Concentration Imbalance-Driven Nucleotide Circuit for Tick-Borne Encephalitis Virus Detection via Hybridization-Responsive DNA-Templated Silver Nanoclusters.

Tick-borne encephalitis virus (TBEV) is a severe neurotropic Flavivirus that requires early and sensitive detection. We developed a concentration imbalance-driven nucleotide circuit (CIDNC) combined with DNA-templated silver nanoclusters (DNA-AgNCs) for enzyme-free and direct RNA detection. The system converts trace TBEV cDNA or RNA into fluorescence signals via strand displacement under isothermal conditions. It achieved detection limits of 0.023 nM for cDNA and 0.035 nM for RNA, with single-base mismatch discrimination. The assay successfully detected viral RNA from plasma and lysed virions.

A Novel Diagnostic Tool for West Nile Virus Lineage 1a and 2 Using a CRISPR-Cas12a System.

The West Nile Virus (WNV), transmitted by Culex mosquitoes as a major vector, has been reported worldwide. Also, West Nile neuroinvasive disease (WNND) caused by WNV lineage 1a and 2 neuroinvasive infections has been constantly reported with high fatality rates. Nevertheless, there are no treatments and vaccinations, so diagnosis in the early stages is important. Recently, a molecular diagnostic technique using DNA endonuclease-targeted CRISPR trans reporter (DETECTR) with the CRISPR-Cas12a system integrated with isothermal nucleic acid amplification has newly emerged. In this study, we design

Auto-Abs against type I IFNs: Strong, common, and global determinants of severe arboviral diseases.

Human-tropic pathogenic arboviruses are spreading worldwide. There is immense interindividual clinical variability following infection with any arbovirus. Autoantibodies (auto-Abs) neutralizing antiviral type I IFNs (AAN-I-IFN) can underlie a small but growing number of severe arboviral diseases, whether transmitted by ticks (tick-borne encephalitis virus, TBEV; Powassan virus, POWV) or mosquitoes (West Nile virus, WNV; Usutu virus, USUV; Ross River virus, RRV) and whether due to flaviviruses (WNV, TBEV, POWV, and USUV) or alphaviruses (RRV). Evidence is documented in large cohort studies for

West Nile Virus Neuroinvasive Disease: A Retrospective Analysis of Hospitalized Cases in a Tertiary Care Center in Southern Europe.

BACKGROUND: West Nile virus (WNV) is a flavivirus primarily transmitted by mosquitoes of the Culex genus and is endemic to Southern Europe. Although infection is usually asymptomatic, it can lead to neuroinvasive syndromes with high morbidity and mortality. Due to the increasing incidence driven by climatic factors, we present a single-center series examining short- and long-term functional outcomes after infection. METHODS: Patients with neurological symptoms and confirmed WNV infection through serology and/or detection in urine and/or cerebrospinal fluid (CSF) between 2017 and 2023 were inc

Fluorescence-based biosensors for West Nile virus detection.

Flaviviruses are emerging pathogens, mostly transmitted by arthropod vectors, responsible for human, animal, and zoonotic diseases. The emergence of flaviviruses has been favored in recent decades by factors related to climate change and globalization, which contribute to the arrival and establishment of their vectors in new geographic areas, thus promoting epidemic outbreaks and facilitating these viruses to become endemic in these areas. This is the case of the West Nile virus (WNV) in Europe, which affects its natural bird host populations and also accidental hosts such as humans and horses

Neutralizing Antibodies against California Serogroup Orthobunyaviruses in Human Serum Samples, Montana, USA.

Viral encephalitis is often underreported and undiagnosed. To understand the potential causes of viral encephalitis in the state of Montana, USA, we examined the relative incidence of human infections for the California serogroup (CSG) of Orthobunyaviruses by screening random convenience serum samples obtained from different hospitals across Montana. We initially screened deidentified samples for neutralizing antibodies against Jamestown Canyon virus (JCV), a CSG virus that has caused encephalitis in Montana. We then analyzed JCV-positive samples for neutralization of other CSG viruses, and de

Structural insights into VLDLR recognition by western equine encephalitis virus.

Western equine encephalitis virus (WEEV), a group of encephalitic alphaviruses that cause severe diseases in humans and equids, historically used the very-low-density lipoprotein receptor (VLDLR) as a receptor during infection. However, current epidemic strains no longer use VLDLR as a receptor. In this study, we identify that LA1, LA2, LA3, and LA5 of VLDLR can directly interact with WEEV. Using cryo-electron microscopy, we investigate the structures of complexes formed between WEEV and VLDLR-LBD or other VLDLR fragments. Our findings show that LA1 and LA2 insert into a cleft formed by two ad

Long-term immunogenicity of a single-dose live recombinant chimeric Japanese encephalitis vaccine in adults.

BACKGROUND: Japanese encephalitis virus is a leading cause of viral encephalitis in Asia, with high case-fatality rate and morbidity. Although the live recombinant Japanese encephalitis chimeric vaccine (Imojev®) offers strong initial immunity, data on long-term efficacy beyond 5 years remain limited. METHODS: We conducted a cross-sectional study on adults vaccinated with Imojev® at a specialist travel clinic in Brisbane, Australia. Participants were stratified based on the time since vaccination: 2-5 years and >5 years. Neutralizing antibody titres were measured using the plaque reduction ne

The detection of Japanese encephalitis virus (JEV) in the Murray region, New South Wales: a public health investigation.

The detection of Japanese encephalitis virus (JEV) in pigs, at four piggeries in the Murray region in February 2022, prompted a public health investigation (PHI) by the New South Wales Department of Health (NSW Health) to identify people at greatest risk of infection. The PHI included three components: a vaccination clinic and accompanying clinic questionnaire; a serological investigation; and a cross-sectional study for consenting Australian-born participants who completed an extended questionnaire after receiving their serological results. The goals were to vaccinate a presumably naïve popul

Development of nucleic acid-based vaccines against dengue and other mosquito-borne flaviviruses: the past, present, and future.

Due to their widespread geographic distribution and frequent outbreaks, mosquito-borne flaviviruses, such as DENV (DENV), Zika virus (ZIKV), Japanese encephalitis virus (JEV), yellow fever virus (YFV), and West Nile virus (WNV), are considered significant global public health threats and contribute to dramatic socioeconomic imbalances worldwide. The global prevalence of these viruses is largely driven by extensive international travels and ecological disruptions that create favorable conditions for the breeding of Aedes and Culex species, the mosquito vectors responsible for the spread of thes

Tracing more than two decades of Japanese encephalitis virus circulation in mainland China.

UNLABELLED: Japanese encephalitis is a viral disease caused by the Japanese encephalitis virus (JEV), primarily affecting rural areas of Asia and western Pacific region. China remains one of the main epicenters, experiencing a significant burden of human and animal cases despite vaccination efforts. The ecology of this arbovirus is complex, involving Culex mosquitoes as primary vectors, wading birds as natural reservoirs, and pigs as amplifying hosts. Given the virus's epidemiological importance in China, combined with the country's expanding pig farming industry and diverse climates, investig

Gorakhpur, India experience of Japanese encephalitis management: A successful One Health approach.

The Japanese Encephalitis Virus (JEV), a zoonotic flavivirus, is responsible for infecting humans and inducing reproductive complications in swine, with transmission primarily facilitated by mosquitoes. Predominantly observed in Southeast Asia, Japanese encephalitis (JE) primarily circulates between amplifying hosts, notably swine, and culicine mosquitoes, particularly Culex tritaeniorhynchus . The wading birds are the reservoir host of this disease. Utilizing real time polymerase chain reaction (RT-PCR) presents a potential approach for detecting JEV in mosquito populations. Manifesting prima

Distribution and frequency of genetic mutations in three insecticide targets in field populations of Culex tritaeniorhynchus in Mianyang City, Sichuan Province, China.

Japanese encephalitis (JE) is an important mosquito borne infectious disease which is mainly transmitted by Culex tritaeniorhynchus Giles (1901) in China. At present, vector control remains an important means to prevent and control vector-borne diseases including JE. The development of insecticide resistance has seriously threatened the effectiveness of insecticide-based vector control programmes. Therefore, understanding insecticide resistance in the target pest is essential to inform evidence-based vector control. In Mianyang City of Sichuan Province of China, Cx. tritaeniorhynchus is the do

RAB4B and Japanese encephalitis virus E protein interaction is essential for viral entry in early endosomes.

RAB4B (Ras-Related GTP-Binding Protein 4b) is essential for intracellular trafficking and endosomal recycling processes. Our previous study, we demonstrated that RAB4B promotes Japanese encephalitis virus (JEV) replication in PK15 cells. However, the exact mechanisms underlying the role of RAB4B in JEV internalization remain unclear. Here, a genome-wide CRISPR/Cas9 library screen was performed, which identified RAB4B, along with other significant hits like ST8SIA4 and ELAVL1, as essential mediators of JEV replication. In vitro validation using RAB4B knockout in U251 and BV2 cells showed a sign

A Review of West Nile Virus as a Cause of Human Disease in Kansas.

Background: In the United States, West Nile virus (WNV) was first identified in 1999 in New York City and in Kansas in 2002. How the virus was introduced remains uncertain, although several potential methods have been hypothesized. This review presents an overview of what has occurred with WNV since 1999 and the potential for establishment of related viruses, such as Japanese encephalitis virus, if they were to be introduced in the United States. Materials and Methods: We have compiled human West Nile case data from Kansas over time with the purpose of illustrating how this virus has become en

Emergence of Japanese encephalitis in a previously non-reported area: Three consecutive annual cases from a tertiary center in Narita, Chiba, Japan.

The number of Japanese encephalitis (JE) cases dramatically decreased after implementing national vaccination program in Japan and the diagnosis requires specific tests in the reference laboratory in most cases. We report three consecutive cases of JE in a single hospital in Narita over three years. Although the PCR test confirmed the diagnosis in one case, the PCR was negative and IgM captured ELISA in the reference laboratory confirmed the diagnosis in the other two cases. These cases suggest that the number of JE patients may be underestimated in Japan. Physicians should consider JE as a di

Integrated duplex reverse transcription-recombinase aided amplification (RT-RAA) and lateral flow assay (LFA) for rapid simultaneous detection of Zika virus and Japanese encephalitis virus in single reaction format.

Zika virus (ZIKV) and Japanese encephalitis virus (JEV), two consequential mosquito-borne flaviviruses, induce severe systemic and neurotropic-encephalitic pathologies with overlapping symptoms profiles, complicating differential diagnosis in co-endemic regions. To address this critical challenge, we developed a dual reverse-transcription recombinase-aided amplification (RT-RAA) coupled with duplex lateral flow assay (LFA) platform for rapid, equipment-free co-detection of ZIKV and JEV in a single reaction tube. The assay achieved isothermal amplification at 39 °C within 40 min using a field-d

Evaluation of in vitro growth and pathogenicity in mice of intra-genotype chimeric strains of genotype IV Japanese encephalitis virus.

Two recombinant intra-genotype chimeric genotype IV Japanese encephalitis virus (GIV JEV) strains with the 5' non-coding and C-prM-E regions (5NCME) of the Vietnamese strain VN113 or the Australian strain sw-22-00722-11.Qld (sw-22) were produced using the Indonesian strain 19CxBa-83-Cv as a backbone. The three strains exhibited similar plaque morphology and growth rate in Vero cells. The virus with sw-22 5NCME was less neuroinvasive than 19CxBa-83-Cv, but the virulence of the virus with VN113 5NCME was similar to that of 19CxBa-83-Cv. Thus, our results suggest that the pathogenicity of sw-22 m

Assessing the endemicity of Japanese encephalitis in central India: A comprehensive study of human and animals in Madhya Pradesh state.

BACKGROUND OBJECTIVES: With an annual prevalence of 2000 cases, India has the highest global burden of Japanese encephalitis (JE). However, the distribution of the disease is not uniform across the country, wherein the states and districts considered endemic are prioritized for vaccination and other control measures. The central Indian state of Madhya Pradesh (MP), which is not yet considered JE-endemic, possesses multiple agents, hosts, and environmental risk factors and is geographically close to several hotspots of transmission. In this study, we explored the potential endemicity of JE in M
