Future threats: Animal orthoflaviviruses that currently infect fewer than 100 people per year.

After long periods of evolution, several orthoflaviviruses, including dengue, Japanese encephalitis, tick-borne encephalitis, West Nile, yellow fever, and Zika viruses, have emerged from nature to infect humans in high numbers. It should be worrisome that there are other orthoflaviviruses now undergoing the same emergence steps, with potential for the same type of outcome as the viruses above. In this review, we document 25 orthoflaviviruses currently infecting fewer than 100 people per year and another 12 for which serological data suggest human exposure, though infection has not been confirm

European Preparedness for Japanese Encephalitis Virus Through Alignment of Animal Health Laboratory Diagnosis.

Outbreaks of Japanese encephalitis (JE) can have severe health and economic impacts in both humans and susceptible animal species and are estimated to cause ~68,000 human disease cases in Asia annually. The disease is caused by infection with the mosquito-borne JE virus (JEV), which continues to expand its geographical range from its endemic region in Asia. Since appropriate vertebrate host and mosquito vector species are present in Europe and average European summer temperatures continue to increase, JEV introduction could lead to the establishment of the pathogen in native mosquito species a

Unveiling viral diversity and dynamics in mosquitoes through metagenomic analysis in Guizhou Province, China.

BACKGROUND: Poverty, disease, and vector ecology intersect to present ongoing health threats, particularly in ecologically sensitive regions. Guizhou Province in China, with its complex karst topography and rich biodiversity, offers a unique environment to study mosquito-borne viral transmission. Despite over 5000 reported cases of Japanese encephalitis in the past two decades and the detection of Zika virus in 2016, the virological landscape of this region remains poorly understood. This study aims to characterize the mosquito-associated virome, assess viral diversity, and identify factors in

Genetic shifts of Japanese encephalitis virus (JEV) in mosquitoes in the Republic of Korea, 2017-2022.

BACKGROUND: The Japanese encephalitis virus (JEV) is transmitted by mosquitoes and circulates in Asia, the western Pacific, and other parts of the world. It is classified into five genotypes (GI-GV) based on the nucleotide sequence of the envelope (E) gene. Despite extensive surveillance, shifts in genotype distribution and mosquito species remain underreported, particularly in the Republic of Korea. METHODOLOGY/PRINCIPAL FINDINGS: We conducted a nationwide mosquito collection from 2017 to 2022, capturing 1,102,031 mosquitoes from 32 sites nationwide. The predominant species were Aedes vexans

Vector competence of European Aedes mosquito species for Japanese encephalitis virus under fluctuating temperature conditions.

The Japanese encephalitis virus (JEV) is a mosquito-borne flavivirus endemic to much of Asia and the Western Pacific, both temperate and tropical regions. Globalisation and the expansion of invasive mosquito species raise concerns about their potential establishment in Europe and other currently non-endemic regions. However, limited knowledge exists regarding the vector competence of European mosquitoes, particularly under the region's characteristic fluctuating temperatures. While Culex species are primary JEV vectors, the role of Aedes mosquitoes remains unclear. This study assessed the vect

Telmisartan, an anti-hypertensive drug, impedes JEV infection, possibly via the AT1/PPARγ axis.

Japanese encephalitis, caused by Japanese encephalitis virus (JEV), is a vector-borne disease with no specific therapeutics available yet. Binding of angiotensin II (Ang II) to angiotensin II type 1 (AT1) receptor induces the release of inflammatory cytokines associated with viral encephalitis. Accordingly, Ang II receptor blockers (ARBs) have been proposed to manage encephalitis. Since telmisartan (TM, antagonist of AT1 and agonist of PPARγ) has relatively better brain access than other ARBs, this investigation aims to evaluate its anti-JEV efficacy in vitro and in vivo. TM reduced JEV titer,

Peripheral nerve injury mediated by JEV strain NX1889 infection and impairment of Schwann cells.

Japanese encephalitis virus (JEV) is a mosquito-borne flavivirus primarily associated with central nervous system disorders. Recent studies have indicated that JEV infection can lead to Guillain-Barré syndrome (GBS), a peripheral nervous system (PNS) disorder; however, the underlying mechanisms remain unclear. In this study, we investigated the tropism of different genotypes of JEV strains for peripheral nerves and their potential impact on peripheral nerves. Four JEV strains-GI NX1889, GI GZ56, GIII P3, and GV XZ0934-representing three prevalent genotypes in China were selected for analysis.

Rutin Attenuates Virus Entry and Replication and Exerts Neuroprotection in Experimental Models of Japanese Encephalitis.

Japanese encephalitis virus (JEV), which belongs to the virus family of Flaviviridae, is a threat to more than three billion people globally. There are no antiviral agents against JEV despite the availability of vaccines. This study considered the need for an effective drug against JEV by evaluating the antiviral and neuroprotective potentials of Chicoric Acid (CA) and Rutin in the in vitro and in vivo models of JE. In the in vitro study, CA and Rutin exhibited variable antiviral potency with IC50 values ranging from 11.03 to 24.04 µM and 16.45 to 26.84 µM in different treatment approaches. Th

Insect-specific viruses: transmission dynamics and biological control strategies against arboviruses.

Mosquitoes are known to vector arthropod-borne viruses (arboviruses) that pose a global public health issue in the form of mosquito-borne viral diseases such as chikungunya fever, dengue fever, Japanese encephalitis, yellow fever, and Zika. Besides, mosquitoes may also carry insect-specific viruses (ISVs), which are evolutionarily alike arboviruses yet do not infect vertebrates. These ISVs have been shown to affect the ability of mosquitoes to transmit arboviruses, as well as potentially inhibit arbovirus infections in vertebrate hosts. Yet, ISVs still constitute a relatively new and little-re

Antibody responses to Japanese encephalitis virus and dengue virus serotype 2 in children from an orthoflavivirus endemic region after IMOJEV vaccination.

BACKGROUND: Japanese encephalitis virus (JEV) is a mosquito-borne pathogen that causes severe neurologic disease. Its endemicity in Asia has prompted its inclusion in nationwide immunization programs. However, the Philippines, which is also endemic for related viruses like dengue (DENV), has not yet adopted this practice. Vaccine hesitancy is a major challenge, exacerbated by concerns over cross-reactive antibodies that may enhance viral infection. This study aimed to determine whether IMOJEV vaccination would induce cross-neutralizing or enhancing antibodies against DENV. METHODOLOGY/PRINCIP

A novel Gaussia luciferase immunoprecipitation assay for the detection of Getah virus antibodies in pigs.

Getah virus (GETV), a mosquito-borne pathogen, causes fever, aseptic meningitis, and abortion across various animal species, including pigs, horses, and cattle. Its widespread prevalence in China has resulted in substantial economic losses to the pig industry and poses a growing threat to public health security. Currently, no effective vaccine or antiviral drug for GETV control in China underscores the urgent need for the development of a rapid and sensitive antibody detection method for large-scale screening. In this study, we established a luciferase immunoprecipitation system (LIPS) assay t

Movement Disorders in West Nile Virus Infection: A Systematic Review of Reported Cases and Conference Abstracts.

BACKGROUND: West Nile virus is the most widely distributed arbovirus and an established cause of neuroinvasive disease. While encephalitis and acute flaccid paralysis are well characterized, the spectrum of associated movement disorders remains insufficiently defined. Similar syndromes are common in Japanese encephalitis and dengue, but evidence in West Nile virus is fragmented. OBJECTIVES: This review aimed to analyze individual patient data on movement disorders, outlining their clinical features, neuroimaging, treatments, outcomes, and underlying mechanisms. METHODS: This review followed

The hidden threats posed by Japanese encephalitis virus genotype V.

Japanese encephalitis virus (JEV) remains a major cause of viral encephalitis in Asia. Among its five genotypes (GI-GV), genotype V (GV) has re-emerged and become the predominant lineage in mosquitoes in the Republic of Korea, with sporadic human cases also reported. GV JEV displays differences from other genotypes in genomic sequence, antigenicity, and pathogenicity, and animal studies indicate higher lethality. Importantly, all currently used vaccines are based on GIII strains and provide partial but insufficient protection against GV JEV, with breakthrough infections documented. These findi

Reliable estimates of Japanese encephalitis virus evolutionary rate supported by a formal test of temporal signal.

The single-stranded RNA viruses exhibit stupendously high evolutionary rate spanned several orders of magnitude. The accurate and reliable estimates of the virus evolutionary rate are of importance for the robust interpretation of outbreak investigations and are critical for understanding short-term virus transmission patterns. In the previous studies, the substitution rate of the Japanese encephalitis virus (JEV) and time to the most recent common ancestor (MRCA) were reported, however, the estimates are substantially different between some of the studies. Importantly, temporal signal in data

National burden of and optimal vaccine policy for Japanese encephalitis virus in Bangladesh: a seroprevalence and modelling study.

BACKGROUND: Bangladesh first reported Japanese encephalitis virus (JEV) in 1977 and has seen regular cases since; however, no JEV vaccination programme currently exists. A barrier to the use of JEV vaccines has been a limited understanding of the underlying burden. METHODS: We conducted a nationally representative serological community study in 70 communities in individuals of all ages from October, 2015, to January, 2016. Serum samples obtained from the community study were tested for IgG antibodies against JEV. We developed spatially explicit binomial regression models to estimate the under

Thioester-Containing Protein TEP27 in Culex quinquefasciatus Promotes JEV Infection by Modulating Host Immune Function.

Thioester-containing proteins (TEPs), which are distinguished by the thioester motif (GCGEQ), are essential to arthropods' defense against infections. Although TEPs have been extensively investigated in Anopheles, Aedes, and Drosophila, their functions in Culex mosquitoes remain inadequately explored. Interestingly, we discovered that Culex TEPs exhibit functional antagonism to their orthologs in other species, actively facilitating viral infection in this vector. In this study, we identified nine TEP genes in Culex quinquefasciatus, three of which were found to critically facilitate Japanese
