODUCTION: Influenza A and Mycoplasma pneumoniae (M. pneumoniae) are common respiratory pathogens, causing severe co-infections in clinical diseases. Current vaccines have failed to provide comprehensive protection against both of the pathogens, highlighting the requirement of integrated solution. METHODS: In this study, a novel dual-target multi-epitope vaccine was developed based on the immunoinformatics method. Based on the consistent sequences, we used the HA and NA proteins of influenza A virus, and the p1, p65, HMW1-3 proteins of M. pneumoniae to determine the immunodominant T- and B

Local and systemic host responses to influenza and concurrent or sequential SARS-CoV-2 infection.

Influenza is an acute respiratory infectious disease caused by the influenza virus, which has been circulating in humans for over a century. In contrast, COVID-19, caused by the novel SARS-CoV-2, emerged recently in December 2019. Following nearly four years of pandemic, the acute phase of SARS-CoV-2 has transitioned towards an endemic state, suggesting a trend of long-term coexistence with humans. Concurrent or sequential coinfection with influenza and SARS-CoV-2 has been clinically observed to exacerbate pulmonary pathology and systemic inflammation in affected individuals. This review discu

Cytokine storm divergence in viral infections of the upper respiratory tract.

Cytokine storm (CS) is a pathological state of dysregulated, hyperactive host immunity that arises in the context of infection, malignancy, or immunotherapy. CS is characterized by the sustained, markedly elevated release of multiple pro-inflammatory mediators, ultimately leading to tissue damage and multi-organ dysfunction. Upper respiratory viral infections, including SARS, MERS, SARS-CoV-2, influenza, adenovirus, and respiratory syncytial virus (RSV), are among the most prominent CS triggers. Inflammatory storms triggered by different pathogens exhibit distinct variations in their cytokine
