A global database of West Nile virus host prevalence and competence.

West Nile virus (WNV) is one of the most widespread arboviruses globally and is maintained primarily through a bird-mosquito-bird transmission cycle, while other vertebrates play more limited roles. Host contributions to transmission depend on both infection evidence in natural populations (reflecting exposure and susceptibility) and reservoir competence, determined by the magnitude and duration of viraemia sufficient to infect mosquitoes. Despite extensive surveillance and experimental research, no comprehensive, standardised resource has integrated evidence on host exposure and infection in

Early-warning surveillance of West Nile and Usutu viruses in wetland and larval habitat waters and in mosquito excreta using digital PCR.

West Nile virus (WNV) and Usutu virus (USUV) are mainly avian mosquito-borne pathogens, increasingly cause human and equine disease in temperate regions. Mostly asymptomatic, current surveillance in France is reactive, based on clinical and veterinary reports, delaying signals for vector control and preventative interventions. We implemented in Southern France, in 2024 and 2025, a proactive surveillance methodology combining molecular xenomonitoring of mosquito excreta and environmental water detection of viral RNA in wetlands and mosquito larval habitat, through a multiplex reverse transcript

Genetic diversity of West Nile virus lineage 2a circulating in Culex pipiens vector populations from Bucharest city during the 2024 and 2025 transmission seasons.

BACKGROUND: West Nile virus (WNV) is an emerging pathogen in Europe with an unprecedented northward expansion. In Romania, the virus has been endemic for at least three decades, and human infection cases are registered yearly, sometimes culminating in outbreak events. Bucharest, the capital city of Romania, is the largest urban area of the country, encompasses a multitude of mosquito and bird habitats, and has been continuously reported as an active hotspot for WNV transmission. The circulation of lineage 2 has been documented in Bucharest since 2010 in mosquito and human populations. Our stud

A Multisystem Presentation of West Nile Neuroinvasive Disease: Lessons in Diagnostic Complexity.

West Nile virus (WNV) is an uncommon but serious case of encephalitis and meningitis, often presenting with nonspecific symptoms that can mimic a wide range of infectious, metabolic, and endocrine disorders. We present the case of a 61-year-old male with multiple comorbidities who arrived with 5 days of fever, chills, profuse diarrhea, black stools, and worsening confusion. Initial evaluation in the emergency department revealed hyponatremia, transaminitis, and concern for tick-borne illness, prompting empiric doxycycline. However, the patient denied any tick bites. Given his known thyroid nod

Isolation and molecular characterization of West Nile Virus with evidence of vertical transmission in the Coastal region, Kenya.

West Nile virus (WNV) is a mosquito-borne flavivirus of global public health importance, maintained in an enzootic cycle between birds and mosquitoes, with humans and other mammals as incidental hosts. Although WNV has been documented in Kenya, surveillance remains inconsistent, geographically fragmented, and largely dependent on serological methods. This limits understanding of circulating lineages, vector diversity, and transmission dynamics, particularly in coastal regions. This study aimed to address these gaps through systematic arboviral surveillance of mosquito populations across four e

Supporting effective anticipatory prescribing in end of life care.

BACKGROUND: West Nile virus (WNV) is a growing threat to public health and a greater understanding of the immune response raised against WNV is important for the development of prophylactic and therapeutic strategies. METHODOLOGY/PRINCIPAL FINDINGS: In a reverse-immunology approach, we used bioinformatics methods to predict WNV-specific CD8(+) T cell epitopes and selected a set of peptides that constitutes maximum coverage of 20 fully-sequenced WNV strains. We then tested these putative epitopes for cellular reactivity in a cohort of WNV-infected patients. We identified 26 new CD8(+) T cell e

Prevalence of human West Nile virus infections in Iran: A systematic review and meta-analysis.

The current systematic review and meta-analysis aims to estimate the prevalence of West Nile Virus (WNV) among humans in Iran. A comprehensive search was conducted in national databases, including SID, Magiran, and Barekat knowledge network and international databases, including Web of Science, MEDLINE, Embase, and Scopus, to identify articles on the prevalence of human WNV in Iran. A random-effects model was used to assess the pooled prevalence. A meta-regression method was applied to determine the factors affecting the heterogeneity among the studied population. Analysis was performed on 11

West Nile virus: epidemiology, prevention, clinical features, diagnosis, treatment, and open research questions.

BACKGROUND: West Nile virus (WNV) is among the most widespread arboviruses and has become a seasonal threat in temperate regions. Sustained in an enzootic bird-mosquito cycle, with humans and horses as incidental hosts, its geographic range has expanded in recent decades due to ongoing climatic and ecological changes. While most infections are asymptomatic or mild, a minority progress to neuroinvasive disease with high morbidity and long-term sequelae. Evidence for Powassan virus deletions and defective RNA in field-collected ticks.

Powassan virus (POWV) is a tick-borne flavivirus endemic to the United States, Canada, and parts of Russia. POWV remains an under-studied pathogen, despite the potential for serious and life-threatening neurologic complications following infection. While prior studies have characterized viral diversity due to single nucleotide polymorphisms, little is known about POWV recombination, defective RNAs (D-RNAs), and functional structural variants (SVs). Understanding POWV recombination in its natural vector can provide important insights into its replication and evolution. We analyzed POWV sequence

Genomic characterization of a severe West Nile Virus transmission season using a single reaction amplicon sequencing approach.

West Nile virus (WNV) is an endemic arthropod-borne virus that has routinely caused seasonal outbreaks in the United States since it was first detected in 1999. While phylogenetic studies have shown how WNV has diversified and undergone genotype replacement since introduction, more geographically focused studies are needed to understand intricate transmission dynamics at local and regional scales. In this study, we validate the IDT xGen WNV panel, a novel single reaction amplicon-based Next-Generation Sequencing approach, to generate high-quality WNV genomes and compare it to the "Primal Schem

Characterization of Culex pipiens cell lines: virus infection and RNAi response.

BACKGROUND: Arboviruses transmitted by mosquitoes pose a global health threat, causing diseases ranging from mild fevers to severe encephalitis and hemorrhagic fevers. Despite their growing impact, arbovirus research is hindered by biosafety constraints and the need of specialized BSL-3 insectariums. To circumvent these challenges, mosquito-derived cell lines have become indispensable tools for investigating virus-vector interactions. However, most available cell lines originate from Aedes and Anopheles spp., creating a critical research gap for other key vectors such as Culex spp. Although a

Detection of La Crosse virus RNA in clinical specimens obtained from children with La Crosse infection.

BACKGROUND: La Crosse virus (LACV), a member of family Peribunyaviridae, genus Orthobunyavirus, is the leading cause of neuroinvasive arboviral infection in children in the United States. Diagnosis relies on detecting specific antibodies (IgG or IgM), a 4-fold titer rise or seroconversion, in patients with compatible presentations. NAAT used for LACV detection has largely been limited to mosquito, animal models or postmortem brain tissue. There is a lack of data on the performance of NAATs in clinical specimens from living patients. METHODS: Children who had positive arbovirus serology tests

Pathogenicity assessment of Spanish West Nile virus isolates of lineages 1 and 2 in a Swiss mouse model.

West Nile virus (WNV) is one of the most widespread emerging arboviruses in the world. Recently, Europe has undergone a significant increase in WNV incidence and geographic extension, with two genetic lineages, lineage 1 and lineage 2, actively circulating. In Spain, lineage 1 was first identified in 2007, and subsequently spread through the southwest and central regions of the country. Since 2010, it has affected both horses and humans (mainly in southern Spain), including two large human outbreaks of WNV meningoencephalitis in 2020 and 2024. Lineage 2 was first identified in Catalonia (north

Divergent roles of Hsp70 chaperones in orthoflavivirus protein secretion and virion formation.

Orthoflaviviruses, such as tick-borne encephalitis virus (TBEV) and West Nile virus (WNV), can cause severe neurological disease and remain without specific antiviral treatments. We found that orthoflavivirus envelope (E) and non-structural protein 1 (NS1) interact with heat shock protein 70 (Hsp70) chaperones, key regulators of protein homeostasis and existing cancer drug targets. We examined how Hsp70 and endoplasmic reticulum-resident BiP contribute to viral protein secretion and infectivity of tick and mosquito-borne orthoflaviviruses. Targeting the Hsp70 nucleotide-binding domain with sma

Serological evidence of West Nile virus in blood donors from a city on the northern border of mexico: a cross-sectional study.

BACKGROUND: West Nile virus (WNV) is primarily transmitted by the bite of Culex mosquitoes, but other mechanisms, such as blood transfusion, have also been described. Since its identification in the Americas in 1999, WNV has circulated consistently in the United States of America (USA); however, although WNV has been detected in humans in South America, no major outbreaks have occurred in more than 20 years. One of the hypotheses to explain this difference is the underdiagnosis of the infection. In Mexico, nine isolated cases have been officially reported since 2003 despite its proximity to th

Discovering Novel Therapeutic VHHs for Emerging Viruses: Perspectives from VEEV Selection Strategies.

Evolution or emergence of a new viral variant is a significant public health concern. Alphaviruses, such as Venezuelan equine encephalitis virus (VEEV), are mosquito-borne viruses which are becoming more prevalent due to expansion of vector habitats. The increased prevalence of such viruses provides opportunities for novel variants to evolve. Key therapeutic molecules that could be developed against viral pathogens are recombinant antibodies or antibody fragments, such as the variable heavy domain of heavy chain antibodies (VHHs). These proteins can neutralize or sequester viral particles, pre
