BACKGROUND: Culex pipiens, the common house mosquito, is the main vector of important medical and veterinary diseases including West Nile fever. This disease is a neglected mosquito-borne disease in Iran, where migratory and resident birds act as the main reservoirs and human and horse act as dead-end hosts of the disease. The Aras River basin is identified as a high-risk area for West Nile fever outbreaks. Chemical vector control stands out as a key method for managing mosquito-borne diseases. So, it is necessary to evaluate the susceptibility of Cx. pipiens to insecticides recommended by the

BACKGROUND: Arboviruses have expanded into new regions in Europe, yet data indicate gaps in disease notifications and a risk of further spread. We aimed to report on prevalence, clinical management, and outcomes of endemic arbovirus infections in southeast Europe.
METHODS: In this prospective observational study (MERMAIDS-ARBO), we enrolled adults (age ≥18 years) hospitalised with an arbovirus-compatible disease syndrome within 21 days of symptom onset across 21 hospitals in seven countries in southeast Europe over four arbovirus seasons (May 1-Oct 31, during 2016-19). We obtained data from c

BACKGROUND: Arboviruses are increasingly affecting Europe, partly due to the effects of climate change. This increase in range and impact emphasises the need to improve preparedness for emerging arboviral infections that often co-circulate and might have overlapping clinical syndromes. We aimed to strengthen surveillance networks for four clinically relevant arboviruses in southeast Europe.
METHODS: This study reports an in-depth analysis of the MERMAIDS-ARBO prospective observational study in adults (ie, aged ≥18 years) hospitalised with an arbovirus-compatible disease syndrome in 21 hospita

Autophagy can defend against infection by delivering viruses to lysosomes for degradation. Semliki Forest virus (SFV) is a positive-sense, single-stranded RNA virus of the alphavirus genus which has been used extensively as a model for arbovirus infection and neuronal encephalitis. Here, we show that autophagy is suppressed during the early hours of SFV infection of neurons. We also show that a switch between autophagy suppression and upregulation between the early and later stages was mediated through modulation of the mammalian target of rapamycin (mTOR) activity during infection. At later s

West Nile (WNV) and Usutu (USUV) viruses co-circulated in a region of Central Italy (Lazio) in 2018, as evidenced by the detection of WNV in the nervous tissues of symptomatic horses and USUV in blood donors and mosquito pools. To assess whether these viruses were endemic in the region, we analysed: 1) diapausing Culex pipiens mosquitoes collected during the winter seasons 2022-2023 and 2023-2024, 2) Cx. pipiens mosquitoes collected during the adult activity period from April to November in 2022 and 2023 across 4 provinces, and 3) sera from 52 horses and tissues from 537 birds. BACKGROUND: Jamestown Canyon virus, a mosquito-borne virus, can cause asymptomatic infection, febrile illness, or neuroinvasive disease in humans. Previous studies have found Jamestown Canyon virus-specific antibodies in a 4%-54% of people in various U.S. regions. To understand baseline seroprevalence in regions with the highest number of reported disease cases, we performed a serosurvey among blood donors.
METHODS: We randomly selected blood donation specimens collected during December 2019-April 2020 from residents of counties reporting ≥2 disease cases in 2019 or 1 case in 2019 and ≥1 case

Oropouche virus (OROV) is a neglected and emerging arbovirus that infects humans and animals in South and Central America. OROV is primarily transmitted to humans through the bites of infected midges and possibly some mosquitoes. It is the causative agent of Oropouche fever, which has high morbidity but low mortality rates in humans. The disease manifests in humans as high fever, headache, myalgia, arthralgia, photophobia, and, in some cases, meningitis and encephalitis. Additionally, a recent report suggests that OROV may cause fetal death, miscarriage, and microcephaly in newborns when women

West Nile virus (WNV) is an arthropod-borne virus primarily transmitted by Culex mosquitoes, among birds, but occasionally infecting humans and equids. Understanding environmental factors on the disease occurrence is crucial for planning effective prevention and control strategies. Our study estimates the short-term effects of ambient temperature, relative humidity and precipitation on weekly cases of WNV in equids in Spain, using a space-time stratified case-crossover design. Because WNV transmission occurs sometime after the weather conditions become favorable for the vector, we aimed to est

PURPOSE OF REVIEW: Arthropod-borne viruses (arboviruses) represent a group of pathogens with increasing global relevance, some of which cause neuroinvasive disease. Transmitted by arthropod vectors and maintained by a variety of primary and amplifying hosts, epidemics are dependent on numerous environmental and anthropogenic factors. This review serves to highlight several important neuroinvasive arboviruses relevant to North America and discuss the neurologic presentations, diagnosis, outcomes, and future trends.
RECENT FINDINGS: Recent shifts in the epidemiology and ecology of arboviruses i

Auto-antibodies (auto-Abs) that neutralize type I interferons (IFNs) have been implicated in severe viral infections, including ~ 40% of cases of West Nile virus (WNV) neuroinvasive disease (WNND). Developing robust in vitro models to evaluate the protective effects of type I IFNs against viral infection, as well as the disruptive effects of auto-Abs, is essential for understanding disease pathogenesis and identifying patients at risk. In this study, we used Vero E6 and ARPE-19 cell lines to investigate the ability of type I (IFN-α, IFN-β, IFN-ω), type II (IFN-γ), and type III (IFN-λ1) IFNs to

BACKGROUND: La Crosse virus (LACV) is a mosquito-borne arbovirus responsible for pediatric encephalitis in North America, predominantly affecting children under 16 years. Early and accurate diagnosis is critical to reducing morbidity in this vulnerable population. However, existing molecular and serological methods are limited in sensitivity, specificity, and accessibility.
METHODS: To address these limitations, we developed a reverse transcription recombinase polymerase amplification (RT-RPA) assay for LACV detection. Primers targeting the divergent M segment of the LACV genome were designed

The Chandipura virus (CHPV) is an emerging arbovirus in India, transmitted primarily by sandflies and associated with outbreaks of acute encephalitis in children. First identified in 1965 in Maharashtra, CHPV has since been linked to Acute Encephalitis Syndrome (AES) in several states, including Maharashtra, Gujarat, Telangana, and Madhya Pradesh. The virus predominantly affects children under 15 years, particularly in rural areas with poor housing conditions, and is most prevalent during the monsoon season when sandfly activities are high. Its clinical presentation includes a rapid-onset febr

The Oropouche virus (OROV) is a new zoonotic arbovirus that mostly affects Brazil and nearby countries. Since its discovery in 1955, it has caused more than 500,000 infections, with symptoms ranging from fever and headache to serious neuroinvasive disorders such as meningitis and encephalitis. The virus spreads through urban and sylvatic cycles via vectors such as Culicoides midges and Culex mosquitos, with humans and some vertebrates serving as amplifying hosts. The manuscript aims to analyze the transmission dynamics, clinical manifestations, diagnostic approaches, and potential preventive s

Alphaviruses, including chikungunya virus (CHIKV), pose a significant global health threat, yet specific antiviral therapies remain unavailable. We evaluated combinations of three oral directly acting antiviral drugs (sofosbuvir (SOF), molnupiravir (MPV), and favipiravir (FAV)), which are approved for other indications, against CHIKV, Semliki Forest virus (SFV), Sindbis virus (SINV), and Venezuelan Equine Encephalitis virus (VEEV) in vitro and in vivo. We assessed antiviral efficacy in human skin fibroblasts and liver cells, as well as in a mouse model of CHIKV-induced arthritis. In human skin

Systemic lupus erythematosus (SLE) is an autoimmune disease that can involve the central nervous system (CNS), manifesting as a neuropsychiatric syndrome. Differentiating between neuropsychiatric systemic lupus erythematosus (NPSLE) and infections, such as West Nile encephalitis (WNE), as well as other etiologies, presents a significant diagnostic challenge and complicates the management of these patients. We present two cases of SLE patients with neuropsychiatric symptoms ultimately diagnosed as WNE and conducted a targeted literature review using bibliographic databases. We report two cases

In the United States, arthropodborne viruses (arboviruses) are primarily transmitted by infected mosquitoes or ticks. Most infections are asymptomatic; symptomatic infections range from mild febrile illness to severe neuroinvasive disease. This report summarizes 2023 data for nationally notifiable domestic arboviral diseases. Forty-eight states and the District of Columbia reported 2,770 human arboviral disease cases, including 2,022 (73%) hospitalizations and 208 (8%) deaths. As in previous years, West Nile virus (WNV) was the most commonly reported domestic arboviral disease in 2023, account

Orthoflavivirus encephalitidis (tick-borne encephalitis virus, TBEV) is of high concern due to its ability to cause severe neurological manifestations. Despite the fact that the role of NS1 proteins from various mosquito-borne flaviviruses in pathogenesis and their ability to affect human endothelial permeability have been investigated, TBEV NS1 has thus far been insufficiently studied. In this study, human endothelial permeability was assessed using TEER and transwell permeability assays. Signaling pathways were determined by RNAseq. The ability of the NS1 protein of TBEV to affect human endo
